  • Objective : To suggest a scientific evidence of Aconitum carmichaeli Debx. (Aconiti Lateralis Radix Preparata: ALRP) as one of cooling and heating medicines on the regulation of body temperature, we investigated the effects of ALRP water extract on hypothyroidism. Methods : Hypothyroidism was induced by intradermal injection with PTU for 4 weeks in SD rats. ALRP extract or L-thyroxine as a control drug was orally administrated for 2 weeks with PTU injection in rats. The physiological and serological parameters were measured in rats. The histological change of thyroid tissues was observed by H&E staining, and also the expression of thermo-regulating proteins was determined by Western blot in dorsal root ganglia and brain tissues of rats. Results : The administration of ALRP extract in PTU-induced hypothyroidism rats was significantly increased body temperature, but did not changes on body weight, food and water intake. ALRP extract did not effect on the levels of TSH and T4 in the hypothyroidism rats. ALRP extract significantly decreased the levels of GPT, glucose, HDL-cholesterol, LDL-cholesterol, and total cholesterol in the hypothyroidism rats. In histological observation, the enlarged epithelium and atrophic follicles with higher concentration of follicular cells on hypothyroidism were improved by ALRP extract. In addition, ALRP extract increased the expression of TRPV1 and TRPM8 ion channel proteins in hypothyroidism rats. Conclusion : These results indicate that ALRP extract can improve PTU-induced hypothyroidism through regulation of body temperature and lipid accumulation. The action mechanism of ALRP extract is related with body temperature control by thermoregulation with TRP ion channels.

  • Genetic divergence and phylogenetic relationships among the major Korean fireflies (Hotaria papariensis, Luciola lateralis, and Pyrocoelia rufa) were studied. A portion of mitochondrial COI (403 bp) and 165 rRNA (490~504 bp) genes were sequenced, and the GenBank-registered, homologous 165 rRNA sequences of Japanese fireflies were compared (27 species of Lampyridae, one of Lycidae, and one of Rhgophthalmidae). Greatest DNA and/or amino acid sequence divergence was found when P rufa, belonging to Lampyrinae was compared with H. papariensis and L. lateralis, both belong-ing to Luciolinae, confirming the current taxonomic status of the species. In the PAUP and PHYLIP analyses with 165 rRNA data, grouping of the two geographic samples of H. papariensis with H. tsushimana validate the use of generic name, Hotaria. Nevertheless, lack of sister-group relationship of the two geographic samples of H. papariensis renders further investigation on this group . Although the Korean and Japanese L. lateralis formed a strong monophyletic group, a substantial genetic differentiation was detected between them (2.9% of 165 rRNA gene sequence divergence). Finally, the geographic samples of Korean p. rufa strongly formed a group with Japanese p. rufa, warranting the use of generic name, Pyrocoelia, but the genetic distance observed between the Cheju-Island individual and all others requires further investigation on this subject. Summarized, this study supports the current taxonomic status of the Korean fireflies in that each respectively formed a strong monophyletic group with its own species or genus.

  • Effects of temperature on the development and reproduction of the Luciola lateralis were investigated at various temperatures. The development time of eggs, larvae, and pupae were shorter at higher temperatures than at lower ones. The insect did not develop at 10$^{\circ}C$ and 35$^{\circ}C$. The hatchability was 61.5% at 15$^{\circ}C$, 73.9% at 20$^{\circ}C$, 93.3% at 23$^{\circ}C$, 91.8% at 25$^{\circ}C$, 74.0% at 27$^{\circ}C$, and 46.0% at 30$^{\circ}C$, indicating the best hatchability rate at the temperature condition of 23 DC. Larval periods were 341.5:t 23.2 days at 15$^{\circ}C$, 265.5${\pm}$17.5 days at 20$^{\circ}C$, and 250.9${\pm}$11.7 days at 25$^{\circ}C$. Pupal periods were 94.7${\pm}$11.5 days at 15$^{\circ}C$, 41.7${\pm}$9.1 days at 20$^{\circ}C$, and 18.5${\pm}$7A days at 25$^{\circ}C$. Emergence rate was 23.3, 89.3 and 80.7%, respectively at the above temperatures. Adult longevity of female was 18.0 days at 15$^{\circ}C$, 2004 days at 20$^{\circ}C$, 10.7 days at 25$^{\circ}C$, and 5.8 days at 30$^{\circ}C$. Mean fecundity per female was higher at 20$^{\circ}C$ compared with at other temperatures. The developmental zero point temperatures (1) and the total effect temperatures (I<) of egg, larva, pupa, and complete development were 10.6, 14.0, and l3.1$^{\circ}C$ and 214.8, 1,564.8, and 229.2 degree-days, respectively. Mean generation time in days (T) was shorter at higher temperature. Net reproductive rate per generation (Ra) was the lowest at the highest temperature as well as at the lowest, and it was 177.19 which was the highest at 23$^{\circ}C$. The intrinsic rate of natural increase (r$\sub$m/) was highest at 27$^{\circ}C$ as 0.019. As a result, optimum range of temperature for L. lateralis growth was between 20-25$^{\circ}C$.

