1-(4-methylpiperazinyl)-3-phenylisoquinoline (CWJ- a-5)의 Sprague-Dawley(SD) 랫드를 이용한 2주간 반복 경구투여 독성시험 (Two -week Oral Toxicity Study of 1- (4-methylpiperazinyl) -3- phenylisoquinoline (CWJ-a-5) in sprague-Dawley (SD) Rats)

  • The subacute oral toxicity of 1-(4-methylpiperazinyl)-3-phenylisoquinoline (CWJ- a-5) was investigated in Sprague-Dawley (SD) rats. Five groups of 5 males and 5 females were orally administered at doses of 0, 37.5, 75, 150 and 200 mg/kg with CWJ-a-5 for 2 weeks. In clinical signs, Salivation was observed in the 75, 150 and 500 mg/kg male and female groups. Loss of fur was observed in the 500 mg/kg male and female group. Body weight were significantly decreased in the 150 and 500 mg/kg male groups and in the 500 mg/kg female group. Food consumption was significantly decreased in the 300 mg/kg male group. In serum biochemistry, total cholesterol and phospholipid were significantly increased in 500 mg/kg male and female group. Aspartate aminotransferase was significantly increased in the 500 mg/kg female group. In histopathological examination, vacuolar degeneration of renal tubules in the kidney, vacuolar degeneration of hepatocytes in the liver vacuolar degeneration of myocytes in the heart, vacuolar degeneration of histiocytes in the spleen and thymus, atrophy of seminiferous tubule and degeneration of germinal epithelium in the testis, vacuolar degeneration of corpus luteum, granulosa cell and theca cell in the ovary were observed in the 150 and 500 mg/kg male and female groups. Based on these results, the no observed adverse effect level (NOAEL) with CWJ-a-5 was considered to be 75 mg/kg and the absolute toxic dose was considered to be 150 mg/kg in this study

Effect of Graded Levels of Wheat Bran Supplementation on Intake, Nutrient Digestibility, Microbial N Yield and Growth Rate of Native Bulls Fed Rice Straw Alone

  • The dose response effects of different levels of wheat barn (WB) supplementation to a rice straw based diet has been studied in growing native (Bos indicus) bulls of Bangladesh. Twelve bulls of $266{\pm}29.6kg$ live weight and $32{\pm}9.5$ months old were given either of three diets of ad libitum untreated rice straw alone (T1) or straw supplemented with 2 (T2) or 3 (T3) kg WB. Besides, the experimental animals also received a mineral mixture. In 4 weeks, data were recorded on the intake, digestibility, microbial N (MN) yield, N balance (NB) and growth rate (GR). In the three diets, WB was 0 (T1), 29.6 (T2) and 42 (T3) percent of the total DM intake. With the increasing levels of WB supplementation, the total DM and OM intake increased, but the straw intake decreased linearly. WB supplementation increased the digestibilities of DM, OM and N but had no effect on ADF digestibility. The urinary purine derivative excretion and hence the MN yield increased with WB supplementation. The total MN yield were 7.26 and 35 g/d respectively for 0.2 and 3 kg WB supplementation. However, the efficiency of MN yield was highest (13 g/kg DOMR) at 2 kg WB level. Despite the increase DOM intake, the dietary ME content remain below 6 MJ/kg DM even at 3 kg WB supplementation. The NB were -84,467 and 1,620 mg/kg $W^{0.75}/d$ which were reflected on the GR of -186,346 and 554 g/d for 0, 2 and 3 kg WB supplementation respectively. Depending on the cost effectiveness, on an untreated rice straw diet, WB may be supplemented by up to 3 kg/d (42% of the diet) or more. However, if the maximum utilization of roughage is the main concern, the optimum WB level would probably be around 2 kg (30% of the diet) daily.

Effects of Restricted Feeding on Performance, Carcass Quality and Hormone Profiles in Finishing Barrows

