HBV-X 단백질과 간세포 단백질간의 상호작용

  • 당해년도에는 HBV-X와 결합하는 상대 단백질을 탐색하는 것이 주목적이다. 이 목적으로 1) 우선 생물학적 활성을 보유하는 X-단백질을 E. coil system에서 다량으로 생산하는 공정을 확립하였으며 2) 이 X-단백질을 labelling한 후 Probe로 사용하여 liver cell내에 존재하는 43Kd, 48Kd, 55Kd, 100Kd의 단백질이 HBV-X에 결합하는 것을 확인하였으며 3) liver cell expression library를 screening하여 HBV-X와 결합하는 단백질을 coding하는 유전자를 cloning하여 현재 각 clone들을 규명하고 있는 중이다. 4) 또한 암억제 유전자 산물인 p53과 X-단백질과의 상호작용을 발견하였다. 이러한 결과는 X-단백질이 간암의 발생에 작용하는 기작을 설명할 수 있는 중요한 발견이다.

당근 세포의 단백질 생합성에 대한 몇가지 식물생장조절제의 영향 (Effects of Some Plant Growth Regulators on Protein Biosynthesis of Carrot Cells)

  • 당근에서 cytokinin, auxin 또는 GA에 의해 조절되는 polypeptide 및 단백질들을 동정하기 위하여, kinetin, BA, IAA, NAA 혹은 $GA_3$가 각각 $10^{-4},\;10^{-5},\;10^{-6}M$인 배지에서 suspension culture 한 당근 callus들의 추출물의 전기영동양상을 조사하였다. 생장조절제를 처리하지 않은 callus에서 15개 polypeptide band가 관찰되었는데, 이들의 분자량은 각각 $18._4,\;20._2,\;24._0,\;34._9,\;35._7,\;37._4,\;40._3,\;42._2,\;44._1,\;44._4,\;49._3,\;55._0,\;56._6,\;58._1$ 그리고 $59._9KD$였다. Kinetin, BA, IAA, NAA 그리고 $GA_3$에 의하여 합성이 촉진되는 것으로 보이는 polypeptide band는 각각 2개, 6개, 1개, 2개 그리고 4개였고, 합성이 억제되는 것으로 보이는 것은 각각 5개, 3개, 4개, 3개, 그리고 2개였다. 분자량 $40._3KD$$42._2KD$의 polypeptide들은 cytokinin류 생장조절제에 의하여 합성이 촉진되는 것으로 나타났고, $44._1KD$의 것은 auxin류에 의하여 합성이 촉진되며 $37._4,\;44._4,\;56._6KD$의 것은 auxin류에 의하여 합성이 억제되는 것으로 나다났다. Kinetin, BA, IAA, NAA 및 $GA_3$처리에서 상대 이동도가 0.56, 0.84, 0.92인 단백질 band들이 증가된 것이 관찰되었으나, 각 생장조절제들에 특유한 단백질은 동정할 수 없었다.

Anisakis 감염(感染) 가토(家兎)의 시기별(時期別) 항체검출(抗體檢出)에 대한 각종 항원(抗原)의 적용성(適用性) (Application of Various Antigens on the Detection of Antibody in Rabbits Infected with Anisakid Larvae)

  • Antibody changes in experimental anisakiasis were observed by ELlSA and SDS-PAGE/EITB using various antigens : whole worm extract antigen(WWE), somatic antigen(SOM), excretory-secretory antign(ES), and hemoglobin antigen(HB) of Anisakis Type 1. The results obtained were as follows. l) Serum levels of IgG antibody by ELISA increased from 1st week of infection and reached their maximum titer at 5th week after infection, and decreased gradually thereafter. 2) The best result expressed as positive/negative ratio could be obtained when ES antigen was used. 3) Silver stained SDS-PAGE of each antigen showed at least 20 protein bands : In WWE, 286, 278, 262, 38, 18 Kd bands ; In SOM, 38 Kd band : In ES, 286, 65, 13 Kd bands ; In Hb, 61, 55, 38, 28, 26, 22, 20, 16, 15 Kd bands iepntibied as were major bands. 4) By EITB using WWE, Serum antibody recognized major protein with molecular weight of 86 Kd and 16 Kd. Using ES, 69, 59, 16 Kd bands were observed and using Hb, 28 Kd band was observed as specific band. In conclusion, excretory-secretory antigen(ES) of Anisakis larvae was most usable for ELISA.

