Effects of Kinetin on Alleviating Manganese Chloride Toxicity during Rice(Oryza sativa L.) Germination (벼 발아중 염화망간 독성경감에 미치는 Kinetin의 효과)

  • The experiment was conducted to find changes of growth response, free proline content and organic acids on manganese chloride toxicity($4,000mg\;l^{-1}$) for germination and early growth in rice. Root growth was increased in kinetin $10^{-3}M$ compared with control and germination rate was also increased in kinetin $10^{-3}M$ as 89%. Chlorophyll contents was slightly increased in kinetin $10^{-3}M$. Free proline content at 3days in Mn $4,000mg\;l^{-1}$ was higher than at 7 and 10 days, and all kinetin concentrations promoted free proline content. In organic acids, particularly, malic acid was remarkably increased in kinetin $10^{-3}M$ compared with control and Mn $4,000mg\;l^{-1}$.

  • The optimum liquid culture conditions were investigated for cell growth and polysaccharide production from liquid culture of Lentinus edodes. In flask culture, the optimal medium compositions for the polysaccharide production contained glucose 60 g/L, yeast extract 10 g/L, $KH_2PO_4$ 2.0 g/L, and $MgSO_4{\cdot}7H_2O$ 1.0 g/L. The maximum mycelial growth and polysaccharide production were 11.01 g/L and 1.64 g/L, respectively. In bioreactor, through the variation of aeration in order to increase mycelial growth and polysaccharide production, the maximum mycelial growth and polysaccharide production were 55.9 g/L at 8th day and 7.34 g/L at 7th day of cultivation with 1.5 vvm, respectively.

  • 본 연구에서는 홍천 자운지구 고랭지 농업지역을 대상으로 장기간의 유역조사와 하천 모니터링을 통해 관측된 축척 데이터를 이용하여 비점오염저감 효과를 정량화하고 추후 모니터링 자료로서 탁수와 비점오염원 저감시설의 저감효과와 비점오염원의 효율적인 관리를 위한 기초자료를 제공하고자 하였다. 모니터링 결과 소양호 유역의 오염부하(1차 강우사상) 중 홍천군 자운지구의 유역단위 비점오염 저감효과의 분석에는 강우량과 단위면적당 오염부하를 이용하였으며, 자운천은 SS 5,396,761 kg, COD 82,261 kg, BOD 57,329 kg, T-N 68,711 kg, T-P 3,091 kg이었으며, 오염부하(2차 강우사상)는 SS 320,293 kg, COD 34,588 kg, BOD 22,350 kg, T-N 48,954 kg, T-P 640 kg으로 나타났다. 또한 소양호 유역의 EMC(1차 강우사상) 중 자운천은 SS 829.9 mg/L, COD 12.7 mg/L, BOD 8.8 mg/L, T-N 10.567 mg/L, T-P 0.475 mg/L 이었으며, EMC(2차 강우사상)는 SS 68.6 mg/L, COD 7.4 mg/L, BOD 4.8 mg/L, T-N 10.487 mg/L, T-P 0.137 mg/L로 나타났다. 소양호 유역의 단위면적당 오염부하(1차 강우사상) 중 자운천은 SS 402.0 kg/ha/event, COD 6.1 kg/ha/event, BOD 4.3 kg/ha/event, T-N 5.118 kg/ha/event, T-P 0.230 kg/ha/event 이었으며, 오염부하(2차 강우사상)는 SS 23.9 kg/ha/event, COD 2.6 kg/ha/event, BOD 1.7 kg/ha/event, T-N 3.646 kg/ha/event, T-P 0.048 kg/ha/event로 나타났다. 오염부하에서는 1차 강우에 비해 2차 강우에서는 SS 5,076,468 mg/L, COD 47,673 mg/L, BOD 34,979 mg/L, T-N 19,757 mg/L, T-P 2451 mg/L로 1차 강우사상에 비해 오염부하가 낮은 것으로 나타났다. 또한 EMC에서는 1차 강우에 비해 2차 강우에서는 SS 761.3 mg/L, COD 5.3 mg/L, BOD 4.0 mg/L, T-N 0.080 mg/L, T-P 0.338 mg/L로 1차 강우사상에 비해 EMC가 저감되었다. 단위면적당 오염부하는 1차 강우에 비해 2차 강우에서는 SS 378.1 mg/L, COD 3.5 mg/L, BOD 2.6 mg/L, T-N 1.472 mg/L, T-P 0.182 mg/L로 1차 강우사상에 비해 EMC가 저감되었다. 강우량과 강우강도 그리고 영농활동의 시기에 따른 EMC와 단위면적당 오염부하는 큰 차이를 보였으며, 예년에 비해 적은 강수량으로 인해 탁수와 비점오염부하의 배출이 상대적으로 적었던 것으로 판단된다. 소양호 유역과 같은 넓은 유역에서 시행되는 비점오염원의 저감연구는 오랜 시간 동안 자료를 축적해야 유의미한 평가가 이루어질 수 있으므로 장기적이고 지속적인 모니터링을 통한 유량 데이터 자료구축과 수질분석뿐만 아니라, 발생되는 비점오염 물질의 관리를 위한 체계적인 연구가 지속되어야하며, 아울러 농민의 인식도 변화 등을 포함하는 많은 인자들을 정밀히 조사하고 다각도로 분석하여 저감효과에 대한 지속적인 연구가 필요하다.

