Isolation and Characterization of a Chitin Synthase Gene Fragments from Pleurotus sajor-caju (여름느타리의 Chitin synthase 유전자 단편분리 및 발현 특성 분석)

  • We isolated three amplified DNA fragments from P. sajor-caju by Polmerase chain reaction (PCR) using the chithin synthase specific primers. Since the sequence analysis of the these fragments showed significant homology to the other known chitin synthase gene, we regarded these cloned fragments as PsCHS1, PsCHS2, and PsCHS3 according to their size. The PsCHS3, which showed the highest sequence homology (83% identity in amino acid level with ChsI of Rhizopus oligosporus in conserved region), was selected to see expression pattern of the corresponding gene. The result of RT-PCR using internal primer of the PsCHS3 fragment revealed that PsCHS3 gene was only expressed in cap and mycelium but not in stipe. In order to see whether the PsCHS3 gene was to be induced by wounding, the comparison of the mRNA level of this gene between wounded and unwounded mature cap showed at least two times induction of this gene by wounding treatment.

Nutrition Practice to Alleviate the Adverse Effects of Stress on Laying Performance, Metabolic Profile and Egg Quality in Peak Producing Hens: II. The Probiotic Supplementation

  • In this experiment, the effects of cage density (CD) and probiotic supplementation (PS) on laying performance, metabolic profile, and egg quality in peak-producing hens were evaluated. After blocking according to the cage location, Lohman layers (n = 180, 46 wks of age) were allocated randomly to two levels of CD (540 vs. 360 $cm^2$/hen) and three levels of PS (0, 0.15, and 0.30%). Probiotic contained Enterococcus faecium culture (10${\times}$10$^9$ cfu/g). Egg production (EP) and feed consumption (FC) were measured daily; egg weight (EW) was measured bi-weekly; BW was measured before and after the experiment; and blood samples were obtained at the end of the experiment. The data were analyzed using two-way ANOVA. Increasing CD decreased FC (125.0 vs. 120.8 g/d, p<0.0001) and FCR (1.93 vs. 1.87, p<0.0001) and did not alter EP, EW, and BW. Increasing level of PS linearly decreased FC (p<0.02) and FCR (p<0.006). Averages were 123.9, 123.2, and 121.6 g/d for FC and 1.91, 1.92, and 1.86 for FCR in hens supplemented with 0, 0.15, and 0.30% probiotic, respectively. Hens placed in high-density cages had greater serum corticosterone concentration than hens placed in normal-density cages (12.8 vs. 11.3 $\mu$g/dL, p<0.04); CD did not affect concentrations of other metabolites. Increasing level of PS linearly increased serum glucose, albumin, and creatine concentrations and quadratically increased total protein, globulin, Ca, and P concentrations. Average concentrations (mg/dL) were 260, 297, and 305 for glucose; 6.28, 8.09, and 7.58 for total protein; 1.98, 2.48, and 2.38 for albumin; 4.30, 5.62, and 5.19 for globulin; 0.40, 0.52, and 0.54 for creatine; 16.0, 16.5, and 16.3 for Ca; and 6.27, 8.14, and 7.17 for P in hens supplemented with 0, 0.15, and 0.30% probiotic, respectively. There was no effect of CD on egg quality. Increasing level of PS linearly improved yolk color (YC) and quadratically increased albumen index (AI) and Haugh unit (HU). The mean values were 9.67, 9.75, and 10.58 for YC; 8.94, 6.93, and 8.72% for AI; and 85.6, 74.9, and 82.9 for HU for hens supplemented with 0, 0.15, and 0.30% probiotic, respectively. There was also CD by PS effect on FC, EP, and serum glucose, total protein, albumin, globulin, creatine, Ca and P concentrations. In conclusion, increased CD partially depressed laying performance and caused stress. Probiotic supplementation improved laying performance and metabolic profile. It also partially alleviated the adverse effects of stress resulting from increased caging density.