Clinical usefulness of serum procalcitonin level in distinguishing between Kawasaki disease and other infections in febrile children

  • Purpose: The aims of this study were to compare serum procalcitonin (PCT) levels between febrile children with Kawasaki disease (KD) and those with bacterial or viral infections, and assess the clinical usefulness of PCT level in predicting KD. Methods: Serum PCT levels were examined in febrile pediatric patients admitted between August 2013 and August 2014. The patients were divided into 3 groups as follows: 49 with KD, 111 with viral infections, and 24 with bacterial infections. Results: The mean PCT level in the KD group was significantly lower than that in the bacterial infection group ($0.82{\pm}1.73ng/mL$ vs. $3.11{\pm}6.10ng/mL$, P=0.002) and insignificantly different from that in the viral infection group ($0.23{\pm}0.34ng/mL$, P=0.457). The mean erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP) level in the KD group were significantly higher than those in the viral and bacterial infection groups (P<0.001 and P<0.001 for ESR, P<0.001 and P=0.005 for CRP, respectively). The proportion of patients in the KD group with PCT levels of >1.0 ng/mL was significantly higher in the nonresponders to the initial intravenous immunoglobulin treatment than in the responders (36% vs. 8%, P=0.01). Conclusion: PCT levels may help to differentiate KD from bacterial infections. A combination of disease markers, including ESR, CRP, and PCT, may be useful for differentiating between KD and viral/bacterial infections.

케톤식이로 발생한 체중감소 및 내당능장애에 침치료 효과 (Effects of Acupuncture Treatment on Weight Loss and Glucose Intolerance Induced by a Ketogenic Diet)

  • Objectives : This preliminary study investigated the effect of acupuncture treatment on short-term ketogenic diet-induced weight loss and glucose intolerance in mice. Methods : Six-week old male C57BL/6J mice were randomly assigned to 3 groups: normal, KD, and KD+ACU. All the mice except normal group were fed with ketogenic diet formula for 7 weeks and mice in KD+ACU group received acupuncture treatment three times a week. Body weights were measured three times a week, and glucose level was measured on week 1,3,5, and 7. Ketone level was measured on week 3,5, and 7. Results : Ketogenic diet showed short-term weight loss effect, however, acupuncture treatment did not affect on the weight loss. Ketone level was increased in KD fed mice compared to normal diet fed mice and the level was decreased in KD+ACU group on week 3. However, the change was not significantly different compared to KD group on week 7. Glucose intolerance was improved in KD+ACU group compared to KD group. Conclusions : Acupuncture treatment was effective in relieving glucose intolerance, and the results suggest that combining acupuncture treatment with ketogenic diet may complement each therapeutic intervention by improving glucose intolerance but not effecting on weight loss. This study provides meaningful evidence as a preliminary study of acupuncture treatment on ketogenic diet.

Detection rate and clinical impact of respiratory viruses in children with Kawasaki disease

  • Purpose: The purpose of this prospective case-control study was to survey the detection rate of respiratory viruses in children with Kawasaki disease (KD) by using multiplex reverse transcriptase-polymerase chain reaction (RT-PCR), and to investigate the clinical implications of the prevalence of respiratory viruses during the acute phase of KD. Methods: RT-PCR assays were carried out to screen for the presence of respiratory syncytial virus A and B, adenovirus, rhinovirus, parainfluenza viruses 1 to 4, influenza virus A and B, metapneumovirus, bocavirus, coronavirus OC43/229E and NL63, and enterovirus in nasopharyngeal secretions of 55 KD patients and 78 control subjects. Results: Virus detection rates in KD patients and control subjects were 32.7% and 30.8%, respectively (P=0.811). However, there was no significant association between the presence of any of the 15 viruses and the incidence of KD. Comparisons between the 18 patients with positive RT-PCR results and the other 37 KD patients revealed no significant differences in terms of clinical findings (including the prevalence of incomplete presentation of the disease) and coronary artery diameter. Conclusion: A positive RT-PCR for currently epidemic respiratory viruses should not be used as an evidence against the diagnosis of KD. These viruses were not associated with the incomplete presentation of KD and coronary artery dilatation.

Clinical implications in laboratory parameter values in acute Kawasaki disease for early diagnosis and proper treatment

  • Purpose: This study aimed to analyse laboratory values according to fever duration, and evaluate the relationship across these values during the acute phase of Kawasaki disease (KD) to aid in the early diagnosis for early-presenting KD and incomplete KD patients. Methods: Clinical and laboratory data of patients with KD (n=615) were evaluated according to duration of fever at presentation, and were compared between patients with and without coronary artery lesions (CALs). For evaluation of the relationships across laboratory indices, patients with a fever duration of 5 days or 6 days were used (n=204). Results: The mean fever duration was $6.6{\pm}2.3days$, and the proportions of patients with CALs was 19.3% (n=114). C-reactive proteins (CRPs) and neutrophil differential values were highest and hemoglobin, albumin, and lymphocyte differential values were lowest in the 6-day group. Patients with CALs had longer total fever duration, higher CRP and neutrophil differential values and lower hemoglobin and albumin values compared to patients without CALs. CRP, albumin, neutrophil differential, and hemoglobin values at the peak inflammation stage of KD showed positive or negative correlations each other. Conclusion: The severity of systemic inflammation in KD was reflected in the laboratory values including CRP, neutrophil differential, albumin, and hemoglobin. Observing changes in these laboratory parameters by repeated examinations prior to the peak of inflammation in acute KD may aid in diagnosis of early-presenting KD patients.

