Inhibitory Effects of Triterpenoids on Interleukin-8/CINC-1 Induction in LPS-Stimulated Rat Peritoneal Macrophages

  • The CINC-1 is a member of rat interleukin-8 with chemotactic and activating properties to neutrophils. The CINC-1 induction in LPS-stimulated rat peritoneal macrophages was analyzed using a sensitive enzyme-linked immunosorbent assay. The peritoneal macrophages contained about 3 ng/ml as a basal level, and induced to maximal 18 ng/ml of CINC-1 by stimulation with 5 ${\mu}g/ml$ of LPS. Antiinflammatory steroids of dexamethasone and triamcinolon significantly suppressed the CINC-1 induction, where as aspirin and idomethacin did not show suppression. Inhibitory effects on the CINC-1 induction by natural triterpenoids having steroidal structures were analyzed. Among the 39 kinds of triterpenoids isolated from herbal medicines, acacigenin B and nigaichigoside F1 exhibited the highest suppression on the CINC-1 induction.

Inhibitory Effects of Herbal Extracts on Cyclooxygenase Activity of Prostaglandin $H_2$ Synthase from Sheep Seminal Vesicle

  • Prostaglandin $H_2$ synthase is the pharmacological target site of nonsteroidal antiinflammatory drugs. Inhibitory effects on cyclooxygenase activity of the synthase by extracts prepared from herbal medicines and wild plants in Korea have been estimated. Sixteen species out of 612 species exhibited more than 50% of inhibition on the enzyme activity. The active extracts prepared from Carex humilis, Celastrus orbiculatus, Eugenia caryophyllata, Gleditsia japonica var. koraiensis, Glycyrrhiza grabra, Glycyrrhiza uralensis, Gyrophora exculenta, Lespedeza maximowiczii, Morus alba, Persicaria conspicua, Prunus salicina, pterocarya stenoptera, Rheum undulatum, Vitis amurensis, and Vitis coignetiae have been sequentially washed with methylene chloride, ethyl acetate, n-butanol. Among the solvent fractions of the active herbal extracts, ethyl acetate fraction of Carex humilis exhibited the highest inhibitory effect on the cyclooxygenase activity of prostaglandin $H_2$ synthase.

Pediococcus spp.-fermented chicken meat for dogs

  • An experiment was conducted to evaluate Pediococcus spp.-fermented chicken meat as a snack for dogs. The fermented or non-fermented snacks used in this study were prepared through the following process; meat mixtures containing 52.8% MDCM, 35.2% chicken breast meat (CBM) and 9.7% corn starch were inoculated with or without Pediococcus spp., incubated at 37℃ for 24 h and then sterilized at 121℃ for 20 min. During the 24-h fermentation, the pH of fermented chicken snack dropped rapidly with concomitant increase in number of lactic acid bacteria. The nutritional composition was not altered by fermentation. In vitro pepsin nitrogen digestibility was higher (p < 0.05) in the fermented snack compared with the non-fermented snack. Upon storage at room temperature for 14 days, bacteria grew slowly in fermented vs. non-fermented snack samples. In a palatability trial, dogs preferred non-fermented over fermented snack food. In 12-d-long feeding trial, fecal ammonia content was lowered, but fecal lactic acid content was increased in dogs fed the fermented vs. non-fermented snack food. Our study shows that the fermented MDCM-based snack exhibited good preservability upon storage, and improved in vitro nitrogen digestibility and fecal characteristics in dogs.

  • Since a power supply substation of the magnetic levitation train system includes a inverter which is necessary to change the frequency of power, it has a different characteristic comparing to any other railway system. Nowaday a study for determining rating of facilities constituting a power supply system and modeling that system is undergoing. In this paper, a analysis model for the power supply system of the magnetic levitation train using the 3-level inverter is proposed. And the analysis for output characteristic is performed using the PSCAD/EMTDC program. The control method about the output of 3-level inverter for supplying power to loads in magnetic levitation train system is also described.

