Construction and Differential Screening of a cDNA Library Specific to Osmotic Stress of Haloxylon ammodendron Seedlings

  • A subtracted cDNA library specific to osmotic stress of Haloxylon ammodendron (Mey.) Bge seedlings was constructed by suppression subtractive hybridization (SSH) and T/A cloning. SSH was performed between two groups of H. ammodendron seedlings, one was cultivated in Hoagland (H) solution as a driver and the other group was treated with osmotic stress of the Hoagland solution by the addition of 400 mM mannitol (M), as a tester. The library consisted of about 400 recombinant clones, with the average size being of 500 bp, ranging from 300 bp to 1500 bp. Using a PCR-select differential screening kit, 100 recombinant clones were randomly chosen from the subtracted cDNA library and hybridized with forward,reverse subtracted and unsubtracted probes for two rounds. As a result, 21 positive clones specific to osmotic stress were obtained and some of them were verified by Northern blot analysis. The sequencing analysis of 6 positive clones and the following homology comparison to GenBank [blastx] non-redundant databases characterized that two sequences obtained in this experiment may contribute to novel drought-related genes.

A Study on the Quality Control of 2-site Immunoradiometric Assay of Serum Ferritin (혈청(血淸) ferritin 측정(測定)의 정도관리(精度管理)에 관(關)한 고찰(考察))

  • A 2-site immunoradiometric assay for serum ferritin was evaluated with commercially available kit. The assay required 6 hours. The slope of the standard curve kept up ideal range with the calculation of maxium binding instead of total dose until expire date. The stage II washing was more important than the stage I washing on the modified washing procedure as the bead keeping to remain in the tube. With this modified mothod, three times of tube. washing was sufficient to reduce the significant errors The measured values of serially diluted sample with standard diluting buffer was proportional to the predicted values. In the experiment of serum effect on the assay. a linear relationship from 5 to 50% serum, but beyond 50% there was reduction in measured ferritin concentration. It has a sensitivity of 2.77 ng/ml, within-assay precision (CV) of 8.0%, and between-assay reproducibility(CV) of 7.4% (mean 174.8 ng/ml).

Pregnancy Diagnosis in Sows by Using an On-Farm Blood Progesterone Test

  • To improve animal production, a simple and accurate pregnancy diagnosis plays a very important role. Therefore, the purpose of this study was to develop an on-farm blood progesterone enzyme immunoassay (EIA) system for monitoring the early pregnancy in sows. Star tubes coated with mouse monoclonal anti-progesterone antibody were used for this proposed EIA system which was tested in field trials. The results could be obtained within 30 minutes either by spectrophotometry or the naked eye. Heparinized fresh blood samples collected from the ear vein of sows 17-22 days after breeding (day 0) were tested qualitatively to diagnose sows as pregnant or non-pregnant with high ( > 3 ng/ml) or low ($${{\leq_-}}3ng/ml$$) progesterone in the blood. To provided a double check data, plasma progesterone levels were also measured quantitatively by the same EIA system with some modification. Total agreement of diagnosis by the on-farm EIA kit and by farrowing or abortion from 128 tested sows was found to be 92.2% accuracy (93.1% on pregnant diagnosis and 83.3% on non-pregnant diagnosis). It was concluded that the on-farm EIA blood progesterone test is a very useful method for monitoring the early pregnancy status of sows.

Isolation and In vitro Culture of Pig Spermatogonial Stem Cell

  • The present study identified the favorable conditions for isolation, enrichment and in vitro culture of highly purified, undifferentiated pig spermatogonial stem cell (SSC) lines that proliferate for long periods of time in culture. The colonies displayed morphology similar to miceSSC and were positive for markers of SSC (PGP9.5), proliferating germ cell (PigVASA), pre-meiotic germ cell (DAZL) and pluripotency (OCT4, SSEA-1, NANOG, and SOX2) based on immuno-cytochemistry and RT-PCR. The purity of these colonies was confirmed by negative expression of markers for sertoli cell (GATA4 and SOX9), peritubular myoid cell (${\alpha}$-SMA), differentiating spermatogonial and germ cells (c-KIT). The colonies could be maintained with undifferentiated morphology for more than two months and passaged more than 8 times with doubling time between 6-7 days. Taken together, we conclude that pigSSC could be successfully isolated and cultured in vitro and they possess characteristics similar to miceSSC.

Isolation and Characterization of Putrescine-producing Bacteria in Commercially Available Sauces Made from Salted and Fermented Sand Lance Ammodytes personatus (시판 까나리(Ammodytes personatus) 액젓에서 Putrescine 생성균의 분리 및 특성)

  • Bacterial decarboxylation of amino acids in food leads to the production of biogenic amines, which can cause reactions in human that include headaches, nausea, palpitations, chills, and severe respiratory distress. The amine putrescine is an especially effective inhibitor of metabolizing enzymes and amplifies histamine intoxication and tyramine poisoning. Using an L-ornithine decarboxylating medium, we isolated 14 putrescine-producing bacteria from sand lance, Ammodytes personatus, sauces. The isolates were identified, using an API kit and 16S rRNA analysis, as Lysinibacillus fusiformis (1 strain), Lysinibacillus xylanilyticus (6 strains), Lysinibacillus macroides (1 strain), Lysinibacillus sphaericus (3 strains), Bacillus fusiformis (1 strain), Paenibacillus favisporus (1 strain), and Staphylococcus caprae (1 strain). These strains produced between 1.66 to 236.97 μg/mL of putrescine after 48 h incubation. Lysinibacillus spp. were the dominant putrescine-producing bacteria in sand lance sauces, which produced 236.97 μg/mL of putrescine from a culture broth containing 0.5% L-ornithine. This is the first report on the isolation and identification of putrescine-producing bacteria from sand lance sauces.