  • To investigate the effects of feeding restricted on growth, carcass characteristics and plasma profiles in an attempt for optimum responses, a total of 108 cross-bred finishing barrows [(Landrace${\times}$Yorkshire)${\times}$Duroc]weighing an average of $46.88{\pm}0.52kg$ were assigned in a randomized complete block (RCB) design to one of four treatments with three replicates and nine pigs per pen. Feeding regimens were, 1) ad libitum from 50 kg to market weight (Ad 3/3), 2) restricted feeding from 90 kg to market weight (Ad 2/3), 3) restricted feeding from 70 kg to market weight (Ad 1/3), and 4) restricted feeding from 50 kg to market weight (Ad 0/3). During the experimental period, average daily feed intake (ADFI) was decreased from 2.53 kg (AD 3/3) to 2.09 kg (AD 0/3) with increasing restricted feeding duration of (p<0.05). Average daily gain (ADG) of AD 3/3 (0.79 kg) was significantly higher (p<0.05) than those of AD 1/3 (0.74 kg) or AD 0/3 (0.72 kg). Feed efficiency was not influenced by restriction regimens. Blood IGF-I concentrations were increased from 74.14 to 134.25 (167.36-115.66) ng/ml as body weight increased. Blood leptin concentrations were affected by feed intake level and coincided with blood IGF-I concentrations. Most of carcass characteristics were not significantly affected by restricted feeding, however cooking losses in AD 1/3 and Ad 0/3 treatment diet were higher than those in Ad 3/3 and Ad 2/3. In addition, there was a trend that backfat thickness was lowered in proportional to decreasing feed intake (p>0.05). In conclusion, restricted feeding improved feed efficiency after 50 kg body weight without deteriorating the pork quality of barrows.

음식물 및 농업폐기물 열분해장치 개발 (Development of A Torrefaction Unit of High Moisture Food & Agricultural Wastes)

  • 2012년 국내 총 폐기물(지정폐기물 제외) 발생량은 382,009 톤/일 으로 이 중 12.8%를 차지하는 생활폐기물 중 음식물 채소류폐기물 발생량은 13,209 톤/일 으로 대부분 소각 후 매립 처리되고 있다. 본 연구에서는 고수분 음식물 및 농업 폐기물을 재활용한 고형연료 제조에 필요한 열분해장치를 개발하고 실험을 통해 그 성능을 확인하고자 하였다. 본 연구를 위해 건조용량 50 kg/hr인 실험실용 열분해장치를 제작하였다. 건조 처리된 농업폐기물과 음식물 쓰레기를 열분해용 실험 원료로 사용하였다. 원료종류, 열분해 온도, 열분해 시간에 따른 농업폐기물과 음식물 쓰레기의 열분해 특성을 파악하였다. 농업부산물 건조물의 열분해 처리 결과, 열분해 처리능력은 평균 55.35 kg/hr, 저위발열량은 평균 3,333 kcal/kg으로 측정되었다. 농업부산물을 단순 건조 처리한 경우 고위발열량은 3,400 kcal/kg, 저위발열량은 3,090 kcal/kg으로 측정되어 열분해처리로 발열량이 향상됨을 알 수 있다. 음식물 쓰레기 건조물의 열분해 처리조건 및 결과, 열분해 처리능력은 평균 88.27 kg/hr, 저위발열량은 평균 4,016 kcal/kg으로 측정되었다. 음식물 쓰레기를 단순 건조 처리한 경우 고위발열량은 4,040 kcal/kg, 저위발열량은 3,686 kcal/kg으로 측정되어 열분해처리로 발열량이 향상됨을 알 수 있다. 열분해 처리능력은 연구목표치인 50 kg/hr보다 높게 나타났으며, 저위발열량은 연구목표치인 4,000 kcal/kg 보다 다소 높게 나타났다. 다만 저위발열량 측정 기준 함수율이 습량기준으로 약 10%로 추정되는 바 5%로 조절하고, 열분해 열풍온도를 $200^{\circ}C$ 까지 상승시키면 발열량이 훨씬 향상될 것으로 판단된다.

Nonylphenol과 Bisphenol A의 지렁이 독성시험 및 토양 중 생태 위해성평가 (Risk Assessment of Soil through Earthworm Toxicity Test of Nonylphenol and Bisphenol A)

  • Earthworm (Eisenia fetida) acute toxicity test was carried out and ecological risk assessment in soil was performed with national monitoring data. 14 day - $LC_{50}$ of nonylphenol and bisphenol A were 288.1 mg/kg and 90.1 mg/kg, respectively. And NOECs of nonylphenol and bisphenol A were 250 mg/kg and 50 mg/kg, respectively. Significant weight decrement was appeared at 70 mg/kg of bisphenol A, however, nonylphenol at concentrations tested did not severe adverse effect on the weight decrement. The environmental monitoring has been carrying out by NIER since 1999. Exposure levels of nonylphenol in soil were ND$\sim$10.55 $\mu$g/kg and those of bisphenol A were ND$\sim$15.50$\mu$g/kg in National Monitoring data which had been performed from 2000 to 2004. The measured soil exposure level was applied to evaluate the environmental risk assessment. The values of PNEC for bisphenol A and nonylphenol were determined as 0.5 mg/kg and 2.5 mg/kg, respectively using the safety factors which were suggested in EU and OECD. The values of HQ (PEC/PNEC) were determined to be below I for bisphenol A and nonylphenol when the maximum exposure levels for bispheol A (15.50$\mu$g/kg) and nonylphenol (10.55$\mu$g/kg) were applied. Conclusively, the environmental risk assessment of bisphenol A and nonylphenol was not critical in soil.