The Activity of 5-Lipoxygenase and the Inhibitor of Leukotriene $B_4$ Biosynthesis (5-Lipoxygenase의 활성과 Leukotriene $B_4$ 생합성 억제물질)

  • Polymorphonuclear leukocyte (PMNL) was obtained from peritoneal cavity in rat treated with casein. Effects of divalent cations and drugs on leukotriene $B_4(LTB_4)$ formation from arachidonic acid in the PMNL were determined by HPLC assay. 5-Lipoxygenase, a key enzyme for $LTB_4$ formation from arachidonic acid, exhibited $V_{max}$ at $1\;{\times}\;10^{-5}M$, and $K_m$ at $9.89\;{\times}\;10^{-5}M$ of arachidonic acid. Optimal $Ca^{++}$ concentration for the enzyme activity was $1\;{\times}\;10^{-5}M$ but $Zn^{++}$ did not show any significant effects on the $LTB_4$ formation. Indomethacin and caffeic acid exhibited inhibitory effects on the $LTB_4$ formation at $1\;{\times}\;10^{-5}M$ and $4\;{\times}\;10^{-5}M$, respectively. However, 6-aminohexanoic acid did not show any significant effects on the $LTB_4$ formation.

  • The wireless visible light communication technology has received great attention for high-data rate services in the room and underwater. However, performance of a visible light seawater link is limited by multiple constraints from the current light source and detector technology, and underwater channel conditions. In this paper, performance of the line of sight underwater link was analyzed in terms of signal to noise ratio and bit error rate of the detector. Roles of different parameters such as data rate, transmission distance, and attenuation coefficient, are studied. Through the modeling and simulation of the OOK and L-PPM modulation technologies for undersea environment application, the advantages and limitations are described in detail.

Suppressive Effects of Triterpenoids on CINC-1 Induction in Interleukin-1{\betha}-stimulated rat fibroblast NRK-49F cells

  • CINC-1 is a member of chemokine family with chemotactic and activating properties to neutrophils. CINC-1 induction in IL-$1{\beta}$-stimulated rat fibroblast NRK-49F cells was quantitated by a sensitive ELISA. CINC-1 production was increased up to 135 ng/ml from basal 2-6 ng/ml by stimulation with IL-$1{\beta}$.Steroidal anti-inflammatory drugs including dexamethasone and prednisolone exhibited potent suppressive effects on IL-$1{\beta}$-induced CINC-1 production. Among 39 kinds of natural triterpenoids tested, acacigenin B exhibited the highest suppressive effects with about $10{\mu}M$ to be 50% of inhibition on the CINC-1 induction. The suppressive potency of acacigenin B on IL-$1{\beta}$induced CINC-1 production was about 10-fold less than that of the steroidal anti-inflammatory drugs.

  • The interaction between poly(HEMA) which is material of contact lens containing about 45% water and amino acids (alanine, arginine, methionine, proline, glycine, serine, lysine) have been studied by using the thermal analysis methods through DSC and TGA. The experimental results of DSC and TGA were shown that lysine, arginine and alanine have the most stable interaction between poly(HEMA) and amino acids.

  • Giant cell-rich osteosarcoma is a rare tumor that is hard to distinguish from giant cell tumors of bone. The location and radiographic aspect of these tumors can be identical. The histologic differentiation between a giant cell-rich osteosarcoma and a giant cell tumor can be difficult. Due to the different prognosis and treatment strategies of these tumors it is essential to make the right diagnosis at presentation. This article reports a case of a giant cell-rich osteosarcoma of the proximal tibia that initially was diagnosed histologically as a giant cell tumor by needle biopsy.

  • The urinalysis, which is universally accessible to the general public, has disadvantages of being less objective using sight and purchasing a separate portable urinalysis machine. However, due to the high penetration rate and performance improvement of smartphone created by the development of mobile communication technology, research on urinalysis services using smartphone has been conducted. In this paper, a new urinalysis screening application based on smartphone was developed by supplementing the limitations of the previously studied urinalysis services. The key technology of the application is urinalysis recognition algorithm and urinalysis pad color determination algorithm through image-processing and contour detection. In order to confirm the performance of the developed application, urinalysis strip was photographed and analyzed from various backgrounds and angles.
