Chitinase from an Antarctic Bacterium, Sanguibacter sp. KCTC10714

  • A chitinase-producing bacterium strain KCTC10714 was isolated from sea sand around the King Sejong Station, King George Island in Antarctica. It was identified as Sanguibacter sp., based on the biochemical properties and 16S rRNA gene sequence. KCTC10714 chitinase showed enzyme activity in broad range of temperature from 0 to $70^{\circ}C$. At $0^{\circ}C$, it showed 70.9% of relative activity in comparison with 100%. The chitinase gene of KCTC10714 was cloned using inverse PCR cloning method. KCTC10714 chitinase gene was designated as chi21702. The ORF of chi21702 consisted of 1,449 bp (482 amino acid), and contained ChtBD3 (a chitin/cellulose binding domain) and an active site for chitinase family 18.

Physico-chemical Characteristics of Soil in the Vicinity of King Sejong Station, King George Island, Antarctica (남극 킹조지섬 세종기지 주변지역 토양의 물리화학적 특성)

  • In order to collect basic data of soil environment in the Vicinity of King Sejong Station, King George Island, Antarctica, the physico-chemical characteristics of soils were investigated. Soil samples were collected in Barton Peninsula from 13 sites according to direction from the Sejong Cape. Soils from 13 sites were divided into three groups. The sand percentage of soils were much higher as above 90% than silt and clay percentages of soils at the all sites. Soil texture was classified sandy (10 sites) and loamy sand (3 sites). In distribution characteristics at different soil particles according to direction, large particles ($>500{\mu}m$) were higher in the order of Group 1 (Marian cove coast regions) > Group 2 (Inland regions) > Group 3 (Maxwell bay coast regions). On the other hand, small particles ($<355{\mu}m$) were higher in the order of Group 3 > Group 2 > Group 1. Chemical characteristic of soils showed significant differences at different areas. pH ranged 4.5-6.7, showing it was slightly acid and EC ranged $0.06-0.16dS\;m^{-1}$. T-N, OM and T-C contents were high at #6, #8, #12 and #13 sites. T-P and P2O5 contents were high at #9 and #12 sites. The results of this study will be helpful to understand soil environment in the Antarctic Peninsula and surrounding islands.

  • Antarctic King Sejong Station was established in King George Island, the South Pole in 1988, and has been executing the monitoring studies on the change of antarctic natural environment. As an available power, the wind energy generator has been used in the form of hybrid with mainly diesel generator. Because the wind generation power sharply changes by wind energy, it must be careful during the system operation. When the power system becomes stable, the output performance of wind energy generator becomes stable. But, in case of unstable system, the errors frequently occur on the wind energy generator and it badly impacts the power system by output of wind energy generator. The purpose of this paper is to analyze suitability while operating the system of 10kW wind energy generator at Antarctic King Sejong Station, an isolated area, and to analyze the problem and improvements by power quality.

  • Wind/diesel hybrid operation can be one of the most effective option for electrical power production at a remote area such as Antarctica. The king Sejong station at Antarctica relies its power production on diesel engines and diesel oil is supplied every other year by ships. However, the oil transportation processes are liable to potential oil spillage caused by the floating ice around the King George island. The long-term storage of the oil at the station can also contaminate the surrounding soils. A l0kW wind turbine has been installed to save oil consumption and operated in connection with the diesel generators since 2006. The diesel engine that operated poorly during the first year of installation was replaced in 2008 to enhance power production an recent measurements indicate that both diesel power quality and the wind turbine availability have been dramatically improved by the replacement. This report discusses electrical power qualities of wind/diesel hybrid system operating at an isolated micro gird located in the king Sejong station. Our experience reveals that the similar technologies can be applied to domestic islands, for example, in the south sea.

Lipid and Fatty Acid Composition of the Antarctic Krill Euphausia superba

  • Total lipid content, lipid class and fatty acid composition of the Antarctic krill Euphausia superba collected from the water of King George Island, Antarctica during austral summer of 1997-1998, were investigated. The overall lipid content of 5. superba was 72 mg/g dry mass similar to the reported values for most temperate species. The neutral lipid of E. superba was 29% of the total lipid and that of phospholipid was 71%. The majority of neutral lipid was triacylglycerols(31.6% of neutral lipids) while phosphatidyl choline (44% of phospholipid) was the most abundunt in phospholipids. The quantitative composition of the fatty acid in E. superba show consisting mostly of the saturated 16:0 (16.6% -22.1%), along with the polyunsaturated 20:5(n-3) (20.6% -22.1%) and 22:6(n-3) (14.9% -16.9%) acids.

