Sixteen New Records of Ascomycetes from Crop Field Soil in Korea

  • The present study reports 16 species of Ascomycota that were previously unknown in Korea, namely Acremonium cellulolyticus (KNU14-25), Acremonium zonatum (KNU14-15), Chaetomium madrasense (KNU14-9), Cladosporium silenes (KNU 14-18-1), Humicolopsis cephalosporioides (KNU15-3), Leptosphaerulina chartarum (KNU14-16), Paecilomyces marquandii (KNU14-8), Paecilomyces tenuis (KNU14-18-2), Paraphaeosphaeria sporulosa (KNU15-2), Penicillium rubidurum (KNU14-12), Pochonia suchlasporia (KNU15-6), Sporothrix inflata (KNU15-8), Thielavia hyrcaniae (KNU15-1), Thielavia terricola (KNU14-23-1), Xylogone sphaerospora (KNU15-7), and Zopfiella longicaudata (KNU15-5). These fungal species were isolated from soil samples collected from various regions of Korea and identified based on their morphological characteristics and rDNA internal transcribed spacer sequence data. Full descriptions and illustrations for each species are provided.

Studies on collection and spawn manufacture of Armillaria spp. for production of Gastrodia tuber (천마생산을 위한 천마버섯균의 수집과 우량종균 제조에 관한 연구)

  • Armillaria isolates (KNU-A110, KNU-A234, KNU-A1022 and KNU-A1030) were excellent isolates for producing Gastrodia tuber in farm cultivation. Depth of soil between $10\;cm{\sim}19\;cm$ was favorable for producing Gastrodia tuber and rhizomorph at cultivated area. Eighty nine isolates were collected from 5 countries; 16 from Japan, 22 from USA, 26 from France, 4 from Africa and 21 from Korea. Mycelial fan of most isolates were better formed on basal medium with lemon extract than without lemon.A. mellea (KNU-A997) were strongly pathogenic to Gastrodia but A. gallica (KNU-A110) were excellent symbiotic to Gastrodia tuber. Mycelial growth were good on basal medium containing $0.25%{\sim}0.5%$ ethanol and sawdust spawn added wheat bran and corncob. Liquid culture inoculation were not only fast growth of mycelium but also reduction of contamination.

Effect of NaCl Adaptation on the Thermotolerance and Alcohol Fermentation in Saccharomyces cerevisiae KNU5377. (Saccharomyces cerevisiae KNU5377의 NaCl에 대한 적응이 고온내성과 알코올발효에 미치는 영향)

  • Saccharomyces cerevisiae KNU5377 is a constitutively thermotolerant, fermentative strain at high temperatures over 4$0^{\circ}C$. The exposure to 0.5 M NaCl caused S. cerevisiae KNU5377 to be lost its constitutive thermotolerance. Furthermore, the NaCl adaptation beyond 0.3 M during the overnight culture forced the strain-specific fermentation ability of S. cerevisiae KNU5377 to be disappeared. However, these phenomena did not occur in the reference, Saccharomyces cerevisiae ATCC24858. As a result, this adaptation led both strains to show the closely similar thermotolerance level and alcohol fermentation ability, implying the NaCl adaptation eliminated its strain-specific characteristics of S. cerevisiae KNU5377 Therefore it indicated that the superior intrinsic characteristics of S. cerevisiae KNU5377 must be related to the NaCl adaptation. On the other hand, the heat adaptation elevated alcohol productivity for both strains, but surprisingly did it for KNU5377 at the rate of two times higher than the reference's one; this suggests that KNU5377 possesses more efficient system enough to cause the difference. Consequently, these characteristics of S. cerevisiae KNU5377 must be interesting targets for further study to understand on how KNU5377 could acquire the constitutive thermotolerance and the outstanding fermentative capacity at high temperatures.

Morphological and phylogeny of Plenodomus sinensis and P. collinsoniae, two unreported species isolated from soil in Korea

  • Two unreported fungal isolates, KNU-GW1901 and KNU-AP100C, were collected from soil sample in Gyeongsangbuk-do, Korea. Their cultural and morphological characteristics were examined after 4 weeks of incubation at 25℃ on potato dextrose agar (PDA), malt extract agar (MEA), and oatmeal agar (OA). The conidial shape of KNU-GW1901 was aseptate, hyaline, globose to ellipsoidal, oblong, and reniform to pyriform and 2.61-4.97×1.93-3.61 ㎛ in size, whereas no conidial structures were observed in KNU-AP100C. The internal transcribed spacer (ITS) regions, large subunit (LSU), and small subunit (SSU) sequences were used to determine the taxonomic positions of the strains using the maximum likelihood phylogenetic tree. The isolate KNU-GW1901 was closely clustered with Plenodomus sinensis MFLUCC 17-0767, and KNU-AP100C was closely matched with P. collinsoniae CBS 120227. Based on the findings of morphological, cultural, and phylogenetic analysis, the isolates KNU-GW1901 and KNU-AP100C were identical to the previously described P. sinensis and P. collinsoniae isolates, respectively, which are first reported in Korea.

