Analysis of Cone and Seed Characteristics from Different Mating Design Strategies of Pinus densiflora for. multicaulis

  • This study aimed to enhance seed productivity and secure genetic resources for Pinus densiflora for. multicaulis. We analyzed the characteristics of cone and seed generated by control pollination between Pinus densiflora (PD) and Pinus densiflora for. multicaulis (PDM). The highest number of cone scales (63.0) was obtained from the self-pollinated (sp) PDM clone B (PDM-sp-B), whereas the lowest number of cone scales (44.7) was obtained from two combinations designated as PDM-A×PD-075 and PDM-A×PD-0111. Both female parents of the hybrids were PDM-A. The highest seed production capacity (80.8) was obtained from the open-pollinated (op) PDM clone B (PDM-op-B). The seed potentials of PDM-B×PD-0111, PDM-op-A, and PDM-sp-B were 67.4, 66.5, and 63.1, respectively. The highest number of fertile scales (41.5) was obtained from PDM-op-B, and the lowest number of fertile scales (28.8) was obtained from PDM-A×PD-075. The total number of aborted ovules and 1st aborted ovules was not statistically significant in the mating design. The cross combination of PDM-B×PD-0111 had the highest number (34.8) of filled seeds and the lowest number of 2nd aborted ovules (5.2) and empty seeds (9). PDM-op-B had the highest number of developed seeds (47.6), although the number of empty seeds was the highest (41.2). Therefore, we conclude that the mating design of PDM-B×PD-0111 is useful for future breeding programs to improve seed yield of PDM. Our results showed that there was a strong correlation between the following two parameter pairs: number of scales and number of fertile scales, and the number of fertility scales and seeds potential (r=0.89 and r=0.84, respectively; both p<0.01).

Assessing Trees Diversity in Jebel Elgarrie Forest Reserve in the Blue Nile State, Sudan

  • The study aims to examine population indices of mature trees in Jebel Elgarrie forest, Blue Nile State, Sudan. We used remote sensing techniques to stratify the forest into vegetation classes depending on tree density. We distributed 97 circular sample plots (0.1 ha) proportionally to the area of the vegetation classes. In each sample plot we identified, counted and recorded all mature trees (DBH ≥10 cm). We calculated frequency, density, abundance, richness, evenness and diversity for each species and we drew abundance rank curve of mature trees. We used One-Way ANOVA to test for differences (α=0.05) in mean density (No./ha) of mature trees between vegetation classes. Results revealed that the forest was conveniently sub-divided into high density (C1), medium density (C2), low density (C3) and bare farm land (C4) classes. We identified fifteen tree species that belong to 10 families and 14 genera. Combretaceae and Fabaceae were the common families while Anogeissus leiocarpa was the most frequently occurring species. While species diversity varied between vegetation classes, diversity of the forest as a whole is low. While mean density of mature trees in C1, C2, C3 and C4 it was 100, 74, 10, and 0, respectively, it was 54 for the whole forest indicating low stocking, Following One-Way ANOVA, multiple comparisons revealed significant differences in mean density of mature trees between C1 & C3 and C2 & C3. The study provided empirical results on population indices of mature tree species, which would be of importance for successful management and conservation of the forest.

Evaluation of Drought Tolerance for Biomass Production of Salix gracilistyla Miq.

  • Salix gracilistyla is widely distributed along riversides in Korea and very good for biomass production by SRC because of its excellent germination ability, but it is necessary to measure drying tolerance for cultivation. The drought tolerance of S. gracilistyla was tested using cuttings, and growth and physiological analysis were performed after irrigation was stopped. The growth inhibition of S. gracilistyla was observed from the day irrigation was stopped, and the soil moisture content decreased to less than 10% on the 25th day after irrigation was stopped. Over 50% of the seedlings turned brown 25 days after watering was stopped. The chlorophyll content of S. gracilistyla decreased dramatically after 25 days of stopping of irrigation. RWC values were unchanged until day 12 after irrigation was stopped but decreased rapidly until day 21, but there was a slightly decreasing trend after that. RWL levels increased slightly during irrigation stops. The proline content of plants subjected to drought stress was 0.91-2.63 mg/0.05 g, 2.75 times higher than that of the control treatment. The sugar content of the drought stress treatment group was 29.77 to 350.66 mg/0.05 g, which increased 12.24 times that of the control treatment. As a result of this study, S. gracilistyla was found to have a drought tolerance almost comparable to that of evergreen broad-leaved trees growing on the land. This study is expected to contribute to the resource utilization S. gracilistyla, a native willow tree of Korea, and the mass production of biomass by SRC.