  • 해양으로 유출된 5 mm 이하의 크기로 분해된 미세플라스틱이 해양 환경 오염의 주요 원인으로 자리잡았다. 최근에는 청정해역으로 알려진 남극해에서도 발견되고 있어 남극해에 잔류하는 미세플라스틱 오염 수준을 이해하기 위해 노력하고 있다. 하지만, 파랑의 효과를 고려한 남극해의 해수 순환 구조와 미세플라스틱의 고유 특성을 반영한 미세플라스틱의 거동 및 공간적 분포에 대한 복합적 이해는 상대적으로 부족하다. 남극해에서 발견된 미세플라스틱은 과학기지들의 방류수나, 조사선 등과 같은 인위적인 활동으로 인해 집적될 수 있으며, 특히 영구적으로 거주하는 과학기지에서 흘려보내는 방류수에 포함된 미세플라스틱은 과학기지 주변 해수 오염에 직접적인 영향을 줄 것으로 예상된다. 따라서, 본 연구에서는 파랑 효과에 따른 남극 킹조지 섬(King George Island)에 위치한 세종과학기지의 방류수에 포함된 미세플라스틱의 이송에 대해 모의하였다. 세종과학기지가 위치한 킹조지 섬과 넬슨 섬(Nelson Island) 사이의 멕스웰 만(Maxwell Bay)의 해수 흐름을 재현하기 위하여 해수 유동 모델(Delft3D-FLOW)이 사용되었다. 또한, 해수 유동 모델에 파랑 모델(Delft3D-WAVE)을 결합하여 파랑의 효과가 미세플라스틱의 이송에 미치는 영향을 확인하였다. 세종과학기지의 방류수가 흘러나가는 마리안 소만(Marian Cove)의 유속장을 바탕으로 이송, 확산, 입자의 침강 속도를 고려하여서, 세종과학기지에서 밀물 시 방출한 입자를 라그랑지안 입자 추적(Lagrangian Particle Tracking) 방법을 이용해 추적하였다. 해수의 밀도보다 가벼운 플라스틱의 경우 해수 표층의 흐름을 따라 소만 내부로 이송되어 해안선에 도달하고, 해수의 밀도보다 무거운 플라스틱의 경우 소만 내부로 이송되나 입자의 침강 속도로 인해 방출 위치 근처에서 집적된다. 파랑의 효과를 고려하게 되면, 고려하기 전보다 두 종류의 미세플라스틱 모두 소만 내부로 더 멀리 이송되는데, 이는 파랑으로 인한 힘(wave-induced force)이 해수 유동 모델의 운동방정식에 추가되며 파랑 에너지 분산으로 인해 해수 흐름에 변화를 준 것으로 보인다.

  • To determine subglacial topography and internal features of the Fourcade Glacier on King George Island in Antarctica, helicopter-borne and ground-towed ground-penetrating radar (GPR) data were recorded along four profiles in November 2006. Signature deconvolution, f-k migration velocity analysis, and finite-difference depth migration applied to the mixed-phase, single-channel, ground-towed data, were effective in increasing vertical resolution, obtaining the velocity function, and yielding clear depth images, respectively. For the helicopter-borne GPR, migration velocities were obtained as root-mean-squared velocities in a two-layer model of air and ice. The radar sections show rugged subglacial topography, englacial sliding surfaces, and localised scattering noise. The maximum depth to the basement is over 79m in the subglacial valley adjacent to the south-eastern slope of the divide ridge between Fourcade and Moczydlowski Glaciers. In the ground-towed profile, we interpret a complicated conduit above possible basal water and other isolated cavities, which are a few metres wide. Near the terminus, the GPR profiles image sliding surfaces, fractures, and faults that will contribute to the tidewater calving mechanism forming icebergs in Potter Cove.

Seasonal Variation of Microalgae in the Surface Water of Marian Cove, King George Island, the Antarctic 1998/1999 (1998/1999 남극 킹조지섬 마리안소만 표층수에 서식하는 미세조류의 계절적 변동)

  • We investigated seasonal variation of microalgal assemblages, sea water temperature, salinity and suspended solid and the parameters measured daily from January 1998 to October 1999 at a nearshore shallow-water in Marian Cove, Maxwell Bay, King George Island, the Antarctic. Annual mean surface water temperature was -0.3$0^{\circ}C$ and the highest water temperature was 4.53$^{\circ}C$ (22 January 1999) and the lowest water temperature was -2.07$^{\circ}C$ (23 August 1998). Annual mean salinity was 33.38 psu, ranging from 42.80 psu (6 January 1999) to 19.50 psu (6 June 1999). Annual mean suspended solid (SS) during two years was 34.14 mgㆍ1$^{-1}$, ranging from 60.62 mgㆍ1$^{-1}$(7 March 1998) to 12.90 mgㆍ1$^{-1}$ (26 December 1998). Chlorophyll $\alpha$ (Chl $\alpha$) concentrations were measured in order to know seasonal variations of microalgae in the surface seawater. Annual mean of total Chl a concentration was 0.55$\mu\textrm{g}$ㆍ1$^{-1}$, the highest Chl $\alpha$ concentration (12.16$\mu\textrm{g}$ㆍ1$^{-1}$) appeared in 4 October 1998, the lowest Chl $\alpha$ concentration appeared 0.19$\mu\textrm{g}$ㆍ1$^{-1}$, Monthly mean total Chl $\alpha$ concentration was high in October 1998 (1.32$\mu\textrm{g}$ㆍ1$^{-1}$) and low in July on 1998 (0.28$\mu\textrm{g}$ㆍ1$^{-1}$). Annual mean nano-sized Chl $\alpha$ concentration was 0.40$\mu\textrm{g}$ㆍ1$^{-1}$, monthly mean nano -sized Chl $\alpha$ concentration was high in November 1998 (0.90$\mu\textrm{g}$ㆍ1$^{-1}$), and low in July 1999 (0.22$\mu\textrm{g}$ㆍ1$^{-1}$). Annual mean micro-sized Chl $\alpha$ concentration was 0.15$\mu\textrm{g}$ㆍ1$^{-1}$ monthly mean micro-sized Chl $\alpha$ concentration was high in October 1998 (0.81$\mu\textrm{g}$ㆍ1$^{-1}$), and low July 1998, January, February and September 1999 (0.05$\mu\textrm{g}$ㆍ1$^{-1}$). More than 65% of total Chl $\alpha$ was concentrated during spring and summer time between October and March. Microalgal variation appeared to be due to physical factors of seawater in the Antarctic nearshore from 1998 to 1999. The reason why micro-sized Chl $\alpha$ did not increase during austral summer was the bay had been frozen by decrease of water temperature. We think that total microalgal abundance was decreased because the summer microalgal abundance was determined by variation of water temperature during winter season. The Community Structure of Meiofauna in Marian Cove, King George Island, Antarctica (남극 킹조지섬 마리안소만의 중형저서동물 군집구조)

