Rapid Drug Susceptibility Testing for Isoniazid and Rifampicin by Reverse Hybridization Assay (역교잡반응법을 이용한 아이소니아지드 및 리팜피신 신속감수성검사)

  • Background : Development of rapid drug susceptibility testing provides the opportunity for rapid identification of individuals with drug resistant tubercle bacilli, allowing selection of appropriate therapeutic regimens. Methods : A total of 502 drug resistant isolates were subjected to reverse blot hybridization assay to detect mutations within genes (rpoB, katG, inhA, and ahpC) associated with rifampicin (RMP) and isoniazid (INH) resistance. Results : Among the 264 RMP resistant strains ($RMP^R$) tested, the most prevalent mutation was the Ser531Leu seen in 121 strains (46%). The second common mutation occurred in 84 strains (32%) at codon 526. And 27 strains (10%) showed the mutation at codon 516. Among all 469 INH resistant strains ($INH^R$), the katG mutation was responsible for INH. The inhA mutation was present in 88 strains (19%). In 11 isolates (2%), coexisting of the katG and inhA mutations were identified. Reverse hybridization assay successfully detected over 80% of $INH^R$ and over 92% of $RMP^R$ among Korean isolates. CONCLUSION: Reverse hybridization was useful for rapid detection of $INH^R$ and $RMP^R$.

A Duplex PCR for Detection of Phytophthora katsurae Causing Chestnut Ink Disease (밤나무 잉크병균, Phytophthora katsurae의 검출을 위한 Duplex PCR)

  • Phytophthora katsurae is a fungal pathogen responsible for chestnut ink disease. We designed two duplex primer sets (SOPC 1F/1R+KatI 3F/5R, SOPC 1-1F/1-1R+KatI 3F/5R) to detect P. katsurae. SOPC 1F/1R and SOPC 1-1F/1-1R primer pairs were designed for sequence characteristic amplification regions (SCAR) marker, and KatI 3F/5R primer pair was used for P. katsurae-specific primer designed from internal transcribed spacer (ITS) region. To assess the sensitivity of duplex PCR, genomic DNA was serially diluted 10-fold to make the final concentrations from 1 mg/ml to 1 ng/ml. The sensitivity for two primer sets were 1 ${\mu}g/ml$ and 100 ng/ml, respectively. To find detection limits for zoospores of P. katsurae, each zoospore suspension was serially diluted 10-fold to make the final concentrations from $1{\times}10^6$ to $1{\times}10^2$ cells/ml, and then DNA was extracted. The limits of detection for all of two primer sets were $1{\times}10^5$ cells/ml. All of two primer sets were specific to P. katsurae in PCR detection and did not produce any P. katsurae-specific PCR amplicons from other 16 Phytophthora species used as the control. This study shows that duplex PCR using two primer sets might be a useful tool for rapid and efficient detection of P. katsurae.

Relationships between Teaching Professional Rank, Course Taking, Teaching Experience and Knowledge of Algebra for Teaching

  • In this study, we examined the relationships among years of teaching experience, professional rank, number of courses taken, and knowledge of algebra for teaching (KAT). 338 in-service and 376 pre-service secondary mathematics teachers in China completed a KAT questionnaire. Various statistical techniques were employed to examine these relationships. The pre-service participants teachers performed statistically significantly higher in advanced mathematics knowledge than their in-service counterparts. Among the inservice teachers, senior teachers had scored higher in school mathematics and teaching mathematics, compared with junior teachers. Yet participants' advanced mathematics knowledge decreased as their professional rank advanced or their teaching experience increased. The number of courses taken has significantly positive correlation with school mathematics knowledge and advanced mathematics knowledge. The implications of these findings for mathematics teacher education are discussed.

  • Gossi-cave, which is Natural monument No. 219, originally was called Norigok-cave, but happened Imjin-Japanese war, aborigine and Gossi family took refuse to Norigok-cave. Japanese soldiers set fire to this cave, in result lots of people had been died. And then, this cave was called Gossi-cave because Gossi family only survived during the war. Stalactite, Stone pillar that was created from four hundred million ago Harmonized with the others. The length is 6.3km in 1969.6.4. this cave was appointed natural monument as a result of investigating Korean Speleological Society. Kim SaKat a Master Poet(1807-1863) : A Genius with wit and eccentric conduct, a poet who wrote a refined poems with his own dramatic path of life destructed the format of Chines poem and created a new folk literature. He is Kim SaKat - a wizard of poet Kim Sakat - a poet wearing conical bamboo hat. Kim Sakat was born in 1807, in the latter part of Chosun period, his antonym was Kim Byung-Yun. He passed the state examination in the first place when he was 20. Two years later, he set out his life as a wanderer when he realized his winning composition was a criticism on his grandfather.

