Cloning and Organization of the Ribosomal RNA Genes of the Mushroom Trichloma matsutake

  • A portion (7.4 kb) of ribosomal DNA tandem repeat unit from a genome of the mushroom T. matsutake has been cloned. A 1.75 kb EcoRI fragment was cloned first using S. cerevisiae 255 rRNA gene as a probe, and this was then used for further cloning. A chromosomal walking experiment was carried out and the upstream region of the 1.75 kb fragment was cloned using SmaI/BamHI enzyme, the size was estimated to be 5.2 kb in length. Part of the downstream region of the 1.75 kb fragment was also cloned using XbaI/BamHI enzymes. Restriction enzyme maps of three cloned DNA fragments were constructed. Northern hybridization, using total RNA of T. matsutake, and the restriction fragments of three cloned DNAs as probes, revealed that all four ribosomal RNA genes (large subunit[LSU], small subunit [SSU], 5.85 and 5S rRNA genes) are present in the cloned region. The gene organization of the rDNA are regarded as an intergenic spacer [IGS]2 (partial) - SSU rRNA - internal transcribed spacer [ITS]1 - 5.8S rRNA - ITS2 - LSU rRNA - IGS1 -5S rRNA - IG52 (partial).

Taxonomy and phylogeny of the genus Cryptomonas (Cryptophyceae, Cryptophyta) from Korea

  • The genus Cryptomonas is easily recognized by having two flagella, green brownish color, and a swaying behavior. They have relatively simple morphology, and limited diagnostic characters, which present a major difficulty in differentiating between species of the genus. To understand species delineation and phylogenetic relationships among Cryptomonas species, the nuclear-encoded internal transcribed spacer 2 (ITS2), partial large subunit (LSU) and small subunit ribosomal DNA (rDNA), and chloroplast-encoded psbA and LSU rDNA sequences were determined and used for phylogenetic analyses, using Bayesian and maximum likelihood methods. In addition, nuclear-encoded ITS2 sequences were predicted to secondary structures, and were used to determine nine species and four unidentified species from 47 strains. Sequences of helix I, II, and IIIb in ITS2 secondary structure were very useful for the identification of Cryptomonas species. However, the helix IV was the most variable region across species in alignment. The phylogenetic tree showed that fourteen species were monophyletic. However, some strains of C. obovata had chloroplasts with pyrenoid while others were without pyrenoid, which used as a key character in few species. Therefore, classification systems depending solely on morphological characters are inadequate, and require the use of molecular data.

Seven Unrecorded Indigenous Fungi from Mudeungsan National Park in Korea

  • Fungi act as important decomposers in the forest environment. They recycle essential nutrients, promote plant growth through mycorrhizal relationships, and act as food for small animals. Samples of 265 indigenous fungal species were collected from Mudeungsan National Park in 2020. These species were identified based on morphological, molecular, and phylogenetic analyses using the internal transcribed spacer (ITS), nuclear large subunit rRNA (LSU), and RNA polymerase II second largest subunit (rpb2) regions. Subsequently, seven species were identified as unrecorded species in Korea: Cordyceps cicadae, Dentocorticium bicolor, Hymenochaete nanospora, Physisporinus crataegi, Rigidoporus piceicola, Russula raoultii, and Scutellinia crinita. This study reveals their detailed macro- and microscopic morphological characteristics with phylogenetic trees to report them as unrecorded species in Korea.

  • 본 연구에서는 민주지산에 서식하는 철쭉(Rhododendron schlippenbachii)의 뿌리에서 내생균을 분리하였다. 형태적 특성의 확인 및 ribosomal DNA의 internal transcribed spacer(ITS) 지역, large subunit (LSU)지역의 분자적 분석을 통해 국내 미기록종인 Engyodontium album, Phaeosphaeria microscopica 2종을 동정하였다. 연구 과정에서 확인된 미기록종 내생균 2종의 형태적, 분자적 특성을 여기에 기술하였다.

Erysiphe cornicola, a Powdery Mildew Occurring on Cornus controversa in Korea

  • In Korea, Cornus controversa and C. florida are known as hosts of Erysiphe pulchra from section Microspharea of the genus Erysiphe. However, recent molecular-phylogenetic analyses on the internal transcribed spacer regions and large subunit gene of the rDNA revealed that the Erysiphe powdery mildew on C. controversa in Japan is in fact E. cornicola. To assess the taxonomic status of Erysiphe-C. controversa association in Korea, isolates collected since 1987 were investigated and consequently identified as E. cornicola based on molecular-phylogenetic analyses and new morphological traits. To our knowledge, this is the first study to confirm the presence of this powdery mildew in Korea.

