Eastern medicine approach led about diagnosis and treatment of the leukemia and with leukemia treatment the medical herb which the eastern medicine possesses in development and room sacrifice an eastern medicine theory and the false eye presented the drug which is therapeutic possibility and they presented. The eastern medicine demonstration and therapeutic method is showing the therapeutic ratio which is effective about the leukemia.   • A class I type 2 metallothionein (CMet2) cDNA from taproot of Codonopsis lanceolata was isolated and characterized. A CMet2 cDNA was 572 nucleotides long and had an open reading frame of 234 bp with a deduced amino acid sequence of 78 residues (pI = 4.99). The deduced amino acid sequence of CMet2 matched to the previously reported type 2 metallothionein-like protein genes and showed 74% identity with that of G. max (BAD18377) and C. arietinum (CAA65009). Expression of CMet2 by the RT-PCR was increased at 1 hr after cadmium and hydrogen peroxide treatment, respectively.

  • GCSB-5 preparation is a purified extract from a mixture of 6 medicinal plants(Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used for the treatment of various bone disorders. The aim of this study was to investigate HPLC analysis method and screening of standard compound on Saposhnikoviae Radix for quality standardization of a medicinal crude drug GCSB-5. Standard compound of Saposhnikoviae Radix was decided with cimifugin by isolation and instrumental analysis such as NMR. HPLC analysis method for the simultaneous determination of cimifugin was established for the quality control of the medicinal plants of Saposhnikoviae Radix species, GCSB-5 raw material and preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline.

Cuttings for Mass Propagation Affecting the Impact of Increasing Reproductive Efficiency of Schisandra chinensis (오미자 대량증식을 위한 삽목번식 효율증대에 영향을 미치는 요인)

  • This experiment was carried out to establish the optimum cutting time, plant growth regulator, and bed-soil for rooting by greenwood cutting of Schisandra chinensis. Approximately 7 cm long-shoots of greenwood cuttings were transplanted by various cutting times, plant growth regulators, and bed-soils in the plastic-film house. The rooting rate of greenwood cutting was 12.5% in the April 5th, 73.5% in the May 8th, and 75.5% in the August 5th. The number and length of primary root in greenwood cutting were more in the early May than those in the early August. For mass propagation of Schisandra chinensis. using greenwood cutting, shoots were treated with plant growth regulators on May to increase rooting rate. Rooting rate was 100% with IAA $50mg/{\ell}$, 92.9% with NAA $100mg/{\ell}$, and NAA $1,000mg/{\ell}$, for 60 min. To select effective media for rooting, various medias for bed-soil were treated by single and mixture form for 100 days after cutting. Rooting rate was 91.8% in the single treatment of peat moss or decomposition of granite soil, and this result was better than those in other treatment. The treatment by 1 : 1 mixture of peat moss and horticulture bed soil was rooting with 94.0% best rooting rate.

Effects of Roasting Conditions on Components of Safflower(Carthamus tinctorius L.) Seed (홍화(紅花) 종실(種實)의 볶음 조건에 따른 성분(成分)의 변화(變化))

  • The main objective of this study was to characterize physico-chemical properties, sensory property and oxidative stability of safflower seed obtained by various roasting temperature and time. The contents of water soluble solids decreased in the higher roasting temperature and time. Sensory evaluation of safflower seed roasted in various conditions showed significant differences in taste, color, flavor and palatability. The safflower seed roasted at $190^{\circ}C$ for 20min had the best palatability. At the change of Hunter's values, L values were decreased, and a, b and ${\Delta}E$ values were increased in the higher roasting temperature and time. The content of free sugars such as sucrose and raffinose were reduced significantly in higher roasting time of $190^{\circ}C$ and $210^{\circ}C$. During the storage period after roasting treatment, peroxide values (POV) were highly increased after eight months at the all treatment except for $150^{\circ}C$. Therefore, it is inadequate over eight months after roasting treatment.

Colchicine-Induced Polyploidy and It's Agronomic Characters in Bupleurum falcatum (배수체 작성에 따른 시호 작물 특성)

  • The effect of colchicine treatment on the agronomic performance and polyploid formation of Bupleurum falcatum using flow cytometry technique was investigated. The roots of 4-leaf stage plants were treated with colchicine (0.5%) for 3, 6, 12, and 24 hours and then transplanted in the field. Agronomic characters (survival rate, plant height, chlorophyll content, bolting rate) were recorded at 4 weeks and 6 months after transplanting while flow cytometry technique was conducted for determination of polyploid formation. Flow cytometry technique revealed polyploid nuclear DNA formation in colchicine treated plants. The highest number of polyploids was obtained at the shortest colchicine treatment time indicating an inverse relationship between colchicine treatment time and polyploid formation. Results also showed that survival and bolting rates were inversely related with the treatment time while plant height and chlorophyll were not significantly affected by the treatment. This study showed a convenient method for determination of colchicine-induced polyploid in B. falcatum and its superior agronomic performance at shorter treatment time.

