The Amundsen Sea in West Antarctica is one of the most affected regions by climate change, but it is one of the least studied realms due to difficulties in access. Korea Polar Research Institute (KOPRI) launched a research project in the Amundsen Sea in 2010 using the icebreaker research vessel (IBRV) Araon and has been conducting various research initiatives. In this paper, previous researches derived from the Amundsen Sea Embayment by Korean researchers are introduced. Through previous studies, researchers have been able to interpret the environmental and biogeochemical changes according to the inflow Circumpolar Deep Water (CDW) and provide information for climate models. In particular, researches using radiocarbon isotopes (14C) were introduced to understand the physical and biogeochemical mechanisms of the carbon cycle in the Amundsen Sea. Opportunely, with the construction of a second icebreaker research vessel, the direction for systematic and long-term polar data acquisition can be presented.

In order to understand the carbon cycle in the Amundsen Sea of the Southern Ocean, the export fluxes of particulate organic carbon from the euphotic zone to deep water estimated using ${\psi}$/${\psi}$ disequilibrium method. Seawaters in 14 water columns were collected during February and March 2012, and analyzed for total and dissolved ${\psi}$, and particulate organic carbon. Total ${\psi}$ activities in the water column showed deficiency and excess relative to those of ${\psi}$ depending on the water depth. Deficiency of total ${\psi}$ in the euphotic zone showed mirror images both with chlorophyll-a and fluorescence, and was consistent with the loss of nitrate, which indicated the effect of biological activity. In addition, deficiency of total ${\psi}$ from deep water was associated with the increase of total dissolvable Fe/Mn concentration. Excess total ${\psi}$ activity presented below the euphotic zone might be related to particulate ${\psi}$ concentrated in this water depth. Mean export flux of ${\psi}$ estimated using the steady state model was $867{\pm}246dpmm^{-2}day^{-1}$. Mean export flux of particulate organic carbon, which were estimated by the product of total ${\psi}$ flux and ratio of POC/${\psi}$ ($7.08{\pm}4.27{\mu}molCdpm^{-1}$) in the sinking particles, was $5.9{\pm}3.9mmolCm^{-2}day^{-1}$. These fluxes were similar levels to those in the Weddell Sea during February and March 2008. Export ratios (ThE) relative to the primary production in the euphotic zone were in the range of 3-54% (av. 28%).

Water vapor isotopes can be excellent tools for understanding complex mechanisms in the water cycle and atmospheric hydrological cycle and they can be applied to various fields of paleoclimatology, atmospheric science, hydrogeology, oceanography, and ecohydrology. Thus, studies of global or local transport of water vapor may be able to provide a very useful clue to better understand the movements of water and energy in the atmosphere, hydrosphere and biosphere. In this study, the isotopic compositions of water vapor have been observed for moisture transport during the passage of Typhoon Bolaven at Korea Polar Research Institute (KOPRI), Incheon, in the western part of Korea, from August 27 to August 29, 2012. In the clear sky, the isotopic compositions of water vapor at KOPRI exhibited relatively higher isotopic ratios, which were near isotopic equilibrium with sea surface water (${\delta}^{18}O$=-14‰). On the other hand, a largely depleted isotopic ratios in surface water vapor were observed in association with the passage of Typhoon Bolaven (approximately 10‰ depleted compared to the clear sky). The fact that the isotopic minima in water vapor are encountered during the onset period of the Typhoon Bolaven with increases of relative humidity, which is consistent with, so called, "the amount effect".

