The Role of Nitric Oxidei in Non-Adrenergic Non-Cholinergic Relaxation in the Guinea-Pig Gstric Fundus

  • The role of nitric oxide (NO) in non-adrenegic non-cholinergic (NANC) neurotransmission was studied on circular muscle strips of the dorsal part of the fuinea-pig gastric fundus. In the presence of atropine and guanethidine, a low frequency-dependent relaxsations which were not affected by adrenergic and cholinergic blockage but abolished by tetrodotoxin. $N^G$-nitro-L-arginine (L-NNA), a stereospecific inhibitor of NO-biosynthesis, inhibited the relaxations induced by electrical stiumulations but not the relaxations to exogenous nitric oxide. The effect of L-NNA was prevented by L-arginine, the precursor of the NO biosynthesis but not by its enantiomer, D-arginine. Exgenous administration of No caused concentration -dependent relaxations which showed a similarity to those obtained with electrical simultaion. Hemoglobin, a NOscavenger, abolished the NO-induced relaxations and also markedly reduced those induced by electrical simultaion. The inhibitory effect os hemoglobin was similar to that of L-NNA. Application of ATP caused weak relaxations compared with those to electrical stimultaion, which were unaffected by L-NNA. Exogenously applied vasoactive intestinal polypeptide (VIP) induced concentration-dependent relaxation which was not affected by L-NNA. These results suggest that NO is produced and released mainly as a neurotransmitter from enteric neurons during NANC relaxation induced by low frequencies and short trains of electrical simulation and has a main role in NANC neurotransmission at relaxation induced by these electrical simultaions in the guinea-pig gastric fundus.

  • 최근 염분의 섭취와 관련하여 건강에 대한 우려가 늘어남에 따라, 염미대체제에 대한 소비자들의 관심이 급증하고 있으며, 세계적으로 경쟁력 있는 천연 저염제품의 개발이 필요한 실정이다. 최근 연구를 통해 식물성 및 동물성 원료 배합을 통해 가수분해물을 조제하여 최적 화합물을 얻는 것에 성공하였다. 본 연구에서는 염미성 펩타이드 분말내 arginine을 함유한 염미성 펩타이드를 규명하고 정량 하고자 하였다. L-arginine 또는 arginine을 포함한 펩타이드를 표준물질로 하여 $^{13}C$-NMR로 분석한 결과 유사한 위치에 구아니딘기 탄소가 시그널(L-arginine: 156.8 ppm, Arg-Ala: 156.4 ppm, Arg-Ser: 156.4 ppm)이 나타남을 확인하였고, 이를 통해 간편하고 신속하게 arginine 함유 펩타이드 정량분석이 가능할 것으로 기대된다.

남자 대학 태권도 겨루기 선수의 8주간 고강도 동계 훈련 시 L-아르기닌 섭취가 경기수행능력, 젖산, 젖산탈수소효소 및 암모니아에 미치는 영향 (Effect of Intake L-Arginine on Athletic Performance, Lactate, Lactate Dehydrogenase and Ammonia in Male College Taekwondo Kyorugi Players during an 8-Week Intensive Winter Training)

  • 본 연구는 8주간의 고강도 동계 훈련 시 L-아르기닌 섭취가 남자 대학 태권도 겨루기 선수의 경기수행능력, 젖산, 젖산탈수소효소 및 암모니아에 미치는 영향을 구명하기 위해 L-아르기닌 섭취군(n=14), 위약군(n=14)으로 구분하여 실시하였다. L-아르기닌 섭취군은 일일 아침 1 g, 점심 1 g, 저녁 1 g으로 총 3 g 섭취하였고 위약군은 말토덱스트린을 동일한 방법으로 섭취하였다. 8주간의 동계 훈련 프로그램은 70-90%HRR로 실시하였다. 측정된 자료의 L-아르기닌 섭취군과 위약군 간의 그룹 및 시기 간 상호작용 효과는 two-way repeated measures ANOVA, 그룹 내 시기 간 차이는 paired t-test를 사용하였으며, 그룹 간 차이는 independent t-test를 사용하여 분석하였다. 그 결과 TAAA (Taekwondo-specific aerobic anaerobic agility) test를 통한 경기수행능력 중 평균 발차기 수에서 그룹×시기 간 상호작용 효과가 나타났으며 그룹 간 주효과가 나타났다(p<.05). 또한, 발차기 피로지수에서 그룹×시기 간 상호작용 효과가 나타났다(p<.05). 한편, 젖산에서는 시기 간 주효과가 나타났으며(p<.05) 젖산탈수소효소에서 상호작용 효과 및 시기 간 주효과 나타났다(p<.05). 암모니아의 경우 그룹×시기 간 상호작용 효과가 나타났다(p<.05). 이러한 결과는 남자 대학 태권도 겨루기 선수의 고강도 훈련 후 피로에 쉽게 노출되는 선수들에게 L-아르기닌 섭취로 인해 체내 피로 유발 물질들을 신속하게 제거하는 데 긍정적인 역할을 할 수 있다고 사료된다. 따라서 고강도 엘리트 태권도 겨루기 운동선수의 경기수행능력 향상과 피로 회복 방법으로 L-아르기닌 섭취를 권장한다.

