Research on the Reformation of the Selection Index for Hanwoo Proven Bull (한우보증씨수소 선발지수 개선에 관한 연구)

  • Hanwoo proven bulls have been selected since 1987 and consequently contributed to farmers for the improvement of beef cattle in Korea. The demand for the quality beef production as well as higher production efficiency was erupted after early 2000 as relatively cheap imported beef released. Therefore the pressure on the reformation of selection index for Hanwoo proven bulls have been piled up to furnish with Hanwoo's competitive. A total of 734 progeny test data were analyzed to select traits and their weights in the selection index to meet the beef market requirement. Regression analysis with stepwise selection method was used to select proper trait and its weight for selection index. A series of computer simulation was carried out to compare the currently using selection index with the alternate two selection indices proposed in this study. New selection index using standardized breeding values of Loin eye Muscle Area (LMA), Backfat Thickness (BFT) and Marbling Score (MS) with weight ratio 1:-1:6 was proposed. Results showed higher performance in improving MS and BFT gain by 22% and 31% still holding 86%~89% of genetic gain achieved by current index in Carcass Weight (CW) and LMA when new selection index was fitted. Because, new index has little consideration for production cost, further research should be performed to build selection index including cost and income simultaneously.

STUDIES ON BRUCELLOSIS 2. Incidence of Brucellosis Reactors (BRUCELLOSIS에 관(關)한 연구(硏究) (제2보(第二報) 가축(家畜) BRUCELLOSIS에 대(對)한 역학적(疫學的) 조사보고(調査報告)))

  • During the period from 1956 to 1959, a serological survey was conducted in au effort to assessing the incidence of brucellosis in various domestic animals in Korea. The following results were obtained. 1. Seventy-six dairy cattle which were imported in the year of 1956 had 9 positive reactors (11.84%). Of 1127 goats imported in 1956 and 1958, eleven(0.98%) were reactor animals. 2. In 1956, of 108 dairy cattle tested 10(9.25%) were reactors. In 1937 and 1958, a total of 155 dairy cattle examined hid ten (6.45%) positive reactors. In 1959, of 127 dairy cattle examined two were reactor animals. 3. In 1958 and 1959, 432 goats in some districts in Korea were subject to test for brucellosis and the test revealed five (1.15%) reactors. In 1958, 683 swine serum samples were examined and eleven (1.04%) were positive. In 1959, 1133 Korean cattle were tested and seven samples (0.64%) showed positive reactions. 4. In the month of July in 1939, 580 beef cattle in Cheju National Ranch and Branch were examined and 111 (19.13%) were found to be reactors. In October of the same year, 157 cattle, consisting of reactor and suspicious cattle groups, were tested, of which 71 samples reacted positive and 47 remained suspicious. 5. In December of the year of 1958, there occurred. an outbreak of brucellosis in a swine herd in the Sachon Branch Experimental Station. Seven serological tests on 438 swine serum samples were conducted, of which 122 (27.87%) were found positive. 6. Dairy cattle No. 33 and No. 35 which had been imported in 1956 and detected as highly positive, were examined for a prolonged period to follow the variation of antibody titers. A marked drop in antibody titer was seen two months after the initial test while the re-increase in titer was observer our months after the first examination.

  • Epidemiological situation of taeniasis in Mongolia was assessed based on mitochondrial DNA identification of the parasite species. Multiplex PCR was used on a total of 194 proglottid specimens of Taenia species and copro-PCR and loop-mediated isothermal amplification (LAMP) assays were utilized for detection of copro-DNA of 37 fecal samples from taeniasis patients submitted to the Mongolian National Center for Communicable Diseases (NCCD) from 2002 to 2012. In addition, 4 out of 44 calcified cysts in beef kept in formalin since 2003 were evaluated for histopathological confirmation of cattle cysticercosis. All proglottid specimens and stool samples were confirmed to be Taenia saginata by multiplex PCR and by copro-PCR and LAMP, respectively. Cysts collected from cattle were morphologically confirmed to be metacestodes of Taenia species. T. saginata taeniasis was identified from almost all ages from a 2-year-old boy up to a 88-year-old woman and most prominently in 15-29 age group (37%, 74/198) followed by 30-44 age group (34.8%, 69/198 ) from 15 of Mongolia's 21 provinces, while cattle cysticerci were found from 12 provinces. The highest proportion of taeniasis patients was in Ulaanbaatar, the capital of Mongolia.

