3종의 한국산 겨우살이 메탄올 추출물의 Tyromyces palustris, Endothia nitschkei 그리고 Trichophyton rubrum에 대한 항균활성 (Antifungal Activities of meOH Extracts from Three Korean Mistletose against Tyromyces palustris, Endothia nitschkei and Trichophyton rubrum)

  • 본 실험에서는 3종의 한국산 겨우살이 메탄올 추출물을 유기용매 분획과 컬럼크로마토그래피를 이용한 분획을 실시한 후, Tyromyces palustris, Endothia nischkei 그리고 Trichophyton rubrum에 대한 항균활성을 평가하여 겨우살이 추출물의 항균활성 자료로 활용하고자 하였다. 그 결과 모든 겨우살이 추출물은 갈색 불후균에 대한 활성은 매우 낮게 나타났다. 그러나 Viscum albumvar. coloratum 추출물중 부탄올 분획은 황색줄기 마름 병원균의 성장을 억제하는 것으로 나타났으며, 이 겨우살이의 EtOAC 분획은 족부백선균에 대하여 높은 포자 발아 억제효과를 나타내었다. 시험된 3종의 한국산 경우살이류 중 V. album var. Coloratum은 병원균에 대하여 황균활성 성분을 함유할 가능성이 가장 높을 것으로 제시되었다.

한국산 겨우살이 전초의 Methanol 추출물로부터 암세포증식 저해성분의 분리 (Active Principles of the Methanol Extract of Korean Mistletoe Responsible for the Inhibitory Effect on the Proliferation of Human Tumor Cell Lines)

  • A bioassay-guided fractionation of the whole extract of Viscum album var. coloratum Ohwi (Loranthaceae) led to the isolation of two triterpenoidal components; oleanolic acid (1) and ${\beta}-amyrin$ acetate (2), and a flavonoid, homoflavoyadorinin B (3) as well as large quantity of free fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble fraction and BuOH soluble fraction of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system), and HCT-15 (colon) in vitro, whereas the remaining water soluble fraction exhibited a poor inhibition. The intensive phytochemical investigation of the EtOAc soluble fraction and BuOH soluble fraction of the extract indicated that the oleanolic acid (1) and large amounts of free fatty acid mixtures might be attributed to the in vitro antitumoral property of the whole extract of Viscum album var. coloratum.

한국산 겨우살이(Viscum album coloratum)로부터 추출된 lectin의 돼지에 대한 독성 및 오제스키병 백신의 면역원성에 미치는 영향 (Toxicity of lectin extracted from Korean mistletoe (Viscum album coloratum) in piglets and its effects on the immunogenicity of Aujeszky's disease virus vaccines)