  • Pulp made from sugarcane bagasse (SCB) was bleached in element chlorine free (ECF) sequence. The peroxide bleaching process for the final bleaching process has been introduced in order to reduce the use of chlorine dioxide. Prior to peroxide bleaching, different chelating chemicals were applied. When 4.5% of the total chlorine dioxide was used, bleached SCB pulp using additional DTPA chelate stage (DEDQP) resulted in 87.0% of the ISO brightness. However, bleached pulp using simultaneous stage of DTPA chelate and chlorine dioxide (DE(DQ)P) was reached at 83.9% of the ISO brightness. The viscosity of DEDQP bleached pulp was 25.6 cPs, and the one of DE(DQ)P bleached pulp was 21.9 cPs. Decreasing of chelate effect by chlorine dioxide led to a decrease in the final brightness and a lower viscosity. But simultaneous stage of EDTA chelate and chlorine dioxide (DE(DQ)P) led to higher final brightness (87.0% ISO) and higher viscosity (25.8 cPs) than those of the $DEDQ_{EDTA}P$ bleached pulp (86.4% ISO, 25.2 cPs).

Effect of Physical Stimulation by Free-Ranging Ducks on Growth, Lodging Resistance and Yield of Paddy Rice (방사오리의 접촉자극이 벼 생육, 내도복성 및 수량에 미치는 영향)

  • This study was conducted to clarify the effects of physical stimulation to the rice plant by free-ranging ducks (Chinese native ducks) on growth, lodging resistance and yield of the rice plants in a rice-duck farming system. Two paddy fields were used for this experiment, one exposed to physical stimulation (PS) and the other with no physical stimulation (NPS). Fifteen days after ducks were introduced into the paddy plot, rice plants in the PS field were significantly (P<0.05) shorter than those in the NPS field. Measurements of lodging characteristics showed that, the 3rd internode was significantly (P<0.05) shorter in the PS treatment than in the NPS treatment, but neither the 4th internode length nor the center of gravity of the rice plants differed between treatments. The breaking strength of $N_4$ in PS rice was significantly (P<0.05) higher than in NPS rice, however the bending moment and the lodging index of $N_3$ (distance between fulcra - 5 cm) was significantly (P<0.05) lower in PS than in NPS. The number of panicles per $m^2$ was significantly (P<0.05) greater in PS rice than in NPS, but there was no such difference in spikelet number per panicle, percentage of ripened grain or weight per 1000 kernels. Paddy rice yield per $m^2$ in the PS treatment was significantly (P<0.05) higher than that in the NPS treatment.

  • Porous silicon (PS) was prepared under different anodization conditions and the photoluminescence (PL) was measrued. In addition PL of the naturally and thermally oxidized PS was measured. It was found that the PL peak was shifted to shorter wavelength as the anodization current density and the extent of the oxidation increased. The absence of correlation between the PL behavior and the surface hydrogen species (Si-H2, Si-H) implies that the mechanism of PL of PS is not likely related to the surface hydrogen species effect but to the quantum confinement effect.

  • This paper presents SVBS-TDC (Semi-Vernier Binary-Search Time-to-Digital Converter) for the noise improvement of ADPLL (All-Digital Phase Locked Loop. We used a Semi-Vernier BS-TDC (Binary-Search TDC) architecture to improve the operation speed more then 10 times compared with the previous conventional BS-TDC and ensured a 510ps wide input range. The proposed Semi-Vernier BS-TDC was designed in a 65ns CMOS process and the simulation results showed 200MHz speed and 4ps resolution with a 1.2V supply voltage, and considerable noise improvement of ADPLL.