Azadirachtin, a Novel Biopesticide from Cell Cultures of Azadirachta indica

  • Callus cultures of Azadirachta indica flower petals were established on MS medium supplemented with naphthalene acetic acid (1 mg/L), kinetin (0.5 mg/L) and sucrose $(3\%\;w/v)$. Cell cultures of Azadirachta indica were established and studied the growth and production kinetics. Half 85 medium supplemented with dicamba (2 mg/L), kinetin (1 mg/L) and sucrose $(3\%\;w/v)$ was found to be suitable for initiation and maintenance of cell cultures from the calli. MS medium supplemented with naphthalene acetic acid (NAA) (1 mg/L), kinetin (0.5 mg/L) and sucrose $(3\%\;w/v)$ was found to be suitable as production medium. Around $80\%\;(0.05\%\;w/v)$ of azadirachtin was found to be intracellular. The effect of various precursors, elicitors, permeabilizing agents and growth retardants in cell cultures was studied. The addition of precursors sodium acetate (10 mg/L), squalene (10 mg/L), isopentenyl pyrophosphate (1 mg/L) and geranyl pyrophosphate (1 mg/L) to the cell cultures on day 3 has shown significant increase in bioproduction of azadirachtin $(64.94{\pm}4.40\;mg/L,\;72.81{\pm}0.04\;mg/L,\;51.63{\pm}1.26\;mg/L\;and\;30.70{\pm}0.28\;mg/L\;respectively)$ over the control cultures $(4.70{\pm}0.27 mg/L)$. $5\%$ v/v cell extracts of Fusarium solani has shown moderate increase in the content of azadirachtin $(5.71{\pm}0.34\;mg/L)$ when compared to control cultures $(2.40{\pm}0.56\;mg/L)$. The addition of methyl jasmonate $(500\;{\mu}M/L)$ on day 3 has shown $\~4$ fold improvement in bioproduction of azadirachtln $(6.92{\pm}0.11\;mg/L)$ when compared to control cultures $(1.63{\pm}0.02\;mg/L)$. There was no significant effect of the studied growth retardants and permeabilizing agents on bioproduction of azadirachtin. Cells are cultivated in large volumes using the effective precursors.