  • In the present study toxicity and immunostimulating activity of the lectin(KML-C), which was extracted from Korean mistletoe(Viscum album coloratum) were investigated in swine. To determine the toxicity, lectin was injected into thigh or cervical muscles of 4-week-old piglets(Landrace) and observed clinically and pathologically. For determination of the immnunostimulating activity, lectin($0.7{\mu}g/kg$ of body weight)-adjuvanted vaccine of Aujeszky's disease virus(ADV)(NYJ1-87) which was inactivated by 0.2% formalin was injected into the cervical muscle of antibody-negative piglets in the same age group. Subpopulation of the immune cells and serum neutralizing(SN) antibodies in the piglets were examined after vaccination, and resistance of the piglets against challenge by virulent NYJ1-87 was further examined. The results were also compared with those from piglets injected with aluminum hydroxide [$Al(OH)_3$]-adjuvanted vaccine of inactivated NYJ1-87 and NYJ1-87 vaccine without adjuvant, and the results are as follows. By injection of lectin with $30{\mu}g/kg$ of body weight to the thigh muscle, all of 12 piglets died after signs such as dyspnea, fever, systemic erythema and subcutaneous hemorrhages, and lesions pertaining to poisonous hepatitis and dysfunction of kidney were observed. By injection of lectin with $7{\mu}g/kg$ of body weight to the thigh muscle, all of 12 piglets showed signs such as edema and cutaneous hemorrhage in the injected area, lameness and depression, and lesions pertaining to poisonous hepatitis and dysfunction of kidney were observed. By injection of lectin with 1, 3 and $5{\mu}g/kg$ of body weight to the thigh muscle of each one piglet, signs such as congestion, induration and grayish coloration in the injected area, depression and inappetence were observed in all piglets. Toxic changes were also observed in the liver and kidney of piglets by lectin of 3 and $5{\mu}g$. By injection of lectin with 0.5 and $0.7{\mu}g/kg$ of body weight to the cervical muscle of each 9 piglets, all piglets were clinically normal and there were no significant changes in blood counts and chemistry values. Whereas, epithelial swelling and vacuolation of convoluted tubules were observed from one piglet injected with lectin of $0.7{\mu}g$, and necrosis and fibrosis of muscular fiber were observed in the muscle of one piglet injected with lectin of $0.5{\mu}g$. Only population of sIgM+ B lymphocytes increased among immune cells in all of 15 piglets immunized with lectin($0.7{\mu}g/kg$ of body weight)-adjuvanted vaccine, while compared to those in $Al(OH)_3$-adjuvanted vaccine and vaccine without adjuvant. No additional stimulation to the immune cells was recognized when lectin was added to $Al(OH)_3$-adjuvanted vaccine. In piglets immunized with lectin-adjuvanted vaccine, SN titers in reciprocal values for loge were 1.3-4.0 at 1-4 weeks after vaccination, which was similar to those with 1.0-3.3 by vaccine without adjuvant but lower than those with 2.0-5.7 by $Al(OH)_3$-adjuvanted vaccine. Also, no additional increase in the SN titers was recognized when lectin was added to $Al(OH)_3$-adjuvanted vaccine. Piglets immunized with lectin-adjuvanted vaccine were resistant to challenge by the virulent NYJ1-87 at 4 weeks after vaccination, and the SN titers reached to 5.0 one week after challenge, which was higher than those with 4.0 by vaccine without adjuvant but somewhat lower than those with 7.7 by $Al(OH)_3$-adjuvanted vaccine.

Which is the More Effective Option for Pleurodesis to Prevent the Recurrence of Malignant Pleural Effusion? Large-Particle Talc or Mistletoe Extract (ABNOVA Viscum Injection)

  • Background: Malignant pleural effusion affects many patients with advanced cancer. When chemotherapy or radiotherapy fails to relieve malignant pleural effusion and related symptoms, drainage and pleurodesis can help. Although surgical talc pleurodesis is the most widely used method, Viscum album, which has been recently used in surgical or bedside procedures, has demonstrated significant results and is as effective as talc. This study aimed to determine the most effective agent and procedure. Methods: Between January 2015 and July 2022, chemical pleurodesis was performed in 137 patients with malignant pleural effusion, using a V. album surgical procedure in 48, a V. album bedside procedure in 55, and a talc surgical procedure in 34 patients. We reviewed patients' clinical responses and disease progression after chemical pleurodesis. Results: The success rate was not significantly different among the V. album surgical procedures (91.7%), V. album bedside procedures (83.6%), and talc surgical procedures (91.2%). However, the total drainage amount and tube insertion duration in both Viscum groups were more effective than those in the talc group. Furthermore, the bedside Viscum group showed significantly lower post-pleurodesis pain scores than the other 2 groups. Conclusion: According to our results, talc and V. album can be considered ideal agents for chemical pleurodesis. However, Viscum pleurodesis showed safer outcomes in terms of ensuring quality of life than talc. Additionally, the bedside Viscum group showed significantly lower pain scores than the other groups. Hence, patients for whom surgical procedures are inappropriate can undergo bedside Viscum pleurodesis without diminishing the therapeutic effect.

