Effect of Bleaching and Permanent Wave Manipulation on the Amino Acid Composition of Hair (모발 탈색 및 퍼머넌트 웨이브가 모발의 아미노산 조성에 미치는 영향)

  • Variations of the seventeen amino acids(aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystein, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, histidine, lysine, arginine) were analyzed in human hair sample by amino acid auto analyzer(AAA). The effect of bleaching and permanent wave manipulation on the amino acid composition of hair were investigated. Hair samples were collected from 10 males in their thirties. Hair samples were treated with 10 mL of 6 N hydrochloric acid at $110^{\circ}C$ for 24 h and analysed by AAA. The results showed that the amino acid content of normal hair(73.9%) decreased to 71.5% and 69.3% after bleaching and permanent wave treatment, respectively. Furthermore, mean contents of lysine and tyrosine in amino acids showed obviously decreased about 25% by permanent wave and bleaching treatment. On the other hand, serine, cystein, leucine and histidine were not changed.

Effects of Amino Acids on Larval Settlement and Metamorphosis in Haliotis discus hannai

  • The compositions of amino acid in 6 monocultured benthic diatoms used in aquaculture of Haliotis discus hannai were analyzed, and effects of 15 artificial synthetic amino acids on the settlement and metamorphosis of H. discus hannai larvae. Results showed that the content of L-aspartic acid was highest in all diatoms, and that of L-glutamic acid was second high. In experiment using settlement slat without benthic diatom attached, the highest settlement rate (33.3 ${\pm}$ 8.8%) was obtained with L-glutamic acid, and a higher value (16.7 ${\pm}$ 3.3%) was found with L-aspartic acid at 24 h after experimental commencement, compared to that of control (8.6 ${\pm}$ 5.1%). 80 h later the metamorphosis rates of L-glutamic acid (86.7 ${\pm}$ 10.7%) and L-aspartic acid (80.0 ${\pm}$ 3.3%) groups were higher than control group(0) and other amino acids significantly. The response rate of L-glutamic acid was the highest (62.0%), and those of L-aspartic acid (30.0%) and L-threonine (25.3%) groups were also significantly higher than control group. In the experiment using settlement slat with benthic diatom attached, the best effect of various amino acids on induction of larval settlement was obtained with L-glutamic (82.0 ${\pm}$ 6.9%) and L-aspartic acid (78.7 ${\pm}$ 5.1%) at 24 h after experimental commencement. The settlement rates of L-histidine, L-leucine, L-lysine, L-methionine, L-phenylalanine, and L-tyrosine groups were significantly lower than control group. The same differences in the metamorphosis rate at 56 h after experimental commencement and in the response rate were found. It should be noted that after 80 h the metamorphosis rates of L-histidine (74.0 ${\pm}$ 12.0%) and L-lysine (87.0 ${\pm}$ 8.8%) declined rapidly compared to those of 56 h (8.0 ${\pm}$ 12.0%; 7.7 ${\pm}$ 12.0%).

The Glucose Repression of Aerial Mycelium Formation in Streptomyces (Streptomyces의 Aerial Mycelium 형성에 대한 Glucose 억제 기작에 관한 연구)

  • We have demonstrated that both L-histidine as an amino acid factor and dextrin as a carbon source were required for the glucose repression. 1% glucose was sufficient to the glucose repression of aerial mycelium formation in Streptomyces lavendulae and Streptomyces aureofacience. the synthesized medium, KK, which is lack of all orgnic nutrients except dextrin was able to induce glucose repression, but the addition of 0.003% or more L-histidiner recovers the capacity of glucose repression. 0.02% or more histidine was reuqired for glucose repression of aerial mycelium formation in the absence of dextrin. Treatments of $5{\mu}M$ ormore ethidium bromide (EtBr0 gave rise to bald mutants at high frequency in Streptomyces aureofaciens, and it is probable that the gene(s) for the function of aerial mycelium formation is linked to plasmed DNA in this species.

  • L-Histidine tetrafluoroborate single crystals have been grown from the aqueous solution. The profitable pH value to grow large crystals, the relative flow rate to get clear crystals, crystals habit and the orientation of the obtained crystal have been clarified. We have also demonstrated that the LHBF crystal has very high damage threshold which is potentially good for generation of the phase conjugated waves.

