알루미늄 용액과 갈근 열수 추출액의 급여가 흰쥐의 Aldosterone, rennin의 호르몬과 GOT, GPT. GOT는 농도를 달리한 알루미늄 단독 급여군은 $258.60{\sim}395.80IU/L$인데 비하여 알루미늄과 갈근 열수 추출물 병합 급여군이 $314.20{\sim}320.20IU/L$으로 유의적으로 감소하였다. GPT는 농도를 달리한 알루미늄 급여군은 $100.40{\sim}108.20IU/L$인데 비하여 갈근 열수 추출액 급여군과 알루미늄 병합 급여군은 $70.80{\sim}81.00IU/L$으로 유의적으로 감소하였다. LDH는 농도를 달리한 알루미늄 급여군은 $3157.20{\sim}3726.20IU/L$인데 비하여 갈근 열수 추출액 급여와 알루미늄 동시 병합 급여군은 $2846.60{\sim}2993.20IU/L$으로 유의적으로 감소하여 갈근 열수 추출액 급여군에 의한 알루미늄 중독 완화 효과를 볼 수 있었다.
표고버섯균사체 고체배양물 분말과 액체배양물 분말을 2:1 (w/w)로 혼합한 시료 (LED로 명명)가 $CCl_4$로 유발된 숫컷 쥐의 간독성을 보호하는 효과에 대해 연구하였다. Vehicle 처리군(0.2 ml 증류수), Control 처리군(0.2 ml 증류수), LED 처리군(LED 100, 200, 300, 400 mg/kg BW을 0.2 ml 증류수에 각각 혼합), 그리고 Silymarin 처리군(200 mg/kg을 0.2 ml 증류수에 용해)의 각 처리군(군당 여섯 마리)에 매일 2주간 투여한 다음, Vehicle 처리군을 제외한 모든 군에 $CCl_4$ ($CCl_4$:corn oil, 1:1 v/v; 0.5 ml/kg BW)를 복부에 주사하였다. 하루가 지난 후에 생화학적 지표성분을 분석하기 위해 혈액과 간 시료를 채취하였다. 모든 LED 처리에 의해 간 SOD, catalase, 그리고 GSH peroxidase의 활성이 증가하였으며, TBARS, tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin-$1{\beta}$ (IL-$1{\beta}$) 그리고 interleukin-6 (IL-6)가 감소하였다. 그 결과, 혈청중의 GOT, GPT, 그리고 LDH의 활성이 감소하였다. 본 연구에서는 젖산발효를 통해서 얻어진 목이버섯과 녹각 추출액 혼합물의 항산화 및 생리활성평가를 수행하였다. 녹각 추출액의 GABA생성 최적조건에서 probiotics 및 기능성 강화를 위해 목이버섯을 2.5% 첨가하여 $30^{\circ}C$에서 7일간 젖산발효를 하였다. 발효 7일 pH 5.06, 산도 0.77%로 나타났으며 $1.3{\times}10^8CFU/mL$로 높은 균수를 유지하였고 GABA를 1.4% 생성하는 것으로 나타났다. 목이버섯을 첨가했을 때 젖산균 발효물의 물성이 개선되며 단기간에 고농도의 GABA를 생성하는 것으로 나타났다. 녹각 추출액의 젖산발효물의 세포 생존율을 실험한 목이버섯 2.5% 조건에서 발효 전 6 mg/mL에서 독성이 나타났으나 발효 후 독성이 완화되는 것으로 나타났다. 발효 후 6 mg/mL 농도에서는 $5.58{\mu}M$로 발효 후 NO 생성이 감소되는 것으로 나타났다. 결론적으로 녹각 추출액과 목이버섯 혼합물의 젖산균을 이용한 정치배양을 통해 단기간에 고농도의 GABA 생산이 가능하였으며, 젖산 발효물은 세포독성 완화 효과 및 NO 생성을 저해하는 것으로 나타났다. 따라서 발효물은 GABA, probiotic, 식이섬유 등을 함유하여 기능성 식품소재로 이용이 가능할 것으로 기대된다.
