This study investigated the physical properties of polymers and antimicrobial activities of organic acids on Listeria monocytogenes to develop hydrogels. ${\kappa}-carrageenan$ (1, 2, and 3%), carboxymethylcellulose (CMC; 1, 3, and 5%), and agar (1.5 and 3%) were mixed with cross-linkers ($Na^+$, $K^+$, $Ca^{2+}$, and $Al^{3+}$) or each other by stirring or heating to form cross-linkage, and their physical properties (hardness, elasticity, and swelling) were measured. The hydrogels formulated with organic acid (1, 3, and 5%) were analyzed by spot assay against L. monocytogenes. ${\kappa}-carrageenan$ formed hydrogels with high hardness without other cross-linkers, but they had low elasticity. The elasticity was improved by mixing with other cross-linkers such as $K^+$ or other polymer, especially in 3% ${\kappa}-carrageenan$. CMC hydrogel was formed by adding cross-linkers $Al^{3+}$, $Na^+$, or $Ca^{2+}$, especially in 5% CMC. Thus, stickiness and swelling for selected hydrogel formulations (two of ${\kappa}-carrageenan$ hydrogels and three of CMC hydrogels) were measured. Among the selected hydrogels, most of them showed appropriate hardness, but only 3% ${\kappa}-carrageenan-contained$ hydrogels maintained their shapes from swelling. Hence, 3% ${\kappa}-carrageenan+0.2%$ KCl and 3% ${\kappa}-carrageenan+1%$ alginate+0.2% KCl+0.2% $CaCl_2$ were selected to be formulated with lactic acid, and showed antilisterial activity. These results indicate that 3% ${\kappa}-carrageenan$ hydrogels formulated with lactic acid can be used to control L. monocytogenes on food surface.

This paper is to induce design floods through L-moment with 3 and 4 parameter Kappa distributions including test of independence by Wald-Wolfowitz, homogeneity by Mann-Whitney and outlier by Grubbs-Beck on annual maximum flood flows at 9 water level gaging stations in Han, Nakdong and Geum Rivers of South Korea. After analyzing appropriateness of the data of annual maximum flood flows by Kolmogorov-Smirnov test, 3 and 4 Kappa distributions were applied and the appropriateness was judged. The parameters of 3 and 4 Kappa distributions were estimated by L-moment method and the design floods by water level gaging station was calculated. Through the comparative analysis using the relative root mean square errors (RRMSE) and relative absolute errors (RAE) calculated by 3 and 4 parameter Kappa distributions with 4 plotting position formulas, the result showed that the design floods by 4 parameter Kappa distribution with Weibull and Cunnane plotting position formulas are closer to the observed data than those obtained by 3 parameter Kappa distribution with 4 plotting position formulas and 4 parameter Kappa distribution with Hazen and Gringorten plotting position formulas.

Background : The importance of pro-inflammatory cytokines, especially tumor necrosis factor $\alpha$ (INF-$\alpha$) and interleukin-1$\beta$ (IL-1$\beta$), have been extensively documented in the generation of inflammatory lung disease. Lung epithelial cells are also actively involved in initiating and maintaining inflammation by producing pro-inflammatory mediators. Understanding the mechanism of pro-inflammatory cytokine expression in lung epithelial cells is crucial to the development of new therapeutic modalities for inflammatory lung disease. Transcription of most pro-inflammatory cytokines is dependent on the activation of NF-${\kappa}B$. However, the relationship between pro-inflammatory cytokine expression and NF-${\kappa}B/I{\kappa}B$ pathway in lung epithelial cells is not clear. Methods : BEAS-2B, A549, Na-H157, NCI-H719 cells were stimulated with IL-$1{\beta}$ or TNF-$\alpha$ at various times, and then IL-8 and TNF-$\alpha$mRNA expressions were assayed by Northern blot analysis. IL-$1{\beta}$ or TNF-$\alpha$-induced NF-${\kappa}B$ activation was assessed by the nuclear translocation of p65 NF-${\kappa}B$ subunit. The degradation of $I{\kappa}B{\alpha}$ and $I{\kappa}B{\beta}$ by IL-$1{\beta}$ or TNF-$\alpha$stimulation was assayed by Western blot analysis. The