Effect of Medicinal Plants on Cytokine-induced Apoptosis in Thyroid Cells (생약혼합물이 사이토카인에 의한 갑상선세포의 Apoptosis에 미치는 영향)

  • Apoptosis plays an important role in autoimmune chronic (Hashimoto's) thyroiditis, a disorder that often results in hypothyroidism. The goal of this study was to induce apoptosis by the combination of inflammatory cytokines, interferon $(IFN)-{\gamma}$ and tumor necrosis factor $(TNF)-{\alpha}$, and to investigate a potential role of medicinal plants in the thyroid follicular cells (FRTL) in vitro. The apoptosis was evaluated by cellular viability, DNA fragmentation, and terminal deoxynucleotidyl transferase-mediated deoxy-UTP nick end labeling (TUNEL) assay. Extract of Gamgung-tang (GGT, Glycyrrhizae Radix, black beans, Angelicae Radix, and Cnidii Rhizoma) $(0.3{\sim}9.0mg/ml)$ was shown to maintain the viability of cells treated with $IFN-{\gamma}(100U/ml)$ and $TNF-{\alpha}$ (0.5 ng/ml). FRTL cells were found to undergo DNA fragmentation with the inflammatory cytokines. The extract of GGT inhibited DNA fragmentation in dose-dependent manner. The cells with TUNEL-positive nuclei were detected with $IFN-{\gamma}$ and $TNF-{\alpha}$ treatment. The number of TUNEL-positive cells decreased with the treatment of extract of GGT. These results indicate that medicinal plants inhibit the occurrence of apoptosis in thyroid follicular cells, therefore, may have therapeutic potential in the treatment of autoimmune chronic thyroiditis.

Antioxidant and Xanthine Oxidase Inhibitory Activities of Hot Water Extracts of Medicinal Herbs (한약재 열수추출물의 항산화 활성 및 Xanthine Oxidase 저해 활성)

  • In our study, as many as 29 edible medicinal herbs were selected for testing their ability in the effective treatment of gout based on oriental medicine theory. We extracted each medicinal herb (135 g) with 4 L of distilled water at $100{\sim}105^{\circ}C$ for 210 min. Thereafter, we evaluated both the antioxidant and xanthine oxidase inhibition activities of the extracts obtained. Among all the edible medicinal herbs used in our study, only the extract from Scutellaria baicalensis Georgi (Korean name: hwang-geum) showed (1) the maximum total phenolic content (TPC) (2.25 mg gallic acid equivalent/mL), (2) DPPH radical scavenging activity (94.04%), and (3) xanthine oxidase inhibition activity (87.75%). We also observed that TPC was relatively highly correlated with both the DPPH radical scavenging activity (r=0.63) and xanthine oxidase inhibition activity (r=0.77). Our results suggest that S. baicalensis G. may be a potent antioxidant source for the extraction and development of nutraceuticals that may be utilized for effective treatment of gout.

Analysis of Existing Guidelines and Randomized, Controlled, Clinical Trials for Development of [Guideline of Clinical Trial with Herbal Medicinal Product for gastric cancer] (위암 한약제제 임상시험 가이드라인 개발을 위한 한약제제 무작위배정 대조군 임상시험 고찰)

  • Objectives: This study aimed to learn what should be considered in [Guideline of Clinical Trial with Herbal Medicinal Product for Gastric Cancer)] by analyzing the existing guidelines and clinical trials. Methods: The development committee searched guidelines for herbal medicinal product or gastric cancer developed already. Then, clinical trials for gastric cancer using herbal medicine were searched. The searched trials were analyzed in terms of inclusion and exclusion of participants, intervention, comparator, outcomes and trial design. Then, we compared the results of analysis with the regulations and guidelines of Ministry of Food and Drug Safety to suggest the issue that we will have to consider when making the [Guideline of Clinical Trial with Herbal Medicinal Product for Gastric Cancer]. Results: As a result, few guidelines for anti-tumor agent and clinical trial with herbal medicinal product were searched in the national institution homepage. In addition, 10 articles were searched by using the combination following search term; 'stomach neoplasm', 'herbal medicine', 'Medicine, Korean traditional', 'Medicine, Chinese Traditional', 'TCM', 'TKM', 'trial'. Most trials included gastric cancer participants with medical history of operation. The type of intervention was various such as decoction, granules, and fluid of intravenous injection. Comparators were diverse such as placebo, conventional treatment including chemotherapy and nutritional supplement. The most frequently used outcome for efficacy was quality of life. Besides, the symptom score, tumor response, and survival rate were used. Safety was investigated by recording adverse events. Conclusion: We found out some issue by reviewing the existing guidelines and comparing it with clinical trials for gastric cancer and herbal medicinal products. These results will be utilized for developing [Guideline of Clinical Trial with Herbal Medicinal Product for Gastric Cancer].

Selection of Oriental Medicinal Plants for Screening of Anticancer Agents (항암소재의 발굴을 위한 한방소재의 선별)

  • An oriental medicinal database was used from medicinal plants for screening of anticancer agents. The prescription frequency and dosage of medicinal plats in 170 anticancer prescriptions were analyzed. From the total prescription score of each medical plant, it was found that high-scored medicinal plants have been widely studied in the previous research on the cancer treatment. Consequently, dangui, hwangui, banha, bachul, jinpy, insam, bacjakyak, deahwang, chungoong, jimo, chunnamsung, omija, hyunggae, huekchook, banggi, boclyung, osooyou were selected as raw materials for the screening of anticancer agents.