20 kGy 감마선으로 조사된 겨우살이 냉수 추출물의 90일 반복투여 독성평가 (90-day Repeated-dose Oral Toxicity Study of 20 kGy Irradiated Cold Water Extract Powder of Mistletoe)

  • 연구에서는 임상에서 암 치료의 보조제로 사용하고 있는 겨우살이의 독성성분의 구조적인 변화를 유도하여 겨우살이의 기능성을 향상시키는 하나의 방법으로, 감마선을 조사한 겨우살이 냉수 추출물의 안전성을 검토하고자 ICR계 열의 암수 마우스에 시험물질을 0, 20, 100 및 500 mg/kg/day의 용량으로 90일간 반복 경구투여한 후, 사망률, 일반증상, 체중변화, 혈액 및 혈액생화학적 변화, 부검소견, 조직학적인 변화를 관찰하였다. 시험기간 중 암수 모든 군에서 시험물질 투여에 기인한 일반적인 증상변화는 관찰되지 않았고, 시험물질의 반복 투여로 인한 사망례 역시 관찰되지 않았다. 시험물질의 투여에 기인한 유의적인 체중감소 또한 나타나지 않았으며, 상기 이외의 육안적인 부검소견에서도 시험물질 투여에 기인한 어떠한 이상소견도 발견되지 않았다. 혈액학적 분석 결과 일부 시험물질 투여군에서 총 백혈구 수와 림프구, 중성구, 단핵구, 헤모글로빈, 혈중 적혈구 비율, 평균 헤모글로빈의 함량, 평균헤모글로빈 농도, 적혈구 크기 분포폭, 혈소판의 수치가 유의적인 변화를 보였으나, 정상범위 내에서의 변화로 감마선조사에 의해 야기된 독성이라고 판단하기는 어렵다. 또한 혈청 중의 alanine aminotransferase, aspartate aminotransferase, 중성지방, 콜레스테롤 수치가 대조군과 비교할 때에 통계학적으로 유의성 있는 변화를 나타냈으나 정상 생리 범위 내에서의 변화로 시험물질에 의한 독성이라 판단하기는 어렵다. 간장과 신장의 조직학적인 관찰에서 시험물질 투여에 의한 변화는 관찰되지 않았다. 이상의 독성 시험 결과, 20 kGy 감마선 조사 겨우살이 냉수 추출물을 3개월간 ICR 마우스에 섭취시 킨 경우, 시험한 최고 농도인 $500{\mu}g$/kg/day에서는 독성이 없는 것으로 판명되었다.

한국산 겨우살이과 식물의 형태와 기주별 흡기 특징 (Anatomy of the Korean mistletoe and their haustorial features in host plants)

  • 한국에 자생하는 겨우살이과 4종, 겨우살이, 참나무겨우살이, 동백나무겨우살이, 꼬리겨우살이 등에 대해 잎과 줄기의 해부학적 특징과 기주식물별 2차 흡기구조의 해부학적 특징을 조사하였다. 그 결과 잎과 줄기의 해부학적 특징으로 한국산 겨우살이과 4 분류군을 종과 속 수준에서 구별할 수 있는 것으로 파악되었다. 잎에서는 향축면과 배축면의 축, 중앙맥에 발달된 유관속 형태, 후벽세포의 발달 여부 등이, 줄기에서는 횡단면에서 줄기의 전체적인 형태, 큐티클층의 발달 정도, 유관속의 배열, 후벽세포와 수의 발달 여부 등이 표징형질로 조사되었다. 기주별 2차 흡기의 특징을 확인하기 위해 참나무겨우살이와 동백나무겨우살이에서 각각 7종류 및 5종류의 기주식물을 대상으로 지상부 흡기 발달 정도, 종단면에서 2차 흡기의 모양과 플랜지 발달 여부, 기주내 침투 지역과 흡기 축의 발달 여부, 2차 흡기에서 흡기세포의 위치와 짧은 흡기 가도관의 발달 여부 등 형질을 조사하였다. 조사된 형질에서 참나무겨우살이와 동백나무겨우살이는 뚜렷하게 구별되었다. 한편, 기주식물의 강도와 흡기의 발달 정도에 따라 2차 흡기의 전체적인 형태가 기주별로 다소 차이가 있었으나, 침투 지역, 흡기 짧은 가도관 세포의 발달 등 정성적인 형질들은 기주와 발달 정도에 영향을 받지 않고 항상 일정하여 계통학적으로 의미 있는 특징으로 파악되었다.