마우스에서 계면활성제 LAS-Na와 MES의 급성 경구독성 (Acute Toxicity of Surfactants LAS-Na and MES in Mice)

  • ICR 마우스를 이용하여 계면 활성제인 LAS-Na 와 MES의 급성 경구독성을 실시하였다. 6주령된 마우스 LAS-Na는 0, 1,000, 1,320, 1,780, 2,280, 3,000 mg/kg의 용량으로 MES는 0, 1,000, 1,560, 2,450, 3,830, 6,000mg/kg의 용량으로 1회 경구 투여한 후 14일간 관찰하였다. 두 물질 모두 관찰 기간동안 대조군에 비하여 유의성 있는 체중변화는 관찰되지 않았다. 시험물질과 관련된 주요 임상증상으로는 설사, 활동성 감소, 입모 등이 관찰되었다. 사체에 대한 부검결과 시험물질에 의한 것으로 판단되는 소장 점막의 충혈이 소수례 관찰되었으며, 생존 동물에서는 특이한 만한 이상소견은 발견되지 않았다. 이상의 결과로부터 $LD_{50}$값은 LAS-Na의 경우 수컷과 암컷에서 각각 1,319 mg/kg, 1,402 mg/kg이었으며, MES의 경우 수컷에서는 2,040 mg/kg, 암컷에서는 2,546 mg/kg으로 산출되었다.

비음과 질소시비가 Orchargrass의 건물생산에 미치는 영향 (the Effects of Shade and N Fertilization on the Dry Matter Production of Orchargrass)

  • The purpose of this experiment was to evaluate the effects of shade and levels of N fertilization on the dry matter yield and chemical compositions of orchardgrass grown under floor of chestnut tree. Shading conditions consist of $S_0$, (full light), $S_1$, (about 6OC4 shade) and $S_2$, (about 70% shade). And, nitrogen fertilizer was applied at 3 levels, O($N_0$), 12($N_1$), and 30($N_2$) kg per 10a, respectively. The results are may be summarized as follows: 1. Maximum total dry matter yield of $S_0$, was obtained about 1.28 ton/lOa at $N_2$, level. But, total dry matter yields of N levels in $S_1$, and $S_2$, were decreased about 42-45% compared with $S_0$. 2. The response of the dry matter yield to N fertilization were differences between shading and levels of N. Thus, the dry matter yield of $S_1$, increased almost linear up to about 30 kg/l0a level, while the dry matter yield of S, was increased slightly up to 30 kgIl0a. But. $S_2$, was increased up to 12 kg/lOa and then decreased slightly with N fertilization over the 12 kg/l0a. 3. Average increase in total dry matter yield to N fertilization were 23.85 kg, 7.97 kg and 5.08 kg DM for $S_0$, $S_1$, and $S_2$, respectively. 4. The level of 12 kg N/lOa is the limiting N level to obtain dry matter production under 60-709 shading conditions. 5. The contents of crude protein arid nitrate nitrogen were increased with shading and incremental N fertilization up to 30 kg/l0a. But, water soluble carbohydrate content was decreased greatly with high shading and high levels of N. 6. Nitrate nitrogen content indicated highly significant positive correlation with crude protein, but significant negative correlation with water soluble carbohydrate content. 7. At 30 kg N level with $S_1$, was necessary to exceed the potentially toxic nitrate nitrogen level of 0.20%.