phosphorylation of $I{\kappa}B{\alpha}$ was evaluated by Western blot analysis after pre-treating cells with proteasome inhibitor followed by IL-$1{\beta}$ or TNF-$\alpha$ stimulation. The basal level of IKK $\alpha$ expression was evaluated by Western blot analysis. Results: $I{\kappa}B{\alpha}$ and $I{\kappa}B{\alpha}$ was rapidly degraded after 5 minutes of incubation with IL-$1{\beta}$ or TNF-$\alpha$ in BEAS-2B, A549, and NCI-H157 cells. The activation of NF-${\kappa}B{\alpha}$ and the induction of IL-8 and TNF-$\alpha$ mRNA expression were observed by IL-$1{\beta}$ or TNF-$\alpha$ stimulation in these cells. In contrast, neither the changes in NF-${\kappa}B/I{\kappa}B$ pathway nor IL-8 and TNF-$\alpha$mRNA expression was induced by IL-$1{\beta}$ or TNF-$\alpha$ stimulation in NCI-H719 cells. IL-$1{\beta}$ and TNF-$\alpha$-induced $I{\kappa}B$ phosphorylation was observed in BEAS-2B, A549, and NCI-H157 cells, but not in NCI-H719 cells. The basal level of IKK$\alpha$ expression was not different between cell. Conclusion : NF-${\kappa}B/I{\kappa}B$ pathway plays an important role in the expression of pro-inflammatory cytokine in most lung epithelial cells. The absence of the effect on NF-${\kappa}B/I{\kappa}B$ pathway in NCI-H719 cells sæms to be due to the defect in the intracellular signal transduction pathway upstream to IKK.
The logarithmic form for turbulent flow analysis guarantees the positivity of the turbulence variables as ${\kappa}$ and ${\varepsilon}$ of the ${\kappa}-{\varepsilon}$ model by using the natural logarithm of these variables. In the present study, the logarithmic form is incorporated into the finite element solution procedure for the unsteady turbulent flow analysis. A backward facing step flow using the standard ${\kappa}-{\varepsilon}$ model and a flow around a 2D square cylinder using the modified ${\kappa}-{\varepsilon}$ model (the Kato-Launder model) are simulated. These results show that the logarithmic form effectively keeps adequate balance of turbulence variables and makes the analysis stable during transient or unsteady processes.
Background: This study investigated the anti-inflammatory effects of bee venom (BV) through the inhibition of nuclear factor kappa beta ($NF-{\kappa}B$) expression in macrophages and keratinocytes. Methods: Cell viability assays were performed to investigate the cytotoxicity of BV in activated macrophages [lipopolysaccharide (LPS)] and keratinocytes [interferon-gamma/tumor necrosis factor-alpha ($IFN-{\gamma}/TNF-{\alpha}$)]. A luciferase assay was performed to investigate the cellular expression of $NF-{\kappa}B$ in relation to BV dose. The expression of $NF-{\kappa}B$ inhibitors ($p-I{\kappa}B{\alpha}$, $I{\kappa}B{\alpha}$, and p50 and p65) were determined by Western Blot analysis, and the electromobility shift assay. A nitrite quantification assay was performed to investigate the effect of BV, and $NF-{\kappa}B$ inhibitor on nitric oxide (NO) production in macrophages. In addition, Western Blot analysis was performed to investigate the effect of BV on the expression of mitogen-activated protein kinases (MAPK) in activated macrophages and keratinocytes. Results: BV was not cytotoxic to activated macrophages and keratinocytes. Transcriptional activity of $NF-{\kappa}B$, and p50, p65, and $p-I{\kappa}B{\alpha}$ expression was reduced by treatment with BV in activated macrophages and keratinocytes. Treatment with BV and an $NF-{\kappa}B$ inhibitor, reduced the production of NO by activated macrophages, and also reduced $NF-{\kappa}B$ transcriptional activity in activated keratinocytes (compared with either BV, or $NF-{\kappa}B$ inhibitor treatment). Furthermore, BV decreased p38, p-p38, JNK, and p-JNK expression in LPS-activated macrophages and $IFN-{\gamma}/TNF-{\alpha}$-activated keratinocytes. Conclusion: BV blocked the signaling pathway of $NF-{\kappa}B$, which plays an important role in the inflammatory response in macrophages and keratinocytes. These findings provided the possibility of BV in the treatment of atopic dermatitis.
