음곡 옥미수 약침이 LPS로 유도된 흰쥐의 신장염에 미치는 영향 (Anti-inflammatory Effect of Zeae Stigma Herbal-acupuncture at KI10 on LPS-induced Nephritis in Rats)

  • Objectives: The purpose of this study was to investigate the effects of Zeae Stigma herbal-acupuncture (ZS-HA) at KI10 (Umgok) on nephritis induced by lipopolysaccharide (LPS) in rats. Methods: Rats were assigned to five groups, normal, LPS, NP, saline and ZS-HA. Rats in NP, saline, and ZS-HA groups were treated with needle prick, saline injection, and ZS-HA respectively at KI10, three times a week. All animals except the normal group were injected intra-peritoneally with LPS to induce nephritis. RBC and WBC, neutrophils in blood, TNF-alpha, CINC-1, creatinine in serum, urinal volume, total protein and creatinine in urine, and renal MPO were analyzed. Results: Needle prick at KI10 significantly reduced WBC in blood and CINC-1 in serum of LPS-stimulated rats. Saline injection at KI10 significantly reduced TNF-${\alpha}$ in serum and total protein in urine of LPS-stimulated rats. ZS-HA at KI10 significantly increased RBC in blood, and significantly reduced neutrophils in blood, TNF-${\alpha}$ in serum, and total protein in urine of LPS-stimulated rats. Conclusions: According to these results, it is postulated that ZS-HA at KI10 has anti-inflammatory and renal-protective effects on LPS-induced nephritis in rats, and both acupoint KI10 and herb Zeae Stigma made contributions to this effect. Further studies on the interaction between acupoint KI10 and herb Zeae Stigma may be needed.

  • Objectives : This study was designed to evaluate the effects of Desmodii herb pharmacopuncture (DH-HA) at $KI_{10}$(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rat. Methods : Rats were injected LPS to induce nephritis. DH-HA group was treated with DH-HA at $KI_{10}$ three times for a week, NP group with 26 gauge needle, saline group with normal saline. To evaluate the effects of DH-HA at $KI_{10}$ on nephritis in rats, RBC, WBC, neutrophil in blood, TNF-a, CINC-1 in serum, creatinine and total protein in urine, renal TNF-a, renal MPO were measured and renal tissue was analyzed. Results : WBC in blood, CINC-1 in serum, renal MPO significantly reduced in DH-HA group. Conclusions : DH-HA at $KI_{10}$ is effective for nephritis in LPS-induced rats. Therefore, DH-HA at $KI_{10}$ may be useful for nephritis in clinical field.

  • Objective : This study aimed to evaluate the effects of Benincasae Semen Herbal-acupuncture (BS-HA) at KI10 (Umgok) on nephritis induced by LPS in rat. Methods : Rats with nephritis induced by LPS, were treated with Benincasae Semen Herbal-acupuncture(BS-HA) injection at KI10. Two control groups, N.P. group and saline group, were treated with 26 gauge needle at KI10, 3 times a week. In Saline group normal saline was injected at KI10. To evaluate the effects of Benincasae. Semen Herbal-acupuncture at KI10 on nephritis in rats, WBC, Neutrophils in blood, BUN, Creatinine TNF-$\alpha$, CINC-1 in serum, urinal volume and creatinine and Total protein in urine, reanl TNF-$\alpha$, renal MPO were measured and reanl tissue was also analyzed. Results: BS-HA injected at KI10 significantly inhibited WBC and neutrophil in blood. creatinine in serum, and MPO in kidney of LPS-stimulated rats. BS-HA injected at KI10 reduced concentration of neutrophil in renal tissue of LPS-stimulated rats. Conclusion : BS-HA at KI10 has a therapeutic effect for nephritis in LPS-stimulated rat There fore, it is suggested that BS-HA at KI10 may be an useful therapeutics in clinical field after further researches.

