Antimicrobial drug susceptibility and plasmid profiles of Salmonella species isolated from poultry (가금에서 분리한 Salmonella속 균의 항균물질에 대한 감수성 및 plasmid profile)

  • In this study, we aim to find the presence of virulence-related plasmid in Salmonella isolates from poultry, and the difference between S pullorum and S gallinarum on the plasmid profile and antibiotics resistance. We used seventeen isolates of Salmonella spp that were isolated from poultry. Thirteen isolates, S typhimurium(ST), S pullorum(SP) and S gallinarum(SG), contained virulence-related plasmids. These are 95Kd plasmid in ST and 85Kd plasmid in SP and SG. Three(1/4 of ST, 1/1 of SE, and 1/9 of SP) isolates have no detectable plasmids. The isolates of ST have relatively variable plasmid profile but the isolates of S pullorum except No 12(additional 3.0Kb plasmid) have common 85K6, 8.1Kb, 4.0Kb and 2.3Kb plasmid and two of three isolates of S gallinarum have common 85Kb, 4.0Kb and 2.3Kb plasmid but the rest has only 85Kb plasmid. Interestingly, all of the isolates of SP have 8.1Kb plasmid, and same size of plasmid is also found in one of ST isolates. All of the isolates have the resistance to penicillin, chloramphenicol, rifampicin, streptomycin, sulfamethazine and some isolates show the resistance to ampicillin and tetracycline. There is no relatedness between plasmid profile and antibiotics resistance and no differences between SP and SG in antibiotics resistance. Therefore further differentiation of each isolates by restriction enzyme assay and, if possible, charaterization of each plasmid, especially, 8.1Kb plasmid in SP and ST, may be necessary.

Manipulation of Mini-Yeast Artificial Chromosome Containing Xylan Metabolism Related Genes and Mitotic Stability Analysis in Yeast (Xylan 대사유전자를가진미니효모인공염색체의가공및 Mitotic Stability 분석)

  • In this study, yeast artificial chromosome Insert (YAC) harboring genes which related xylan metabolism was constructed by using chromosome manipulation technique. For efficient chromosome manipulation, each splitting fragment (DNA module) required for splitting process was prepared and these DNA modules were transformed into Saccharomyces cerevisiae strain YKY164. By two-rounds chromosome splitting, yeast chromosome VII (1,124 kb) was split 887 kb-YAC, 45 kb-mini YAC and 198 kb-YAC and YKY183 strain containing 18 chromosomes was constructed. Splitting efficiency for chromosome manipulation was 50- 78% and expression level of foreign genes on 45 kb-mini YAC and enzyme activity were indistinguishable from that of the YKY164 strain. Furthermore, xylan-degraded products by recombinant enzymes were confirmed and mini-yeast artificial chromosome maintained stable mitotic stability without chromosome loss during 160 generations.

KB-BERT: Training and Application of Korean Pre-trained Language Model in Financial Domain (KB-BERT: 금융 특화 한국어 사전학습 언어모델과 그 응용)

  • Recently, it is a de-facto approach to utilize a pre-trained language model(PLM) to achieve the state-of-the-art performance for various natural language tasks(called downstream tasks) such as sentiment analysis and question answering. However, similar to any other machine learning method, PLM tends to depend on the data distribution seen during the training phase and shows worse performance on the unseen (Out-of-Distribution) domain. Due to the aforementioned reason, there have been many efforts to develop domain-specified PLM for various fields such as medical and legal industries. In this paper, we discuss the training of a finance domain-specified PLM for the Korean language and its applications. Our finance domain-specified PLM, KB-BERT, is trained on a carefully curated financial corpus that includes domain-specific documents such as financial reports. We provide extensive performance evaluation results on three natural language tasks, topic classification, sentiment analysis, and question answering. Compared to the state-of-the-art Korean PLM models such as KoELECTRA and KLUE-RoBERTa, KB-BERT shows comparable performance on general datasets based on common corpora like Wikipedia and news articles. Moreover, KB-BERT outperforms compared models on finance domain datasets that require finance-specific knowledge to solve given problems.

Induction of Growth Inhibition by BCH in KB Human Oral Epidermoid Carcinoma Cells (구강 편평세포암종 KB세포에서 아미노산 수송억제제 BCH에 의한 세포성장 억제)

  • Amino acid transporters play an important role in supplying nutrients to normal and cancer cells for cell proliferation. System L is a major transport system responsible for the N $a^{+}$-independent, large neutral amino acids including several essential amino acids. L-type amino acid transporter 1 (LAT1), an isoform of system L amino acid transporter, is highly expressed presumably to support their continuous growth and proliferation in malignant tumors. 2-Aminobicyclo- (2,2,1) -heptane-2-carboxylic acid (BCH) is a model compound for study of amino acid transporter as a system L selective inhibitor. In the present study, we examined whether BCH induced growth inhibition in KB human oral squamous carcinoma cell line or not. The uptake of L-[$^{14}$ C]leucine by KB cells is inhibited by BCH in a concentration dependent manner with a Ι $C_{50}$ value of 75.3$\pm$6.2 ${\mu}{\textrm}{m}$ and a $K_{i}$ value of 98.7$\pm$ 4.1 ${\mu}{\textrm}{m}$. The growth of KB cells is inhibited by BCH in time dependent manner and concentration dependent manner with a Ι $C_{50}$ value of 11.1 $\pm$0.8 mM. In the DNA of KB cells treated with the various concentrations and various periods of BCH, the characteristic ladders associated with DNA fragmentation were not observed. These results suggest that BCH inhibits the growth of KB oral epidermoid carcinoma cells through the inhibition of transport of neutral amino acids into cells without DNA break down. This phenomenon will be a new rationale for anti-cancer therapy.y.