Haraldiophyllum hawaiiense sp. nov. (Delesseriaceae, Rhodophyta): a new mesophotic genus record for the Hawaiian Islands

  • Haraldiophyllum hawaiiense sp. nov. is described as a new mesophotic alga and a new genus record for the Hawaiian Islands. Six specimens were collected at a depth range of 81-93 m from Papahānaumokuākea Marine National Monument, and their morphology investigated, as well as molecular phylogenetic analyses of the plastidial ribulose-1,5-bisphosphate carboxylase-oxygenase large-subunit (rbcL) gene and a concatenated alignment of rbcL and nuclear large-subunit rRNA gene (LSU) sequences. Phylogenetic analyses supported H. hawaiiense sp. nov. as a distinct lineage within the genus Haraldiophyllum, and sister to a large clade containing the type species, H. bonnemaisonii, as well as H. crispatum and an undescribed European specimen. The six Hawaiian specimens were shown to be identical, but unique among other species of the genus as well as the recently segregated genus Neoharaldiophyllum, which comprises half of the species previously included in Haraldiophyllum. The vegetative morphology of H. hawaiiense sp. nov. resembles Neoharaldiophyllum udoense (formerly H. udoensis); however, no female or post-fertilization structures were found in the Hawaiian specimens to allow a more comprehensive comparison. The molecular phylogenies demonstrate that Haraldiophyllum is paraphyletic, suggesting either that the Myriogrammeae tribe includes undescribed genera, including Haraldiophyllum sensu stricto, or that Neoharaldiophyllum species should be transferred into the genus Haraldiophyllum. However, based on vegetative morphology and molecular analyses, and pending resolution of this taxonomic issue, the Hawaiian specimens are placed within the genus Haraldiophyllum. This new record for the Hawaiian Islands highlights the novel biodiversity from mesophotic depths, reaffirming the need for further investigation into the biodiversity of Mesophotic Coral Ecosystems.

First Report of Leptosphaerulina australis Isolated from Soil in Korea

  • The fungal strain KNU16-004 was isolated from a field soil sample collected in Seoul. The isolate was identified as Leptosphaerulina australis based on morphological characterization and phylogenetic analysis using the internal transcribed spacer (ITS), large subunit (LSU) rDNA regions, and ${\beta}-tubulin$ (Tub2). This is the first report of Leptosphaerulina australis in Korea.

First Reports of Unrecorded Mortierellomycetes and Umbelopsidomycetes Fungi from Freshwater Ecosystems in Korea

  • In this study, we isolated several fungal strains from filtered water and sediment collected from rivers and streams. The strains were identified by molecular phylogenetic analyses of rDNA sequences (internal transcribed spacer [ITS], large subunit of ribosomal DNA [LSU]). The morphological characteristics of the fungi were investigated using microscopy, and the culture characteristics of fungi grown on several media were examined. We identified four species previously unknown in South Korea, namely, Dissophora globulifera, Linnemannia exigua, Mortierella rishikesha and Umbelopsis autotrophica.

Magnaporthiopsis poae에 의한 한지형 잔디의 여름잎마름병 보고 (First Report of Summer Patch Caused by Magnaporthiopsis poae on Cool Season Grass)

  • 2015년 6월 초순, 한지형 잔디인 Kentucky bluegrass "Midnight II" 품종이 식재된 서울시 소재 잔디밭에서 잎마름 증상, 뿌리썩음 증상 및 개구리 눈 모양의 전형적인 summer patch 병징이 관찰되었다. 병원균의 정확한 동정을 위해 이병 잎과 뿌리에서 병원균을 분리하고, 배양학적 및 균학적 특징과 분자마커를 이용한 계통학적 유연관계 분석을 실시하였다. 그 결과, 분리된 병원균은 배양 10일 후 약 50~60 mm 가량 생장하였고, 균사는 회백색을 띄며 중앙부터 황록색 혹은 갈색으로 변색되었다. 경자(phialides)의 경우, 그 유사조직이 주로 균사의 말단에서 관찰되었으나, 분생포자는 관찰되지 않았다. 분리된 병원균의 정확한 동정과 계통학적 유연관계 분석을 위하여, large subunit (LSU)와 RNA polymerase II large subunit (RPB1) 유전자를 증폭시킨 후 염기서열 분석을 진행하고 계통학적 유연관계를 분석하였다. 그 결과, 분리된 병원균은 Magnaporthiopsis poae M47 균주와 가장 근연한 관계에 있는 것으로 확인되었다. 또한 potato dextrose agar (PDA) 배지에서 배양된 병원균을 멸균된 메스로 잘라 동일한 품종의 잔디에 접종한 결과, 약 20일 후 잎마름 증상을 확인할 수 있었다. 따라서 본 연구에서는 국내 Kentucky bluegrass에 summer patch병을 일으키는 원인균을 Magnaporthiopsis poae로 보고하고, 위 병원균에 의한 한지형 잔디의 병징을 여름잎마름병으로 제안하고자 한다.

The Genus Acervus from Southwestern China and Northern Thailand

  • Acervus (Pyronemataceae, Pezizales) is a saprobic genus in Pezizomycetes, characterized by colored apothecia, subcylindrical to cylindrical asci and guttulate ascospores. We collected four Acervus samples from China and Thailand. Descriptions and illustrations are introduced for all fresh samples. One new record of A. globulosus from Thailand, one new species, A. rufus, two known species, A. epispartius and A. stipitatus from China are reported. Phylogenetic analysis based on five genes, the large subunit rRNA (LSU), the translation elongation factor-1 alpha (tef1-α), the second largest subunit of RNA polymerase II (rpb2), the largest subunit of RNA polymerase II (rpb1), and the small subunit rRNA (SSU), revealed the distinct position of the new species. The new species is set apart by its red apothecia. A key to Acervus species is also given.
