젓갈산업의 현황 및 발전 방향 (The present condition and development prospect of the fermented fishery products)

  • The traditional Korean fermented fish products are classified into mainly three groups; Jeot-gal, Aek-jeot, and Sik-hae. Jeot-gal is a salt-fermented fish. Aek-jeot (Eoganjang) is actually a liquid part of Jeot-gal. Sik-hae is a salt-fermented whole or part fisheries with adjuncts. The production of jeot-gal products has been increased constantly. However, there is not enough fishery for raw materials. Recently, consumers have been preferred low-salted foods because they have become aware that high levels of salt cause adult diseases such as hypertension or gastric cancers. The main consumers of jeot-gal are adults above 40~50 years old. Young generation and school nutrition teachers dislike fishery products because of distinct fish smell, small bone, as well as food safety. Therefore, in order to increase the consumption of jeot-gal and extend its industry, jeot-gal should be developed to match the preference of new generation with good safety, health-oriented, and new concept.

  • Indigenous fermented foods have played a vital role in human history, and continue to offer a multitude of diverse sensory characteristics. According to earthenware relic, the fermented fisheries products might be consumed in Korean peninsula from Paleolithic period. The traditional Korean fermented fish products are classified into four groups; Jeot-gal (sik-hae), Aek-jeot, Seasoned Jeot-gal, and Seasoned Aek-jeot. Jeot-gal is a fermented fishery with salt. Aek-jeot is a liquid part of Jeot-gal. Sik-hae is manufactured by fementing fishery with salt, cereal, malt powder, etc. Seasoned Jeot-gal is a salt-seasoned fish with red pepper powder, garlic, onion, etc. Seasoned Aek-jeot is a seasoned product by adding "salt-water" or "condiment" to Aek-jeot. The fermented fisheries industry has traditionally been succeeded mainly in a cottage scale to the middle of 20th century. Thereafter, together with the development of pelagic fishery, the fermented fisheries industry also developed constantly to an enterprise size.

저식염 오징어 젓갈의 숙성 중 품질변화 및 최적 유통기한 설정 (Quality Changes in Low-Salted Squid Jeot-gal during Fermentation and Determination of Shelf-life)

  • 본 연구는 저식염 오징어 젓갈에 대한 과학적 자료를 제공함과 동시에 전통수산발효식품 소비의 확산에 기여하기 위하여 염 농도에 따른 발효기간별 품질특성 측정, 발효최적조건, 유통기한 설정 및 발효최적조건에서 제조한 저식염 오징어 젓갈의 생리활성 등을 연구하였으며, 그 결과를 요약하면 다음과 같다. 숙성온도가 증가할수록 pH는 급격히 감소하였으며, 기간에 따라 대조구인 10% 젓갈의 pH 변화는 거의 없는 반면 3 및 5% 첨가군은 급격히 감소하였으나 pH 5 이하에서는 서서히 감소하였다. 아미노질소와 휘발성염기 질소 또한 발효온도가 증가할수록 각각의 함량이 급격히 증가하였으며 식염의 농도가 낮을수록 휘발성 염기질소의 함량은 증가하였다. 아미노질소의 함량이 최고치가 되는 시점을 최적발효조건 지표로 산출하였으며 그 결과 $-1^{\circ}C$에서 저장한 염 농도 3% 젓갈일 때 41일의 최적발효기한을 나타내었다. 저식염 오징어 젓갈의 냉수 및 에탄올 추출물의 항산화 활성, 항당뇨 및 항대장암은 뚜렷한 활성을 보이지 않았으며, 일반성분 함량은 식염과 부재료 첨가로 수분과 회분의 함량에 변화를 보였지만 그 이후 저온저장 하는 기간 동안에는 일정하게 유지되다. 구성아미노산 및 유리아미노산은 glycine, arginine, alanine 및 glutamic acid 등의 함량이 절반 이상을 차지함으로써 맛에서 우수하였고, 관능검사결과는 시중에서 판매하고 있는 젓갈과 비교했을 때 큰 차이를 보이지 않았다. 이상의 결과를 고려할 때 염 농도 3% 젓갈을 $-1^{\circ}C$에서 제조, 숙성시킨다면 최근 소비자의 건강상 기호패턴의 변화에 부응시킴과 동시에 전통 수산발효식품의 보급 및 소비 확산에 기여할 수 있을 것이라 판단된다.

