Simulation of Indian Summer Monsoon Rainfall and Circulations with Regional Climate Model

  • It is well known that there is an inverse relationship between the strength of Indian summer monsoon Rainfall (ISMR) and extent of Eurasian snow cover/depth in the preceding season. Tibetan snow cover/depth also affects the Asian monsoon rainy season largely. The positive correlation between Tibetan sensible heat flux and southeast Asian rainfall suggest an inverse relationship between Tibetan snow cover and southeast Asian rainfall. Developments in Regional Climate Models suggest that the effect of Tibetan snow on the ISMR can be well studied by Limited Area Models (LAMs). LAMs are used for regional climate studies and operational weather forecast of several hours to 3 days in future. The Eta model developed by the National Center for Environmental Prediction (NCEP), the Fifth-Generation NCAR/Penn State Mesoscale Model (MM5) and Regional Climate Model (RegCM) have been used for weather prediction as well as for the study of present-day climate and variability over different parts of the world. Regional Climate Model (RegCM3) has been widely . used for various mesoscale studies. However, it has not been tested to study the characteristics of circulation features and associated rainfall over India so far. In the present study, Regional Climate Model (RegCM-3) has been integrated from 1 st April to 30th September for the years 1993-1996 and monthly mean monsoon circulation features and rainfall simulated by the model at 55km resolution have been studied for the Indian summer monsoon season. Characteristics of wind at 850hPa and 200hPa, temperature at 500hPa, surface pressure and rainfall simulated by the model have been examined for two convective schemes such as Kuo and Grell with Arakawa-Schubert as the closure scheme, Model simulated monsoon circulation features have been compared with those of NCEP/NCAR reanalyzed fields and the rainfall with those of India Meteorological Department (IMD) observational rainfall datasets, Comparisons of wind and temperature fields show that Grell scheme is closer to the NCEP/NCAR reanalysis, The influence of Tibetan snowdepth in spring season on the summer monsoon circulation features and subsequent rainfall over India have been examined. For such sensitivity experiment, NIMBUS-7 SMMR snowdepth data have been used as a boundary condition in the RegCM3, Model simulation indicates that ISMR is reduced by 30% when 10cm of snow has been introduced over Tibetan region in the month of previous April. The existence of Tibetan snow in RegCM3 also indicates weak lower level monsoon westerlies and upper level easterlies.

Diffusion Tensor-Derived Properties of Benign Oligemia, True "at Risk" Penumbra, and Infarct Core during the First Three Hours of Stroke Onset: A Rat Model

  • Objective: The aim of this study was to investigate diffusion tensor (DT) imaging-derived properties of benign oligemia, true "at risk" penumbra (TP), and the infarct core (IC) during the first 3 hours of stroke onset. Materials and Methods: The study was approved by the local animal care and use committee. DT imaging data were obtained from 14 rats after permanent middle cerebral artery occlusion (pMCAO) using a 7T magnetic resonance scanner (Bruker) in room air. Relative cerebral blood flow and apparent diffusion coefficient (ADC) maps were generated to define oligemia, TP, IC, and normal tissue (NT) every 30 minutes up to 3 hours. Relative fractional anisotropy (rFA), pure anisotropy (rq), diffusion magnitude (rL), ADC (rADC), axial diffusivity (rAD), and radial diffusivity (rRD) values were derived by comparison with the contralateral normal brain. Results: The mean volume of oligemia was $24.7{\pm}14.1mm^3$, that of TP was $81.3{\pm}62.6mm^3$, and that of IC was $123.0{\pm}85.2mm^3$ at 30 minutes after pMCAO. rFA showed an initial paradoxical 10% increase in IC and TP, and declined afterward. The rq, rL, rADC, rAD, and rRD showed an initial discrepant decrease in IC (from -24% to -36%) as compared with TP (from -7% to -13%). Significant differences (p < 0.05) in metrics, except rFA, were found between tissue subtypes in the first 2.5 hours. The rq demonstrated the best overall performance in discriminating TP from IC (accuracy = 92.6%, area under curve = 0.93) and the optimal cutoff value was -33.90%. The metric values for oligemia and NT remained similar at all time points. Conclusion: Benign oligemia is small and remains microstructurally normal under pMCAO. TP and IC show a distinct evolution of DT-derived properties within the first 3 hours of stroke onset, and are thus potentially useful in predicting the fate of ischemic brain.

