Application of L-Arginine as a Substitute for Inorganic Polyphosphate in Pork Sausage Production (소시지 제조시 인산염 대체 소재로 L-Arginine의 활용)

  • $\small{L}$-Arginine was applied to processed pork sausages and evaluated as a substitute for commonly used inorganic polyphosphate. Processed pork sausage was prepared with an addition of 0.25, 0.5, and 1.0% (w/w) $\small{L}$-arginine. Quality characteristics of the processed pork sausages were than evaluated in terms of pH, moisture content, emulsion stability and textural properties. The pH of sausage increased with increasing $\small{L}$-arginine concentration. Moisture content of 0.25% $\small{L}$-arginine sausage, emulsion stability of 0.5% $\small{L}$-arginine sausage, and textural properties of 1.0% $\small{L}$-arginine sausage were similar to those of 0.5% polyphosphate sausage, respectively. In terms of sensory evaluation, taste and overall acceptability of 0.5% $\small{L}$-arginine sausage were slightly higher than those of 0.5% polyphosphate sausage and textural properties were reversed. According to sensory score and quality characteristics, $\small{L}$-arginine had a potential as a substitute for inorganic polyphosphate and the optimum concentration of $\small{L}$-arginine was around 0.5%.

Effect of Intake L-Arginine on Athletic Performance, Lactate, Lactate Dehydrogenase and Ammonia in Male College Taekwondo Kyorugi Players during an 8-Week Intensive Winter Training (남자 대학 태권도 겨루기 선수의 8주간 고강도 동계 훈련 시 L-아르기닌 섭취가 경기수행능력, 젖산, 젖산탈수소효소 및 암모니아에 미치는 영향)

  • This study was conducted by dividing L-arginine intake group (n=14) and placebo group (n=14) to investigate the effect of L-arginine intake on athletic performance, lactate, lactate dehydrogenase, and ammonia of men's college Taekwondo Kyorugi players during 8-week of intensive winter training. The L-arginine intake group consumed a total of 3 g, including 1 g for breakfast, 1 g for lunch, and 1 g for dinner, and the placebo group consumed maltodextrin in the same way. The 8-week winter training program was conducted at 70-90%HRR (heart rate reserve). Two-way repeated measures ANOVA was used for the interaction between the L-arginine intake group and the placebo group of the measured data, paired t-test was used for the difference between the periods within the group, and the difference between groups was analyzed using independent t-test. As a result, there was an interaction between groups in the average number of kicks among the performance ability through the TAAA (Taekwondo-specific aerobic anaerobic agility) test , and the main effect between groups was shown (p<.05). In addition, there was an interaction between groups and time in the kick fatigue index (p<.05). In lactate, the time-interaction were shown (p<.05) and interaction effects and inter-time main effects were observed in lactate dehydrogenase (LDH)(p<.05). In the case of ammonia, the interaction effect between the group and time was shown (p<.05). These results show that L-arginine intake can play a positive role in quickly synthesizing nitric oxide in blood vessels and expanding blood vessels to quickly remove fatigue-causing substances in the body for athletes who are easily exposed to fatigue after high-intensity training for male college Taekwondo Kyorugi players. Therefore, it is recommended to take L-arginine as a way to improve the performance of high-intensity elite Taekwondo Kyorugi players and recover from fatigue.

Purification and Characterization of Carboxypeptidase B from Wild Salmon (Salmo Salar) Eggs (연어알중의 Carboxypeptidase B 분리정제 및 그 특성에 관한 연구)

  • Carboxypeptidase B from Salmo Salar eggs was purified by CM-cellulose, 0.5 ammonium sulfate saturation, DEAE-cellulose, and Sephadex G-75 gel filtration and its enzymatic properties were investigated. Optimum temperature was 55$^{\circ}C$, pH optima were 4.0 and 7.0 at 37$^{\circ}C$, and the enzyme was stable at pH 2.0∼3.0 and 5.5∼7.0 for 1.5h. This enzyme showed substrate specificity hydrolyzing the peptide bond of glycyl-L-arginine. Its K$_m$ values was 0.21mM, and the enzyme activity was stimulated by Cu$^{2+}$ and Fe$^{3+}$, while inhibited by Zn$^{2+}$. The lysine was found to be competitive inhibitor and its K$_i$ value was determined to be 4.3mM. Molecular weight of this enzyme was determined to be 36,400 daltons by SDS-PAGE and the enzyme was monomeric protein composed of 19 kinds of amino acid residues.

