Metamorphism of the amphibolites in the Hwanggangri area, the northeastern region of Ogcheon metamorphic belt, Korea (옥천변성대 북동부 황강리 지역내 앰피볼라이트의 변성작용)

  • In the amphibolites of the Hwanggangri area, three metamorphic zones are established like hornblende-actinolite zone (H-AZ), hornblende zone (HZ) and diopside zone (DZ) by the main mineral assemblages. Hornblende zone and hornblende-actinolite zone develope away from the diopside zone that experienced the highest thermal effect. Thus, this pattern identifies the decreasing metamorphic grade of the contact metamorphism with increasing distance from the granitic pluton. The mineral assemblages of this rock are classified into six representative groups such as $\circled1$ actinolite+plagioclase+chlorite, $\circled2$ actinolite+hornblende+plagioclase+chlorite$\pm$epidote$\pm$biotite, $\circled3$ actinolite+hornblende+plagioclass$\pm$biotite$\pm$epidote, $\circled4$ hornblende+plagioclase$\pm$biotite$\pm$chlorite, $\circled5$ hornblende+plagioclase+diopside+actinolite$\pm$epidote$\pm$chlorite, $\circled6$hornblende+plagioclase+diopside$\pm$biotite$\pm$epidote. Two metamorphic events m recognized in the amphibolites of the study area that the first metamorphism is the regional metamorphism dominantly occurred in the whole Ogcheon metamorphic belt and it gave rise to the growth of actinolite at the core or center of the amphibole grains of coarse and medium size. Its metamorphic grade ranges from the greenschist facies to epidote-amphibolite facies. The second metamorphism overlapped is the contact metamorphism caused by the adjacent granitic pluton, and its metamorphic grade is thought to reach to the low pressure part of upper amphibolite facies. According to the calculation by TWEEQU thermobarometry and amphibole-plagioclase thermometry, the metamorphic temperature of initial regional metamorphism is $439-537^{\circ}C$ under pressure of 4.6-7.3 kb and its peak temperature and pressure are considered to reach to the range of 492-537 and 5.2-7.3 kb. And the temperature range of contact metamorphism occurred by intrusion of cretaceous granitic body, is $588-739^{\circ}C$ under pressure of 2.6-5.2 kb and its peak temperature and pressure are estimated as having the range of $697-739^{\circ}C$ and 3.8-5.2 kb that this amphibolites are estimated to pass through the metamorphic evolution of both the rise of temperature and the drop of pressure.

The Influence of Pretreatment Period, 2-Hydroxynicotinic Acid and Anther Co-pretreatment on Embryo Induction in Isolated Microspore Culture of Capsicum annuum L. (고추의 나출 소포자 배양시 전처리 기간, 2-Hydroxynicotinic Acid 및 약-공동전처리가 소포자배 발생에 미치는 영향)

  • Microspores were isolated from pepper (Capsicum annuum L.) anthers by using a micro-blender and cultured in modified NLN medium at $25^{\circ}C$. The influence of pretreatment period at $32^{\circ}C$, adding the 2-hydroxynicotinic acid to a pretreatment medium, and co-pretreatment anthers with microscopes on the induction of embryo were examined. Globular and torpedo embryos were observed from 3 weeks after culture. Embryo development was not synchronized within culture. After 4 weeks in culture, in addition to globular and torpedo embryos, cotyledonary embryos were observed. Normal cotylodonary embryos developed into plantlets when transferred to a solid hormone free B5 medium containing $2\%$ sucrose. Embryo yields were significantly higher after 1- and 2-day pretreatment at $32^{\circ}C$. However the development of embryo ceased at the globular or heart stage. In contrast, embryo yields were lower after 3- to 6-day pretreatment at $32^{\circ}C$ and embryo developed at the cotyledonary stage. After adding the 2-hydroxynicotinic acid to anther pretreatment solution, embryo yields were slightly increased. However most embryos occurred were at the globular or heart stage. Co-pretreatment of microspores with anthers was deleterious for embryo induction and development. AS far as we know, this is the first report of success in obtaining high frequency of embryogenesis and plantlets formation from isolated microspores of pepper. Although the culture conditions have to be optimized further, this promising microspore culture system can be used for genetic transformation, selection for dominant and recessive traits as well as for the production of homozygous doubled haploid plants.