사료내 택사, 겨우살이 및 산수유 생균제 첨가가 비육돈의 생산성 및 면역성에 미치는 영향 (Effects of dietary Alisma canaliculatum(Alismatis rhizoma), Viscum album (Mistletoe) and Cornus officinalis (Corni fructus) probiotics as feed additives on growth performance and immunity in growing pigs)

  • An investigation was conducted to evaluate the effects of supplementing Alisma canaliculatum, Viscum album and Cornus officinalis probiotics on the growth performance and immune response in growing pigs. This experiment was conducted using 120 pigs (crossing of Landrace${\times}$Yorkshire and castrated) which were assigned to 5 treatments in 3 replications with 8 pigs per replications. The dietary treatments were NC group (without antibiotics), PC group (basal+Oxytetracycline 50ppm), AC group (basal+A. canaliculatum 0.5%), VA group (basal+V. album 0.5%) and COP group (basal+C. officinalis probiotics 0.5%). The initial body weights of pigs were 35kg on average and the experiment lasted for 9 weeks. The experimental animals were kept in the pens following a completely randomized design. They were provided the diets adequate for grower stage as recommended by NRC (ME:3,265 kcal/kg and CP:16%). COP fed pigs showed lower weight gain up to 6 weeks of age compared to NC group and other groups without significant differences (P>0.05). The carcass weights of pigs fed VA and COP were significantly higher compared to NC group (P<0.05), Back fat thicknesses groups fed three different additives were higher than NC group and lower then PC group (P<0.05). Crude fat contents in loin meat were significantly lower in groups fed three different additives while moisture contents of those three groups were higher than other groups (P<0.05). The thiobarbituric acid reaction substance (TBARS) value measured at fresh and $2^{nd}$ weeks was lower in additives fed groups but no statistical differences were observed among the treatments (P>0.05). Significantly highest PUFA (16.42g/100g) and ${\omega}$-3 fatty acids (ALA, EPA and DHA) content of meat were observed in COP fed pigs compared to NC group (P>0.05), which might mean that three additives function to enhance serum IgG in pigs. In consequence, it can be suggested that AC, VA and COP may have a potential to replace antibiotics as growth promoter and immune enhancer in the diets for growing pigs.

택사와 겨우살이 급여가 육계의 생산성 및 면역성에 미치는 영향 (Effect of Dietary Supplementation of Alisma canaliculatum (Alismatis Rhizoma) and Viscum album (Mistletoe) on Growth Performance and Immunity in Broiler Chickens)

  • 택사와 겨우살이가 육계의 생산성 및 면역성에 미치는 영향을 규명하기 위하여 실시하였으며, 공시동물은 육계 초생 추 140수를 4처리구 5반복 반복당 7수씩 공시하여 5주간 사양 시험을 실시하였다. 처리구는 무항생제 사료 급여구를 대조구로 하고 항생제 처리구(기초 사료+ OTC 0.05%), 택사 처리구(기초 사료+ 택사 0.5%) 및 겨우살이 처리구(기초 사료+겨우살이 0.5%)로 하였다. 증체량과 사료 섭취량은 처리구간에 유의적인 차이는 없었으며, 사료 요구율은 택사 처리구가 겨우살이 및 기타 처리구에 비해 유의적으로 높았다(P<0.05). 도체의 지방산 분석에서${\omega}$3 및${\omega}$6 계열의지방산인linolenic acid, ${\alpha}$-linolenic acid 및 arachidonic acid의 함량은 겨우살이 및 택사 급여구에서 항생제 처리구에 비해 비교적 높았으며, SFA, USFA 및 USFA/SFA의 함량은 처리구 간에 유의적인 차이는 없었다. 혈중의 IgG의 농도는 처리구 간에 유의적인 차이는 없었으나, 택사와 겨우살이 처리구가 높았으며, IL2의 농도는 택사와 겨우살이 처리구에서 유의적으로 높았다(P<0.05). IL6는 검출되지 않았다. 겨우살이 첨가구에서 GOT, GPT 및 T-billirubin이 유의적으로 높게 검출됨으로써 간장의 손상을 일으킬 수 있는 것으로 판단되었다(P<0.05). 결론적으로 육계 사료에 택사 0.5%와 겨우살이 0.5% 첨가 급여는 생산성은 항생제 첨가와 유사한 효과가 있고 면역성은 증가시켰다. 따라서 항생제 대체제로 사용 가능성이 높은 것으로 사료된다.