$NF{\kappa}B$ and ZAS3 are transcription factors that control important cellular processes including immunity, cell survival and apoptosis. Although both proteins bind the ${\kappa}B$-motif, they produce opposite physiological consequences; $NF{\kappa}B$ activates transcription, promotes cell growth and is often found to be constitutively expressed in cancer cells, while ZAS3 generally represses transcription, inhibits cell proliferation and is downregulated in some cancers. Here, we show that ZAS3 inhibits $NF{\kappa}B$-dependent transcription by competing with $NF{\kappa}B$ for the ${\kappa}B$-motif. Transient transfection studies show that N-terminal 645 amino acids is sufficient to repress transcription activated by $NF{\kappa}B$, and that the identical region also possesses intrinsic repression activity to inhibit basal transcription from a promoter. Finally, in vitro DNA-protein interaction analysis shows that ZAS3 is able to displace $NF{\kappa}B$ by competing with $NF{\kappa}B$ for the ${\kappa}B$-motif. It is conceivable that ZAS3 has therapeutic potential for controlling aberrant activation of $NF{\kappa}B$ in various diseases.
Frictional loss in turbulent regime is abnormally increased compared with in laminar regime. Thus the consideration of temperature rise across fluid film is significant in analysis and conventional isothermal theory loses its usefulness for performance prediction. This paper proposes to the three-dimensional thermohydrodynamic analysis of finite journal bearings operating under turbulent condition using two-equation model($\kappa-\varepsilon$ model) proposed by Hassid & Poreh. The equations are solved numerically by finite difference method. We make the analysis applicable even at large eccentricity when back flow of the lubricants occurs and axial flow is no longer ignored compared to circumferential flow.

The objective of this study is to induce the design floods by the methodology of L-moment including test of homogeneity, independence and outlier of the data of annual maximum flood flows for 12 water level gaging stations of South Korea. To select appropriate distribution of the data for annual maximum flood flows, the distributions of Wakeby and Kappa are applied and the appropriateness is judged by Kolmogorov-smirnov (K-S) test. The parameters of selected Wakeby and Kappa distributions are calculated by the method of L-moment and the design floods are induced. Through the comparative analysis using the relative root mean square errors (RRMSE) and relative absolute errors (RAE) of design floods, the result shows that the design floods by Wakeby distribution are closer to the observed data than those obtained by the Kappa distribution.
Resveratrol (trans-3,4'-trihydroxystillbene), a naturally occurring polyphenolic antioxidant found in grapes and red wine, elicits diverse biochemical responses and demonstrates anti-aging, anti-inflammatory, and anti-proliferative effects in several cell types. Previously, resveratrol was shown to regulate differentiation and inflammation in rabbit articular chondrocytes, while the direct production of nitric oxide (NO) in these cells by treatment with the NO donor sodium nitroprusside (SNP) led to apoptosis. In this study, the effect of resveratrol on NO-induced apoptosis in rabbit articular chondrocytes was investigated. Resveratrol dramatically reduced NO-induced apoptosis in chondrocytes, as determined by phase-contrast microscopy, the MTT assay, FACS analysis, and DAPI staining. Treatment with resveratrol inhibited the SNP-induced expression of p53 and p21 and reduced the expression of procaspase-3 in chondrocytes, as detected by western blot analysis. SNP-induced degradation of I-kappa B alpha ($I{\kappa}B-{\alpha}$) was rescued by resveratrol treatment, and the SN50 peptide-mediated inhibition of NF-kappa B (NF-${\kappa}B$) activity potently blocked SNP-induced caspase-3 activation and apoptosis. Our results suggest that resveratrol inhibits NO-induced apoptosis through the NF-${\kappa}B$ pathway in articular chondrocytes.

Agreement analysis is conducted to assess reliability among rating results performed repeatedly on the same subjects by one or more raters. The kappa statistic is commonly used when rating scales are categorical. The simple and weighted kappa statistics are used to measure the degree of agreement between two raters, and the generalized kappa statistics to measure the degree of agreement among more than two raters. In this paper, we compare the performance of four different generalized kappa statistics proposed by Fleiss (1971), Conger (1980), Randolph (2005), and Gwet (2008a). We also examine how sensitive each of four generalized kappa statistics can be to the marginal probability distribution as to whether marginal balancedness and/or homogeneity hold or not. The performance of the four methods is compared in terms of the relative bias and coverage rate through simulation studies in various scenarios with different numbers of raters, subjects, and categories. A real data example is also presented to illustrate the four methods.