  • The temporal dynamics of the meiofauna community in Marian Cove, King George Island, Antarctica were observed from March 7 to December 21 2007. Nine meiofauna taxa were found, with nematodes the most dominant group, making up 92.97% of the total meiofauna density, followed by harpacticoid copepods (3.18%). Meiofauna abundance ranged from 123 to 874 individuals per 10 $cm^2$ (mean 464 inds.10 $cm^{-2}$), which is lower than that found in some polar and temperate regions. The lowest meiofauna abundance was found in the 26th April sample (III) and the highest meiofauna abundance was found in the March 23rd sample (II). There was no correlation between meiofauna abundance and season. The seasonal changes were likely caused by meltwater runoff, and there were the physical disturbances on the bottom sediment by huge iceberg. Biomass of meiofauna varied between 20.36 and 101.02 ${\mu}gC{\cdot}10\;cm^{-2}$, and overall mean biomass was 54.17 ${\mu}gC{\cdot}10\;cm^{-2}$ during the study periods. More than 80% of meiofauna was concentrated in the upper 2 cm of the sediment, and density decreased with depth. The mean diversity index was 0.37, and the ratio between the abundance of nematodes: and harpacticoids (N/C) ratio ranged from 7.31 to 95.04 (mean 26.39). NMDS analysis divided the community into three groups: A (III, IV, V, VII, VIII), B (II, IX, XI, XII) and C (I, V, X). The results of ANOSIM and SIMPER analysis revealed significant differences in community structure among three groups and major contributed meiofauna taxon in grouping were nematodes and copepods. No significant correlations were observed between major meiofauna taxon and environmental factors. Thirteen species in 12 genera representing nine families of harpacicoids were recorded. Ancorabolidae was the most diverse family, and Heteropsyllidae was the most abundant. The correlation analysis between benthic harpacticoid copepods and environmental factors showed that some species were affected by water temperature, sediment temperature, salinity, chlorophyll a concentration, grain size of the sediments and heavy metal contents of the sediments. These data describe the usefulness of benthic harpacticoid copepods as biological indicator species in Antarctic regions.

Distribution of Nutrients and Phytoplankton Biomass in the Area Around the South Shetland Islands, Antarctica (남극 남쉐틀랜드군도 주변 해역의 영양염과 식물플랑크톤 생물량 분포)

  • Temperature, salinity, nutrients, chlorophyll-a, and primary production were measured within the upper 200 m water column in the area around the South Shetland Islands in January, 2000. Surface temperature was relatively high in the Drake Passage north of the South Shetland Islands and low in the northeastern area of the Antarctic Peninsula. In contrast, surface salinity was low in the Drake Passage and increased toward the Antarctic Peninsula, reaching the maximum value in the northeastern area of the Antarctic Peninsula. Surface nutrients were low in the Drake Passage and high in the area near the South Shetland Islands. Surface chlorophyll-a was also low in the Drake Passage and near the Antarctic Peninsula and high in the area of the northern King George Island. The study area could be classified as four geographical zones based on the characteristic shape of the T/S diagrams;the Drake Passage, the Bransfield Strait, the mixed zone, and the Weddell Sea. Each geographical zone showed apparently different physical, chemical, and biological characteristics. Phytoplankton biomass was relatively low in the Drake Passage and the Weddell Sea and high in the Bransfield Strait and the mixed zone. The low phytoplankton biomass in the Weddell Sea could be explained by the low water temperature and deep surface mixing down to 200 m. The high grazing pressure and low availability of iron could be responsible for the low phytoplankton biomass in the Drake Passage.