The Improvement of the Opacity and Printing Strength of Fancy Paper Overlaid Plywood

  • The purpose of this study is to investigate the opacity and printing strength of MG paper overlaid plywood. The printing strength of ink on MG paper can be evaluated effectively by a formula $E^{*2}=[(L^{*})^{2}+(a^{*})^{2}+(b^{*})^{2}]^{1/2}$ that we proposed. Higher E value indicates good printing strength of ink-on-paper. We also assess the real color of translucent printed MG paper with a formula CIE ${\bigtriangleup}E^{*}$ (color difference between a pile of same paper to be opaque and fancy paper laminated board). In addition, the color difference on paper surface caused by the color of wood-based board (bottom) can be evaluated by a formula of Pc. No. Generally, an acceptable appearance quality of fancy boards is ${\bigtriangleup}E^{*}$ <2.0 and small Pc.No. value. The experimental results showed that Japan-made MG papers -J1, J2 and J3 have better printing strength and gloss than that of Taiwan-made paper (T1). The reason for this was that Taiwan-made paper has poor printing strength and low gloss, which might be correlated to the fiber compositions in paper. Higher printing strength can be seen for short fiber containing handsheets when comparing to that of handsheets. Nonetheless, low-freeness sheets gives better printing strength than that of high-freeness sheets. High-opacity MG paper gives good opacifying effect to the fancy paper laminated wood-based boards. Comparing the surface color of 2 kinds of fancy paper laminated boards, paperboard T1 laminated with high-opacity fancy paper showed slight color difference. The same results can be seen for $??g/m^{2}$ handsheets. Higher-opacity Acacia and Eucalyptus bleached sulfate pulps (short fiber) gives higher opacifying effect on the plywood when comparing to Northan pine and Radiata pine sulfate pulps(long fiber). The former ones also showed small color differences when comparing the color differences between the color of fancy paper and laminated paper board. Additionally, the color of bottom plywood can't be shown through for the high-opacify surface paper adhered to. Besides, the PC No of the base paper laminated board is small as well. Apparently, we can add colorants to the binders for the manufscture of various handsheets ($30g/m^{2}$) with various pulp mix ratios to increase the opacity of paperboards to certain extents. When we using yellow and brown binders in paper laminated board, the color difference between Acacia and Eucalyptus handsheets overlaid boards decreasing to 2.0 (acceptable ${\bigtriangleup}E^{*}$ <2.0, hard to discern), but not much improvement for Northern and Radiata pines. Definitely, show-through defects can be discernible for lower opacity papers. In general, admirable printing strength of fancy paper by which glued to plywood can be made with high-opacity paper and colored binders techniques.