  • In this review, we present the results of our ten years of study on beef cattle production as a means of preserving phytodiversity and landscape in the semi-natural grasslands of the Aso region of Japan. There are about 16,000 ha of semi-natural grasslands in the region. The grasslands are maintained for animal production through such activities as grazing, mowing, and controlled burning. The Shannon's diversity index values, as measured in the present study, were higher in the grazed sites than in the abandoned sites. The grazing activities thus seem to create suitable habitats for coexisting herbaceous species. The grassland grazed moderately over a long term was able to sustain more animals than the grassland unused over a long term.

  • Not many types of roughage cutters have been introduced in Korea so far. However, those machines could not satisfy farmers to cut rice straw or barley wrap-silage properly. Stiffness and firmness of roughage bale were two obstructing factors. In order to solve this problem, a tractor attached cut-feeding machinery for the round baled roughage was developed for dairy and beef cattle farm in Korea. A series of tests were performed and acquired data were analyzed by using the several imported roughage cutters, which have been already introduced in dairy farm in Korea. And, a prototype of forage cutter was selected, analyzed, designed and manufactured to develop a tractor attached roughage cut-feeder for round bale. Also, the prototype machine was tested, modified and improved through revising model. As a result, a tractor attached roughage cut-feeder for round bale was manufactured. In order to evaluate the performance of the model machinery. a series of test were performed by the prototype machinery both at the plant and field. The model machinery developed satisfied in both power requirement and cutting capacity. As a conclusion, one of the obstacles against feeding the round baled roughage in the korea cattle farm can be eliminated by developing the tractor attached round baled roughage cut-feeder.

Comparison of OECD Nitrogen Balances of Korea and Japan

  • The nitrogen (N) balance in Korea during 1985-1997 was calculated according to the surface balance method of the PARCOM guidelines and compared with Japanese N balance. The some differences were founded in the coefficients used on calculating N balance in two nations. Of the important parameters, which can make a big difference in balance, N input by organic fertilizers was not included in Korea different with Japanese, due to absence of reliable statistics and then made lower the input. Nitrogen destruction rate from livestock manure was adjusted differently with 15% in Korea but 28% in Japan. There was some difference in the conversion factors of livestock number into manure N quantity in two nations, but the gap was ignoble scale except beef cattle. Our manure N production rate of beef cattle might be evaluated to be so lower than Japanese. Biological N fixation by pulses was very higher in Korea than in Japan but scarcely affect the increase of total N input, due to small cultivation area. In contrast, N fixation rate by free-living organisms in Korean and Japanese wet paddies showed the big difference with 7.6 and $37.0kg\;ha^{-1}\;yr^{-1}$, respectively, and therefore $29.4kg\;ha^{-1}\;yr^{-1}$ of nitrogen was estimated to be more inputted in Japan. Although there are many points to be more specified and improved, still, Korean N balance was very high with $250-257kg\;ha^{-1}$ in the mid of 1990s, which was the second highest level in OECD countries and furthermore increased continuously during the investigation. In contrast in Japan, which has similar fanning system with Korea, N balance was lower with $130-158kg\;ha^{-1}$ and has decreased continuously since 1993. This high N balance was mainly due to a high usage of chemical fertilizers in our intensive fanning system and the fast increment of livestock feeding. Therefore, the more active action to decrease chemical fertilizer utilization and reduce livestock feeding density is required in the government and farmer sides.

Nutrient composition and in vitro fermentability of corn grain and stover harvested at different periods in Goesan, a mountainous area