Molecular Analysis of Isoniazid-Resistance Related Genes of Mycobacterium tuberculosis Isolated from Korea

  • Resistance to isoniazid (INH), which is one of the most important drugs in Mycobacterium tuberculosis chemotherapy, has been associated with mutations in genes encoding the mycobacterial catalse-peroxidase (katG), the enoyl acyl carrier protein (ACP) reductase (inhA), alkyl hydroperoxide reductase (ahpC), beta-ketoacyl acyl carrier protein synthase (kasA), and NADH dehydrogenase (ndh). In this study, we examined INH-resistance related genes in 50 INH-resistant and 24 INH-susceptible isolates by PCR-sequence analysis. In brief, mutations at the katG gene were found at codon 315 alone (2/50), at codon 463 alone (19/50), and both at 315 and 463 (29/50). However, while mutations at codon 315 were only detected in INH-resistant isolates, mutations at codon 463 were also detected in INH-susceptible isolates indicating mutations at 463 alone do not seem to confer resistance to INH. Similar to the case of katG 463, some of inhA mutations were also found among INH-susceptible isolates. For example, whereas mutations at 8 upstream of the start codon (UPS) and 15 UPS of the inhA gene were detected only in INH-resistant isolates, mutations at 101, 115, and 125 UPS were detected only in INH-susceptible isolates. Many different kinds of mutations were detected in INH­resistant isolates at ahpC, oxyR gene, and intergenic region of the oxyR-ahpC genes. Howerver, the mutations were not found oxyR and the intergenic regions in INH-susceptible isolates. No mutations were found at either kasA or at ndh gene among INH-resistant isolates. In conclusion, some of mutations such as katG 315, inhA promotor region, and oxyR-ahpC seem to be strongly related to INH-resistance. Currently we are developing a molecular diagnostic method based on these results.

Pyrosequencing Based Detection of Rifampicin or Isoniazid Resistant in Mycobacterium tuberculosis (Pyrosequencing 분석법을 이용한 Rifampicin과 Isoniazid 결핵약제내성의 빠른 검사법)

  • Rifampicin (RIF) and isoniazid (INH) are the most important drug for the treatment of Mycobacterium tuberculosis. Mutations correlated to rifampicin and isoniazid-resistance have been detected in rpoB gene and katG gene, respectively. Of the rifampicin-resistant isolates, 90% showed mutations in rpoB gene at codon 507 to 533. Isoniazid-resistant isolates analysed had a mutation in katG at codon 315. The aim of this study is to develop a pyrosequencing-based approach for rapid detection of ripampin or isoniazid resistant M. tuberculosis based on characterization of all possible mutation in the target region. For this study, the DNA selected from 35 cases of MTB PCR positive clinical sample such as bronchial washing, sputum, and pleural fluid. RIF or INH resistant was analyzed by pyrosequencing data of rpoB and katG gene. 28 (80%) and 7 (20%) of 35 MTB PCR positive DNAs were occured rifampicin-sensitivity and resistant, respectively. For INH, 30 (85.7%) and 5 (14.5%) cases were detected isoniazid-sensitivity and resistant, respectively. When pyrosequencing analysis was compared with ABI sequencing analysis, both analysis were presented same result, but pyrosequencing analysis was more rapid than ABI sequencing analysis. In conclusion, we found that pyrosequencing technology offers high accuracy, specificity, short turn around time and a high throughput in detection of rifampicin or isoniazid resistance in M. tuberculosis.

The Effect of Halliwick 10 Point Program on the Balance Control (Halliwick 10 Point Program이 균형능력에 미치는 영향)

  • The purpose of this study was to evaluate the effect of Halliwick 10 point program on the balance control. Fourteen undergraduate students participated in the experiment, and classified 7 experiment and 7 control group, randomly. Halliwick 10 point program was applied in the experiment group for 4 weeks. Balance index was measured using KAT 2000. Balance index were measured before, during(2 weeks), and after(4 weeks) the training for 4 weeks. 2-way repeated measures ANOVA was used to further distinguish between the groups. The following results were obtained; 1. The total balance index scores from a KAT were significantly decreased after 4 weeks compared with before the training passing of time. However there was no significant difference between experiment and control group. 2. The left and anterior shifting balance index scores from a KAT were significantly decreased after 4 weeks compared with before the training passing of time. However there was no significant difference between experiment and control group. These results lead us to the conclusion that the balance ability of women twenties increase as the water is higher than that of the ground applied Halliwick 10 point program of the water specific therapy province. Therefore, A further direction of this study will be to provide more evidence for Halliwick 10 point program in the water specific therapy.