Effects of L-Arginine Supplementation and Regular Exercise in D-Galactose Induced Aging Rat Aorta: Study on Inflammatory Factors, Vasodilation Regulatory Factors (노화유도 쥐의 대동맥에서 L-arginine 투여와 규칙적인 운동의 효과: 염증인자와 혈관이완조절 인자의 변화)

  • The purpose of this study was to identify the effects of an L-arginine supplementation and regular exercise training on NF-${\kappa}B$, TNF-${\alpha}$, iNOS, Cav-1, eNOS and Ang II in the aortas of D-galactose (D-gal) induced aging rats. The male Strague-Dawley rats were treated with a D-galactose aging inducing agent; the D-gal injection (50 mg/kg) was given intraperitoneally for 12 wk. Experimental groups were divided into five groups: (1) Young control group (Y-Con, n=8), (2) Aging control group (A-Con, n=8), (3) Aging exercise group (A-Ex, n=8), (4) Aging exercise group with L-arginine supplementation group (A-Ex+A, n=8), and (5) Aging with L-arginine supplementation group (A-A, n=8). The exercise consisted of running on a treadmill for 60 min/day at 20 m/min for 6 day/wk, at 0% gradient for 12 wk. The L-arginine supplementation was given orally at a dose of 150 mg/kg/day for 12 wk. The findings of this study were as follows: 1. NF-${\kappa}B$, TNF-${\alpha}$, iNOS, Cav-1 and Ang II proteins in the aortas of D-gal induced rats were significantly increased, however, L-arginine supplementation and regular exercise resulted in a significant inhibition in the expression of NF-${\kappa}B$, TNF-${\alpha}$, iNOS, Cav-1 and Ang II proteins. 2. eNOS protein in the aortas of D-gal induced rats was significantly decreased, however, L-arginine supplementation and regular exercise resulted in a significant increase in the expression of eNOS proteins. In conclusion, the findings of the present study reveal that L-arginine supplementation alone or regular exercise alone or in combination with L-arginine supplementation for 12 wk increases anti-inflammatory effects by decreasing NF-${\kappa}B$, TNF-${\alpha}$, and iNOS protein expressions within the aortic tissue. In addition, L-arginine supplementation alone or regular exercise alone or in combination with L-arginine supplementation may prevent endothelial function by up-regulation of eNOS protein in the aortas of D-gal induced aging rats.

Effect of L-arginine on the Stability of Omeprazole (L-아르기닌이 오메프라졸의 안정성에 미치는 효과)

  • To investigate the effect of L-arginine as stabilizing agent for omeprazole, the degradation rate constant of omeprazole in aqueous solution was determined at 30, 40 and $50^{\circ}C$ with various ratios of L-arginine to omeprazole. The pH of omeprazole solutions was also determined. As the amount of L-arginine increased, the pH of omeprazole solution also increased, and the solution appeared to be more stable. The omeprazole in aqueous solution could be stabilized by more than 15:1 molar ratio of L-arginine to omeprazole. The stability of omeprazole in commercial products using L-arginine or sodium phosphate dibasic as stabilizing agent was investigated. Among the commercial products, the omeprazole product prepared with L-arginine (molar ratio of L-arginine to omeprazole, 20:1) was most stable.

Renal Action of $N^G$-Nitro-L-arginine, Nitric Oxide Synthase Inhibitor, in Dog and Rabbit (니트릭옥사이드의 합성 억제제인 $N^G$-니트로-L-아르기닌의 신장작용)

  • This study was performed in order to investigate the effect of renal function of NG-nitro-L-arginine (L-NOARG), inhibitor of nitric oxide (NO) synthase, in dog and ra bbit. L-NOARG, when given intravenously in dogs, exhibited the decrease in urine flow (vol), renal plasma flow (RPF), osmolar clearance ($C_{osm}$) and amounts of sodium and potassium excreted in urine($E_{Na},\;E_K$). These renal functions of L-NOARG showed the same aspect in rabbit, too. L-NOARG, when administered into a renal artery, showed the same pattern as was obtained when given intravenously in both experimental and control kidney in dog. L-NOARG administered into the carotid artery showed the decrease in Vol, RPF, $E_{Na}$, in a low doses that did not show any effect when given intravenously. Above results suggest that L-NOARG produces antidiuretic action in dog and rabbit, and these antidiuretic actions may be mediated by central action.