천연식물추출물을 첨가한 저염 오징어젓갈의 품질특성, 유통기한 및 생리활성 (Quality Characteristics, Shelf-life, and Bioactivities of the Low Salt Squid Jeot-gal with Natural Plant Extracts)

  • 본 연구는 염농도의 저하에 따른 저염 오징어젓갈의 유통기한 감소에 대하여 유통기한의 연장과 기능성 제품화의 가능성을 검토하기 위하여 천연식물추출물 첨가에 따른 저장기간별 품질특성, 최적발효기한, 유통기한 설정 및 생리활성을 연구하였으며, 그 결과는 다음과 같다. 첨가군 모두 저장온도가 증가할수록 pH는 급격히 감소하였고, 아미노질소, 휘발성염기질소의 함량은 급격히 증가하였으며, 총균수 역시 증가하였다. 유통기한 및 최적발효기한은 Arrhenius equation의 결과에 따라 $10^{\circ}C$에서 대조구 34 및 28일, 월계수첨가 35 및 29일, 녹차첨가 34 및 30일, 솔잎첨가 34 및 29일 저장한 것이었다. 구성 및 유리아미노산은 첨가군 모두가 glutamic acid, glycine, alanine 등의 감칠맛, 단맛을 내는 아미노산의 함량이 대부분을 차지하여 맛에서 우수하다고 판단되었고, 핵산관련물질은 첨가군 모두 쓴맛을 나타내는 hypoxanthine이 대부분을 차지하였으며, 그 뒤 inosine 및 정미성분인 IMP 순이었다. 관능검사결과 월계수첨가군이 종합적으로 가장 뛰어났다. 생리활성은 첨가군이 대조구에 비해 활성이 높았으며, 기능성 제품으로서의 상업화가 가능하다고 판단된다. 그러므로 천연식물추출물을 첨가한 저염 오징어젓갈은 웰빙 등 시대의 변화에 맞추어 현대인의 식기호에 적합한 식품으로 이용 가능하며, 젓갈의 고급화 및 부가가치 창출이 가능하다고 사료된다.

  • A bacteial strain which can produce the extracellular fibrinolytic enzyme was isolated from Jeot-Gal (anchovy) that was Korean traditional salt-fermented fish. The isolated bacterium was identified to be a strain of Bacillus sp. The optimal medium for fibrinolytic enzyme production was determined to consist of 5 g maltose, 10 g defatted soybean, 20 g sodium chloride, 1.75 g K2HPO4 per liter (pH 7.0)

송이버섯 추출액의 항균력 확인과 저염젓갈 생산에 적용 가능성 (Determination of Antibacterial Activity from Tricholoma matsutake Extract and Its Application to Low Salted Jeot-gal)

  • 송이버섯 추출액의 박테리아에 대한 항균력을 측정하기 위하여 최소저해농도를 확인한 결과 젓갈 제조시 빈번하게 발생하는 부패균, 어병원인 균등 9가지 박테리아에 대해 성장이 저해되는 것을 확인하였다. 확인된 송이버섯 추출액의 항균력을 저염 젓갈의 제조 공정에 적용하기 위해 식품보존제인 사과산나트륨과 아질산나트륨을 함께 첨가하여 S. aureus에 대한 최소저해농도를 확인하였다. 그 결과 사과산나트륨과 아질산나트륨의 최소저해농도가 각각 0.25 g/L와 0.025 g/L로 크게 낮아지는 것을 확인할 수 있었다. 본 연구를 통해 항균력을 갖는 송이버섯 추출액을 첨가함으로써 일반적으로 첨가되는 식품보존제의 농도를 크게 낮출 수 있었을 뿐 아니라 송이버섯의 다양한 기능성이 추가된 고급화된 저염 젓갈을 개발할 수 있을 것으로 예상된다.

