Effects of Dietary Fiber Sources on the Composition Intestinal Microflora in Aged Rate (노화 흰쥐의 장내 균총 조성에 대한 식이섬유질원의 영향)

  • This study was performed to investigate the influence of vadrious dietary fiber sources such as mugwort, apple, sea mustard, butterbur on the intestinal in aged rats. Nine groups of aged rats (12months after weanling) were fed each experimental diet containing 5% and 15% dietary fibers for 4 weeks. The contents in large intestine of aged rats used for the measurement of intestinal microflora. Total viable counts and the numbers of Bifidobacterium, Lactobacillus, Clostridium, E. coli and Staphylococcus were determined by nonselective medium and various selective media. The intake of dietary fiber sources employed did not show any differece in the number of total microorganisms, Lactobacillus and Bifidobacterium. It was remarkable that Clostridium perfringens appeared in the colon of aged rats. The prpulations of Peptococcus and Clostridium were significantly larger in the apple 5% group than in other dietary fiber groups while the number of Staphylococcus was significantly smaller in the dietary fiber groups than in cellulose group. And the Streptococcus populations decreased in the mugwort 15% group. Accordingly, the mugwort 15% group is effective in improving intestinal flora of aged rats.

PAH로 오염된 토양의 미생물 분해 가속화 연구

  • Bioremediation of hazardous hydrophobic organic compounds, such as polycyclic aromatic hydrocarbons (PAHs), is a major environmental concern due to their toxic and carcinogenic properties. Bue to their low solubility in water, the compounds are microbiologically persistent. This work investigates optimal conditions to enhance the biodegradation of phenanthrene in water and soil-slurry systems. Biodegradation tests were performed with three different types of supplements: glucose as a general carbon source, salicylate as an enzyme inducer, and Triton X-100 as a surfactant. The tests indicate that glucose and Triton X-100 were not very effective to increase biodegradation rate, even though the number of microorganisms are highly increased in the case of glucose addition. Salicylate accelerated biodegradation of phenanthrene, but the addition above optimal concentration inhibited microbial growth. Salicylate is considered to be an attractive alternative for the successful bioremediation of PAH-contaminated soil.

Antibacterial Activity of Euphorbia humifusa Extracts on Food-Borne Pathogenic Bacteria (비단풀 추출물의 식중독세균에 대한 항균활성)

  • This study was performed to investigate the antimicrobial effects of Euphorbia humifusa ethanol-extract against food-borne pathogens. The growth inhibitory effects of the extract at a concentration of 250, 500, 1,000 or 2,000 mg/L on food poisoning microorganism were determined against Salmonella typhimurium, Listeria monocytogenes, Yersinia enterocolitica, Escherichia coli O157:H7 and Staphylococcus aureus. The microorganisms growth was not affected by the extract at the concentration up to 250 mg/L, but was significantly (p<0.05) inhibited by the extract at a concentration higher than 1,000 mg/L. The extract of Euphorbia humifusa had strong antimicrobial activity against all test strains at a concentration of 2,000 mg/L. The results in the present study demonstrate antimicrobial effects of Euphorbia humifusa ethanol-extract against food-borne pathogens, suggesting that Euphorbia humifusa could be an effective natural antibacterial agent in food.

Effects of Ozone Treatment and Gamma Irradiation on the Microbial Decontamination and Physicochemical Properties of Red Pepper Powder (고춧가루의 오염미생물 제거 및 이화확적 특성에 관한 오존처리와 감마선 조사의 영향)

  • The comparative effects of ozone treatment and gamma irradiation on the sterilization, physicochemical properties and sensory quality of red pepper powder were investigated. As for the sterilization of microorganisms, 7.5~10 KGy of gamma irradiation completely eliminated the coliforms, yeast and molds, and total aerobic bacteria. On the other hand, ozone treatment failed to eliminate the highly contaminated microbial load, especially total aerobic bacteria. The physicochemical properties including capsaicin, capsanthin, browning, fatty acid compositions and sensory quality were not significantly changed by gamma irradiation up to 10 kGy, whereas ozone treatment caused significant changes in fatty acid compositions and destruction of natural pigments (p<0.05). The above results led us to conclude that gamma irradiation was more effective than ozone treatment for the sterilization and maintenance of physicochemical and sensory qualities of red pepper powders.

  • Vaccination has been considered to be the most effective way to control infectious diseases. Currently, many vaccines used in humans are live-attenuated or killed microorganisms. Polio, mumps, and measles vaccines are live-attenuated. Killed vaccines include cholera and pertussis vaccines, These conventional vaccines, however, suffer from some problems. In the case of live-attenuated vaccines, reversion to virulence is observed in a small but significant number of clinical cases each year. In killed vaccines, due to the possible hazard to employees working with live pathogens, the cost of preparation is high. Killed vaccines also need to be given in multiple doses, Furthermore, both live-attenuated and killed vaccines have possible presence of cellular materials leading to side effects. Moreover, there are diseases such as malaria and hepatitis for which conventional attenuated and killed vaccines are not available because the pathogens cannot be grown in sufficient amounts to allow the classical methods to be used.

