The Effect of TGF-{\beta}_1 on Cellular Activity of Periodontal Ligament Cells activated by PDGF-BB (PDGF-BB에 의한 치주인대세포활성에 대한 TGF-{\beta}의 효과)

  • The purposes of this study is to evaluate the combination effects of TGF-${\beta}_1$ and PDGF-BB on the periodontal ligament cells to use as a regeneration promoting agent of periodontal tissue. Human periodontal ligament cells were prepared from the first premolar tooth extracted for the orthodontic treatment and were cultured in DMEM/100% FBS at the $37^{\circ}C$, 5% $CO_2$ incubator. Authors measured the DNA synthesis, total protein, collagen and noncollagenous protein synthesis according to the concentration of TGF-${\beta}_1$,(1,5ng/ml) and PDGF-BB (1,10 ng/ml) in combination. To explore further this delayed effect of TGF-${\beta}_1$, we preincubated human periodontal ligament cells with TGF-${\beta}_1$ for 4 or 24 hours before PDGF-BB stimulation. The results were as follows: The DNA synthetic activity was increased dose dependently by TGF-${\beta}_1$, PDGF-BB. The combination of TGF-${\beta}_1$ and PDGF-BB consistently enhanced the DNA synthetic activity to PDGF-BB alone. The ability of TGF-${\beta}_1$ to enhance DNA synthetic activity in PDGF-BB treated periodontal ligament cells was dose dependent. The maximum mitogenic effect was at the 5ng/ml of TGF-${\beta}_1$ and l0ng/ml of PDGF-BB. Preincubation of cell with TGF-${\beta}_1$ resulted in significantly greater response to PDGF-BB at all TGF-${\beta}_1$ concentration studied, and may be useful for clinical application in periodontal regenerative procedures. The total protein, collagen and noncollagen synthesis was increased dose pendently by TGF-${\beta}_1$, PDGF-BB. The % of collagen was slightly decreased according to the concentration of TGF-${\beta}_1$, PDGF-BB. The effect of TGF-${\beta}_1$, PDGF-BB were not specific for collagen synthesis since it also increased noncollagenous protein synthesis. This study demonstrates that PDGF-BB is major mitogens for human periodontal ligament cells in vitro, and supports a role for TGF-${\beta}_1$ as a regulation of the mitogenic and total protein formation to PDGF-BB in these cells.

Antimicrobial Activity of Salvia miltiorrhiza Bunge Extract and Its Effects on Quality Characteristics in Sulgidduk (단삼 추출물의 향균 활성 및 첨가 설기떡의 품질 특성)

  • In this study, Salvia miltiorrhiza Bunge(Danshen) powder was extracted with ethanol, and its antimicrobial activity was investigated. The ethanol extract of the Danshen had antimicrobial activity against Bacillus subtilis, Escherichia coli, and Staphylococcus aureus. The inhibition zones of the Danshen extract(3 mg/disc) against B. subtilis, E. coli and S. aureus were 13, 12 and 8.5 mm, respectively. To test the food preservation effect of Danshen and determine the optimal ratio of the Danshen extract in a formulation, Sulgidduk samples were prepared with substitutions of 0, 0.25, 0.5, 0.75 and 1% Danshen extract, and then their quality characteristics were investigated over 4 days of storage. According to the results, total cell counts showed a decreasing trend with an increasing amount of added Danshen extract. Moisture contents were not significantly different among the Sulgidduk samples. As the content of Danshen extract increased, the L-values of samples decreased and the a- and b- values increased. For the textural characteristics, the hardness, gumminess, and chewiness of the Sulgidduk samples decreased as the amount of Danshen extract increased; however, they increased with the progression of storage time. In the sensory evaluation, the control group had significantly higher scores for color, flavor and after-taste as compared to the Danshen extract-added groups. With increasing Danshen extract contents, flavor and overall acceptability decreased, while Danshen flavor, bitterness and off-flavor increased. Chewiness was not significantly different among the samples. In conclusion, the results indicate that substituting 0.5% Danshen extract in Sulgidduk is optimal for quality and provides a product with reasonably high overall acceptability.

