  • Development of hematite-coated sand was evaluated for the application of the PRB (permeable reactive barrier) in the arsenic-contaminated subsurface of the metal mining areas. The removal efficiency of As(III) and As(V), the effect of anion competition and the capability of arsenic removal in the flow system were investigated through the experiments of adsorption isotherm, arsenic removal kinetics against anion competition and column removal. Hematite-coated sand followed a linear adsorption isotherm with high adsorption capacity at low level concentrations of arsenic (< 1.0 mg/l). When As(III) and As(V) underwent adsorption reactions in the presence of anions (sulfate, nitrate and bicarbonate), sulfate caused strong inhibition of arsenic removal, and bicarbonate and nitrate caused weak inhibition due to specific and nonspecific adsorption onto hematite, respectively. In the column experiments, high content of hematite-coated sand enhance the arsenic removal, but the amount of the arsenic removal decreased due to the higher affinity of As(V) than As(III) and reduced adsorption kinetics in the flow system, Therefore, the amount of hematite-coated sand, the adsorption affinity of arsenic species and removal kinetics determined the removal efficiency of arsenic in the flow system. arsenic, hematite-coated sand, permeable reactive barrier, anion competition, adsorption.

  • Removal characteristics of Mn and Co was studied from the contaminated solutions via surface reaction with various calcium carbonate (calcite). Synthetic calcium carbonates which has different surface morphology as well as surface areas were prepared by a spontaneous precipitation method and used. Mn and Co removal behavior by the different solid surface demonstrate characteristic sorption behaviors depend on the type of calcite used, such as surface area or surface morphology. Calcium carbonate crystals (mostly calcite) which exhibit complicated surface morphology (c-type) shows strong sorption affinity for Mn and Co removal via sorption than on the a-type or b-type calcite crystals of less complicated surfaces. The applicability of two kinetic models, the pseudo-first-order kinetic equation and the Elovich kinetic model was examined on these sorption behavior. Elovich kinetic model was found more suitable to explain the very early stage adsorption kinetics, while the pseudo-first-order kinetic equation was successfully fitted for the adsorption kinetics after 50 hours.

Retarding Retrogradation of Korean Rice Cakes(Karedduk) with a Mixture of Trehalose and Modified Starch Analyzed by Avrami Kinetics (Avrami Kinetics에 적용한 트레할로스와 변성 전분 혼합 사용 떡의 노화 억제 분석)

  • Retarding retrogradation of Korean rice cakes(Karedduk) with a mixture of trehalose and Sun-Tender added, after 0, 24, and 48 hr of storage at $5^{\circ}C$, was analyzed by Avrami kinetics. A central composite design was used for arrangement of treatment. The two independent variables selected for retarding retrogradation analysis were amounts of trehalose(x) and Sun-Tender(y). Trehalose was added at 0, 3, 6, 9, and 12% levels, and Sun-Tender added at 0, 0.3, 0.6, 0.9, and 1.2% levels, to dry rice flour. The Avrami exponent(n) for the mixtures of 9% trehalose and 0.3% Sun-Tender, and 9% trehalose and 0.9% Sun-Tender were lower than in the control. The time constant(1/k) for the mixture of trehalose and Sun-Tender was higher than in the control. The effect of retarding retrogradation of Korean rice cakes with added mixtures of trehalose and Sun-Tender showed an increasing trend as the amount of trehalose increased. These results suggest that adding a mixture of 9% trehalose and 0.3% Sun-Tender, or 9% trehalose and 0.9% Sun-Tender to Korean rice cakes(Karedduk) is effective for retarding retrogradation.

Sorption of Chromium Ions from Aqueous Solution onto Chemically Activated Carbons Developed from Maize Cobs

  • Chemically activated carbons were prepared from maize cobs, using phosphoric acid of variable concentration. The textural parameters of the activated carbons were determined from the nitrogen adsorption isotherms measured at 77 K. The chemistry of the carbon surface was determined by measuring the surface pH, the pHPZC and the concentration of the carbon - oxygen groups of the acid type on the carbon surface. Kinetics of Cr(VI) sorption/reduction was investigated at 303 K. Two processes were investigated in terms of kinetics and equilibrium namely; Cr(VI) removal and chromium sorption were studied at various initial pH (1-7). Removal of Cr(VI) shows a maximum at pH 2.5. At pH<2.5, sorption decreases because of the proton competition with evolved Cr(III) for ion exchange sites. The decrease of sorption at pH>2.5 is due to proton insufficiency and to the decrease of the extent of Cr(VI) reduction. The chemistry of the surface of activated carbon is an important factor in determining its adsorption capacity from aqueous solutions particularly when the sorption process involves ion exchange.

  • Magnesium and its alloys have the high potential as hydrogen storage materials because of their highest hydrogen storage capacity, low density and abundant resources. But poor kinetic properties of hydriding and dehydriding and high working temperature have limited their practical applications. In this study, the Mg-Ni binary alloys with different amount of Ni were produced by gravity casting and characterized in order to investigate the relationship between the microstructures and hydriding properties. The maximum hydrogen absorption capacity decreased, but the absorption kinetics increased with Ni content. The difference in the absorption kinetics was resulted from the differences in the sort and shape of primary solid phases and eutectic microstructure.