Differentiation of Four Major Gram-negative Foodborne Pathogenic Bacterial Genera by Using ERIC-PCR Genomic Fingerprinting (ERIC-PCR genomic fingerprinting에 의한 주요 식중독 그람 음성 세균 4속의 구별)

  • Widespread distributions of repetitive DNA elements in bacteria genomes are useful for analysis of genomes and should be exploited to differentiate food-borne pathogenic bacteria among and within species. Enterobacterial repetitive intergenic consensus (ERIC) sequence has been used for ERIC-PCR genomic fingerprinting to identify and differentiate bacterial strains from various environmental sources. ERIC-PCH genomic fingerprinting was applied to detect and differentiate four major Gram-negative food-borne bacterial pathogens, Esherichia coli, Salmonella, Shigella, and Vibrio. Target DNA fragments of pathogens were amplified by ERIC-PCR reactions. Dendrograms of subsequent PCR fingerprinting patterns for each strain were constructed, through which relative similarity coefficients or genetic distances between different strains were obtained numerically. Numerical comparisons revealed ERIC-PCR genotyping is effective for differentiation of strains among and within species of food-borne bacterial pathogens, showing ERIC-PCR fingerprinting methods can be utilized to differentiate isolates from outbreak and to determine their clonal relationships among outbreaks.

Study of Electrophoretic Karyotypes of Fusarium Section Liseola (Fusarium 균의 section Liseola에 대한 핵형 연구)

  • CHEF-PFGE(Contour-Clarnped Homogeneous Electric field- Pulsed Field Gel Electrophoresis) was used to identify electrophoretic karyotype for eight strains belonging to the Fzisoriuni section Liseolo. Chromosome numbers were nine to thirteen bands, ranging in size Cram 0.75 to 6.45 Mb. The total genome size was eslimated to range from 38.19 Mb to 43.12 Mb and numerous chromosome-length polymorphisms (CLPs) were observed. For the chromosome localizalion of the gene, 1GS sequence(2.6 Kb) of rDNA from F: moniliforme, chs-2 gene(2.8 Kb) and 4 - 3 gene(3.8 Kb) from Neuuospora cmssa were wed as probes.

Restriction endonuclease mapping of the plasmid pTi12 from agrobacterium tumefaciens (Agrobacterium tumefaciens KU-12 균주에서 분리한 플라스미드 pTi 12의 제한효소 지도)

  • Ti plasmids were isolated from three strains of Agrobacterium tumefaciens in Korea and their types and molecular weights were determined. All of these are octopine-type and their molecular weights are 44Kb (pTi 12), 180Kb (pTi 14) and 172Kb (pti 49), respectively. In order to construct physical map of pTi 12, pTi 12 was digested with restriction endonucleases Sma I and Hind III. Sma I degestion of pTi 12 produce 8 fragments and Hind III produced 10 fragments. Physical arrangements of these fragments was determined by Southern hybridization techniques.

  • MAK18 gene of Saccharomyces cerevisiae, needed for M1 replication, was mapped within 2cM of PET3 on chromosome VIII. From 38kb clone pRE66 carrying SPO11 and PET3, we have localized MAK18 gene whose insert is 2.8kb. MAK18 gene is Iocalized on about 9kb distance from PET3 and about 18kb distance from SPO11 on chromosome VIII.

A Vegetation Characteristics of Native and Introduced Kentucky bluegrass Cultivars Seeded on Cut-Slop (자생 및 도입 켄터키 블루그래스 품종의 훼손비탈면 녹화 특성)

  • Research was initiated to investigate vegetation characteristics of native ('Pureundle' and 'Sewon') and introduced ('Brilliant' and 'Midnight') Kentucky bluegrass (KB, Poa pratensis L.) varieties by soil-seed applying system on a cut-slope in May 17, 2008. There were no statistic differences observed in soil hardness and soil moisture content while significant differences were observed in surface coverage rate, height and disease occurrence in native and introduced KB varieties. The introduced 'Brilliant' KB was highest in the surface coverage rate from the early period of seeding to August 11, 2008. The surface coverage rate of introduced varieties were high when compared to native varieties in their early growth stage. But native varieties reached to equal surface coverage rate with the introduced varieties when three months after seeding. The height of 'Pureundle' and 'Brilliant' KBs were high compared with Midnight and Sewon KBs. 'Pureundle' and 'Brilliant' KBs were damaged by disease whereas there were no disease occurrence in 'Midnight' and 'Sewon' KBs. Probably, the disease occurrence was close related with the height of KBs not with native and introduced KB varieties. These results indicate that the growth of KB on a cut-slope are statistically affected by varieties in early growth stage of this experiment but reach equal rate between native and introduced KB varieties in three months after seeding. This growth characteristics implies the huge potential of native KB varieties seeding for cut-slope vegetation. Considering that native KB varieties are adapted to a environment and have no risk of causing disturbance to the ecosystem compared to the introduced KB varieties, the use of them as native varieties for a cut-slope vegetation can be increased.

A Study on the Quantization Noise in LDM and CFDM Systems (LDM방식과 CFDM방식의 양자화 잡음에 관한 연구)

  • Quantization noise of nonadaptive Linear Delta Modulation(LDM) and adaptive Constant Factor Delta Modulation(CFDM) systems is studied. The formulas for quantization noise of CFDM system are derived on the basis of the rusults of LDM. And the output signal-to-quantization noise ratios(SNR) in LDM and CFDM systems are calculated in the range of bit rates from 16[Kb/s] to 96[Kb/s]. By comparing LDM and CFDM, it is known that the adaptive DM is superior to non-adaptive DM by 8[dB] when bit rate is 20[Kb/s] and SNR advantage increases to 14[dB] when bit rate is 56[Kb/s]. All the theoretical results agree well with the experimental results.
