Effect of Supplementing Betaine on Performance, Carcass Traits and Immune Responses in Broiler Chicken Fed Diets Containing Different Concentrations of Methionine

  • An experiment was conducted with broiler (Cobb 400) male chicks (n = 480) to determine the effect of betaine (Bet) supplementation (0 and 800 mg/kg) to diets containing five concentrations (15, 18, 20, 22 and 24 g/kg crude protein, CP) of methionine (Met) in a $2{\times}5$ factorial study for performance, carcass traits, immune responses, and serum parameters. Each diet was fed ad libitum from 1 to 42 d of age to 8 replicates of 6 chicks. Birds were housed in battery brooders placed in an open-sided poultry shed. Body weight gain, feed intake, feed conversion efficiency and slaughter variables were recorded at 21 and 42 d of age. Serum biochemical profile, antibody production against Newcastle disease (ND) and lymphocyte proliferation ratio (LPR) were analysed at 42 d of age. Supplementing Bet to diets containing sub-optimal concentrations of Met (15 g/kg) improved weight gain and breast yield at 21 d of age (p<0.01), and feed conversion efficiency at 42 d of age (p<0.05). Feed efficiency at 21 d of age, body weight gain at 42 d of age, slaughter variables except breast yield at 21 d of age and ND antibody titres were not affected (p>0.05) by the interaction. LPR increased (p<0.05) with Bet supplementation at 20 g Met/kg CP equal to those broilers fed 24 g Met without Bet. Bet supplementation enhanced the concentrations of protein, globulin and cholesterol in serum of broilers fed sub-optimal concentrations of Met. Results suggested that Bet supplementation (800 mg/kg diet) enhanced growth (21 d), feed conversion efficiency (42 d), breast yield and lymphocyte proliferation in broilers fed a diet containing 15 g Met/kg CP.

Immune Response of Broiler Chickens Fed Diets Supplemented with Different Level of Chromium Methionine under Heat Stress Conditions

  • The objectives of this study were to investigate the immune responses of broiler chickens fed diets supplemented with different level of chromium methionine (CrMet) in heat stress (HS) condition. Two hundred and eighty eight male broiler chickens (Ross 308) were allocated to four treatment groups (supplementation with 0, 200, 400 or 800 ppb Cr in the form of CrMet) in a completely randomized design. The experiment was conducted at heat stressed condition and all birds were kept under temperature of $33{\pm}2^{\circ}C$. Antibody titers against Newcastle disease virus (NDV) and infectious bronchitis virus (IBV), heterophil to lymphocyte ratios (H/L), and concentration of plasma cortisol (CPC) were measured at 21 and 42 d. At 42 days of age two birds were chosen randomly from each replicate, slaughtered, spleen and bursa of Fabricius were collected, weighed and expressed as a percentage of live body weight. Antibody titers against NDV and IBV at 21 and 42 days of age in broiler fed supplemental CrMet were higher than in broiler chickens fed control diet (p<0.05). CPC level in broiler chickens fed CrMet were significantly (p<0.05) decreased. Increases in lymphocyte counts and consequently a decrease in heterophil to lymphocyte ratios in broiler chickens fed 800 ppb Cr were observed at 21 and 42 d. Supplementation with CrMet had no significant effect on lymphoid organs of broilers. The results suggest that dietary CrMet supplementation at a level of 800 ppb can improve some immune responses of broiler chickens under heat stress conditions.