  • This study has been performed for physico-chemical property and composition of sinew, testes, tail, blood and velvet antler in Elk. Amino acid contents in the sinew taurine, alanine, histidine, and lysine were high contained, histidine, glutamic acid, taurine, and lysine were high contained in testes, glutamic acid, lysine, alanine, glycine, and phenylalanine were high contained in tail, histidine, glycine, and lysine were high contained in blood, glutamic acid, lysine, taurine, alanine, and glycine were high contained in velvet antler. And, based on the amount of mineral, $K^{+}$, $Ca^{2+}$, and $P^{+}$ were 444.8, 166.6 and 242.9mg per 100g in sinew, respectively. $K^{+}$, $Ca^{2+}$ and $P^{+}$ were 294.4, 330.5, and 514.3 mg per 100g in testes, respectively, $K^{+}$, $Mg^{2+}$, and $P^{+}$ were 1420.6, 118.4, and 1105.2mg per 100 mg in tail, respectively. Fe3+,-K+, and P+ were 344.1, 1023.6 and 157.2 mg per 100 mg in blood, respectively and $K^{+}$, $Ca^{2+}$ and $P^{+}$ were 888.4, l1533.1 and 14722.0 in velvet antler, respectively. Finaly, difference were found in comparison of composition of free mwtal ion (N $a^{+}$, $K^{+}$, $Mg^{2+}$ and $Ca^2$) bwteen blood, tail, tests, sinew and velvet antler, and composition of free metal in blood higher than those othersd higher than those others.

  • Rice bran was employed as a main medium component for ethanol fermentation by Saccharomyces species. Among the several strains of .Saccharomyces yeasts. S. cerevisiae IF0 2346 was selected as the hest strain in view of the interest in the production of ethanol and amino acids. It was found that S. cerevisiae IF0 2346 showed $3\times 10^8$cells/d and 4.7% (v/v) ethanol production after 72 hr cultivation. Although total amount of free amino acids was decreased from 1.099 mg/l to 829 mg/l during the fermentation, glutamic acid. histidine, and isoleucine were increased considerably. With the supplement of 5% glucose to the ferrnentation medium, both ethanol and amino acid production were increased up to 134% and 264%, respectively. compared to the control case. Glutamic. acid, leucine, alanine. phenylalanint:, and valine were the major amino acids in the fermentation broth.

Experimental Measurement and Correlation of two α-Amino Acids Solubility in Aqueous Salts Solutions from 298.15 to 323.15 K

  • By the gravimetric method at atmospheric pressure, the solubility of two α-amino acids was resolved over temperatures from (293.15 to 323.15) K. The α-amino acids studied were L-arginine and L-histidine. Results showed a salting-out effect on the solubility of the tested amino compounds. It is obvious that there was an increase in the solubility, in aqueous chloride solutions, with the increasing temperature. Results were translated regarding the salt hydration shells and the ability of the solute to form hydrogen-bond with water. The solubility data was precisely associated with a semi-empirical equation. The standard molar Gibbs free energies of transfer of selected α-amino compounds (ΔtrGo) from pure water to aqueous solutions of the chloride salts have been calculated from the solubility data. The decrease in solubility is correlated to the positive (ΔtrGo) value which is most part of the enthalpic origin.

  • Gallena mellonella L. was parasitized by the pupal parasitoid, Brachymerica lasus Walker in the laboratory. Haemolymph total protein and carbohydrate concentrations from the hosts parasitized by B. lasus decreased within 1st day following parasitization. However, haemolymph total free amino adds, glycine, proline, arginine, isoleucine, phenylalanie, leucine, valine, glutamk acid, histidine, serine, and tyrosine increased and free amino acids decreased were threonine, aspartic acid, methionine, cysteine, alanine, and lysine.

  • Changes in contents of general components of comfrey cultured in Korea were experimented and amino acids were analyzed by thin layer chromatography (T.L.C.) and gas chromatography (G.L.C.). The results obtained were as follows; 1. General components of comfrey such as moisture, fat, protein, carbohydrate, crude fiber and total ash were 13.20, 2.22, 22.30, 37.62, 9.38 and 15.06%. respectively, after 60 days growth. 2. The root of comfrey after 60 days contained 6.03% of alanine, 2.24% valine, 10.77% arginine, 2.96% glycine, 4.08% histidine. 1.54% isoleucine, 0.58% cystein, 1.72% methionine, 7.55% aspartic acid, 7.81% glutamic acid and 4.65% lysine in the gas chromatographic analysis of amino acid composition 3. The crude protein was decreased after 60 days of growth whereas the contents of carbohydrate, crude fiber and total ash were increased. 4. The total amount of amino acids in root was greater than that in leaf of comfrey.