  • Objectives : This study aimed to evaluate the effects of Alismatis Rhizoma Herbal-acupuncture( AR-HA) at $KI_{10}$(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rat. Methods : Rats were injected with LPS to induce nephritis. The rats in $KI_{10}$-AR-HA group were treated with AR-HA, $KI_{10}$-NP group with needle prick, $KI_{10}$-saline group with normal saline injection respectively at $KI_{10}$ three times for a week. Several experimental items were performed including measurements of the numbers of RBC, WBC, neutrophil in blood, the levels of creatinine, $TNF-{\alpha}$, CINC-1 in serum, creatinine, total protein in urine, MPO in kidney. Results : Needle prick stimulation at $KI_{10}$ significantly reduced WBC in blood of LPS-stimulated rats. Saline injection at $KI_{10}$ significantly reduced WBC in blood, $TNF-{\alpha}$ in serum, total protein in urine of LPS-stimulated rats. AR-HA at $KI_{10}$ significantly reduced WBC, neutrophil in blood, $TNF-{\alpha}$, CINC-1 in serum, creatinine, total protein in urine of LPS-stimulated rats. Conclusion : AR-HA at $KI_{10}$ has a therapeutic effect on nephritis in LPS-stimulated rat. There may be a synergism between $KI_{10}$(Umgok) stimulation and AR-HA solution.

  • Objectives : The purpose of this study is evaluating the effect of Hoelen Herbal-acupuncture (HO-HA) at $KI_{10}$(Umgok) on Lipopolysaccharide(LPS) induced nephritis in rats. Methods : The experimental rats were assigned to four groups; normal, LPS, saline, HO-HA groups. LPS(2 mg/kg) was administered to the rats in LPS, saline and HO-HA groups to induce acute inflammatory kidney damage. Saline injection and HO-HA were administered at $KI_{10}$ three times a week. Blood samples were taken from the rats for analysis of white blood cell(WBC), neutrophil, blood urea nitrogen(BUN), creatinine TNF-${\alpha}$, CINC-1. Urine samples were taken from the rats for analysis of urinal volume, creatinine and total protein. The kidney samples were taken from the rats for analysis of renal myeloperoxIdase(MPO). Results : HO-HA suppressed the increases of WBC and neutrophils in blood, BUN, creatinine, TNF-${\alpha}$ and CINC-1 in serum, and MPO in kidney of LPS-stimulated rats. In addition, HO-HA inhibited the decrease of urinary volume in LPS-stimulated rats. Conclusions : HO-HA has therapeutic effects on LPS-induced inflammatory kidney damage in rats. Further studies may be needed for clinical use of HO-HA.

To estimate the effects of DS-HA at KI10 on nephritis in rats, WBC, neutrophils in blood, BUN, creatinine TNF-${\alpha}$, CINC-1 in serum, urinal volume and creatinean and total protein in urine, reanl TNF-${\alpha}$, renal MPO were measured and histological analysis of renal tissue was performed. Results : DS-HA at KI10 significantly decreased WBC and neutrophil in blood and BUN and creatinin in serum, MPO in kidney, and significantly increased urinary volume in LPS-stimulated rats. DS-HA at KI10 reduced accumulation of neutrophil in renal tissue of LPS-stimulated rats. Conclusions : Taken together, DS-HA at KI10 has a protective or therapeutic effect for nephritis in LPS-stimulated rat. Therefore, it is suggested that DS-HA at KI10 may be an useful therapeutics in clinical field after further researches.

  • Background and Objectives The representative mucin genes in the human airway are MUC5AC and MUC5B, which are regulated by several inflammatory and anti-inflammatory substances. Triptolide (TPL), udenafil, betulinic acid, changkil saponin, and glucosteroid are some of the many anti-inflammatory substances that exist. TPL is a diterpenoid compound from the thunder god vine, which is used in traditional Chinese medicine for treatment of immune inflammatory diseases, such as rheumatoid arthritis, systemic lupus erythematosus, nephritis and asthma. However, the effects of TPL on mucin expression of human airway epithelial cells have yet to be reported. Hence, this study investigated the effect of TPL on lipopolysaccharide (LPS)-induced MUC5AC and MUC5B expression in human airway epithelial cells. Subjects and Method The NCI-H292 cells and the primary cultures of human nasal epithelial cells were used to investigate the effects of TPL on LPS-induced MUC5AC and MUC5B expression using real-time polymerase chain reaction, enzyme immunoassay, and Western blot. Results TPL significantly decreased the LPS-induced MUC5AC and MUC5B mRNA expression and protein production. TPL also significantly decreased the nuclear factor-kappa B (NF-kB) phosphorylation. Conclusion These results suggest that TPL down regulates MUC5AC and MUC5B expression via inhibition of NF-kB activation in human airway epithelial cells. This study may provide important information about the biological role of triptolide on mucus-secretion in airway inflammatory diseases and the development of novel therapeutic agents for controlling such diseases.