Measurement of the Levels of IgG Subclasses Reactive to Salmonella typhi in the Sera of Patients with Typhoid Fever (장티푸스환자의 혈청내 Salmonella typhi에 대한 IgG subclass항체의 분포)

  • To diagnose the typhoid fever rapidly and accurately in clinically suspected patients, the levels of IgG subclass antibody were measured by enzyme-linked immunosorbent assay(ELISA). With symptom, blood culture and agglutination test, tested persons were categorized into 6 groups as typhoid fever, FUO, paratyphi A or B, other bacterial infctions, cancers, and control. ELISA was performed on the polyvinyl chloride plates coated with killed whole cell($10^8\;cell/ml$) of S. typhi 0901W by poly-L-lysine applied as binding substance (and polyvinyl chloride as solid phase). The distribution of the level of IgG subclass antibodies in each group was analyzed and compared with other groups. The results obtained were summarized as follow: 1. The optimal dilution of the sera from patients with typhoid fever was 1:160, and those of the sheep anti-human IgG subclass and the peroxidase conjugated rabbit anti-sheep IgG were 1:4000 and 1:5000, respectively. 2. The absorbance levels of IgG subclass in the sera of typhoid fever patients were as follows; a) IgG1 value is $0.439{\pm}0.110$ b) IgG2 value is $0.416{\pm}0.165$ c) IgG3 value is $0.449{\pm}0.145$ d) IgG4 value is $0.525{\pm}0.154$ IgG subclass levels in the sera of typhoid patients were much higher than in control group and patient with paratyphi A or B as well as other infectious diseases. The sensitivity and the specificity in differential diagnosis of typhoid fever and other febrile diseases were 92% and 79% in the assay of IgG1 respectively, whereas those in the assay of IgG2 were 97% and 72%, respectively (above absorbance 0.3). 3. The absorbance levels of IgG subclass in the serial sera of typhiod fever patients tend to decrease to the level of absorbance 0.3 in 10 months from the onset of illness. 4. The order of absorbance levels of IgG subclass in the serum of each group were typhoid fever, paratyphi A or B, other infectious diseases, control and cancer. 5. For the serodiagnosis of typhoid fever against other febrile diseases, the sensitivity and the specificity in the assay of IgG2 activity were 76% and 93% in absorbance 0.4, respectively. 6. In the distribution of the level of each IgG subclass in the sera of FUO patients which were suspected of typhoid fever, the positive rate was ranged from 36% to 82%. This suggest that more than 50% of FUO patients are caused by S. typhi.

Effects of Cutivation Method and Planting Date in Growth and Yield of Momordica charantia L. in Spring Season (여주 봄 작형의 재배방법과 정식기가 생육과 수량에 미치는 영향)

  • This study was conducted in order to investigate the effects that the difference of planting time by a method of cultivation in the non-heated greenhouse and the open field with spring planting had on growth and yield. With regard to the tested variety of Momordica charantia, variety 'Dragon' (Japan Yae 農藝) was selected. And 3 treatments on March 20, April 5 and April 20 for the greenhouse cultivation and 3 treatments on April 20, May 5 and May 20 for the open-field cultivation 1 month later than those for the greenhouse cultivation were planted by the randomized complete block design, and 4 secondary vines were trained. In the results of examining 15-day average atmospheric temperature after planting according to the methods of greenhouse and open-field cultivation and planting time, it was shown that there was a tendency for atmospheric temperature inside the greenhouse to decrease as the planting time was moved up. In particular, the average atmospheric temperature was $16.7^{\circ}C$ when seedling was planted on April 20 in the open-field cultivation, which was approximately equal to $17.0^{\circ}C$ of the average atmospheric temperature when a seedling was planted on March 20 in the greenhouse cultivation. With regard to the date of first harvest by the method of cultivation, it was shown that there was a tendency for the date of first harvest to be earlier in the greenhouse cultivation than in the open-field cultivation, and the date of first harvest was moved up as a seedling was planted earlier for the planting period. The number and weight of harvested fruits per plant showed a tendency which was almost similar to that of total number of harvest days and number of harvests. Thus, the number of fruits was 189 and the weight of fruits was 31,649g in case of the greenhouse cultivation and planting on March 20, which were maximum. In case of planting on the latest planting date : May 20 in the open-field cultivation, the number of fruits was 77 and the weight of fruits was 12,502g, which were at a level of 40% of those of planting on March 20 in the greenhouse cultivation 2 months earlier. The total yield per 10a was 10,228kg in the greenhouse cultivation and was 2.2 times as heavy as 4,607kg in the open-field cultivation with regard to the method of cultivation. For the planting period in the greenhouse cultivation, it was 10,539kg and 10,517kg in planting on March 20 and April 5, which was higher by 9% than 9,629kg in planting on April 20. And in the open-field cultivation, it was 4,785kg in planting on April 20 and 4,872kg in planting on May 5, which was higher by 15~17% than 4,163kg in planting on May 20. Taking the above results into account, it is considered proper to plant Momordica charantia from March 20 to April 5 for the greenhouse cultivation and from April 20 to May 5 or thereabouts when a risk of late frost is gone for the open-field cultivation in southern area.