  • Arginine methylation is a common post-translation protein modification in eukaryotic cells. Protein-arginine N-methyltransferase transfer methyl groups from S-adenosyl-L-methionine to the guanidino group of arginine residues. However. The significant of this modification has been questionable. because it occurs rarely and is present at very low abundance. Recently, the discovery of two protein arginine methyltransferase, PRMT1 and CARM1, as cofactors required for responses to muclear Hormone receptors provided an indicationthat arginine methylationhave an important role in transcriptional regulation. 오수유가 N-nitro-L-arginine methyl ester로 유발한 고혈압흰쥐의 심혈관계에 미치는 영향 (Studies on the Effects of Evodiae Fructus on the Cardiovascular System in N-nitro-L-arginine Methyl Ester-induced Hypertensive Wistar Rats)

  • The present study examined the effect of a methanol extract of Evodiae Fructus on the cardiovascular function in N-nitro-L-arginine methyl ester (NAME)-induced hypertensive Wistar rats after treatment over 6 weeks. In rats treated with NAME, blood pressure, weight of heart, aorta media thickness and media/lumen ratio significantly (p<0.05) increased, whereas coronary flow and heart rate of isolated heart significantly (p<0.05) decreased compared with control group at 6 weeks. In rats treated with NAME and Evodiae Fructus, blood pressure, aorta media/lumen ratio significantly(p<0.05) decreased compared with NAME treated group at 6 weeks. These results suggest that Evodiae Fructus is applicable to the treatment of hypertension and vascular hypertrophy.

맹꽁이 간의 산소소비량에 미치는 수종 아미노산의 영향 (Effect of Amino Acids on Oxygen Consumption of the Mang-Kong Frog (Gallula tornieri VOGT) Liver)

  • The present experiment was conducted to investigate the rate of oxygen consumption in relation to the several amino acids in the Mang-Kong frog ,Gallula tornieri VOGT. 1. When the liver tissue was suspended in the Ringer's solution (0.85% NaCl), the amount of oxygen consumption per hour was 0.043$\mu$l/mg , whereas when it was suspended in M/15, pH 5.8 phosphate buffer the oxygen consumption reached 0.93$\mu$l/mg. 2. When 0.5ml of 1% alanine and 0.5ml of 0.5% arginine were used as respiratory material the amount of oxygen consumption increased remarkably, while on the other hand glycine showed little influence on the amount of oxygen consumption. 3. When 0.5ml of 0.1% , 0.5% alanine-arginine mixture, 0.5ml of 0.1% , 0.5% alanine-glycine mixture and 0.5ml of 1 % arginine-glycine were added the oxygen consumption in the liver tissue increased somewhat in comparison to the case in which single amino acid was used. 4. Glycine appeared to have influence on the respiration of the liver tissue but the amount of oxygen consumption increased as the concentration of arginine-glycine mixture increased, whereas the rate of oxygen consumption increased remarkably as alaninie-glycine mixture decreased. It was evidenced that in the combination of alanine-arginine the rate of oxygen consumption increased as the concentration of the mixture decreased and vice versa.

일부 아미노산과 식품 추출물의 에탄올 간독성에 대한 보호효과 (Protective Effect of Selected Amino Acids and Food Extracts on Ethanol Toxicity Decrement in Rat Liver)

  • 본 실험에서는 aspartate, arginine, glutamate가 에탄올로 처리한 간세포에 대한 lipid peroxidation의 변화를 비교분석하고 예로부터 숙취제거 식품으로 잘 알려진 콩나물, 북어 및 영지버섯 추출액의 in vitro 실험을 통하여 에탄올로 처리된 간세포에 대한 lipid peroxidation 변화를 보았다. 또한 에탄올처리에 대한 숙취제거 식품의 처리순서에 따른 lipid peroxidation의 변화를 보았다. S9분획(간세포)에 에탄올(0.56 M)을 $50\;{\mu}L$씩 처리한 후 아미노산용액과 식품추출물을 6.25, 12.5, 25, 50 mg/mL 농도로 처리하였다. 이 실험의 결과는 다음과 같다. Aspartate, arginine, glutamate중 arginine이 가장 큰 간 보호 효과를 나타내었다. 콩나물, 북어, 영지의 단백질 함량은 콩나물이 $8.916\;{\mu}g/mL$으로 가장 많았다. 콩나물, 북어, 영지중에서 콩나물 추출물이 가장 큰 간 보호 효과를 나타내었다. 각각의 식품추출물을 에탄올 다음에 처리하였을 경우 전처리보다 간 보호 효과가 더욱 증가하는 경향을 나타내었다.