In vitro Effects of Mono- and Dimethylarginines on the Contractility of Rat Thoracic Aorta (쥐 흉부대동맥 수축에 미치는 모노- 및 디메칠아르기닌의 영향)

  • In order to study the functions of vascular endothelial nitric oxide(NO) generating system, we examined the effects of monomethylarginine(MMA) and dimethylarginine(DMA)(asym., sym.), arginine analogues, on modulation of vascular tone. Also, the concentrations of endogenous arginie and MMA were measured by HPLC in rat aortic tissues. The results were as follows. 1. The maximum relaxation induced by Ach (1.5$\times$10$^{-6}$M) was 80% of the contractility of rings of rat aorta induced by phenylephrine and L-Arg causes endothelium-dependent relaxation of the aorta precontracted with phenylephrine and these relaxation were concentration-dependent. 2. Endothelium-dependent contractility of rings of rat aorta induced by MMA (100 $\mu{M}$), DMA (asym., 100 $\mu{M}$) and DMA (sym., 100 $\mu{M}$) were 25.5%, 27.5% and 16.5% of that induced by phenylephrine respectively. 3. The relaxation of rat aortic ring induced by L-Arg was inhibited by MMA, DMA(asym.) and DMA(sym.). The degrees of inhibition induced by MMA, DMA(asym.) and DMA(sym.) were 45.7%, 37.1% and 18.3%, respectively. 4. The endogenous arginine and monomethylarginine contents in rat aorta were 83 pmoles/mg wet tissue, and 34.9 pmoles/mg wet tissue. After stimulation with Ach, the concentrations of L-Arg and MMA were significantly decreased. These results suggest that there are the strong relationships between the endogenous L-Arg and methylated arginines and NO-generating system.

  • Essential amino acid composition of seventy eight medicinal herbs marketed in Korea was analyzed of the basis of their dried weight. From the results analyzed, it was shown that Alisma orisntale (S.) J. (1,911.5 mg%), Foeniculum vulgare M. (1,090.8 mg%), Artemisia princeps P. (1,073.6 mg%) and Typha latifolia L.(956.1 mg%) contain in threonine. Alisma orisntale (S.) J. (2,523.3 mg%), Nelumo nucifera G. (1,984.7 mg%) and Foeniculum vulgare M. (1,553.8 mg%) contain in lysine. Nelumo nucifera G. (738.3 mg%), Alisma orisntale (S.) J. (635.2 mg%) and Foeniculum vulgare M. (507.7 mg%) contain in methionine. Nelumo nucifera G. (2,496.9 mg%), Alisma orisntale (S.) J. (2,295.6 mg%) and Coix lachrymajobi L. var. M. S. (2,222.5 mg%) contain in leucine. Nelumo nucifera G. (1,624.6 mg%), Alisma orisntale (S.) J. (1,432.7 mg%) and Xantnium strumarium L. (1.371.7 mg%) contain in isoleucine. Alisma orisntale (S.) J (1,791.0 mg71), Nelumo nucifera G. (1,570.6 mg%) and Xantnium strumarium L. (1,498.7 mg%) contain in phenylalanine.

Effect of Poly-L-arginine on the Mucin Release from Airway Goblet cells of Hamster and on the Mucosubstances of Airway Goblet cells of Rat (폴리-엘-아르기닌이 햄스터 기도 배상세포에서의 뮤신 유리 및 흰쥐 기도 배상세포내 함유된 점액에 미치는 영향)

  • In this study, we tried to investigate whether poly-L-arginine (PLA) (MW 10,800) significantly affect mucin release from cultured hamster airway goblet cells and the mucosubstances of hypersecretory air-way goblet cells of rats. Confluent primary hamster tracheal surface epithelial (HTSE) cells were metabolically radiolabeled with $^3$H-glucosamine for 24 hr and chased for 30 min in the presence of varying concentrations of PLA to assess the effects on $^3$H-mucin release. Possible cytotoxicities of PLA were assessed by measuring both Lactate Dehydrogenate (LDH) release and by checking the possible changes on the morphology of HTSE cells during treatment. For in vivo experiment, hyperplasia of rat airway goblet cells and increase in intraepithelial mucosubstances were induced by exposing rats to SO$_2$ for 3 weeks and varying concentrations of PLA were administered inhalationally to assess the effects on the mucosubstances of airway goblet cells of rats. The results were as follows : (1) PLA significantly inhibited mucin release from cultured HTSE cells in a dose-dependent manner; (2) there was no significant release of LDH and no significant change on the morphology of cultured HTSE cells during treatment; (3) PLA also affected the intraepithelial mucosubstances of hypersecretory rats and restored them to the levels of control animals. We conclude that PLA inhibit mucin release from airway goblet cells without significant cytotoxicity and possibly normalize the hypersecretion of airway mucosubstances in vivo. This finding suggests that PLA might function as an airway mucoregulative agent.

  • Bovine testicular hyaluronidase PH-20 was cloned into pPIC9 vector and expressed in Pichia pastoris. Recombinant PH-20 was 75 kDa MW and 7460 units/L activity. Extracellular hyaluronidase activity was two times higher than that of intracellular activity. Non-buffered medium and $30^{\circ}C$ cultivation was favorable for PH-20 production. 1M sorbitol as an osmotic pressure and 0.3% methanol inducer increased cell growth and enzyme activity. 0.4 M arginine augmentation decreased the proteolytic degradation of recombinant hyaluronidase.