Analysis of Suspended Solids Reduction by Vegetative Filter Strip for Cultivated Area Using Web GIS-Based VFSMOD (VFSMOD를 이용한 경작지의 고형물질 유출 저감효과)

  • The study was performed to simulate the reduction efficiency of suspended solids (SS) for cultivated land located at riverine area at the Namhan River and the Bukhan River watershed sites (site A, B, C) under the rainfall conditions using HUFF & SCS UH-based VFS Design module of Web GIS-based VFSMOD System. The study indicates that the field 5% sloped, located at Bukhan River watershed (site A), requires at least 0.5 m width of Vegetative Filter Strip (VFS) to reduce 70% of SS while the field 10% sloped requires the at least 1.0~1.5 m width of VFS to reduce 70% SS, under the condition 106.2 mm of rainfall event and bell pepper or corn of crops. Against the conditions 95.1 mm of rainfall event and sweet potato or soy bean of crops, the field 5% sloped, located at Namhan River watershed (site B) requires at least 0.5 m width of VFS to reduce 70% of SS while the field 10% sloped requires at least 1.0 m width of VFS to reduce 50% SS. The crops sweet potato and soy bean are cultivated in the site C, located at Namhan River watershed, 1 m of VFS is capable of 64.0% and 62.0% of SS reduction against 102.6 mm and 151.2 mm rainfall conditions respectively, for the 5% sloped field. The result supports that VFS is one of most potential methods to reduce SS from cultivated area, which is environment-friendly hydrologic structure. The VFS design, however, needs to be simulated before its installation in the field, the simulation needs to consider not only various characteristics of the field but also different conditions affecting the VFS, using a model capable to consider a lot of factors.

  • Metallothionein (MT) family of metal-binding proteins are involved in maintaining homeostasis and heavy metal poisoning. Recently, MT has been considered as a biomarker that can identify a particular species, very similar to the use of cytochrome oxidase I (COI) gene. Satsuma myomphala species of land snails have been reported from North-East Asia, including South Korea and Japan. In particular, the land snail species have been known from only a limited area of Geoje Island, Gyeongsangnam-do province of South Korea. Genetic studies of S. myomphala has been limited with only 6 nucleotide, 2 protein registered on the NCBI server. For elucidating the genetic information of S. myomphala, we conducted RNA sequencing analysis using Illumina HiSeq 2500 next-generation platform. We screened the MT gene from the RNA-Seq database to confirm the molecular phylogenetic relationship. After sequencing, the de novo analysis and clustering generated 103,774 unigenes. After annotation against PANM database using BLAST program, we obtained MT sequence of 74 amino acid residues containing the coding region of 222 bp. Based on this sequence, we found about 53 sequences using the BLAST program in NCBI nr database. Using ClustalX alignment, Maximum-Likehood Tree of MEGA program, we confirmed the molecular phylogenetic relationships that showed similarity with mollusks such as Helix pomatia and H. aspersa, Megathura crenulata.

An Comparison of Storability of Several Cultivars Chicon Grown in Different Regions (품종과 재배지역에 따른 치콘의 저장성 비교)