  • With South Korea's limited capability of feed production because of its relatively small cultivable area, the country is pushed to depend on foreign feed imports despite the immensely fluctuating price of corn. Hence, intensive efforts to increase the total cultivable area in Korea like extending of farming to mountainous area is being practiced. Corn was planted in Goesan County, a mountainous area in the country. Grain and stover were harvested separately in three harvest periods: early-harvest (Aug 8), mid-harvest (Aug 18), and late-harvest (Aug 28). The nutrient composition such as dry matter (DM), crude protein (CP), crude fat (EE), organic matter (OM), neutral detergent fiber (NDF), acid detergent fiber (ADF), and non-fibrous carbohydrates (NFC) was determined after harvest. Effective degradability (ED) of the major nutrients (DM, NDF, ADF, and CP) were measured through in vitro fermentation of rumen fluid from Hanwoo (Korean cattle). pH, ammonia-N concentration, volatile fatty acid (VFA) concentration, and gas production were periodically measured at 0, 3, 6, 12, 24, 48, and 72 h. Corn grain showed higher nutrient content and ED than stover. It also had higher gas production but its pH, ammonia-N, and total VFA concentration were lower than corn stover. The best nutrient composition of corn grain was observed in early-harvest (high CP, EE, NDF, OM, NFC, and low ADF). Early-harvest of corn grain also had high effective degradability of dry matter (EDDM), effective degradability of neutral detergent fiber (EDNDF), effective degradability of acid detergent fiber (EDADF), and total VFA concentration. On the other hand, the best nutrient composition of stover was observed in mid-harvest (high DM, CP, NDF, and low ADF). EDDM, EDNDF, and EDADF were pronounced in early-harvest and mid-harvest of stover but the latter showed high total VFA concentration. Hence, early and mid-harvested corn stover and grain in a mountainous area preserved their nutrients, which led to the effective degradation of major nutrients and high VFA production.

Regional Differences of Proteins Expressing in Adipose Depots Isolated from Cows, Steers and Bulls as Identified by a Proteomic Approach

  • Adipose tissue in the loin muscle area of beef cattle as a marbling factor is directly associated with beef quality. To elucidate whether properties of proteins involved in depot specific adipose tissue were sex-dependent, we analyzed protein expression of intramuscular adipose tissue (IMAT) and omental adipose tissue (OMAT) from Hanwoo cows, steers, and bulls of Korean native beef cattle by liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteomic analysis, quantitative polymerase chain reaction (PCR) and western blot analysis. Two different adipose depots (i.e. intramuscular and omental) were collected from cows (n = 7), steers (n = 7), or bulls (n = 7). LC-MS/MS revealed a total of 55 and 35 proteins in IMAT and OMAT, respectively. Of the 55 proteins identified, 44, 40, and 42 proteins were confirmed to be differentially expressed in IMAT of cows, steers, and bulls, respectively. In OMAT of cows, steers, and bulls, 33, 33, and 22 were confirmed to be differentially expressed, respectively. Tropomyosin (TPM) 1, TPM 2, and TPM3 were subjected to verification by quantitative PCR and western blot analysis in IMAT and OMAT of Hanwoo cows, steers, and bulls as key factors closely associated with muscle development. Both mRNA levels and protein levels of TPM1, TPM2, and TPM3 in IMAT were lower in bulls compared to in cows or steers suggesting that they were positively correlated with marbling score and quality grade. Our results may aid the regulation of marbling development and improvement of meat quality grades in beef cattle.

A study on the needs to improve Korea abattoir's critical control point of HACCP system

  • These days abattoirs' hygiene of Korea is regulated by Hazzard Analysis Critical Control Point (HACCP). Although 20 years have been left since first HACCP was adjusted in Korea, 12% of abattoirs got inconsistence on evaluations. Food poisoning caused by bacteria feces like pathogenic Escherichia coli and Salmonella has not decreased. These bacteria on meat cross-contaminate at the abattoir. Therefore, field verification of abattoir's critical control point (CCP) and experiments to find alternative ways of the CCP were conducted. The aerobic bacteria were measured before and after high-pressure water based washing process set as CCP in most abattoirs. Four parts of cattle carcasses were selected to apply sponge-swab method. The effects were < 1 log reduction which is not significant. Lactic acid (LC), chlorine dioxide (ClO2) and slightly acidic electrolyzed water (SAEW) were used to measure the effect of reducing bacteria on beef by the different time. LC has 1.24-2.02 log reduction for aerobic bacteria. ClO2 has 1.44-1.96 log reduction for aerobic bacteria. SAEW has 1.1-1.91 log reduction for aerobic bacteria. There was significant difference according to concentrations (p < 0.05). This study presents legitimacy for hygiene improvement of CCP by field verification. In addition, chemical disinfectants that can be mechanically applied have better reduction effects of high-pressure washing.