  • POLARBEAR is a ground-based experiment located in the Atacama desert of northern Chile. The experiment is designed to measure the Cosmic Microwave Background B-mode polarization at several arcminute resolution. The CMB B-mode polarization on degree angular scales is a unique signature of primordial gravitational waves from cosmic inflation and B-mode signal on sub-degree scales is induced by the gravitational lensing from large-scale structure. Science observations began in early 2012 with an array of 1.274 polarization sensitive antenna-couple Transition Edge Sensor (TES) bolometers at 150 GHz. We published the first CMB-only measurement of the B-mode polarization on sub-degree scales induced by gravitational lensing in December 2013 followed by the first measurement of the B-mode power spectrum on those scales in March 2014. In this proceedings, we review the physics of CMB B-modes and then describe the Polarbear experiment, observations, and recent results.

Changes of pH, Meat Color, Cooking Loss, Shear Force and Sensory Evaluation on Hanwoo Meat Fed with Supplemental Fig Fermentation during Storage Period (무화과 발효물을 급여한 한우고기의 저장기간 중 pH, 육색, 가열감량, 전단력 및 관능평가의 변화)

  • This study was conducted to investigate changes of pH, meat color, cooking loss, shear force and sensory evaluation on Hanwoo meat fed with supplemental fig fermentation(SFF) during storage period. A total of 10 Hanwoo bulls were subjected to one of two treatment diets (control and 10% SFF) from live weight of 400kg far 6 months. After slaughter, longissimus muscles were removed, zipper-wrapped and stored at 4$\^{C}$ for 21 days. The pH, meat color, cooking loss, shear force and sensory evaluation of the samples were measured at 1, 3, 9, 15, 21 days of storage. There were no differences in pH, meat objective color(Hunter L, a and b) and cooking loss (%) of longissimus muscles between control and SFF treatment during storage. Shear force values of longissimus muscle from SFF treatment showed lower level in 1, 3 and 9 days and tended to decrease during storage. No differences in odor and appearance of sensory evaluation were observed between control and SFF treatment during storage. The taste induced by SFF was increased(f<0.05) at 1, 3 and 9 days of storage. These results indicate that the SFF may improve meat quality of Hanwoo during storage.

Germination Characteristics of Chinese Milk Vetch(Astragalus sinicus L.) Seeds Produced in China and Korea (수입 및 국내 채종 자운영 종자의 발아 특성)

  • Germination characteristics of Chinese milk vetch(CMV) seeds produced from Hanam Province, China and Milyang, Korea were investigated to give basic information on the stability of seedling establishment in the CMV cultivation. The germination percentage of the imported CMV seed from China varied according to importation year and seed collection site ranging from 79~95%. The germination of black colored seed coat was lower than the light green colored ones and germination by seed weight was not significantly different. Although the seed germination was lower under dark than in the presence of light, it was not significantly different. The germination of the imported CMV seed slightly declined to only less than 6% after one-year of storage under natural environment conditions but it significantly decreased after two years. However, when the seed was stored at the $5^{\circ}C$, the seed germination was the same as after two years of storage. On the other hand, fresh CMV seed produced in Milyang, Korea had only 8% germination due to seed coat dormancy but the germination increased to 73~85% after breaking seed dormancy after a year of storage. The high germination percentage of 72~82% was still maintained even after 27 months of seed storage unlike the CMV seed produced from China. These results indicate that CMV seeds do not require light for germination and the seed from China should be used within one-year after importation while the seeds produced from Korea can be used even after two years from harvest for stable CMV seedling establishment in the CMV-rice cropping system.