  • This study was conducted to compare the storability of 6 chicory cultivars for producing chicon; 'Vintor', 'Focus', 'Metafora', 'Kibora', 'Nobus', and 'Redoria Red' grown in 2 regions: Chuncheon (plain region) and Pyeongchang (high land region). Chicons were produced from chicory roots that grown for 120days and then stored for over 120days at $2^{\circ}C$ and 90% of RH conditions. To produce chicon, chicory root was forced at $18^{\circ}C$ for 22days with suppling the nutrient solution ($KNO_30.54g{\cdot}L^{-1}$, $Ca(NO_3)_2\;1.02g{\cdot}L^{-1}$, $MgSO_4\;0.36g{\cdot}L^{-1}$, $KH_2PO_4\;0.21g{\cdot}L^{-1}$, $K_2SO_4\;0.10g{\cdot}L^{-1}$, pH 7.0). Chicons produced from 6 different chicory cultivars packed with $25{\mu}m$ ceramic film and stored for 25days at $8^{\circ}C$. The fresh weight of chicon in MAP was maintained to 99.5% of pre-storage weight. The fresh weight of 'Redoria Red' was lowest in all cultivars, and that of Chuncheon region cultivated treatment was lower than Pyeongchang treatment. The $CO_2$ and $O_2$ concentration in chicon MAP were 2% and $10{\sim}17%$. There were not significantly different among cultivars and between regions, although 'Redoria Red' cultivar showed highest $CO_2$ and lowest $O_2$ concentrations. The ethylene concentration in chicon MAP was $1.0{\mu}{\iota}{\cdot}{\iota}^{-1}$ and also didn't show any significant difference among all treatments. Chicon detoriorated visual quality with appearing russet spotting that result from ethylene gas. The visual quality of 'Redoria Red' cultivar decreased faster than the other cultivars. 'Metafora', 'Focus', and 'Kibora' maintained higher firmness of their leaf than the others, and the firmness was higher grown in Pyeongchang region cultivated treatments than in Chuncheon region.

The Evaluation of Meteorological Inputs retrieved from MODIS for Estimation of Gross Primary Productivity in the US Corn Belt Region (MODIS 위성 영상 기반의 일차생산성 알고리즘 입력 기상 자료의 신뢰도 평가: 미국 Corn Belt 지역을 중심으로)

  • Investigation of the $CO_2$ exchange between biosphere and atmosphere at regional, continental, and global scales can be directed to combining remote sensing with carbon cycle process to estimate vegetation productivity. NASA Earth Observing System (EOS) currently produces a regular global estimate of gross primary productivity (GPP) and annual net primary productivity (NPP) of the entire terrestrial earth surface at 1 km spatial resolution. While the MODIS GPP algorithm uses meteorological data provided by the NASA Data Assimilation Office (DAO), the sub-pixel heterogeneity or complex terrain are generally reflected due to coarse spatial resolutions of the DAO data (a resolution of $1{\circ}\;{\times}\;1.25{\circ}$). In this study, we estimated inputs retrieved from MODIS products of the AQUA and TERRA satellites with 5 km spatial resolution for the purpose of finer GPP and/or NPP determinations. The derivatives included temperature, VPD, and solar radiation. Seven AmeriFlux data located in the Corn Belt region were obtained to use for evaluation of the input data from MODIS. MODIS-derived air temperature values showed a good agreement with ground-based observations. The mean error (ME) and coefficient of correlation (R) ranged from $-0.9^{\circ}C$ to $+5.2^{\circ}C$ and from 0.83 to 0.98, respectively. VPD somewhat coarsely agreed with tower observations (ME = -183.8 Pa ~ +382.1 Pa; R = 0.51 ~ 0.92). While MODIS-derived shortwave radiation showed a good correlation with observations, it was slightly overestimated (ME = -0.4 MJ $day^{-1}$ ~ +7.9 MJ $day^{-1}$; R = 0.67 ~ 0.97). Our results indicate that the use of inputs derived MODIS atmosphere and land products can provide a useful tool for estimating crop GPP.

