Objective: To determine the killing effects on extracorporeal HepG2 cells under different temperatures, pressures of permeability and lengths of treatment time. Method: According to different temperatures, pressures of permeability and lengths of treating time, extracorporeal HepG2 cells of human hepatoma cell-line were grouped to 80 groups. Cell index (CI) as the measurement of killing effect were calculated by monotetrazolium (MTT) methods, i.e., CI =1- (the OD value in treated group - the OD value in blank control group) / (mean of untreated control group - mean of blank control group). According to the factorial design, data were fed into SPSS 10.0 and analyzed by three-way ANOVA (analysis of variance). Result: Temperature, pressure of permeability and length of treating time all had effects on the CI (cell index) level. Length of treating time was the most influential factor of the three. Additionally, any two of them all had statistically significant interactive effects on the CI level. When treated for 5-30 min, destilled water at $46^{\circ}C$ stably generated the highest CI. Conclusion: The "$46^{\circ}C$-destilled water-60 min" was considered as the optimal combination of conditions which lead to highest CI. We suggest exerting celiac lavage for 15 min with stilled water at $40^{\circ}C-43^{\circ}C$ in surgical practice as a hyperthermia treatment to achieve ideal killing effects on free cancer cells, which is feasible, practical, and clinically effective.
Effects of stress-induced suppression of humoral immunity on scuticociliate killing activity of olive flounder plasma were investigated. Changes in glucose level. alternative complement activity and Iysozyme activity of plasma by handling stress were analysed in relation to in vitri parasiticidal activity of plasma. The plasma glucose level was about two times higher in fish after a handling stress than in control fish. Plasma lysozyme activity and natural haemolytic activity were decreased in suessed fish.The scuticociliate killing activity of plasma was significantly lower in stressed fish than in non-stressed control fish. The present results indicated that stress-induced immunodepression could be a cause of scuticociliatosis occurrence in olive flounder.
Enrofloxacin-colistin combination, widely used in Gram negative infections in veterinary sector, was investigated in terms of MIC and initial killing rate using E coli k 88ab, Salmonella typhimurium, Pasteurella multocida type A, Bordetella bronchiseptica and Staphylococcus aureus as test organisms. On the basis of MICs of enrofloxacin-colistin combination against the above bacteria, killing rates of the combination of enrofloxacin and colistin at the ratio of 5:0, 4:1, 3:2, 1:1, 2:3, 1:4 and 0:5, indicated high and rapid antibacterial acitivities against all but Staphylococcus aureus R-209, with the number of bacteria reducing to less than one percent within two hours. At the MIC of enrofloxacin or colistin, both antibacterials showed the highest killing rates during 2-4 hours against Gram negatives such as E coli K88ab,Pasteurella multocida type A and Bodetella bronchiseptica but allowed the regrowth of the same pathogens thereafter. On the while, the combination of two antibacterials at a fourth MIC resulted in high killing rate without bacterial regrowth during 24 hours, suggesting the synergistic antivacterial effects. The combination, however, did not show favourable activity against Gram negatime S typhimurium and Gram positive S aureus ergistic antibacterial activity against Gram negatime pathogens but also colistin showed LPS-neutraization, we could suggest the combination should provide clinically positive therapeutic armarium in Gram negative infections.
We have isolated a bacterial strain that tends to kill P. micans from the mixed culture of p. minns plus seawater filtrate (poresize, 0.8 $\mu$m) collected at Masan bay in July 1996, in which the mixed culture grown in the f/2 medium. According to the experimental results of the isolated bacterium such as fatty acids analysis, morphological and biochemical characteristic tests, the strain was supposed to be a Pseudomonas and then it was named as Pseudomonas sp. LG-2. The killing effect of Pseudomonas sp. LG-2 against P. micans was proportionally increased with the concentrations of culture filtrate (pore size, 0.8 $\mu$m) is well as with the number of bacterium inoculated. In the mixed culture inoculated with $1.3\times10^6$ cells/ml of Pseudomonas sp. LG-2, the number of P. micans (2,000 cells/ml) was gradually decreased and then killed below 100 cells/ml within 7 days. In addition, the culture filtrate with $30\%$ of final concentration revealed a significant killing effect against P. micans around 3 days after culture. In the relationship between killing effects and growth stage of Pseudomonas sp. LG-2, the culture filtrate at lag phase has little effects on P. micans. In constant, the culture filtrate at mid-log phase showed the killing effect by decreasing P. micans to 112 in number within 5 days. In particular, the culture filtrate at stationary phase showed a significant killing effect against P. micans in which the majority of it was killed after 3 day culture. The species specificity of killing effects of Pseudomonas sp. LG-2 against 5 species of dinoflagellate was only found in P. micans and Scrippsiella trochoidea.

Protective roles of antioxidant enzymes, copper-zinc superoxide dismutase (CuZnSOD), manganese superoxide dismutase (MnSOD), and catalase of Candida albicans against exogenous reactive oxygens and oxidative killing by macrophages were investigated. The initial growth of C. albicans was inhibited by reactive, oxygen-producing chemicals such as hydrogen peroxide, pyrogallol, and paraquat, but it was restored as the production of antioxidant enzymes were increased. The growth inhibition of C. albicans by reactive, oxygen-producing chemicals was reduced by treating the purified candidal SOD and catalase. Also, in the presence of SOD and catalase, the oxidative killing of C. albicans by macrophages was significantly inhibited. These results suggest that antioxidant enzymes, CuZnSOD, MnSOD, and catalase of C. albicans may play important roles in the protection of C. albicans not only from exogenous oxidative stress but also from oxidative killing by macrophages.