Persicaria senticosa Ameliorates Imiquimod-induced Psoriasis-like Skin Lesions in Mice via Suppression of IL-6/STAT3 Expression and Proliferation of Keratinocytes

  • Psoriasis is one of the most common inflammatory skin disorders, with a global prevalence of 2% - 3%. It is an autoimmune skin disorder characterized by excessive generation of plaques on the skin with typical long-lasting red, itchy, and scaly lesions. In this study, we aimed to elucidate the anti-psoriatic effect of the methanolic extract of Persicaria senticosa (PS), a bioactive edible plant extract used in traditional medicine, using a mouse model of imiquimod (IMQ)-induced psoriasis. The daily topical application of IMQ could induce human psoriasis-like lesion. The extract ameliorated IMQ-induced psoriasis. Furthermore, hematoxylin and eosin staining and the Psoriasis Area and Severity Index (PASI) scores indicated that topical application of PS led to an improvement in erythema, scaling, and thickness scores of the mouse dorsal skin and a considerable decrease in the epidermal thickness of the ear and dorsal skin in the IMQ-induced psoriatic mouse model. We also studied the effect of PS on the proliferation of keratinocytes using HaCaT cells. The extract inhibited cell proliferation and IL-6 and pSTAT3 expression induced by M5 cocktail (comprising interleukin [IL]-1α, IL-17A, IL-22, oncostatin M, and tumor necrosis factor-α) in HaCaT cells. Thus, PS might serve as a potential therapeutic agent for the treatment of psoriasis.

Prevalence of Lower Urinary Tract Symptoms and Association of Hypertension with I-PSS (국제 전립선 증상 점수(I-PSS)를 이용한 하부요로증상의 유병률 및 고혈압과의 관계)

  • Benign prostatic hyperplasia (BPH) is a highly prevalent, age-related disorder in men which place a considerable burden on health care resources worldwide. While BPH and hypertension are apparently diverse disease processes, they have some features in common(e.g. underlying etiology of the sympathetic nervous system). The purpose of this study is to estimate the prevalence of lower urinary tract symptoms using I-PSS(international prostate symptom score) and to investigate the association of hypertension with I-PSS. This study was carried out to 390 men(40~86 year old) at 3 Myun in Chungju City from July to August, 1997. Subjects answered on questionnaire for I-PSS and were checked age, education, marital status, annual income, blood pressure. The mean age of subjects was 59.8 year old. The mean of I-PSS were increased by age decades(40~49, 50~59, 60~69, 70~86) and it's scores were 4.4, 7.0, 8.2, 12.3 respectively. Strong correlations were observed between the I-PSS and the QUL(quality of life)(P=0.0001). I-PSS(mean-value) were 7.85 in hypertensive group and 8.39 in normotensive group but there was no statistically significance between the two groups(P>0.05). The proportion of mean scoring greater than 8 was 42.5%. There have been reports of association between lower urinary tract symptoms and hypertension, but there was no consistent suggestion that such an association could be casual. The need for high-quality epidemiological information and consequent increased prospects for prevention is obvious.

Piriformis Syndrome in Knee Osteoarthritis Patients after Wearing Rocker Bottom Shoes

  • Background: Rocker bottom shoes (RBS) are popular among patients with different foot, leg, or back problems in Korea. Patients with knee osteoarthritis concurrent weakness in the quadriceps femoris muscle, who wear these shoes, are often assumed to develop piriformis syndrome (PS). This study was performed to improve the understanding about the effect of wearing such shoes on duration of the syndrome in knee osteoarthritis. Methods: We randomly assigned 150 patients with PS, who had used RBS daily for at least 6 months, to 2 groups, the S (stopped wearing) and K (kept wearing) groups. Both the groups were subdivided into the O and N groups, comprising patients with and without knee osteoarthritis, respectively. The effects of the treatment, including piriformis muscle injections and a home exercise program, were compared between the 2 groups by using a flexion-adduction-internal rotation (FAIR) test, a numeric rating scale (NRS), and the revised Oswestry disability index (ODI) during the 12-week follow-up. Results: The positive FAIR test ratios, mean NRS scores, and revised ODIs were higher in the KO group than the SN group from 4-12 weeks after treatment. Conclusions: RBS may extend duration of the PS in osteoarthritis patients.