Pollutants Unit Loadings of the Stormwater Runoff in Industrial Complex (강우시 산업단지에서의 유출오염부하량 원단위 산정)

  • The objective of this paper is to estimate the pollutant load based on event mear concentrations(EMC) in industrial complex. Eight sub-basins in the Chongju industrial complex were selected for sampling and study with different characteristics during the period from June 1997 to August 1998. During the storm events, measured EMC ranges of $BOD_5$, COD, SS, TKN, TP, HEM, $NO_3-N$, $PO_4-P$, Cr, Pb, Cu and Fe in the industrial complex watersheds were 11~176mg/L, 40~502mg/L, 23~633mg/L, 104~20.9mg/L, 0.22~7.51mg/L, 12.7~548.7mg/L, 0.06~2.66mg/L, 0.12~3.39mg/L, 0.01~0.50mg/L. 0.02~0.42mg/L, 0.01~0.15mg/L and 1.29~11.51mg/L respectively. And the calculated annual average pollutant unit loadings of $BOD_5$ COD, SS, TKN, $NO_3-N$, $PO_4-P$, TP, HEM, Cr, Pb, Cu and Fe were 374.4kg/ha/yr 924.1kg/ha/yr, 983.6kg/ha/yr, 48.8kg/ha/yr, 8.1kg/ha/yr, 9.7kg/ha/yr, 17.8kg/ha/yr 943.0kg/ha/yr, 0.7kg/ha/yr, 0.9kg/ha/yr, 0.3kg/ha/yr and 28.9kg/ha/yr, respectively.

Occurrence of UV Filters in Nakdong River Basin : Mainstreams, Tributaries and STP Effluents (낙동강 수계에서의 자외선 차단제류 검출 특성 : 본류, 지류 및 하수처리장 방류수)

  • This study was investigated occurrence and distribution patterns of UV filters in Nakdong River basin (mainstream, tributaries and sewage treatment plant (STP) effluents). 5 (EHS, BP-3, 4-MBC, BZC and EHMC) out of 7 UV filters were detected in 5 out of 20 sampling sites (mainstream and tributaries), 7 UV filters were not detected in mainstream samples, and the EHS, BP-3, 4-MBC, BZC and EHMC concentration levels in tributary samples were ND~60.8 ng/L, ND~72.1 ng/L, ND~57.2 ng/L, ND~60.1 ng/L and ND~85.2 ng/L, respectively. 5 (EHS, BP-3, 4-MBC, BZC and EHMC) out of 7 UV filters were detected in effluents of 11 STPs around the Nakdong River basin. The EHS, BP-3, 4-MBC, BZC and EHMC concentration levels in 11 STP effluents were ND~89.3 ng/L, ND~90.8 ng/L, ND~88.1 ng/L, ND~118.5 ng/L and ND~104.4 ng/L, respectively. According to the sampling season, distribution patterns and detected concentrations of 5 UV filters were similar in June and September 2014, but change ranges of distribution patterns and detected concentrations of 5 UV filters were highly variable in April and November 2014.

Synthesis of 2-(5,6-dimethoxy-l-indenyl)ethyl amine (2-(5,6-디메톡시-1-인데닐)에틸 아민의 합성)

  • In order to search the new serotonin bioisoster, 2-(5,6-dimethoxy-l-indenyl)ethyl amine(1) was synthesized. 3,4-Dimethoxybenzaldehyde, as starting material, was condensed with malonic acid in the presence of pyridine and piperidine to form 3,4-dimethoxycinnamic acid(2). Compound 2 was performed catalytic hydrogenation with 10% Pd-C to give propanoic acid derivative 3, which was cyclized by Friedel-Crafts acylation to afford 5,6-dimethoxyindan-l-one(4). Compound 4 was reduced with NaBH$_4$ in ethanol to obtain l-indanol 5, and it was dehydrated to give 5,6-dimethoxy-l-indene(6). This compound was lithiated with 2.5M n-butyllithium and reacted with 1,2-dibromoethane to give 2-(5,6-dimethoxy-l-indenyl)ethyl bromide(7), and which was treated with anhydrous ammonia to synthesize compound 1.