Interaction between Lactobacillus acidophilus and Kluyveromyces fragilis on the Metabolism of Amino Acids in Soymilk (Lactobacillus acidophilus와 Kluyveromyces fragilis의 혼합배양에 의한 두유의 젖산발효중 아미노산 대사의 상호작용)

  • The interaction between Lactobacillus acidophilus and Kluyveromyces fragilis on the utilization of amino acids in soymilk was investigated. K. fragilis assimilated relatively well various amino acids such as Met., ILe., Phe., Leu., Thr., Lys., Val., Arg., Tyr., Ser., Asp., Ala. and Glu. that existed only in trace amounts in soymilk. K. fragilis did not utilized Gly., while hi accumulated His. L. acidophilus hydrolyzed soyprotein to liberate various amino acids. Among various amino acids, it utilized Met., ILe., Thr., Tyr., Ser., Val. and His. as growth factors and accumulated Leu., Phe., Lys., Arg., Glu., Asp. and Ala. among the essential amino acids required by K. fragilis and Gly. These results implied that K. fragilis grew on amino acids that existed only in trace amounts in soymilk, but it's growth was stimulated by amino acids such as Leu., Phe., Lys., Arg., Glu., Asp. and Ala. ac-cumulated by L. acidophilus.

Development of cooking method for senior-friendly food using fruits suitable for older adults with masticatory dysfunction

  • BACKGROUND/OBJECTIVES: The purpose of this study was to establish a fruit-cooking method suitable for older adults with masticatory dysfunction. MATERIALS/METHODS: Five types of fruits were selected to make fruit jelly and puree: apple, sweet persimmon, mandarin, Korean melon, and watermelon. Recipes were selected based on the Korean Industrial Standard (KS) for senior-friendly foods (KS H 4897), which classifies foods into 3 levels (L1-L3) based on their hardness and viscosity. RESULTS: In South Korea, senior-friendly foods are classified into 3 stages based on their hardness. Stage 1 is for foods that are able to eat with teeth (hardness greater than 50,000 N and less than 500,000 N), Stage 2 is for foods that are able to eat with gums (hardness greater than 20,000 N and less than 50,000 N), and Stage 3 is for foods that are able to eat with the tongue (hardness less than 20,000 N). As a result of measuring the hardness by varying the shape of the fruit, it was found that nearly all fruits could be eaten fresh by chewing with the teeth (L1) but did not meet the KS for mastication using the gums (L2) or tongue (L3), so the cooking method was selected as fruit jelly and fruit puree. Only sweet persimmon, which had a hardness of 61,624-496,393 N, was not suitable for consumption in fresh fruit, unprocessed form. Based on their hardness measurements, fruit jellies (27,869 to 36,343 N) and fruit purees (315 to 1,156 N) met the L2 and L3 requirements, respectively. The viscosity results of all fruit purees met the L3 requirement. CONCLUSION: These results offer a simple cooking method to prepare texture-modified fruits suitable for safe consumption by older adults living with masticatory difficulties in general households and nursing facilities.

Purification and characterization of the extracellular alginate lyase from Streptomyces sp. MET 0515 (Streptomces sp. MET 0515의 균체외 Alginate lyase의 정제 및 특성)

  • We isolated a new extracellular alginate lyase-producing microorganism, which displayed alginate-depolymerizing activity in plate assays, from coastal soils in Wando, Jeollanam-do, Korea. This alginate-depolymerizing bacterium belonged to the genus Streptomyces and it was named Streptomyces sp. MET 0515. An extracellular alginate lyase(ALY1) secreted by Streptomyces sp. MET 0515, was purified to homogeneity by a combination of acetone precipitation, anion-exchange chromatography (Q-Sepharose and DEAE-Sepharose) and Sephacryl S-200 HR gel filtration chromatography. Its molecular mass was 26 kDa as determined by SDS-PACE analysis. The enzyme had an optimal temperature of $70^{\circ}C$ for its activity, and was most active at pH 7.5. The thermal and pH stability were $0-50^{\circ}C$, and pH 6.0-9.0, respectively. The enzyme activity was stimulated by 1mM $Mn^{2+}$, and inhibited by 1mM $Fe^{3+}$, 1mM EDTA and 1mM $Zn^{2+}$. Preliminary analysis of substrate specificity showed that this alginate lyase had activity on both poly-alpha 1,4-L-guluronate and poly-beta 1,4-D-mannuronate in the alginate molecule.