Energy Requirements for Maintenance and Growth of Male Saanen Goat Kids

  • The aim of study was to determine the energy requirements for maintenance and growth of forty-one Saanen, intact male kids with initial body weight (BW) of $5.12{\pm}0.19$ kg. The baseline (BL) group consisted of eight kids averaging $5.46{\pm}0.18$ kg BW. An intermediate group consisted of six kids, fed for ad libitum intake, that were slaughtered when they reached an average BW of $12.9{\pm}0.29$ kg. The remaining kids (n = 27) were randomly allocated into nine slaughter groups (blocks) of three animals distributed among three amounts of dry matter intake (DMI; ad libitum and restricted to 70% or 40% of ad libitum intake). Animals in a group were slaughtered when the ad libitum-treatment kid in the group reached 20 kg BW. In a digestibility trial, 21 kids (same animals of the comparative slaughter) were housed in metabolic cages and used in a completely randomized design to evaluate the energetic value of the diet at different feed intake levels. The net energy for maintenance ($NE_m$) was $417kJ/kg^{0.75}$ of empty BW (EBW)/d, while the metabolizable energy for maintenance ($ME_m$) was $657kJ/kg^{0.75}$ of EBW/d. The efficiency of ME use for NE maintenance ($k_m$) was 0.64. Body fat content varied from 59.91 to 92.02 g/kg of EBW while body energy content varied from 6.37 to 7.76 MJ/kg of EBW, respectively, for 5 and 20 kg of EBW. The net energy for growth ($NE_g$) ranged from 7.4 to 9.0 MJ/kg of empty weight gain by day at 5 and 20 kg BW, respectively. This study indicated that the energy requirements in goats were lower than previously published requirements for growing dairy goats.

Amygdalin의 경구투여독성에 미치는 Cysteine과 Streptomycin의 영향 (Effects of Cysteine and Streptomycin on the Toxicity of Amygdalin in Rats orally Administration)

  • Amygdalin has been used for a long time as an anticancer agent. But because of its toxicity, it is difficult to administer continuously for treatment of cancer. This paper was attempted to reduce the side effect and toxicity of amygdalin. That is, effects of cysteine and streptomycin on the toxicity of amygdalin were investigated in rats orally administered amygdalin. 1. The group administered only amygdalin 400 mg/kg was effected on the lung and body weight, hematocrit, hemoglobin, clotting time, SGOT and albumin value. That is, lung and body weight, hematocrit hemoglobin and albumin value were significantly decreased. SGOT and clotting time were significantly increased compared with those of normal group. 2. Weight of lung was significantly increased in the C group (administred amygdalin 400 mg/kg and cysteine 300 mg/kg), D group (amygdalin 400 mg/kg and streptomycin 10 mg/kg), E group (amygdalin 400 mg/kg, streptomycin 10 mg/kg and cysteine 200 mg/kg)and F group (amygdalin 400 mg/kg, streptomycin 10 mg/kg and cysteine 300 mg/kg). 3. Values of hematocrit and hemoglobin were significantly increased, and clotting time was significantly decreased, in the I group and F group compared with those of A group. 4. SGOT was significantly decreased in the C group, E group and F group compared with that of A group. 5. The blood cyanide concentration was significantly decreased in the E group and F group compared with that fo A group. 6. In short, coadministration of cysteine and streptomycin are considered to reduce the toxicity of amygdalin in rats orally administered.

STUDY ON THE DEVELOPMENT OF A TECHNIQUE FOR PRESERVING STRAW UNDER WET CONDITION IN BANGLADESH

  • About 7.7 million tons of straw dry matter are being rotten during the monsoon. The objective of this trial was to develop a technique to preserve straw under wet condition. To simulate the moisture content of wet straw, a dry straw was deeped overnight in water. After draining the excess water, the wet straw(668 g moisture $kg^{-1}$) was divided into twenty fractions and preserved with 0, 30, 50 ro 70 g urea $kg^{-1}$ dry matter for either of 30, 60, 90, 120 or 180 days in sealed plastic container. Considering the colour, smell, fungal infestation and pH, the wert straw was preserved excellently up to 180 days when 50 or 70 g of urea per kg DM was used. Urea preservation increased the crude protein contents of straw by 3.6 to 6.4 times (174 to $364g{\cdot}kg^{-1}$) over that of the dry straw ($48g{\cdot}kg^{-1}$). Although the NDF content of straw was not effected by the level of urea or by the length of the preservation period, but the ADF content increased (p > 0.05) by 0.086 to $0.889g{\cdot}kg^{-1}$ straw DM for each g increase in the urea level. At 48 hours, the DM degradability of dry straw was $350g{\cdot}kg^{-1}$, which increased to $633g{\cdot}kg^{-1}$ when preserved with 50 g urea $kg^{-1}$ for 180 days. For the same straw, both the rate(0.0388 vs. 0.0136 fraction $h^{-1}$), the extent(717 vs. $631g{\cdot}kg^{-1}$) of straw degradation and the estimated ME(9.55 vs. $6.51MJ{\cdot}kg^{-1}$ straw DM) were higher in the preserved than the dry straw.