  • To establish the hematological reference values in the healthy adults visited our hospitals, following examination were done on 2823 persons by Coulter Counter Model S-plus II ; white blood cell count: (WBC), red blood cell count(RBC), hemoglobin(Hb), hematocrit(Hct), mean corpuscular volume (MCV), mean corpuscular hemoglobin(MCH), mean corpuscular hemoglobin concentration(MCHC), red cell distribution width(RDW), platelet, plateletcrit, mean platelet volume(MPV) and platelet distribution width(PDW). Somatic Embryogenesis in Immature Zygotic Embryo Cultures of Korean Soybean (Glycine max L.) Culitivars and Effect of 2,4-Dichlorophenoxyacetic Acid on Somatic Embryo Morphology (한국 품종 대두(Glycine max L) 미숙배로부터 체세포배발생과 배지의 2,4-Dichlorophenoxyacetic Acid 농도가 체세포배의 형태에 미치는 영향)

  • Immature zygotic embryos of five Korean soybean cultivars cultured on Murashige and Skoog's (MS) medium supplemented with various concentrations of 2,4-Dichlorophenoxyacetic acid (2,4-D) produced somatic embryos without forming an intervening callus. The highest frequency (up to 90%) of somatic embryo formation was obtained when zygotic embryos were cultlued on medium containing 1 to 2 mga 2, 0-D in four cultivars. However the frequency was highly variable to the cultivars. Transversely sliced primary somatic embryo halves were also capable of forming secondary embryos at frequencies of up to 70% when cultured on medium containing 0.1 to 1 mg/L 2,4-D. Somatic embryos formed on zygotic embryos cultured on medium containing 0.1 to 0.2 mg/L 2,4-D had two cotyledons more frequently than one horn-type cotyledon and those on medium containing 0.5 to 4mg/L 2,4-LD had a horm-type cotyledon at a prominently higher freequency. However somatic embryos on medium containing 10mg/L or higher concentrations of 2,4-D were usually shunted at the globular stage even after transfer to medium containing lower concentrations of 2,4-D or other growth regulators. non somatic embryos with one or two cotyledons or a hem-type cotyledon were transferred to medium containing $GA_3$, those with two cotyledons converted to plantlets at a higher frequency (25%) than the others.

  • A new type of DTPA-bis-amides (L1-L4) and their Gd(III)-complexes of the type $[Gd(L)(H_2O)]{\cdot}nH_2O$ (5: L1; 6: L2; 7: L3; 8: L4) have been prepared and characterized by analytical and spectroscopic techniques. The X-ray crystal structure of 8 has been determined for structural confirmation. The coordination geometry adopts a tricapped trigonal prism geometry with L4 acting as a chelate octadentate and a water molecule in the coordination sphere. Crystals are monoclinic, $P2_1$, a = 14.468(3), b = 19.235(4), c = 13.527(2) $\AA$ $\beta$ = $107.245(3)^{\circ}$, V = 3595.2(11) $\AA^3$, Z = 4, $D_{calc}$ = 1.570. Significant increases in relaxivities are observed with 6 and 7 as compared with that of $Omniscan^{(R)}$, a commercial MR agent: R1 = 12.46 $mM^{-1}\;s^{-1}$, R2 = 8.76 $mM^{-1}\;s^{-1}$ for 6; R1 = 12.77 nm-1 s-1, R2 = 7.60 mM-1 s-1 for 7; R1 = 4.9 $mM^{-1}\;s^{-1}$, R2 = 4.8 $mm^{-1}\;s^{-1}$ for $Omniscan^{(R)}$. In the case of 5, however, both R1 and R2 are found to be lower to show 2.09 $mM^{-1}\;s^{-1}$, and 1.82 $mM^{-1}\;s^{-1}$, respectively.