황칠나무 추출물의 순차적 복합발효를 통한 복합 기능성 물질들을 강화시킨 발효소재를 개발하였다. 황칠 추출물에 MSG와 glucose를 첨가한 후 1차 B. subtillis HA 발효를 통해 γ-PGA와 단백질 분해효소를 생성시켰다. 이후 skim milk와 glucose, yeast extract를 추가적으로 첨가하여 30℃에서 L. plantarum KS2020에 의한 2차 복합 발효를 통해서 기능성 물질인 GABA를 생산하였다. 황칠 추출물의 고초균 발효에서 생성된 점질물 및 점조도은 발효 2일 4.21±0.04 Pa·s, 4.1%±0.48%로 높은 점조도 값과 점질물을 함량을 보였다. pH 및 산도는 각각 8.9±0.01, 0.00±0.00이었으며 생균수는 7.27 log CFU/mL에서 발효 2일에 9.50 log CFU/mL로 높은 생균수를 보였다. 순차적 복합 발효가 진행되면서 고초균의 생균수는 4.30 log CFU/mL로 크게 감소하였으며 젖산균은 9.12 log CFU/mL로 증가하였다. Protease 활성 및 tyrosine 함량은 각각 0.8 unit/g, 57 mg%로 측정되었다. 이후 연속적 복합 발효 과정에서 발효 5일 차 106 mg% 정도로 증가되었으며, SDS-PAGE를 이용하여 casein 단백질이 분해된 것을 확인하였다. GABA 정성분석 결과 skim milk 5%만을 첨가한 조건을 제외하고 glucose, yeast extract가 추가로 첨가된 모든 조건에서 전구물질 MSG가 소진되면서 GABA로 전환되었다. 이중 최적 조건을 골라 정량분석을 진행하였으며 18.29 mg/mL의 GABA 함량을 나타내었다. 최종 황칠 복합 발효물은 γ-PGA, peptides, 고농도 GABA를 함유한 발효소재로써 다양한 식품의 원료 활용이 기대된다.
본 연구를 통해 BAGGT를 재조합 B. subtilis를 이용하여 대량 생산하기 위하여 유전자 클로닝, 발현 시스템 구축 및 배지 최적화를 진행하였다. 이중프로모터 시스템을 이용하여 야생형 균주에 비해 42배 효소 생산성이 향상된 발현 시스템을 구축하였다. 또한 PBD 분석을 통해 당밀과 CSL이 재조합 B. subtilis 시스템에서 BAGGT의 생산성에 큰 영향을 주는 인자임을 확인하였으며, 염류의 첨가에 의한 효소 생산성 증대 효과는 미비하거나 부정적이었다. 탄소원으로 당밀을 선택하고 고가의 질소원인 트립톤을 저가의 CSL로 교체한 후 CCD 분석을 통해서 결정된 최적배지 사용 시 최적화 이전의 LB 배지 대비 4.3배의 생산성 증대를 이루었으며, 이는 LB 배지에서 야생형 균주의 BAGGT 생산성 대비 180배의 효소 생산성 개선에 해당하였다. 본 연구를 통해 식품용 효소로서 BAGGT의 대량생산을 위한 공정을 구축하였으며, 이후 정미성 소재 생산에 활용할 수 있을 것으로 기대한다.
본 연구는 국내 각 지역의 목장에서 수거한 원유에서 분리된 젖산균을 대상으로 monosodium glutamate 1%가 첨가된 10% 환원탈지유에 $37^{\circ}C$에서 18시간 배양한 후 GABA 양이 $711.40{\mu}g/g$ D.W을 생산하는 우수한 균주를 선발하였다. 선정된 균은 Gram 양성, rod형태의 동종발효균이며, 당 발효실험과 16S rDNA 분석결과 Lactobacillus acidophilus로 판명되었고, L. acidophilus RMK567로 명명하였다. 발효유에 적합한 starter인지 확인하기 위해 생리적 특성을 조사하였다. L. acidophilus RMK567은 배양온도 $40^{\circ}C$에서 빠른 생장을 보였고, pH 4.3에 도달하는데 15시간이 소요되었다. 16종의 항생제 중 kanamycin, neomycin, streptomycin에 대해 내성이 높았으며, 효소활성실험에서 leucine arylamidase와 ${\beta}$-galactosidase의 활성도가 높았다. 담즙 첨가시 약간의 영향을 받았으나 담즙에 대한 내성이 있는 것으로 나타났으며, pH 내성 실험결과 pH 2에서 큰 변화가 없음에 따라 내산성이 있었다. 항균력 시험에서는 Escherichia coli에 대해 29.2%, Salmonella typhimurium와 Staphylococcus aureus에 대해 각각 39.1%, 51.4%의 억제력을 보였다. 이러한 결과를 토대로 GABA 생성능이 우수한 기능성 발효유 제품의 스타터로 L. acidophilus RMK567은 적합하다고 할 수 있다.
