Tolfenamic Acid Suppresses Inflammatory Stimuli-Mediated Activation of NF-κB Signaling

  • Tolfenamic acid (TA) is a traditional non-steroid anti-inflammatory drug (NSAID) and has been broadly used for the treatment of migraines. Nuclear factor kappa B (NF-${\kappa}B$) is a sequence-specific transcription factor and plays a key role in the development and progression of inflammation and cancer. We performed the current study to investigate the underlying mechanisms by which TA suppresses inflammation focusing on NF-${\kappa}B$ pathway in TNF-${\alpha}$ stimulated human normal and cancer cell lines and lipopolysaccharide (LPS)-stimulated mouse macrophages. Different types of human cells (HCT116, HT-29 and HEK293) and mouse macrophages (RAW264.7) were pre-treated with different concentrations of TA and then exposed to inflammatory stimuli such as TNF-${\alpha}$ and LPS. Transcriptional activity of NF-${\kappa}B$, $l{\kappa}B-{\alpha}$-degradation, p65 translocation and mitogen-activated protein kinase (MAPK) activations were measured using luciferase assay and Western blots. Pre-treatment of TA repressed TNF-${\alpha}$- or LPS-stimulated NF-${\kappa}B$ transactivation in a dose-dependent manner. TA treatment reduced degradation of $l{\kappa}B-{\alpha}$ and subsequent translocation of p65 into nucleus. TA significantly down-regulated the phosphorylation of c-Jun N-terminal kinase (JNK). However, TA had no effect on NF-${\kappa}B$ signaling and JNK phosphorylation in HT-29 human colorectal cancer cells. TA possesses anti-inflammatory activities through suppression of JNK/NF-${\kappa}B$ pathway in different types of cells.

Licochalcone B Exhibits Anti-inflammatory Effects via Modulation of NF-κB and AP-1

  • The present study investigated the mechanisms of licochalcone B (LicB)-mediated inhibition of the inflammatory response in murine macrophages. RAW264.7 murine macrophages were cultured in the absence or presence of lipopolysacharide (LPS) with LicB. LicB suppressed the generation of nitric oxide and the pro-inflammatory cytokines interleukin (IL)-$1{\beta}$, IL-6 and tumor necrosis factor-${\alpha}$. LicB also inhibited the expression of mRNA for inducible nitric oxide synthase and pro-inflammatory cytokines induced by LPS. Moreover, LicB inhibited nuclear factor-${\kappa}B$ (NF-${\kappa}B$) and activator protein-1 translocation into the nucleus in a dose-dependent manner. Thus, LicB mainly exerts its anti-inflammatory effects by inhibiting the LPS-induced NF-${\kappa}B$ and activator protein-1 signaling pathways in macrophages, which subsequently diminishes the expression and release of various inflammatory mediators. LicB shows promise as a therapeutic agent in inflammatory diseases.

Effects of Citri Reticulatae Viride Pericarpium on 4-Hydroxynonenal-Induced Inflammation in PC12 Cells (청피의 항염증효과)

  • Objectives: The purpose of this study was to observe the effects of Citri Reticulatae Viride Pericarpium (CP) on 4-Hydroxynonenal (4-HNE)-induced inflammation in PC12 cells. Methods: 4-HNE was treated in PC12 cell to cause inflammatory response, and then treated with CP water extract at 25, 50, and $100{\mu}g/ml$. The phosphorylation of Jun N-terminal kinase (JNK) and the expression of $NF-{\kappa}B$ in PC12 cells were determined by Western blot, respectively. Results: The phosphorylation of JNK was significantly decreased in 4-HNE-stimulated PC12 cell by the treatment of CP extract at $25{\mu}g/ml$. The 4-HNE-induced expression of nuclear factor kappa-light-chain-enhancer of activated B cells ($NF-{\kappa}B$) p65 in nuclear of the cells was significantly decreased in PC12 cell by treatment with CP extract at 25, 50, and $100{\mu}g/ml$. Conclusions: These results suggest that CP water extract has an anti-inflammatory activity through suppressing the JNK and $NF-{\kappa}B$ activation.