Evaluation of MODIS-derived Evapotranspiration at the Flux Tower Sites in East Asia (동아시아 지역의 플럭스 타워 관측지에 대한 MODIS 위성영상 기반의 증발산 평가)

  • Evapotranspiration (ET) is one of the major hydrologic processes in terrestrial ecosystems. A reliable estimation of spatially representavtive ET is necessary for deriving regional water budget, primary productivity of vegetation, and feedbacks of land surface to regional climate. Moderate resolution imaging spectroradiometer (MODIS) provides an opportunity to monitor ET for wide area at daily time scale. In this study, we applied a MODIS-based ET algorithm and tested its reliability for nine flux tower sites in East Asia. This is a stand-alone MODIS algorithm based on the Penman-Monteith equation and uses input data derived from MODIS. Instantaneous ET was estimated and scaled up to daily ET. For six flux sites, the MODIS-derived instantaneous ET showed a good agreement with the measured data ($r^2=0.38$ to 0.73, ME = -44 to $+31W\;m^{-2}$, RMSE =48 to $111W\;m^{-2}$). However, for the other three sites, a poor agreement was observed. The predictability of MODIS ET was improved when the up-scaled daily ET was used ($r^2\;=\;0.48$ to 0.89, ME = -0.7 to $-0.6\;mm\;day^{-1}$, $RMSE=\;0.5{\sim}1.1\;mm\;day^{-1}$). Errors in the canopy conductance were identified as a primary factor of uncertainty in MODIS-derived ET and hence, a more reliable estimation of canopy conductance is necessary to increase the accuracy of MODIS ET.

Projection of Potential Cultivation Region of Satsuma Mandarin and 'Shiranuhi' Mandarin Hybrid Based on RCP 8.5 Emission Scenario (RCP 8.5 기후변화시나리오에 근거한 온주밀감과 '부지화'의 잠재적 재배지 변화 예측)

  • The potential change of the cultivation area of main citrus cultivars, satsuma mandarin (Citrus unshiu Marc.) and 'Shiranuhi' mandarin hybrid [(Citrus unshiu ${\times}$ C. sinensis) ${\times}$ C. reticulata] were determined with base year (1981 to 2010) to 2090. The meteorological data provided by the Korea Meteorological Administration (KMA), and the digital agricultural climate map of 30m-solution based on the Representative Concentration Pathways (RCP) 8.5 was used for projection of potential cultivation area. As a result, the potential suitable region of satsuma mandarin included almost Jeju region during base year. At the 2030s, the potential suitable region of satsuma mandarin increased and the cultivable region also increased focused on the coast region of Jeonnam province. From the 2060s, the suitable area spread out to mountain area of Jeju, Jeonnam, Gyeongnam, and the coast region of Kangwon, and the cultivable region expanded to the area of Gyeongbuk, Chungnam, and Jeonbuk. In the case of 'Shiranuhi' mandarin hybrid, the suitable region included only the partial coast area of Jeju, and cultivable area covered Jeju region and the partial southern coast of Jeonnam during the standard period. At the 2030s, the suitable region of 'Shiranuhi' included the current cultivation area of satsuma mandarin, and the cultivable region moved to northward by the partial southern coast region. At the 2090s, the slightly increased suitable region covered all Jeju regions, Jeonnam, Gyeongnam, and the coast area of Kangwon, and the cultivable region proceeded northward focusing on the coastline. In conclusion, the prediction of the potential land for citrus cultivation based on the RCP 8.5 showed that the suitable region of satsuma mandarin decreased, whereas that of cultivation of 'Shiranuhi' increased. Moreover, it was forecasted that citrus cultivation area would extend to Kangwon region at the end of the $21^{st}$ century.

  • Serpins are a group of proteins involved in the regulation of serine and other type of proteases, and have been identified in many kinds of organisms from invertebrates to vertebrates. Serpins are known to regulate the proteolytic cascades of the innate immune pathways in addition to their roles in blood coagulation, angiogenesis, fibrinolysis, inflammation and tumor suppression. In this study, we have isolated two partial serpin gene fragments from expressed sequence tags (ESTs) of Nesiohelix samarangae. Dotplot analysis indicates that they are of two different types, Ns-serpin type 1 and Ns-serpin type 2. Ns-serpin type 1 has 819 bp coding region (272 amino acids), whereas Ns-serpin type 2 has 555 bp coding region (185 amino acids). Molecular phylogenetic analysis shows that the identified serpins have high similarities to their counterparts in the California see slug, Aplysia californica. Yet, the precise biological and immunological roles of these Ns-serpins remain to be further investigated using RNA interference and other molecular techniques.