Escheriachia coli pSL 2-1 클론과 Bacillus sphaericus 1593 균주가 생산한 모기치사 단백질 (Mosquitocidal Proteins from Escheriachia coli pSL 2-1 Clone and Bacillus sphaericus 1593)

  • Escheriachia coli pSL 2-1 clone은 Bacillus sphaericus 1593의 모기살충 유전자를 클로닝한 재조합 DNA이다. 이 클론이 생산하는 살충독소 단백질의 분자량을 SDS-polyacrylamide gel을 이용하여 측정했다. B. sphaericus 1593균이 생산하는 독소결정체를 분리하여 전기영동을 한 결과는 6개의 단백질밴드(43, 58, 64, 100, 113, 130Kd)가 형성되었으나, 독소결정체를 알칼리 pH로 용해하여 전기영동을 하면 2개의 단백질 밴드(43과 64Kd)만이 나타났다. 그러나 대장균 pSL2-1균이 생산하는 독소단백질을 Sephadex G-200으로 정제하여 모기유충에 살충력이 있는 단백질을 전기영동한 결과는 42Kd만이 나타났다. LC50은 2 $\mu\textrm{g}$/$m\ell$이었다. B. sphaericus와 pSL2-1 clone 생산하는 살충단백질은 42Kd 단백질인 것으로 생각된다.

Pilocarpine으로 유발된 간질중첩증에서 케톤생성 식이요법에 의한 Nitric Oxide의 변화 (Effect of Ketogenic Diet on the Nitric Oxide of Pilocarpine-induced Status Epilepticus)

  • Backgrounds and Objectives: Despite of enormous clinical and laboratory researches focused on the useful markers in status epilepticus(SE), clinically applicable methods are not yet available. Although ketogenic diet (KD) is an old method of treating epilepsies, its outstanding antiepileptic effect in some epileptic patients needs re-evaluation of this methods. This study was performed to evaluate the effect of KD on the change of nitric oxide(NO) during the SE. Methods: After the determination of critical EEG stages in the pilocarpine-induced SE model, serum NO levels were measured with Griess reaction. Open cardiac puncture was done immediately after the four different EEG stages of SE in the KD rats and regular diet (RD) rats. Cessation of SE was done with the 10~20 mg/Kg of diazepam i.p. injection in each stages of SE in KD and RD rats. Results: Pilocarpine-induced SE showed reliable EEG and behavioral patterns in all rats. Also, KD did not affect the SE induced by pilocarpine in terms of the SE induction time and SE severity. Serum NO was consistently higher in KD rats than RD rats in all SE stages. Conclusions: KD significantly increases NO during the pilocarpine-induced SE. These finding might contribute the neuroprotective effect of KD in the SE.

저분자량 폴리에틸렌이민을 이용한 유전자 송달 및 발현 유효성 연구 (Cellular Uptake and Transfection Efficiency of Plasmid DNA Using Low Molecular Weight Polyethylenimine)

  • Branched and linear polyethylenimines (PEIs) have been studied as efficient and versatile agents for gene delivery in vitro and in vivo. PEIs exist in a linear or branched topology and are available in a wide range of molecular weight (Mw). Most studies have been done using PEIs with Mw higher than 10Kd. This study was aimed to test the transfection efficiency and the cell viability following gene delivery using PEI of Mw 2Kd, a relatively lower Mw cationic polymer. We used murine interleukin-2(mIL-2) plasmid DNA complexed with branched PEI 2Kd or 25Kd, and transfected them into a myoblast muscle cell line, C2C12. The cellular uptake of mIL-2 plasmid DNA was determined using quantitative polymerase chain reaction. RNA transcript levels were studied in the myoblast cells. Our results show that PEI 2Kd was as effective as PEI 25Kd in celluar gene delivery and transfection efficiency in C2C12 cells. Moreover, MTT assay indicated that PEI 2Kd/DNA complexes did not significantly reduce the cell viability regardless of N/P ratios. These results suggest that PEI of Mw 2Kd might play a role as effective and low toxic nonviral vector systems for muscular cell lines.