An Ecological Study on the Process of Plant Community Formation in Tidal Land (간척지 식물군락형성 과정에 관한 연구 -목포지방을 중심으로-)

  • Information this research an attempt was made to investigate the plant community structure and the process of its formation in the tidal area surrounding Mokpo City the examined area included the stand in Sam-Hak Do wherein sands have inflowed, and the stand in Kat-Ba-Woo which has been left as tidal soil land. Two hundred stands were sampled by making 100m line transects 5m apart and establishing 30cm$\times$30cm quadrates 10m apart on each tape. These data were used to establish frequency, cover degree, density, standing crop (g/$m^2$), contained Cl in the soil, and pH. Results of the research are as following: 1. There were 25 species of plants in the stands from Sam-Hak Do, 18 in the stands from Kat-Ba-Woo, and so 28 species in all. Regardings the importance value, the rank of dominant species is Salicornia herbacea L., Suaeda maritima Dumorties, Diplachne fusca L., Echinochola hispidula Nakai, Cyperus iria L., Setaria lutescens Hubbard in Sam-Hak Do, and Suaeda maritima Dumorties, Echinochloa hispidula Nakai, Aster tripolium L., Scripus maritimus L., Salicornia herbacea L., Diplachne fusca L., in the lands from Kat-Ba-Woo. Among them are five kinds of halophytes Salicornia herbacea L., Suaeda maritima Dumorties, Atriplex Gmelini C.A. Meyer, Aster tripolium L., and Limonium tetragonum Bullock; and two Kinds of naturalized plants Diplachan fusca L., Aster subulata. M. In the stands from Sam-Hak Do there was evidence of a secondary succession phenomena in the presence of Cyperaceae such as Cyperus liria L., Juncus decipiens Nakai, and Fimbristylis longispica Steudal, which could not be found in the stands from Kat-Ba-Woo. 2. On the number of species of vegetation, the further inland from the floodgate the higher the number; that is, the lower the content of Cl the higher the number of species. 3. On the distribution of the vegetation; comparing DFD index and Cl content, the main plants are Salicornia herbacea L., Suaeda maritima Dumorties, Atriplex gmelini C.A. Meyer, Diplachne fuscal, Echinochola hispidula Nakai where the Cl content of soil is more than 13.2%. 4. On the standing crop; Salicornia herbacea L., which has high resistance to salt, was half the total was production of 1090/$m^2$ while Suaeda maritima Dumorties was 1/4, Diplachne fusca L. 1/8, and Echinochloa the least. Analyzing the result, the research indicates that the main factor in plant community formation in tidal land is tolerance for Cl, and that a plan for utilizing the halophytes which are abundant in tidal land should be devised.

KAT8/MOF-Mediated Anti-Cancer Mechanism of Gemcitabine in Human Bladder Cancer Cells

  • Histone acetylation is a well-characterized epigenetic modification controlled by histone acetyltransferases (HATs) and histone deacetylases (HDACs). Imbalanced histone acetylation has been observed in many primary cancers. Therefore, efforts have been made to find drugs or small molecules such as HDAC inhibitors that can revert acetylation levels to normal in cancer cells. We observed dose-dependent reduction in the endogenous and exogenous protein expression levels of KAT8 (also known as human MOF), a member of the MYST family of HATs, and its corresponding histone acetylation at H4K5, H4K8, and H4K16 in chemotherapy drug gemcitabine (GEM)-exposed T24 bladder cancer (BLCA) cells. Interestingly, the reduction in MOF and histone H4 acetylation was inversely proportional to GEM-induced γH2AX, an indicator of chemotherapy drug effectiveness. Furthermore, pGL4-MOF-Luc reporter activities were significantly inhibited by GEM, thereby suggesting that GEM utilizes an MOF-mediated anti-BLCA mechanism of action. In the CCK-8, wound healing assays and Transwell® experiments, the additive effects on cell proliferation and migration were observed in the presence of exogenous MOF and GEM. In addition, the promoted cell sensitivity to GEM by exogenous MOF in BLCA cells was confirmed using an Annexin V-FITC/PI assay. Taken together, our results provide the theoretical basis for elucidating the anti-BLCA mechanism of GEM.

Analysis of the Stress Effects of Endocrine Disrupting Chemicals (EDCs) on Escherichia coli

  • In this study, three of the representative EDCs, $17{\beta}$-estradiol, bisphenol A, and styrene, were employed to find their mode of toxic actions in E. coli. To accomplish this, four different stress response genes, recA, katG, fabA, and grpE genes, were used as a representative for DNA, oxidative, membrane, or protein damage, respectively. The expression levels of these four genes were quantified using a real-time RT-PCR after challenge with three different EDCs individually. Bisphenol A and styrene caused high-level expression of recA and katG genes, respectively, whereas $17{\beta}$-estradiol made no significant changes in expression of any of those genes. These results lead to the classification of the mode of toxic actions of EDCs on E. coli.