Effects of Fiber Characteristics on the Greaseproofing Property of Paper

  • Grease barrier food containers are commonly used for packaging of fast food, cooked food, and food in general. Greaseproofing is also used for certificate paper and label paper etc. Different pulp raw materials, due to their different fiber morphology and chemical compositions, produce papers of varying characteristics. We used optical photomicroscopy and fiber analysis data to evaluate fiber morphology and traits under various beating conditions in order to understand which pulp raw materials produced superior greaseproofing property when a fluorinated greaseproofing agent was added internally. The experiment studied 9 species of pulps, including 2 softwood (northern pine and radiata pine) bleached kraft pulps which were beaten to 550 and 350 mL CSF, respectively; 3 hardwoods (eucalypts, acacia, mixed Indonesian hardwoods) bleached kraft pulps which were beaten to 450 and 250 mL CSF, respectively; and nonwood fibers of reed, bagasse, and abaca. A fluorinated greaseproofing chemical at 0.12% dosage with respect to dry pulp was added to each pulp preparation and formed handsheets. A total of 67 sets of handsheets were prepared, and their basis weights, thickness, bulks, opacities, wet opacities, air resistance, water absorption and degrees of greaseproofing were measured for an overall evaluation of pulp and freeness on greaseproofing papers. The experimental fiber length, coarseness and distribution characteristics and the greaseproofing results suggest that softwood pulps (radiate pine > northern pine) were superior to hardwood pulps (eucalypts > acacia > mixed Indonesian hardwoods). The unbeaten pulps gave papers with high porosities and nearly devoid of greaseproofing property. Greaseproofing is proportional to air resistance. Among the nonwood fibers, bagasse had the best greaseproofing property, followed by reed and abaca was the poorest. With regards to waterproofing property, hardwood pulps (mixed Indonesian hardwoods > acacia > eucalypts) were better than softwood pulps (northern pine > radiate pine). Among the Nonwood fibers, reed had the highest waterproofing property, and it was followed by abaca, while bagasse had the poorest waterproofing characteristic. In summary, bleached kraft northern pine, eucalypts and reed pulps were best suited for making greaseproofing papers, Freeness of the pulps should be kept at $200{\sim}280mL$ CSF for optimal performance.

인삼 추출물에 의한 Cylindrocarpon destructans의 주화성 반응 연구 (Chemotactic Response Study of Cylindrocarpon destructans towards Ginseng Root Exudates)

  • Background: Cylindrocarpon destructans (Zins) Scholten is an important pathogenic fungus that causes ginseng root rot in many ginseng growing areas in China. Although C. destructans have been studied worldwide, research on its chemotaxis towards ginseng (Panax ginseng C. A. Meyer) root exudates in the rhizosphere remains limited. Methods and Results: In this study, we collected ginseng root exudates with three different polarities from three-year-old ginseng roots, and performed chemotaxis and spore germination assays to investigate the ability of these exudates to induce the response in C. destructans. The results showed that, compared with other conditions, when C. destructans cultivated at $20^{\circ}C$ and a pH of 6 exhibited a strong positive chemotactic response toward $2mg/{\ell}$ aqueous phase, $20mg/{\ell}$ butanol phase, and $0.2mg/{\ell}$ petroleum ether from ginseng root exudates, the chemotactic moving indexes were 0.1581, 0.1638 and 0.1441, respectively. In addition, the spore germination rate with optimal chemotactic parameters were 48%, 53%, and 41% in the aqueous phase, butanol phase and petroleum ether groups, respectiviely, which were significantly higher than that in the control group (23%) (p < 0.05). The mycelial growth rate with optimal chemotactic parameters increased with culture time, and the maximum growth rates in the aqueous phase, butanol phase and petroleum ether groups were 0.425, 0.406 and 0.364 respectively, on the 4th day. The optimal chemotactic parameters were $39.73mg/50mg/{\ell}$, $48.93mg/50mg/{\ell}$, and $31.43mg/50mg/{\ell}$, in aqueous phase, butanol phase and petroleum ether respectively, from ginseng root exudates, compared with $5.5mg/50mg/{\ell}$, in the control group. Conclusions: The present study revealed that certain ginseng root exudates containing chemical attractants act as nutritional sources or signals for C. destructans and support its colonization of ginseng roots.

Virtual Screening and Testing of GSK-3 Inhibitors Using Human SH-SY5Y Cells Expressing Tau Folding Reporter and Mouse Hippocampal Primary Culture under Tau Cytotoxicity