Objectives: Recently Ciprofloxacin, used in the treatment of mastitis, showed many serious side effects. The object of this study was to recognize whether TNS and KWT can be used in the treatment of mastitis by observing the in vitro antibacterial effects of TNS and KWT aqueous herbal extracts against S. aureus. Methods: Antibacterial activities of TNS and KWT aqueous extracts against S. aureus ATCC 25923 were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were monitored at MIC and $MIC{\times}2$ levels. The effects on the intracellular killing and bacterial invasion of individual test materials were also observed using Raw 264.6 and MCF-7. The results were compared with Ciprofloxacin, a second generation of quinolone antibiotics in the present study. Results: MIC of aqueous extracts of TNS and KWT against S. aureus were detected as ($0.313{\pm}0.107$) and ($0.137{\pm}0.053$) mg/ml, respectively. MIC of Ciprofloxacin was detected as ($0.469{\pm}0.297$) ${\mu}g/ml$ at same conditions. In addition, TNS, KWT aqueous herbal extracts and Ciprofloxacin were also showed marked dosage-dependent inhibition of bacterial growth, and dramatical inhibitions on the both intracellular killing assays and bacterial invasion using Raw 264.6 and MCF-7 cells were detected. Conclusions: The results obtained in this study suggest that TNS and KWT aqueous herbal extracts showed antibacterial effects against S. aureus, and they also showed dosage-dependent inhibitory effects on the bacterial growth. And they showed the significant intracellular killing and bacterial invasion effects. It means, KWT and TNS may show more potent anti-infectious effects against S. aureus in vivo.
This experiment was performed to investigate effects of zinc containing solution on the major normal flora Staphylococccus aureus, Streptococus mutans and Candida albicans and to observe the variation according to anionic change and concentration difference. Zinc chloride, zinc iodide and zinc acetate solution were added to werially diluted broth culture so that each final concentration might be 0.25%, 0.5%. 1%. After that, 100ul of each aliquot was spreaded on each selective media plate( Mannitol Salts Agar plate for Staphylococcus aureus, Mitis Salivarius Agar plate for Streptococcus mutans and Sabouraud Destrose Agar plate for Candida albicans). The % killing was calculated bu CFU count after incubation under the appropriate condition. 1. zinc iodide, zinc chloride, and zinc acetate solutions showed inhibitory effects on Staphylococcus aureus, Streptococcus mutans and Candida albicans. 2. The inhibitory effects on Staphylococcus aureus were ranked in order of ainc iodide, zinc chloride and zinc actate. 3. The inhibitory effects on Streptococcus mutans were ranked in orfer of zinc iodide, zinc chloride and zinc acetate. 4. the inhibitory effects on Candida albicans showed no difference among zinc iodide, zinc chloride and zinc acetate. 5. The inhibitory effects of zinc chloride and zinc acetate on Staphylococcus aureus and Streptococcus mutnas showed increasing pattern as the concentration increase. But the inhibitory effects of zinc iodide on Staphylococcus aureus and Streptococcus mutans showed no apparent difference according to concentrations and it was the case with the inhibitory effects of zinc iodide, zinc chloride and zinc acetate on Candida albicans.
The in vitro effects of trisodium phosphate and cetylpyridinium chloride on E. coli 0157:H7 and L. monocytogenes were investigated. The trisodium phosphate and cetylpyridinium chloride was bactericidal toward E. coli 0157:H7 and L. monocytogenes. The killing effects of the $1{\times}10^{-2}\;M$ trisodium phosphate on E. coli 0157:H7 and L. monocytogenes were 30~40%, 40~50%, respectively. The killing effects of the $5{\times}10^{-7}\;M$ cetylpyridinium chloride on E. coli 0157:H7 and L. monocytogenes were 90~95%, 95~99%, respectively. The killing effects of the trisodium phosphate was $10^5$ times that of the cetylpyridinium chloride. Factors effecting the bactericidal action of trisodium phosphate and cetylpyridinium chloride were investigated and the action depended on temperature and pH.
The present experiment was perf'3rmed to investigate the protective effects of ginseng total saponin (GTS) and possible mechanisms on the hepatocytotoxicity induced by tert-butylhydroperoxide (t-BuOOH), 4-Bromo-calciumu ionophore A23187 (Br-A23187) and KCN. Hepatocytes were isolated by collagenase perfusion of livers from fasted male Sprague Dawley rats and cultured overnight. After various treatments in Krebs-Ringer-HEPES buffer at pH 7.4, cell viability was determined by propidium iodide using fluorocytometry. GTS (5-20 ${\mu}$M) inhibited cell killing induced by t-BuOOH, and KCN, dose-dependently. However, GTS did not inhibit Br-A23187-induced cell killing. These findings support that GTS could protect the hepatocytoxicity induced by some toxic chemicals. The mechanisms of these protective effects by GTS seem to be associated with antioxidant activity and increase of cellular ATP.
Many scolicidal agents have been used to destroy fertile protoscolices, but these scolicidal agents have side effects, highlighting the need for research on effective and non-toxic replacement scolicidal agents. Gold nanoparticles (AuNPs) are biocompatible and non-toxic. The current study examined the effects of AuNPs in killing the protoscolices of Echinococcus granulosus in vitro using eosin staining. The protoscolices were treated with 0.2, 0.4, 0.8, or 1.0 mg/mL of AuNPs for 15, 30, 45, or 60 minutes. A concentration of 1.0 mg/mL was the most efficient in killing the protoscolices after 60 minutes exposure, reaching 96%, followed by 0.8 mg/mL (84.5%), whereas 0.4 and 0.2 mg/mL of AuNPs achieved a death rate of 76.8% and 68.5%, respectively. The loss of the protoscolices was lower at shorter exposure times with the same concentration of AuNPs and increased as the AuNP concentration was increased at the same exposure time. Significant differences were found between the different groups compared to the control group.