  • In order to study on the relative significance of nanoplankton in Chonsu Bay, nanoplankton samples were collected and analyzed monthly from September, 1985 to August, 1986. A total of 33 taxa representing 6 phyla, 8 classes, 13 orders, 17 families, 25 genera, 33 species have been identified. Micromonas pusilla, Pedinomonas mikron, Pyramimonas grosii, Chroomonas lateralis, Pyrenomonas salina (=Chromonas salina), chroomonas sp., Cyclotella sp., Gonyaulax sp., unidentified sphericl monads (2-5um and 6-8um in size), and unidentified naviculiod form were common species. the distribution of nanoplankton standing crops showed a great temporal and spartial variations. Nanoplankton standing crops was highest in October, 1985 and lowest in September, 1985. The abundance of nanoplankton in Chonsu Bay may be within the range of that of most coastal areas. Unidentified spherical monada (2-5um and 6-8um in size) were most dominant. Chlorophyll-a concentrations of nanoplankton ranged from 0.81 to 4.78ug/l and daily primary productivity by nanoplankton, 16.4 to 767.2 mgC/m$^2$/day. Nanofraction of total phytoplankton cell number accounted for 38% to 93% (average 6%), chlorophyll-a and primary productivity of nanoplankton 25 to 87% (average 64%) and 9 to 87% (average 53%), respectively. The results implied that nanoplankton could be a considerable contribution to phytoplankton biomass and primary productivity in Chonsu Bay phytoplankton community.

  • Purpose: The purpose of this study was to assess the effects of robo-horseback riding with changes of pelvic tilting and speeds on muscle activities of trunk and lower limb in healthy adults. Methods: Thirty healthy adults were recruited for this study. Subjects performed robo-horseback riding at three pelvic postures (anterior tilting pelvic posture: AP, neutral pelvic posture: NP, posterior pelvic posture: PP) and different speeds (moderate and fast speed). Muscle activities were recorded from the trunk muscles (rectus abdominis [RA], T2 paraspinalis [T2 PS], L2 paraspinalis [L2 PS]) and lower limb muscles (rectus femoris [RF], vastus medialis [VM], vastus lateralis [VL], biceps femoris [BF], gastrucnemius [GCM]). Results: Significantly higher RA activities were observed under the PP condition than under the AP and NP conditions at both speeds. Significantly higher L2 PS activities were observed under the AP condition than under the PP and NP conditions at both speeds. Significantly higher T2 PS activities were observed under the AP condition than under the PP and NP conditions at fast speed. However, the muscle activities of the VM and GCM did not show significant changes with changes in pelvic posture at both speeds. All muscle activities were significantly higher under the fast speed than under the moderate speed in three pelvic postures. Conclusion: There were significant differences in the changes of pelvic tilting on muscle activities of trunk and lower limb were observed in healthy adults. NP condition can be introduced the most efficienct posture for muscle activities of trunk and lower limb in robo-horseback riding.

  • Objectives: As the use of herbal medicinal products (HMPs) increases worldwide, systematic verification of the safety of HMPs is required. The induction of cardiotoxicity is one of the major factors in post-approval withdrawal of medicinal products, and drug-induced cardiotoxicity assessment is emerging as an important step in drug development. In the present study, we evaluated human ether-à-go-go-related gene (hERG) potassium channel-related cardiotoxicity to predict the risk of cardiac arrhythmia in thirteen herbal medicines known to have cardiac toxicity. Methods: We measured the inhibition rate of hERG potassium channel activity of 13 medicinal herbal extracts in hERG-expressing HEK 293 cells using an automated patch-clamping system. Quinidine was used as a positive control for inhibition of hERG activity. Results: Extracts of Evodiae Fructus, Strychni Semen, and Corydalis Tuber potently inhibited the activity of hERG, and IC50 values were 3.158, 19.87, and 41.26 ㎍/mL, respectively. Cnidi Fructus, Ephedra Herba, Lithospermi Radix, Polygoni Multiflori Radix, Visci Ramulus et Folium, Asiasari Radix et Rhizoma, and Scolopendra weakly inhibited hERG activity, and the IC50 value for each herbal medicine was more than 400 ㎍/mL. Aconiti Kusnezoffii Tuber and two types of Aconiti Lateralis Radix Preparata (Po and Yeom) had weak inhibitory activity against hERG, and the IC50 values were more than 700 ㎍/mL. The IC50 value of quinidine against hERG was 1.021 𝜇M. Conclusion: Evodiae Fructus, Strychni Semen, and Corydalis Tuber acted as potent inhibitors against hERG. These herbal medicines may cause cardiac arrhythmia through QT prolongation, so care should be taken when taking them.