유용미생물(Effective Microorganisms ; EM)에 의한 돈육 보존의 효과

  • EM이 돼지 사양에서 육질개선에 미치는 효과를 알아보기 위해 사양실험을 하였다. 일반적으로 사양한 돼지와 EM 축산사료첨가제와 EM 활성액을 정기적으로 급이하여 사양한 EM Pork를 도축한 후 각각의 돈육을 $4^{\circ}C$ 냉장온도와 $-18^{\circ}C$ 냉동온도에서 저장하였다. 그후 일반 돈육과 EM Pork의 일반성분 및 육질의 보존성에 관한 실험을 실시하였다. 단백질 변성에 관한 실험결과 일반 돈육을 5일간 냉동저장한 것은 3.72 mg%를 보였고 EM Pork는 3.2 mg%를 나타내 일반 돈육보다 15% 정도 단백질 변성을 지연시켰다. 또한, 일반 돈육을 5일간 냉장저장한 것은 4.9 mg%를 나타냈으나 EM Pork는 3.19 mg%를 나타내 일반 돈육보다 35% 단백질 변성 억제 효과를 보였다. 또한, 냉동 및 냉장 처리한 일반 돈육의 조단백질 함량은 각각 21.41%와 19.73%인데 비해 냉동 및 냉장 처리한 EM Pork는 각각 22.21%와 21.41%를 나타냈다. 이러한 결과는 단백질 변성과 관련된 것이라 볼 수 있다. 결론적으로 EM Pork육은 일반 돈육에 비해 단백질 변성 억제 효과가 15${\sim}$35% 정도 우수한 것으로 사료된다.

EM(Effective Microorganisms)과 관련된 유산균을 첨가한 발효육의 산화억제 효과

  • 유산균을 첨가한 발효육의 지방산패도를 측정하여 EM의 항산화 효과와의 관련성을 살펴보기 위해 실험을 실시하였다. 발효육의 생산에 사용된 Lactobacillus casei KCTC 3109 공시균주는 한국생명공학연구원 유전자지원센터 한국유전자은행에서 주문 구입하였다. 실험에 사용된 돈육은 도살 후, $4^{\circ}C$ 냉장고에 저장하였던 랜드레이스의 대퇴부 육을 사용하였다. 발효육의 항산화 정도를 알아보기 위해 TBA 방법으로 실험하였다. 대조구는 저장 0일부터 저장 3일까지 TBARS 치가 지속적으로 상승하는 것을 보였다. 그러나 저장 4일째부터는 TBARS 치가 감소하는 것을 볼 수 있다. 반면, 유산균을 첨가한 육에서는 저장 0일째부터 2일째까지는 약간 TBARS 값이 증가하였으나 저장 3일째는 가장 낮은 수치를 보였다. 그러나 저장 4일째부터는 TBARS 치가 점차적으로 증가하였다. 대조구의 저장 3일째의 TBARS치가 0.19 mgMDA/Kg을 나타낸 반면, 유산균을 첨가하여 $37^{\circ}C$에서 3일간 정치 배양한 발효육의 TBARS치는 0.02 mgMDA/Kg을 나타내 8배 정도의 항산화 효과를 나타냈다. 결론적으로 유산균 자체가 항산화 효과가 있음을 알게 되었고 EM의 항산화 효과에도 영향을 주는 것으로 사료된다.

  • In order to control the quality and safety of cooked foods, production and holding methods of foods should be carefully studied and applied to the foodservice industry. Therefore studies on microbiological, nutritional, physicochemical and sensory quality are essential in this area. The purpose of this study was to evaluate the chemical and microbial qualities of cooked foods during preparation and holding From the results, the following guidelines should be adhered to give effective quality control when holding foods after cooking in foodservice institutions, as well as to provide quality foods when selling cooked foods at commercial establishments. Sauteed and simmered foods such as sauteed chicken meat & vegetables and simmered pork in soy sauce satisfy the standard for microorganisms till 1-3 hours of room temp. holding, 6-18 hours at 60$^{\circ}C$ steam table, and 12-18 hours at 80$^{\circ}C$ heating table.

Isolation of Antagonistic Microorganism for Biological Control to Apple Diseases, Bitter Rot (사과 탄저병 방제를 위한 길항미생물 분리)

  • In order to acquire microbial agents that can be utilized for biological control of bitter rot(Glomerella cingulata), the major airborne disease to apple, the effective microorganisms were isolated, tested for antagonistic activity to the pathogen. Through the screening of more than 1,000 species of microorganisms collected in nature, 11 species of antagonists were selected. On of the 11 species, one species designated as CH141 demonstrated outstanding activity. The bacterial strain, CH1141 exerted antagonistic efficiency of 65% on Glomerella cingulata. The CH1141 was identified as a bacterial strain to Bacillus subtilis based on morphology, culture conditions, and physiobiochemical characteristics.

Improvements in Storage Stability of Undaria pinnatifida after Gamma Irradiation (감마선 조사에 의한 미역의 저장성 개선)

  • The effect of gamma irradiation on the storage stability of Undaria pinnatifida was investigated. A three-log reduction in total aerobic bacteria in U. pinnatifida samples was obtained by irradiation with 3 kGy. During storage for 2 weeks, the viable counts of microorganisms in non-irradiated U. pinnatifida increased to 6 log CFU/g. However, irradiation of U. pinnatifida by 5 kGy eliminated microorganisms. The Hunter $L^{\ast}-value$ of U. pinnatifida was slightly increased after 5 kGy of irradiation. The hardness properties of U. pinnatifida decreased during storage, but were not changed by irradiation with up to 3 kGy. These results indicate that low-dose irradiation (3 kGy) could be effective for improvement of the storage stability of U. pinnatifida.