Isolation and Identification of Two Unreported Fungi in Korea: Dothidea insculpta and Metarhizium rileyi

  • In this study, the fungal strains KNU-Gunwi 2B and KNU-SOT5 were isolated from root-soil in a hillside and the cherry tree bark (Prunus serrulata), respectively from Gyeongbuk province in Korea. The strain KNU-SOT5 produced dark brown chlamydospores that were smooth to lightly rough-walled, globose to ellipsoidal, and the conidia were aseptate, guttulate, mostly fusiform with a diameter of 5.3-17.6×4.2-7.0 ㎛. Strain KNU-Gunwi 2B produced phialides that were smooth-walled, cylindrical with semi-papillate apices and the conidia were pale-green, broadly ellipsoid, and sometimes cylindrical with a diameter of 4.4-8.0×2.3-4.0 ㎛. The strain KNU-SOT5 and KNU-Gunwi 2B were resolved based on cultural and morphological characteristics, along with the phylogenetic analysis using the small subunit (SSU), large subunit (LSU), and internal transcribed spacers (ITS) regions. The fungal strains KNU-SOT5 and KNU-Gunwi 2B were identified as Dothidea insculpta and Metarhizium rileyi, which have not been reported in Korea.

Ruvbl1 is Essential for Ciliary Beating during Xenopus laevis Embryogenesis

  • The Ruvb-like AAA ATPase1 (Ruvbl1; also known as Pontin) is an evolutionary conserved protein belonging to the adenosine triphosphates associated with diverse cellular activities (AAA+) superfamily of ATPases. Ruvbl1 is a component of various protein supercomplexes and is involved in a variety of cellular activities, including chromatin remodeling, DNA damage repair, and mitotic spindle assembly however, the developmental significance of this protein is unknown and needs detailed investigation. We investigated the developmental significance of Ruvbl1 in multiciliated cells of the Xenopus laevis epidermis since ruvbl1 is expressed in the multiciliated cells and pronephros during X. laevis embryogenesis. The knockdown of ruvbl1 significantly impaired cilia-driven fluid flow and basal body polarity in the X. laevis epidermis compared to control embryos, but did not affect cilia morphology. Our results suggest that Ruvbl1 plays a significant role in embryonic development by regulating ciliary beating; however, further investigation is needed to determine the mechanisms involved.

Evaluation of Tomato Genetic Resources for the Development of Resistance Breeding Lines against Late Blight (잎마름역병 저항성 육종을 위한 토마토 유전자원의 저항성 평가)

  • Occurrence of tomato late blight (Phytophthora infestans) has caused significant losses in tomato yield in all over the world. Evaluation of the level of resistance in tomato gene resources for main breeding and initiation of the resistance breeding program are important for control of this disease. Resistant assay of 78 tomato cultivars/lines to late blight in pots and field experiment was carried out under controlled and natural conditions in 2009. All commercial cultivars including 'Legend' were susceptible. However, 10 lines including KNU-2, KNU-6-1, KNU-11, KNU-13, KNU-14-1 lines distributed from University of California, Riverside and L3708, $AV107-4{\times}L3708$, $07-15{\times}L3708$, $BS67{\times}L3708$ lines which have resistant gene Ph-3 and $06-9-62A{\times}06-9-62A$ were highly resistant to late blight. These highly resistant lines can be used as resources of resistance to late blight in a tomato breeding program in future.

PRISM-KNU Development and Monthly Precipitation Mapping in South Korea (PRISM-KNU의 개발과 남한 월강수량 분포도 작성)

  • In this study, the parameter-elevation regressions on independent slopes model-Kongju National University(PRISM-KNU) system was developed to interpolate monthly precipitation data. One of the features of PRISM-KNU is that it can adjust the allowable range of slope according to the elevation range in the equation representing a linear relationship between the precipitation and elevation. The parameter value of the model was determined by using the optimization technique, and the result was applied to produce monthly precipitation data with a spatial resolution of $1{\times}1km$ from 2000 to 2014 in South Korea. In the result, the Kling-Gupta Efficiency for model evaluation was over 0.7 in 86% of the total cases simulated. In addition, a dramatic change in the spatial pattern of precipitation data was observed in the output of the Modified Korean PRISM, but such a phenomenon did not occur in the output of the PRISM-KNU. This study confirmed the appropriateness of the PRISM-KNU, and the result showed that the spatial consistency of the data produced by the model improved compared with that produced by the Modified Korean PRISM. It is expected that the PRISM-KNU and its output will be utilized in various studies in the future.