The Korea Polar Research Institute (KOPRI) has assembled a culture collection of cold-adapted bacterial strains from both the Arctic and Antarctic. To identify excellent protease-producers among the proteolytic bacterial collection (874 strains), 78 strains were selected in advance according to their relative activities and were subsequently re-examined for their extracellular protease activity on $0.1{\times}$ ZoBell plates supplemented with 1% skim milk at various temperatures. This rapid and direct screening method permitted the selection of a small group of 15 cold-adapted bacterial strains, belonging to either the genus Pseudoalteromonas (13 strains) or Flavobacterium (2 strains), that showed proteolytic activities at temperatures ranging between $5-15^{\circ}C$. The cold-active proteases from these strains were classified into four categories (serine protease, aspartic protease, cysteine protease, and metalloprotease) according to the extent of enzymatic inhibition by a class-specific protease inhibitor. Since highly active and/or cold-adapted proteases have the potential for industrial or commercial enzyme development, the protease-producing bacteria selected in this work will be studied as a valuable natural source of new proteases. Our results also highlight the relevance of the Antarctic for the isolation of protease-producing bacteria active at low temperatures.
A time-series sediment trap was deployed at 1,034 m water depth in the eastern Bransfield Strait for a complete year from December 25, 1998 to December 24, 1999. About 99% of total mass flux was trapped during an austral summer, showing distinct seasonal variation. Biogenic particles (biogenic opal, particulate organic carbon, and calcium carbonate) account for about two thirds of annual total mass flux $(49.2\;g\;m^{-2})$, among which biogenic opal flux is the most dominant (42% of the total flux). A positive relationship (except January) between biogenic opal and total organic carbon fluxes suggests that these two variables were coupled, due to the surface-water production (mainly diatoms). The relatively low $\delta^{13}C$ values of settling particles result from effects on C-fixation processes at low temperature and the high $CO_2$ availability to phytoplankton. The correspondingly low $\delta^{l5}N$ values are due to intense and steady input of nitrates into surface waters, reflecting an unlikely nitrate isotope fractionation by degree of surface-water production. The $\delta^{l5}N$ and $\delta^{l3}C$ values of sinking particles increased from the beginning to the end of a presumed phytoplankton bloom, except for anomalous $\delta^{l5}N$ values. Krill and the zooplankton fecal pellets, the most important carriers of sinking particles, may have contributed gradually to the increasing $\delta^{l3}C$ values towards the unproductive period through the biomodification of the $\delta^{l3}C$ values in the food web, respiring preferentially and selectively $^{12}C$ atoms. Correspondingly, the increasing $\delta^{l5}N$ values in the intermediate-water trap are likely associated with a switch in source from diatom aggregates to some remains of zooplankton, because organic matter dominated by diatom may be more liable and prone to remineralization, leading to greater isotopic alteration. In particular, the tendency for abnormally high $\delta^{l5}N$ values in February seems to be enigmatic. A specific species dominancy during the production may be suggested as a possible and speculative reason.
Korea Polar Research Institute (KOPRI) has successfully installed two autonomous very broadband three-component seismic stations at the King George Island (KGI), Antarctica, during the 24th KOPRI Antarctic Summer Expedition (2010 ~ 2011). The seismic observation system is originally designed by the Incorporated Research Institutions for Seismology Program for Array Seismic Studies of the Continental Lithosphere Instrument Center, which is fully compatible with the Polar Earth Observing Network seismic system. The installation is to achieve the following major goals: 1. Monitoring local earthquakes and icequakes in and around the KGI, 2. Validating the robustness of seismic system operation under harsh environment. For further intensive studies, we plan to move and install them adding a couple more stations at ice shelf system, e.g., Larsen Ice Shelf System, Antarctica, in 2013 to figure out ice dynamics and physical interaction between lithosphere and cryosphere. In this article, we evaluate seismic station performance and characteristics by examining ambient noise, and provide operational system information such as frequency response and State-Of-Health information.
As there is a growing interest in remote sensing applications, the Korean Society of Remote Sensing has published several special issues in which articles on specific research topics were collected. In this special issue, we first review the research topics on several special issues published in the Korean Journal of Remote Sensing by 1) the National Institute of Agricultural Sciences (NAS), the Korea Environment Institute (KEI), the Ministry of Environment (ME) and the Korea Aerospace Research Institute (KARI) in 2017 and 2) the Korea Institute of Ocean Science and Technology (KIOST), the Korea Polar Research Institute (KOPRI), KARI, and Korea Meteorological Administration (KMA) in 2018. Then, research articles on the remote sensing applications in Korea are introduced.
ICESat launched in Jan. 2003 has a capability to monitor polar regions with its inclination of 94 degree. ICESat carries GLAS (Geosciences Laser Altimetry System) to measure Earth's topography in unprecedented accuracy, and thus it can be applied for glacier variation due to recent climate changes. Here we present a new method to estimate velocity structure of Amery Ice-Shelf using ICESat altimtery data. ICESat data shows horizontal displacement of Amery Ice-Shelf, which can be directly used for velocity estimation. This method is expected to extend to other ice-shelves in Antarctica.

The filamentous cyanobacterium Anabaena sp. Strain PCC 7120 produces a developmental patten of single hete- rocysts separated by approximately 10 vegetative cells. Heterocysts differentiate from vegetative cells and are spe- cialized for nitrogen fixation. The patS gene, which encodes a small peptide that inhibits heterocyst differentiation, is expressed in proheterocysts and plays a critical role in establishing the heterocyst pattem. Another key regulator of heterocyst development is the hetR gene. hetR mutants fail to produce heterocysts and extra copies of hetR on a plas- mid cause a multiple contiguous heterocyst phenotype. To elucidate the relationship between these two counter act- ing factors in the genetic regulatory pathway during heterocyst differentiation, the expression patterns of a patS-gfp and a hetR-gfp fusion were examined in a patS deletion and a hetR deletion strain. The results, in combination with the result from a hetR and patS double deletion strain, suggest patS and hetR are mutually antagonistic and the bal- ance between these two factors in tow different cell types (heterocysts and vegetative cells) may be critical during the decision making process on their cell fates.
To better understand the gene expression of the cold-adapted polar diatom, we conducted a survey of the Chaetoceros neogracile transcriptome by cDNA sequencing and expression of interested cDNAs from the Antarctic diatom. A non-normalized cDNA library was constructed from the C. neogracile, and a total of 2,500 cDNAs were sequenced to generate 1,881 high-quality expressed sequence tags (ESTs) (accession numbers EL620615-EL622495). Based on their clustering, we identified 154 unique clusters comprising 342 ESTs. The remaining 1,540 ESTs did not cluster. The number of unique genes identified in the data set is thus estimated to be 1,694. Taking advantage of various tools and databases, putative functions were assigned to 939 (55.4%) of these genes. Of the remaining 540 (31.9%) unknown sequences, 215 (12.7%) appeared to be C. neogracile-specific since they lacked any significant sequence similarity to any sequence available in the public databases. C. neogracile consisted of a relatively high percentage of genes involved in metabolism, genetic information processing, cellular processes, defense or stress resistance, photosynthesis, structure, and signal transduction. From the ESTs, the expression of these putative C. neogracile genes was investigated: fucoxanthin chlorophyll (chl) a,c-binding protein (FCP), ascorbate peroxidase (ASP), and heat-shock protein 90 (HSP90). The abundance of ASP and HSP90 changed substantially in response to different culture conditions, indicating the possible regulation of these genes in C. neogracile.