  • Glycogen synthase kinase-3β (GSK-3β) is an important serine/threonine kinase that implicates in multiple cellular processes and links with the neurodegenerative diseases including Alzheimer's disease (AD). In this study, structure-based virtual screening was performed to search database for compounds targeting GSK-3β from Enamine's screening collection. Of the top-ranked compounds, 7 primary hits underwent a luminescent kinase assay and a cell assay using human neuroblastoma SH-SY5Y cells expressing Tau repeat domain (TauRD) with pro-aggregant mutation ΔK280. In the kinase assay for these 7 compounds, residual GSK-3β activities ranged from 36.1% to 90.0% were detected at the IC50 of SB-216763. In the cell assay, only compounds VB-030 and VB-037 reduced Tau aggregation in SH-SY5Y cells expressing ΔK280 TauRD-DsRed folding reporter. In SH-SY5Y cells expressing ΔK280 TauRD, neither VB-030 nor VB-037 increased expression of GSK-3α Ser21 or GSK-3β Ser9. Among extracellular signal-regulated kinase (ERK), AKT serine/threonine kinase 1 (AKT), mitogen-activated protein kinase 14 (P38) and mitogenactivated protein kinase 8 (JNK) which modulate Tau phosphorylation, VB-037 attenuated active phosphorylation of P38 Thr180/ Tyr182, whereas VB-030 had no effect on the phosphorylation status of ERK, AKT, P38 or JNK. However, both VB-030 and VB-037 reduced endogenous Tau phosphorylation at Ser202, Thr231, Ser396 and Ser404 in neuronally differentiated SH-SY5Y expressing ΔK280 TauRD. In addition, VB-030 and VB-037 further improved neuronal survival and/or neurite length and branch in mouse hippocampal primary culture under Tau cytotoxicity. Overall, through inhibiting GSK-3β kinase activity and/or p-P38 (Thr180/Tyr182), both compounds may serve as promising candidates to reduce Tau aggregation/cytotoxicity for AD treatment.

Treatment Response Evaluation by Computed Tomography Pulmonary Vasculature Analysis in Patients With Chronic Thromboembolic Pulmonary Hypertension

  • Objective: To quantitatively assess the pulmonary vasculature using non-contrast computed tomography (CT) in patients with chronic thromboembolic pulmonary hypertension (CTEPH) pre- and post-treatment and correlate CT-based parameters with right heart catheterization (RHC) hemodynamic and clinical parameters. Materials and Methods: A total of 30 patients with CTEPH (mean age, 57.9 years; 53% female) who received multimodal treatment, including riociguat for ≥ 16 weeks with or without balloon pulmonary angioplasty and underwent both non-contrast CT for pulmonary vasculature analysis and RHC pre- and post-treatment were included. The radiographic analysis included subpleural perfusion parameters, including blood volume in small vessels with a cross-sectional area ≤ 5 mm2 (BV5) and total blood vessel volume (TBV) in the lungs. The RHC parameters included mean pulmonary artery pressure (mPAP), pulmonary vascular resistance (PVR), and cardiac index (CI). Clinical parameters included the World Health Organization (WHO) functional class and 6-minute walking distance (6MWD). Results: The number, area, and density of the subpleural small vessels increased after treatment by 35.7% (P < 0.001), 13.3% (P = 0.028), and 39.3% (P < 0.001), respectively. The blood volume shifted from larger to smaller vessels, as indicated by an 11.3% increase in the BV5/TBV ratio (P = 0.042). The BV5/TBV ratio was negatively correlated with PVR (r = -0.26; P = 0.035) and positively correlated with CI (r = 0.33; P = 0.009). The percent change across treatment in the BV5/TBV ratio correlated with the percent change in mPAP (r = -0.56; P = 0.001), PVR (r = -0.64; P < 0.001), and CI (r = 0.28; P = 0.049). Furthermore, the BV5/TBV ratio was inversely associated with the WHO functional classes I-IV (P = 0.004) and positively associated with 6MWD (P = 0.013). Conclusion: Non-contrast CT measures could quantitatively assess changes in the pulmonary vasculature in response to treatment and were correlated with hemodynamic and clinical parameters.