  • Recently, one of the hydrogen production methods has attracted using dense metallic membrane. It has high hydrogen permeation and selectivity which hardly could adopt industrial product because of high cost, hydrogen embrittlment and thermal stability. Meanwhile, vanadium has high hydrogen solubility and it use to instead of Pd-Ag amorphous membrane. Aluminum carried out blocking hydrogen diffusion on grain boundary therefore protecting hydrogen embrittlement. Most of dense metallic membrane is solution diffusion mechanism. The solution diffusion mechanism was very similar hydrogen storing steps such as steps of metal hydride. Thus, V-Al composites were fabricated to use hydrogen induced mechanical alloying. The fabricated V-Al composites were characterized by XRD, SEM, EDS and simultaneous TG/DSC analyses. The hydrogenation behaviors were evaluated using a Sievert's type automatic PCT apparatus. The hydrogenation behaviors of V-Al composites was evaluated too low hydrogen stored capacity and fast hydrogenation kinetics. In PCI results, V-Al composites had low hydrogen solubility, in spite of that, hydrogen kinetics was calculated very fast and hydrogen absorption/desorption contents were same capacity.

글루타치온 생산효소( $\gamma$-Glutamylcysteine Synthetase)와 그 변이효소의 구조분석 및 반응 Kinetics 연구

  • Two mutant enzymes of $\gamma$-glutamylcysteine synthetase ($\gamma$-GCS) which catalyzed the synthesis of $\gamma$-glutamylcysteine from L-glutamic acid and L-cysteine in the presence of ATP, were prepared bypoint mutation of $\gamma$-GCS gene with site-directed mutagensis in E. coli. Conformational structuresand catalytic reaction kinetics of mutant enzymes were compared with wild type $\gamma$-GCS afterpurification. The S495F mutant enzyme (serine at 495 residue was substituted with phenylalanine),which had no catalytic activity for $\gamma$-glutamylcysteine synthesis, rarely folded even in neutral pH.However, the mutant A494V (alanine of 494 residue was replaced by valnine) which showed 50 %increase of activity, had a high folding structure. The folding structure of A494V also more stable athigh temperature and extreme pH compared to wild type and S495F. Reaction kinetics of wild typeand A494V were also investigated, Km value of A494V was smaller than that of wild type, while itshowed a little difference at Vmax values. This result evolved that alanine at 494 may be involved inbinding site of substrate rather than catalytic site. In addition, change of catalytic activity by onepoint mutation was highly correlated with the folding structure of enzyme.

Effect of Hydroquinone on Ruminal Urease in the Sheep and its Inhibition Kinetics in vitro

  • Effect of hydroquinone (HQ) on rumen urease activity was studied. Hydroquinone at concentrations of 0.01 ppm, 0.1 ppm, 1 ppm, and 10 ppm inhibited urease activity of intact rumen microbes in vitro by 25%, 34%, 55% and 64% respectively. In the presence of low concentrations of $\beta$-mercaptoethanol, rumen urease could be solubilized and partially purified. The Km for the enzyme was $2{\times}10^{-3}$ M with Vmax of $319.4{\mu}moles/mg$ min. The kinetics of inhibition with partially purified rumen urease was investigated. The result showed that the inhibitory effect was not eliminated by increasing urea concentrations indicating a noncompetitive effect in nature with an inhibition constant $1.2{\times}10^{-5}$ M. Hydroquinone at the concentration of 10 ppm produced 64% urease inhibition, did not affect ruminal total dehydrogenase and proteolytic enzyme (p>0.05), but increased cellulase activity by 28% (p<0.05) in vitro. These results indicated that hydroquinone was a effective inhibitor of rumen urease and could effectively delay urea hydrolysis without a negative effect. The inhibitor appeared to offer a potential to improve nitrogen utilization by ruminants fed diets containing urea.

A Study on Hydration kinetics and Mechanical Properties of Cement Paste Incoporating Limestone Filler (석회석 미분말을 혼입한 시멘트 페이스트의 수화반응 및 역학적 특성 분석에 관한 연구)

  • The addition of a limestone filler(LF) to fill into the voids between cement and aggregate particles can reduce the cementitious paste volume. This paper aim to evaluate the influence of LF contents on the hydration kinetics and compressive strength. Hydration kinetics were evaluate using heat of hydration, ignition loss and thermal analysis. The heat of hydration was measured using Isothermal Calorimetry. The degree of hydration was measured using ignition loss. Hydration product analysis was carried out by Thermal Gravimetric and Differential Thermal Analysis. The results show that the addition of LF reduces not only the initial setting time and heat of hydration peak, also degree of hydration and rate of strength development at early age increase with the addition of LF. It can be concluded the LF fills the pore between cement particles due to formation of carboaluminate, which may accelerate the setting of cement pastes.

  • As the consumption of galvanized steels in cans and automobiles and the quantity of scraps increase, the recycling problems of EAF dust become a important problem. Valuable metals such as Fe, Zn, Pb are of continued interest to metallurgists. To recover the valuable metal and to remove the toxicity of EAF dust, high temperature smelting process is or researching as a pilot scale. The Reduction kinetics of Zn in EAF dust is so important in a view of the economic consideration of the process. In this study, the kinetics behavior of Zn in EAF dust were measured as a point of application in high temperature smelting process. The rate control step in ZnO and franklinite is revealed to be chemical reaction on the reaction surface.