Effects of Cloned Genes on the Stability of Shuttle Vectors between Escherichia coli and Corynebacterium glutamicum (Escherichia coli와 Corynebacterium glutamicum간의 shuttle vectors의 C. glutamicum에서의 안정성에 대한 클론된 유전자의 영향)

  • Escherichia coli/Corynebacterium glutamicum shuttle vectors, pECCG1 and pECCG2 were constructed by joining a 3.00 kb cryptic plasmid pCB 1 from C. glutamicum and a 3.94 kb plasmid pACYC 177 from E. coli. By trimming unessential parts and introducing mulitiple cloning site into the plasmid pECCG 1, a plasmid pECCG122(5.1kb) was constructed. All the shuttle vectors were stably maintained in C. glutamicum up to about 40 generations irrespective of kanamycin addition in the medium. Threonine operon (homoserine dehydrogenase/homoserine kinase) and dapA gene (dihydrodipicolinate synthetase) of C. glutamicum were cloned into the plasmid pECCG122, and the resultant plasmids were designated pTN31 and pDHDP19, respectively. They were used to study the effect of cloned foreign gene on the stability of the plasmid pECCG122. Plasmids pTN31 and pDHDP19 were segregated rapidly from C. glutamicum when cultured in the medium without kanamycin. In medium with $50\mu${\g/ml} of kanamycin, their segregation rates were much slower than those in medium without kanamycin, but the danamycin addition didn't guarantee the complete maintenance of the plasmids in C. glutamicum.

Antioxidative Activities and Antiproliferation Effects on Oral Carcinoma KB Cell of the Brazilin from Caesalpinia sappan L. (소목(Caesalpinia sappan L.)에서 분리한 Brazilin의 항산화 활성과 구강상피암 KB 세포주의 증식억제효과)

  • Caesalpinia sappan L. has long been commonly used in oriental folk medicines and as dyes materials. To investigate the antioxidative activities and antiproliferation effects of brazilin from C. sappan heart wood, the MeOH soluble extract was successively fractionated by using hexane, $CHCl_{3}$, EtOAc, BuOH, MeOH, and $H_{2}O$. In these fractions, we were purified brazilin from EtOAc fraction which partitioned to 3.94% of the highest yields. The effects of brazilin and the extracts on human oral carcinoma cells (KB) by MTT assay and their antioxidant activities by DPPH, TCA assay and Fenton reaction were tested. The results showed that the brazilin could inhibits the proliferation of KB cells and obviously decreased the production of nitric oxide of the cells. When the concentration of the brazilin reached to $100\;{\mu}g/ml$, the inhibition percentage of the cell growth was about 60%. In assay on antioxidant activities, The results showed that brazilin exhibit the highest capacity of DPPH free radical scavenging effects among tested extracts. When the concentration of brazilin reached to 1 mg/ml, the lipid peroxide inhibition and radical inhibition activities were determined to be 65.0% and 85.8%, respectively. These results are suggest that brazilin have stronger antiproliferation effect on KB cell and antioxidant properties.

  • The cytotoxic metabolites, against the KB cell line, halenaquinone, epispongiatriol and aldisin were isolated from the sponge Spongia sp. collected in September 1992, Manado Bay, Sulawesi in Indonesia. Their structures were elucidated by 1H, 13C NMR, 1H 13C(1 bond) Heteronuclear Multiple Quantum Coherence Spectroscopy (HMQC), 1H 13C(2 and 3 bond) Heteronuclear Multiple Bond Correlation Spectroscopy (HMBC), Electron Impact Mass Spectroscopy (EI ms) and Infrared Spectroscopy (IR).

  • A 5.7Kb EcoRI fragment containing alkaline amylase gene of Bacillus sp. AL-8 obtained in the previons experiment (10) was transformed in B. subtilis via plasmid pUB110. The enzymatic proper-ties of the amylase produced by the transformants were Identical to those of the donor strain. Thus, the alkaline amylase activity from the transformant was maximum at pH 10 and 5$0^{\circ}C$. And the enzyme was very stable over the ranges of alkaline pH. In order to determine the location of the alkaline amylase gene within the 5.7Kb DNA fragment, the fragment was subcloned in E. coli. It was found that the alkaline amylase gene was located k EcoRI fragment of 3.7Kb.

Role of Chitinase Produced by Chromobacterium violaceum in the Suppression of Rhizoctonia Damping-off (모잘록병(Rhizoctonia solani)의 억제에 있어서 Chromobacterium violaceum이 생산하는 Chitinase의 역할)

  • To determine whether chitinolytic enzymes from Chromobacterium violaceum C-61 play an important role in the suppression of Rhizoctonia damping-off, Tn5 insertion mutants deficient in chitinolytic activity (Chi a- mutants) were selected and their chitinolytic and disease suppression were compared with those of the parental strain. Four Chi a- mutants selected from about 2,000 transconjugants did not inhibit mycelial growth of Rhizoctonia solani on nutrient agar-potato dextrose agar (BA-PDA) and their abilities to suppress Rhizoctonia damping-off were much lower than the parental strain. However, population density in the eggplant rhizosphere did not differ significantly between the parental strain and four Chi a- mutants. The crude enzyme of the parental strain inhibited growth of R. solani on NA-PDA and its chitinase activity was much higher than that of Chi a- mutants. But the N,N' -diacetylchitobiase activity between these isolates were not significantly different. The chitinase of Chi a- mutants was defective in 2 isoforms of 52- and 37-kDa among four isoforms of 54-, 52-, 50- and 37-kDa. A Tn5 element was inserted into one site of 10 kb EcoRI fragment of chromosomal DNA in three Chi- mutants, C61-C1, -C2, and -C3. In C61-C4 mutant, a Tn5 element was inserted into two sites of 10 kb and 4.4 kb EcoRI fragments. These results suggest that the chitinase of C. violaceum C-61 play an important role in the suppression of Rhizoctonia damping-off of cucumber and eggplant.