Association of Six Susceptibility Loci with Prostate Cancer in Northern Chinese Men

  • Background/Aim: Six prostate cancer (PCa) susceptibility loci were identified in a genome-wide association study (GWAS) in populations of European decent. However, the associations of these 6 single-nucleotide polymorphisms (SNPs) with PCa has remained tobe clarified in men in Northern China. This study aimed to explore the loci associated with PCa risk in a Northern Chinese population. Methods: Blood samples and clinical information of 289 PCa patients and 288 controls from Beijing and Tianjin were collected. All risk SNPs were genotyped using polymerase chain reaction (PCR)-high resolution melting curve technology and gene sequencing. Associations between PCa and clinical covariates (age at diagnosis, prostate-specific antigen [PSA], Gleason score, tumor stage, and level of aggressiveness) and frequencies of alleles and genotypes of these SNPs were analyzed using genetic statistics. Results: Among the candidate SNPs, 11p15 (rs7127900, A) was associated with PCa risk (P = 0.02, odds ratio [OR] = 1.64, 95% confidence interval [CI] = 1.09-2.46). Genotypes showed differences between cases and controls on 11p15 (rs7127900, A), 11q13 (rs7931342, T), and HNF1B (rs4430796, A) (P = 0.03, P = 0.01, and P = 0.04, respectively). The genotype TG on 11q13 (rs7931342, T) was positively associated with an increased Gleason score (P = 0.04, OR = 2.15, 95% CI = 1.02-4.55). Patients carrying TG on 17q24 (rs1859962, G) were negatively associated with an increased body mass index (BMI) (P = 0.03, OR = 0.44, 95% CI = 0.21-0.92) while those with AG on HNF1B (rs4430796, A) were more likely to have PSA increase (P = 0.002). Conclusion: Our study suggests that 11p15 (rs7127900, A) could be a susceptibility locus associated with PCa in Northern Chinese. Genotype TG on 11q13 (rs7931342, T) could be related to an increased Gleason score, AG on HNF1B (rs4430796, A) could be associated with PSA increase, and TG on 17q24 (rs1859962, G) could be negatively associated with an increased BMI in Chinese men with PCa.

8q24 rs4242382 Polymorphism is a Risk Factor for Prostate Cancer among Multi-Ethnic Populations: Evidence from Clinical Detection in China and a Meta-analysis

  • Background: Evidence supporting an association between the 8q24 rs4242382-A polymorphism and prostate cancer (PCa) risk has been reported in North American and Europe populations, though data from Asian populations remain limited. We therefore investigated this association by clinical detection in China, and meta-analysis in Asian, Caucasian and African-American populations. Materials and Methods: Blood samples and clinical information were collected from ethnically Chinese men from Northern China with histologically-confirmed PCa (n=335) and from age-matched normal controls (n=347). The 8q24 (rs4242382) gene polymorphism was genotyped by polymerase chain reaction-high-resolution melting analysis. We initially analyzed the associations between the risk allele and PCa and clinical covariates. A meta-analysis was then performed using genotyping data from a total of 1,793 PCa cases and 1,864 controls from our study and previously published studies in American and European populations, to determine the association between PCa and risk genotype. Results: The incidence of the risk allele was higher in PCa cases than controls (0.222 vs 0.140, $P=7.3{\times}10^{-5}$), suggesting that the 8q24 rs4242382-A polymorphism was associated with PCa risk in Chinese men. The genotypes in subjects were in accordance with a dominant genetic model (ORadj=2.03, 95%CI: 1.42-2.91, $Padj=1.1{\times}10^{-4}$). Presence of the risk allele rs4242382-A at 8q24 was also associated with clinical covariates including age at diagnosis ${\geq}65$ years, prostate specific antigen >10 ng/ml, Gleason score <8, tumor stage and aggressive PCa, compared with the non-risk genotype ($P=4.6{\times}10^{-5}-3.0{\times}10^{-2}$). Meta-analysis confirmed the association between 8q24 rs4242382-A polymorphism and PCa risk (OR=1.62, 95%CI: 1.39-1.88, $P=1.0{\times}10^{-5}$) across Asian, Caucasian and African American populations. Conclusions: The replicated data suggest that the 8q24 rs4242382-A variation might be associated with increased PCa susceptibility in Asian, Caucasian and African American populations. These results imply that this polymorphism may be a useful risk biomarker for PCa in multi-ethnic populations.