Synthetic Peptide-Based Enzyme-Linked Immunosorbent Assay for Human $\alpha$-Fetoprotein

  • $\alpha$-Fetoprotein(AFP) is a good marker for the detection of several diseases such as hepatocellular carcinoma, gonadal germ cell tumor, gastric tumor, and Down's syndrome. In this study, we developed ELISA, using synthetic peptides corresponding to the epitopes of AFP. Five kinds of peptides were synthesized from AFP to produce antibodies in rats that recognize AFP in human plasma as well as amniotic fluid and do not cross-react with serum albumin. All five kinds of antibodies showed good reactivities with their peptide-keyhole limpet hemocyanin conjugates. Anti-synthetic peptide 1 (R-N-E-Y-G-I-A-S-I-L, 4-13) antibody, in particular, reacted well with AEP as well as synthetic peptide 1-KLH but not with human serum albumin. The binding affinity(Kd) was 2.7$\times$10$^{-9}$M for peptide 1 and 6.8$\times$10$^{-8}$M for AEP. The range for measurement of AFP was 10~1,000 ng/ml. The within-assay and between-assay coefficients of variance(CV) were 4.83% and 10.97%, respectively. In a sample of 31 sera and 33 amniotic fluids, there was a good correlation between AFP values determined in this assay and those in a commercial kit. These results indicate that the antibodies against synthetic peptides corresponding to the epitopes of AFP are highly specific to APP and synthetic peptide-based ELISA would be useful for the measurement of human AFP.

Optimal Harvest-Use-Store Design for Delay-Constrained Energy Harvesting Wireless Communications

  • Recent advances in energy harvesting (EH) technology have motivated the adoption of rechargeable mobile devices for communications. In this paper, we consider a point-to-point (P2P) wireless communication system in which an EH transmitter with a non-ideal rechargeable battery is required to send a given fixed number of bits to the receiver before they expire according to a preset delay constraint. Due to the possible energy loss in the storage process, the harvest-use-and-store (HUS) architecture is adopted. We characterize the properties of the optimal solutions, for additive white Gaussian channels (AWGNs) and then block-fading channels, that maximize the energy efficiency (i.e., battery residual) subject to a given rate requirement. Interestingly, it is shown that the optimal solution has a water-filling interpretation with double thresholds and that both thresholds are monotonic. Based on this, we investigate the optimal double-threshold based allocation policy and devise an algorithm to achieve the solution. Numerical results are provided to validate the theoretical analysis and to compare the optimal solutions with existing schemes.

Box Cathode Sputtering Technologies for Organic Optoelectronics (유기물 광전소자 제작을 위한 박스 캐소드 스퍼터 기술)

  • We report on plasma damage free-sputtering technologies for organic light emitting diodes (OLEDs), organic thin rim transistor (OTFT) and flexible displays by using a box cathode sputtering (BCS) method. Specially designed BCS system has two facing targets generating high magnetic fields ideally entering and leaving the targets, perpendicularly. This target geometry allows the formation of high-density plasma between targets and enables us to realize plasma damage free sputtering on organic layer without protection layer against plasma. The OLED with top cathode prepared by BCS shows electrical and optical characteristics comparable to OLED with thermally evaporated Mg-Ag cathode. It was found that TOLED with ITO or IZO top cathode layer prepared by BCS has much lower leakage current density ($1\times10^{-5}$ mA/cm2 at -6V) than that ($1\times10^{-1}\sim10^{\circ}mA/cm^2$)of OLED prepared by conventional DC sputtering system. This indicates that BCS technique is a promising electrode deposition method for substituting conventional thermal evaporation and dc/rf sputtering in fabrication process of organic based optoelectronics.

Development of a Quality Manual for a Research and Educational Centre: A Case Study

  • There have been growing concerns about whether proper measures are in place to ensure the delivery of quality services and the operations in research and educational institutions. This paper describes various quality models and discusses their relevance into the total quality concepts and quality management systems (QMS) in education. It then illustrates how a QMS and the accompanied quality manual be developed in a research and educational institution using the Centre for Enterprise Research Integration (ERI) at The University of the West Indies, Trinidad as a case. Through personal interviews with the Centre's personnel, views on the operations and determinants of the QMS were acquired. Results showed that the people's resistance to changes, conflicts on academic freedom, and difficulties on measuring performance were the obstacles to QMS implementation. People involvement and proper communication are determinants of QMS implementation. The system also rests on having a tailored quality manual that entails the documentation of processes and procedures for ascertaining the delivery of quality services and operations in the Centre. The development of quality manual is briefly described in the paper.

Cross-sectional study: prevalence of subclinical ketosis in dairy cattle at Chungnam province

  • This study was conducted to demonstrate the prevalence of subclinical ketosis in dairy cattle based on days in milk and herd level. Four to five blood samples were collected in five different lactation periods and analyzed for ${\beta}$-hydroxybutyrate using a Portable Ketone Test Kit. Subclinical ketosis was observed in 12 herds at prevalence of 6.3~57.1% depending on herd level, with four herds showing more than 40% subclinical ketosis prevalence. The distribution of subclinical ketosis in the early lactation period, high yield lactation period, mid lactation period, late lactation period and dry period was 9.5%, 27.5%, 36.8%, 27.9% and 12.5%, respectively. Clinical ketosis was only observed during early lactation (9.5%) and mid lactation (2%) period.