Isolation of Proteinase Inhibitor II Genes from Potato (감자로부터 단백질분해효소 억제제 II 유전자의 분리)

  • Southern hybridization of genomic DNAs with radioactively labeled cDNA of tomato proteinase inhibitor II revealed that proteinase inhibitor II proteins in potato plants are encoded by a family of about 10 related sequences. Screening of potato EcoRI genomic library with the cDNA resulted in isolation of 13 recombinant phage clones which carry 3 different genomic regions. Of these clones, clones 8, 18, and 39 were subjected to restriction mapping and subcloning. Further characterization of the subclones of clones 8, 18 and 39 indicated that two inhibitor II genes are present on a 8.0 kb EcoRI fragment of clone 8, one on 3.3 and 0.8 kb EcoRI fragments of clone 18 and two genes on a 13.5 kb EcoRI fragment of clone 39.

Metamorphic Evolution of the central Ogcheon Metamorphic Belt in the Cheongju-Miwon area, Korea (청주-미원지역 중부 옥천변성대의 변성진화과정)

  • In the Cheongju-Minwon area which occupies the middle part of the Ogcheon Metamorphic Belt, three metamorphic events(M1, M2, M3) had occurred. Intermediate P/T type M2 regional metamorphism formed prevailing mineral assemblages in the study area. Low PIT type M3 contact metamorphism occurred due to the intrusion of granites after M2 metamorphism. M1 metamorphism is recognized by inclusions within garnet. During M2 metamorphism, the metamorphic grade increased from the biotite zone in the southeastern part to the garnet zone in the northwestern part of the study area. This result is similar to the metamorphic evolution of the southwestern part of the Ogcheon Metamorphic Belt. Garnets in the garnet zone are classified into two types; Type A garnet has inclusions whose trail is connected to the foliation in the matrix and Type B garnet has inclusion rich core and inclusion poor rim. Type A garnet formed in the mica rich part with crenulation cleavage whereas Type B garnet formed in the quartz rich part with weak crenulation cleavage. In some outcrops, two types garnets are found together. Compared to the rim of Type A garnet, the rim of Type B garnet is lower in grossular and spessartine contents but higher in almandine and pyrope contents. In some Type B garnets, the inclusion poor part is rimmed by muddy colored or protuberant new overgrowth. In the inclusion poor part and new overgrowth, a rapid increase in grossular and decrease in spessartine is observed. However, the compositional patterns of Type A and B are similar; Ca increases and Mn decreases from core to rim. Two types garnets formed mainly due to the difference of bulk chemistry instead of metamorphic and deformational differences. The metamorphic P-T conditions estimated from Type A garnets are 595-690 OC15.7-8.8 kb, which indicates M2 metamorphism is intermediate P/T type metamorphism. On the other hand, a wide range of P-T conditions is calculated from Type B garnets. The P-T conditions from most Type B garnet rims are 617-690 OC16.2-8.9 kb which also indicates an intermediate P/T type metamorphism. However, at the rim part with flat end or weak overgrowth, grossular content is low and 573-624OC14.7-5.8 kb are estimated. The P-T conditions calculated from plagioclase and biotite inclusions in garnet are 460-500 0C/1.9-3.0 kb. The P-T conditions from rim part with weak overgrowth and inclusions within garnet, indicate that low P/T type M1 regional metamorphism might have occurred before intermediate P/T type M2 regional metamorphism. The P-T conditions estimated from samples which had undergone low PIT type M3 metamorphism strongly, are 547-610 0C/2.1-5.0 kb.

Granulite xenoliths in porphyroblastic gneiss from Mt. Jiri area, SW Sobaegsan massif, Korea (소백산육괴 서남부 지리산지역의 반상변정질 편마암에서 산출되는 백립암질 포획암)

  • Mafic granulite xenoliths are found in precambrian porphyroblastic gneiss of the Mt. Jiri area, SW Sobaegsan massif, Korea. The xenoliths are rounded to ellipsoidal in shape, 50-100 cm in length and coarse-grained with granoblastic and foliated texture. The xenoliths consist of orthopyroxene, garnet, biotite, plagioclase, quartz, ilmenite and secondary orthoamphibole. Orthopyroxene is mostly resorbed and rimmed by coronitic orthoamphiboles. Garnets occur as porphyblasts and are zoned with higher pyrope content in cores than in rims. Geothermo-barometry results yield conditions of about $800-850^{\circ}C$, 6 kb and $500^{\circ}C$, 4 kb for early and retrograde stages of equilibration, respectively. According to available geochronological data, it is suggested that the granulite facies metamorphism occurred prior to 2.1-1.9Ga and that the area was superimposed by the high-grade (over $600-700^{\circ}C$) metamorphism between 1.9-1.7Ga, followed by cooling during uplift.