창포(Acorus calamus L.)뿌리 건조분말의 식품학적 성분 (I) 일반성분, 총당, 환원당, 유리당 및 아미노산 (Food Nutritional Composition in Dried Powder of Root of Acorus calamus L. ( I ) Pyoximate Compositions, Sugars, Free Sugers and Amino Acids)

  • 창포 뿌리를 주근(main mot)과 측근(lateral root)으로 분리하여 열풍건조 분쇄, 분말화한 후 일반성분, 당 및 아미노산등 화학성분을 분석하여 식품학적 특성을 검토하였다. 주근과 측근 건조분말에는 각각 조단백질 12.76%, 9.84%와 조지방 7.43%, 3.42%로 높게 함유하고 있어 에너지공급원과 식물성 고단백질 원료로서의 우수한 영양학적 가치를 가지고 있음을 확인하였다. 유리당으로는 sucrose, stachyose, raffinose, glucose 및 fructose 등으로, 특히 sucrose가 주근과 측근에 각 10.85%와 12.27%로 높게 함유하고 있었다. 창포뿌리 주근의 건조분말은 총아미노산 함량은 426.81 mg%이었고, 특히 arginine 78.91 mg%와 glutamic acid 69.73 mg%로 높은 함량 수준을 보였다. 측근 건조분말의 총아미노산 함량은 470.13 mg%로 주근 보다 다소 높은 함량을 나타내었고, 특히l arginine이 116.91 mg%로 주근의 arginine 함량보다 매우 높은 함량 수준을 보였다. 주된 유리아미노산으로는 asparagine, arginine, glutamic acid, phenylalanine 등의 순으로 높게 함유되어 있고, 특히, 주근 건조분말에는 asparagine이 128.53mg%, 측근 건조분말에는 arginine이 86.10 mg%로 높은 함량 수준을 나타내었다. 필수아미노산 함량은 유리아미노산의 경우는 phenylalanine, threonine, valine, leucine 등이 높게 함유되어 있었으며, 특히, 주근 건조분말에는 threonine이 15.62 mg%, 측근 건조분말에는 phenylalanine 12.67 mg%로 타 필수 아미노산에 비해 매우 높은 함량 수준을 나타내었다.

Interaction between Renin-Angiotensin and Endothelium-Derived Nitric Oxide Systems in Two-Kidney, One Clip Hypertensive Rats

  • The present study was aimed to investigate the role of endothelium-derived nitric oxide (NO) in the control of renin release and to examine if NO is implicated in the development of two-kidney, one clip (2K1C) hypertension. Male Sprague-Dawley rats $(150{\sim}200\; g)$ were constricted at the left renal artery. They were then supplemented with $N^{G}-nitro-L-arginine\;methyl\;ester\;(L-NAME,\; 5mg/100\;mL)$ or with L-arginine hydrochloride (400 mg/100 mL) in the drinking water. The control group was supplied with normal tap water. The sham-clipped rats were operated as in 2K1C rats except for that no clip was made. The kidneys were taken to examine in vitro release of renin at days 7 and 14 following clipping the renal artery. Northern blot analysis was also done to assess the expression of renin gene in the kidney. In sham-clipped rats, L-NAME caused a sustained increase of the blood pressure, whereas L-arginine was without effect. Neither L-NAME nor L-arginine-supplementation significantly affected the development of hypertension in 2K1C rats. Plasma renin concentration (PRC) measured on day 28 did not significantly differ among the L-NAME, L-arginine and control groups either in 2K1C or in sham-clipped rats. Renin contents (RRC) in the clipped kidney were increased, while those in the contralateral kidney were decreased. The release of renin in vitro from cortical slices was also enhanced in the clipped kidney, whereas it was attenuated in the contralateral. Comparing the RRC and in vitro release, the latter was more rapidly decreased than the former in the contralateral kidney. The renin mRNA levels in the contralateral kidney were almost at their nadir at days 7 and 14 in 2K1C rats. It is suggested that NO does not affect the development of 2K1C hypertension in which the renin-angiotensin system has been activated. The data also confirm that RRC and renin gene expression are increased in the clipped kidney and suppressed in the contralateral kidney in 2K1C rats.

Production of L-Ornithine by Citrulline Auxotrophic Mutants of Glutamate-Producing Bacteria

  • For the purpose of producing L-ornithine by microbial fermentation, mutant strains were developed from glutamate-producing bacteria by mutagenesis using N-methyl-N'-nitro-N-nitrosoguanidine (NTG) and UV irradiation. Brevibacterium ketoglutamicum BK1046, a L-citrulline auxotroph which is also resistant to arginine hydroxamate (Arghx), was isolated and selected as the best producer of L-ornithine. This strain was capable of producing L-ornithine at a concentration of 24 g/l after 69 hours of cultivation in the 21 jar fermentor. The optimum supplementary level of L-arginine, a substitute for L-citrulline, was found to be about 0.2 g/l in the shake-flask fermentation.