Investigation of ITU Radio Regulations Regarding KPS Service Band

  • In order to reserve frequency bands for stable operation of the Korean Positioning System (KPS), it must be demonstrated that the impact of interference caused by KPS on other services operating in the same bands is tolerable. The International Telecommunication Union (ITU) provides the radio regulations and a compatibility assessment methodology for the coexistence of different services in limited frequency resources. The purpose of this paper is to investigate the radio regulations regarding the compatibility issues between the KPS and other services sharing the same frequency bands. The results of the investigation show that the RNSS system can be operated in the L5-band under the condition that the Power Flux Density (PFD) and Aggregated Equivalent PFD (AEPFD) thresholds specified in the radio regulations are met. In addition, the ITU recommends that the interference-to-noise ratio (INR) protection criteria be met to prevent degradation of the performance of radar systems operating in the L2/L6-band. In the case of the S-band, coordination of allocations for RDSS space stations with respect to terrestrial services is not required if the PFD does not exceed regulated thresholds regulated by ITU.

Effects on Testosterone of Splague-Dawley by Methoxychlor (Methoxychlor 투여한 흰쥐에 있어서의 Testosterone의 변화)

  • In order to establish the experimental clinical model for evaluating the influences on the sexual hormones of SD-rat administrated with Methoxychlor(MET), it was dissolved in acetone and olive oil (1:19), which was administrated orally at doses of 200㎎/㎏ body weight/day for 7 days. The Testosterone value was measured in sera by Gamma Count with 1251 isotype, also weights of the body, testis and liver were measured for 15 weeks. Testosterone in the serum of intacted SD-rats was 0.51±0.43ng/㎖(n=5), but it was 0.69±0.18(n=2)ng/㎖l on 1 week. 0.28±0.05ng/㎖(n=2) on 3 week, 1.52±0.95ng/㎖(n=3) on 5 week, and 0.54±1.95 ng/㎖(n=3) 15 week post-dosing MET, respectively. Histologically, the numbers of spermatozoa and Leydig cells were reduced, which might influence to reduce sexual hormone, and AST and ALT was not increased in the serum due to administrating with MET, blood profile was not changed according to the administration of MET excepting MCH(Mean Corpuscular Hemoglobin). This study suggest that change of male sexual hormone by MET be a experimental model for evaluating some drugs associate with sex hormone.

Changes in Amino Acids and Fatty Acids Composition during Germination of Rapeseed (유채(油菜) 발아중(發芽中) 아미노산과 지방산 조성(組成)의 변화(變化))

  • Rapeseeds (Brassica napus L.)were germinated at $25^{\circ}C$ for 60 hours under dark condition in order to investigate the compositional changes in amino acids and fatty acids. The weight loss was most significant after 30 hours of germination and root growth was observed from that time. Analysis showed that Glu, Lys and Asp were the major amino acids and Ile was the limiting one. Germination caused an increase in Lys, Thr, Val and Leu while Met and Phe decreased among essential amino acids, resulting Met to be the limiting amino acid. The major fatty acids were oleic (49.3%), linoleic (22.0%) and eicosenoic (10.5%) acids. The decrease in oleic and stearic acids was measured while linoleic, linolenic, eicosenoic and erucic acids showed initial decrease followed by gradual increase.

  • The purpose of this study was to determine the homocysteine (Hey), methionine (Met) and cysteine (Cys) using solid phase micro-extraction (SPME)/gas chromatography (GC)-mass spectrometry (MS) in human plasma and to correlate between the plasma concentration of homocysteine with coronary artery disease. The homocysteine, methionine and cysteine in blood can be used as biomarkers for the risk assessment of vascular disease. The plasma homocysteine level for the coronary artery disease patients was higher than general patients. The concentration ranges of the Hcy, Met and Cys for coronary artery disease patients were $18.47-33.38{\mu}mol/L$, $30.16-55.72{\mu}mol/L$ and $183.16-387.32{\mu}mol/L$, respectively. This method showed good sensitivity and convenience.