Anti-inflammatory Effects of Scrophularia Koraiensis Nakai via NF-κB and MAPK Signaling Pathways in LPS-induced Macrophages

  • Scrophularia koraiensis Nakai is widely used to remedy fever, edema, and neuritis. S. koraiensis has harpagoside and angoroside C, these compounds have been reported to alleviate inflammation, rheumatic diseases, and analgesic stimulation. We evaluated the anti-inflammatory effects of the ethanol extract of S. koraiensis (SKE) in lipopolysaccharides (LPS)-induced macrophages. At cellular levels, SKE decreased the production of nitric oxide (NO), the expression of inducible nitric oxide synthase (iNOS), and cytokines (IL-1b, TNF-a, and IL-6) under the LPS stimulation. SKE inhibited the phosphorylation of nuclear transcription factor-kappa B (NF-κB) p65 and its inhibitor (IκB-α). In addition, SKE suppressed the phosphorylation of extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 in the mitogen-activated protein kinase (MAPK) pathway. In conclusion, SKE could be considered a potential resource for attenuating inflammation response and it may be utilized in the material for cosmetics, food additives, and tea.

  • Recently, a considerable number of studies have been made on the $RFID^{[1-6]}$ systems. RFID is a technique of identifying an object using radio frequency transmission. The technology can be used to identify, track, sort or detect a wide variety of objects. The RFID system is composed of two main elements: a reader and a tag. Tags can either be active (powered by battery) or passive (powered by the reader field). The passive tags communicate back to the reader with a technique called 'backscatter'. RFID technology can be applied to the supply chain, security, logistics industry and etc. Especially, UHF RFID is worth noticing because of its relatively long identification range and commercial UHF RFID systems are under development. In this paper, we designed and implemented a UHF RFID reader firmware for ISO/IEC 18000-6 Type B specification.

A Case of Nasal Diffuse Large B-Cell Lymphoma in a Cat

  • A 6-year-old spayed female Turkish Angora cat presented with sneezing, nasal discharge, and decreased appetite lasting for 21 days. Skull radiography revealed slightly increased density of soft tissue in the left nasal cavity. Computed tomography (CT) scan revealed an extensive mass with nasal septum destruction and moderate contrast enhancement in the left nasal cavity. After surgical biopsy, histopathological examination confirmed that the mass was an infiltrative round cell neoplasm, composed of sheets of large neoplastic cells. Immunohistochemical analysis revealed that most of the neoplastic cells were strongly positive for CD79a and weakly positive for PAX5. Additionally, numerous mature lymphocytes were found to be positive for CD3. This is the first reported case of nasal diffuse large B-cell lymphoma (DLBCL) in a Turkish Angora cat in Korea.

  • 7,8-Dioxo-A-norerythrinan (A) was synthesized from acid catalyzed cyclization of 6a-hydroxv-1-(2-phenylethyl)-octahydrocyclo-penta[b] pyrrole-3a-carboxylic acid ethyl ester (B) with concomitant deethoxycarbonylation. The intermediate (B) was a hydroxylated compound of N-acyliminium (C). The unstable pyrrolinium (C) was prepared from oxalylation of the enamine of phenethylamine and ethyl 2-oxocyclopentanecarboxylate.

Study of Analysis Technique of Flight Compatibility Test Data using Instrumented Pod for External Store (외부장착 계측포드를 사용한 비행적합성 자료 분석기법 연구)

  • Analysis technique of flight compatibility test results using instrumented pod for external store is studied. Raw data of sensors and MIL-STD-1553B acquired from instrumented pod are analyzed separately. Besides, tuning and synchronization of sensor data enable the analysis of those two type of data simultaneously. Evaluation of analysis result shows that the analyzed data represent maneuvers of the aircraft successfully and agree the values of the real flight tests.