  • We determined the effects of dietary manipulations on messenger RNA of peroxisome proliferators activated receptor isoforms (i.e., PPAR ${\alpha},\;{\beta}/{\delta},\;{\gamma}$) in red vastus lateralis muscle of rats. Total 16 male Sprague-Dawley rats were used, and animals were divided into one of two dietary conditions: either chow diet group (CHOW; n=8) in which animals were 134 with standard rodent chow (61.8% carbohydrate, 15.7% fat, 22.5% protein) or high fat diet group (FAT n=8) in which animals were fed 24.3% carbohydrate, 52.8% fat, 22.9% protein. At the end of the 8 weeks of experimental period, red vastus lateralis muscle was dissected out from all animals, and PPAR ${\alpha},\;{\beta}/{\delta},\;{\gamma}$ mRNA expression was determined. There was no significant difference in body mass (BM) between CHOW and FAT. As expected, blood glucose and free fatty acid (FFA) concentration was higher in FAT than CHOW (p<0.05), and lactate concentration was significantly lower in FAT compared to CHOW (p<0.05). Insulin concentration tended to higher in FAT than CHOW ($67.2{\pm}21.9\;vs.\;27.0{\pm}5.2$ pmol/L), but it did not reach to the statistical significance. Gene expression of PPAR ${\alpha}$ was not significantly different between CHOW and FAT. It was not also significantly different in PPAR ${\beta}/{\delta}$. Interestingly, expression of mRNA in PPAR ${\gamma}$ however, was markedly depressed in FAT compared to CHOW (approximately 3 fold higher in CHOW; p<0.05). Results obtained from present study implies that PPAR ${\gamma}$ (as compensatory function of PPAR ${\alpha}$ is expressed) possibly exerts another major tuning roles in fatty acid transport, utilization, as well as biosynthesis in skeletal muscle cells. The situations and conditions that can be postulated for this implication need to be further examined.

  • The antiserum against locustatachykinin I, originally isolated from brain and retrocerebral complex of the locust Locusta migratoria, has been used to investigate changes in number, localization, and structure of locustatachykinin I-immunoreactive (LomTK I-IR) neurons in the brains of the common cutworm, Spodoptera Iitura, during postembryonic development. These neurons are found at larval, pupal, and adult stages. In the larval stages, the first instar larva shows the first appearance of about 8 LomTK I-IR neurons. These neurons gradually increase in number from the second to fourth instar larvae which have the largest number of about 92 in all postembryonic stages. Thereafter, these neurons decrease to about 28 in number in the 5-day-old pupa. However, they begin to rise again from the 7-day-old pupal stage, eventually reaching to about 90 in the l-day-old adult. The developing larval brains contain cell bodies of these neurons in most neuromeres. After the metamorphosis of larva to pupa and adult, localization of these neuronal cell bodies is confined to the specific cerebral neuromeres. The 7-day-old pupal brain shows the location of these neuronal cell bodies in pars intercerebralis, pars lateralis of protocerebrum, deutocerebrum, tritocerebrum, optic lobe-near region, and subesophageal ganglion. In the l-day-old adult, however, the brain has these cell bodies only in some neuromeres of protocerebrum, deutocerebrum, and subesophageal ganglion. Throughout the postembryonic life, changes in structure of these neurons coincide with changes in number and localization of these neurons. These findings suggest that changes in number, localization, and structure of these neurons reflect differentiation of these neurons to adult type.

  • The aim of the present study is to examine the brainstem sites where the electrical stimulation produces a suppression of dorsal horn neuron responses of neuropathic rats. An experimental neuropathy was induced by a unilateral ligation of L5-L6 spinal nerves of rats. Ten to 15 days after surgery, the spinal cord was exposed and single-unit recording was made on wide dynamic range (WDR) neurons in the dorsal horn. Neuronal responses to mechanical stimuli applied to somatic receptive fields were examined to see if they were modulated by electrical stimulation of various brainstem sites. Electrical stimulation of periaqueductal gray (PAG), n. raphe magnus (RMg) or n. reticularis gigantocellularis (Gi) significantly suppressed responses of WDR neurons -to both noxious and non-noxious stimuli. Electrical stimulation of other brainstem areas, such as locus coeruleus. (LC) and n. reticularis paragigantocellularis lateralis (LPGi), produced little or no suppression. Microinjection of morphine into PAG, RMg, or Gi also produced a suppression as similar pattern to the case of electrical stimulation, whereas morphine injection into LC or LPGi exerted no effects. The results suggest that PAG, NRM and Gi are the principle brainstem nuclei involved in the descending inhibitory systems responsible for the control of neuropathic pain. These systems are likely activated by endogenous opioids and exert their inhibitory effect by acting on WDR neurons in the spinal cord.