'Youho', A New Forage Barley Cultivar with Ruminant-Palatable Hood Spike Type and Non-Scatteredness (가축 기호성이 높은 내탈립 삼차망 청보리 신품종 '유호')

  • 'Youho' (Hordeum vulgare L.), a new ruminant-palatable forage barley cultivar, was developed by the breeding team at the Department of Rice and Winter Cereal Crop, National Institute of Crop Science, RDA in 2008. It was derived from the cross between 'Suwon339' and 'Suwon355'. Among the cross made in 1999, a promising line, SB992047-B-B-B-6-2, showed good characteristics in potential forage yield in the yield trial tested at Iksan from 2005 to 2006. In 2007, it was designated as 'Iksan431' and placed in regional yield trials at eight locations in Korea for two years from 2007 to 2008, and was released as the name of 'Youho'. It has the growth habit of group II, erect plant type, green leaf and hood spike. Its average heading and maturing dates were on Apr. 24 and May 26, respectively, which are similar to check cultivar 'Yuyeon'. 'Youho' also showed weaker winter hardiness, but better resistance to lodging, shattering and BaYMV than those of check cultivar. It showed higher crude protein content, grade of silage quality than those of check cultivar. The average forage dry matter yield in the regional yield trial was about 14.1, $10.9MT\;ha^{-1}$ in upland and paddy field, respectively, which were 1% to 4% lower than that of the check cultivar. This cultivar would be suitable for the area whose daily minimum temperature was above $-8^{\circ}C$ in January in Korean peninsula.

Nutritional Components and Their Antioxidative Protection of Neuronal Cells of Litchi (Litchi chinensis Sonn.) Fruit Pericarp (리치 과피의 영양화학 성분 및 항산화성 신경세포 보호효과)

  • The nutritional components, antioxidant, and neuroprotective effects of water and a 50% methanol extract from litchi fruit pericarp were investigated. The most abundant mineral, amino acid, and fatty acid were K, proline, and palmitic acid, respectively. In addition, the total water phenolics and 50% methanol extracts were 8.02 and 12.28 mg/g, respectively. The DPPH, ABTS radical scavenging activities and ferric reducing antioxidant power of the water and 50% methanol extracts showed dose-dependent antioxidant activity. In a cell viability assay using MTT, almost all extracts showed a protective effect against $H_2O_2$-induced neurotoxicity, and lactate dehydrogenase leakage was also inhibited by the pericarp extracts. In particular, the 50% methanol extract showed a higher cell membrane protective effect than the water extract at the highest concentration. Consequently, these data suggest that litchi fruit pericarp can be utilized as an effective and safe functional food substances for natural antioxidants and may reduce the risk of neurodegenerative disorders.

Changes in Meat Quality Characteristics on Refrigerated Pork Loin Fed with Supplemental Bamboo Vinegar (죽초액을 급여한 돼지고기의 저장 중 육질특성 변화)

  • This experiment was conducted to investigate the changes in meat quality characteristics on refri- gerated(4$^{\circ}C$) pork loin fed with supplemental levels of Bamboo vinegar(BV). Thirty pigs were divided into 3 groups of 10 pigs. Dietary levels of Bamboo vinegar 0%(control), 2% and 4% were included in experimental diets of each of the groups. The pH value was decreased(P<0.05) in 2% and 4% BV compared to the control after 3d of storage. Lightness was increased(P<0.05) in 2% and 4% BV compared to the control at 1d and 3d. Redness was decreased(P<0.05) in 4% VB compared to the control at 1d and 3d. The 2% and 4% BV showed a significant(P<0.05) increase in yellowness throughout the whole experiment compared to control. Cooking loss was increased(P<0.05) in 2% and 4% BV compared to the control after 3d of storage. Shear force was decreased(P<0.05) in 2% and 4% BV compared to control at 1d and 3d. TBA was decreased(P<0.05) in 2% and 4% BV at 6d and 9d. Total microbial counts was decreased(P<0.05) in 2% and 4% BV compared to control. The 2% and 4% BV scored higher(P<0.05) in odor evaluation compared to the control at 1d and 3d. The 2% and 4% BV also scored higher(P<0.05) than the control in the evaluation of appearance at 1d. The taste was increased(P<0.05) in 2% and 4% BV compared to control at 1d, 3d, and 6d. These results showed that the addition of 2% or 4% BV improved the meat quality and storage characteristics of refrigerated pork.