The methanol extracts were prepared from 46 oriental medicines currently used for stroke treatment, and the effects were assessed on the excitotoxic neuronal cell death induced by L-glutamate(Glu) in primary cultured rat cortical neurons. The extracts from Angelicae gigantis Radix, Manitis Squama, Acori graminei Rhizoma, Uncariae Ramulus et Uncus, Alpiniae Fructus, Paeoniae Radix, and Cnidii Rhizoma inhibited the Glu-induced neurotoxicity with the IC$_50$ values of 95.2, 218.6, 263.3, 295.1, 297.9, 310.1, and 446.7 $\mu$g/ m$\ell$, respectively. The extracts from Arisaematis Rhizoma, Loranthi Ramulus, Anemarrhenae Rhizoma, Carthami Flos, Clematidis Radix, Bambusae Concretio Silicea, and Angelicae koreanae Radix also exhibited significant inhibition of the toxicity. In contrast, the extracts from Aconiti Tuber Araliae cordatae Radix, Curcumae Rhizoma, Leonuri Herba, Polygalae Radix, Salviae Radix, and Siegesbeckiae Herba increased the Glu-induced toxicity at the concentrations of 500 and 1000 $\mu$g/m$\ell$. Rest of the extracts evaluated in the present study showed minor or negligible inhibition. liken together the oriental medicines including Angelicae gigantis Radix, Muitis Squama, Acori graminei Rhizoma, Uncariae Ramulus et Uncus, and Alpiniae Fructus appear to exert pharmacological effects through the inhibition of excitotoxic neuronal cell death. BAF53 is an actin-related protein that shuttles between nucleus and cytoplasm. In the nucleus, it constitutes an integral component of many chromatin-modifying complexes such as the SWI/SNF, TIP60, TRRAP, and TIP48/49 complexes. BAF53 is essential for growth, but its function remains elusive. BAF53 homologues from yeast to humans have a conserved N-terminal motif, MS_(G/A)(G/A)__(V/L)YGG, which is unique to these proteins. Previously we showed that over-expression of an N-terminal deletion mutant of BAF53 ($BAF53_-{\Delta}N$) reduced the viability of HEK293 and HeLa cells. When we replaced the serine 2 and tyrosine 6 of this N-terminal motif with alanine, over-expression of the alanine-replaced BAF53 strongly impaired the growth of HEK293 cells whereas replacement with aspartate/glutamate had no effect. The alanine-replaced BAF53 mutants also stimulated p53-dependent transcription, in which the SWI/SNF and TRRAP complexes are involved. Our results demonstrate that serine 2 and tyrosine 6 play important roles in BAF53 activity.
Poly-${\beta}$-hydroxybutyrate (PHB) and enzymes related PHB metabolism have been measured in nitrogen-fixing symbiosis of chickpea and cowpea plants. Bacteroids from chickpea and cowpea contained PHB to 0.8% and 43% of their dry weight, respectively, whereas the free-living cells CC 1192 and I 16 produced $285{\pm}55mg$ and $157{\pm}18mg$ of PHB g (dry weight)$^{-1}$. To further understand why chickpea bacteroids contained little PHB, the enzyme activities of PHB metabolism (3-ketothiolase, acetoacetyl-CoA reductase, PHB depolymerase, and 3-hydroxybutyrate dehydrogenase), the TCA cycle (malate dehydrogenase, citrate synthase, and isocitrate dehydrogenase), and related reactions (malic enzyme, pyruvate dehydrogenase, and glutamate:2-oxoglutarate transaminase) were compared in extracts from chickpea and cowpea bacteroids and the respective free-living bacteria. Significant differences were observed between chickpea and cowpea bacteroids and between the bacteroid and free-living forms of CC 1192, with respect to the capacity for some of these reactions. It is indicated that a greater potential for oxidizing malate to oxaloacetate in chickpea bacteroids could be a factor that favors the utilization of acetyl-CoA in TCA cycle rather than for PHB synthesis.
Certain dietary contents, biological supplements might influence the occurrence or treatment of epilepsy. Some studies have found that the supplementation with individual nutrients reduced seizure frequency or improved other aspects of health in patients with epilepsy. Potentially beneficial dietary interventions include treating blood glucose dysregulations. Identifying and avoiding allergenic foods, and avoiding suspected triggering agents such as alcohol, aspartame, and monosodium glutamate. The Atkins diet (very low in carbohydrates) is a less restrictive type diet that may be effective in some cases. Nutrients that may reduce seizure frequency include vitamin B6, magnesium, vitamin E, manganese, taurine, dimethylglycine, and omega-3 fatty acids. Administration of thiamine may improve cognitive function in patients with epilepsy. Supplementation with folic acid, vitamin B6, biotin, vitamin D, and L-carnitine may be needed to prevent or treat deficiencies resulting from the use of anticonvulsant drugs. Vitamin K1 has been recommended near the end of pregnancy for women taking anticonvulsants. Melatonin may reduce seizure frequency in some cases, and progesterone may be useful for women with cyclic exacerbations of seizures. In most cases, nutritional therapy is not a substitute for anticonvulsant medications. However, in selected cases, depending on the effectiveness of the interventions, dosage reductions or discontinuation of medications may be possible. However, nutrient supplementation may be necessary to prevent or reverse the effects of certain deficiencies that frequently result from the use of antiepileptic drugs.