Purification and Characterization of a New Fibrinolytic Enzyme of Bacillus licheniformis KJ-31, Isolated from Korean Traditional Jeot-gal

  • Jeot-gal is a traditional Korean fermented seafood and has long been used for seasoning. We isolated 188 strains from shrimp, anchovy, and yellow corvina Jeot-gal, and screened sixteen strains that showed strong fibrinolytic activities on a fibrin plate. Among those strains, the strain that had the largest halo zone was chosen and identified as Bacillus licheniformis by using 16S rDNA sequencing and an API CHB kit. The fibrinolytic activity of Bacillus licheniformis was characterized and designated as bpKJ-31. The active component of bpKJ-31 was identified as a 37 kDa protein, designated bacillopeptidase F, by internal peptide mapping and N-terminal sequencing. The optimum activity of bpKJ-31 was shown at pH 9 and $40^{\circ}C$, with a chromogenic substrate for plasmin. It had high degrading activity for the $B{\beta}$-chain and $A{\alpha}$-chain of fibrin(ogen), and also acted on thrombin, but not skim milk and casein. The amidolytic activity of bpKJ-31 was inhibited by 1 mM phenylmethanesulfonyl fluoride, but 1 mM EDTA did not affect the enzyme activity, indicating that bpKJ-31 is an alkaline serine protease, like a plasmin. The bpKJ-31 showed approximately 14.3% higher fibrinolytic activity than the plasmin. These features of bpKJ-31 make it attractive as a health-promoting biomaterial.

Screening and Chacterization of Bacteriocinogenic Lactic Acid Bacteria from Jeot-Gal, a Korean Fermented Fish Food

  • Bacteriocins are classified as proteins which are produced by heterogeneous groups of bacteria, having and antimicrobial effect of the closely related organisms. Recently, bacteriocins derived from lactic acid bacteria and other food-related organisms have been the subject of much research on potential food biopreservatives. The goal of this study was to screen and characterize the bacteriocinogenic lactic acid bacteria from Jeot-gal(commercial fermented fish foods). All bacteriocinogenic isolates were identified as lactic acid bacteria. Isolates NK24, NK34, and SA72 were tentatively identified as Lactobacillus brevis, according to the API 50 CHL kit database. All antimicrobial substances produced from four lactic acid bacteria isolates completely lost their antibacterial activity after being treated with some proteases, indicating to their proteinaceous nature. The bacteriocin produced from isolates NK24, NK34, and SA72 showed a broad spectrum of activity when compared to those produced from isolate SA131. All bacteriocins isolated during the course of this study showed a bactericidal mode of inhibition.

Characterization of Glutamate Decarboxylase (GAD) from Lactobacillus sakei A156 Isolated from Jeot-gal

  • A gamma-aminobutyric acid (GABA)-producing microorganism was isolated from jeot-gal (anchovy), a Korean fermented seafood. The isolate, A156, produced GABA profusely when incubated in MRS broth with monosodium glutamate (3% (w/v)) at 37℃ for 48 h. A156 was identified as Lactobacillus sakei by 16S rRNA gene sequencing. The GABA conversion yield was 86% as determined by GABase enzyme assay. The gadB gene encoding glutamate decarboxylase (GAD) was cloned by PCR. gadC encoding a glutamate/GABA antiporter was located immediately upstream of gadB. The operon structure of gadCB was confirmed by RT-PCR. gadB was overexpressed in Escherichia coli BL21(DE3) and recombinant GAD was purified. The purified GAD was 54.4 kDa in size by SDS-PAGE. Maximum GAD activity was observed at pH 5.0 and 55℃ and the activity was dependent on pyridoxal 5'-phosphate. The Km and Vmax of GAD were 0.045 mM and 0.011 mM/min, respectively, when glutamate was used as the substrate.

Identification and Partial Characterization of Lacticin SA72, a Bacteriocin Produced by Lactococcus lactis SA72 Isolated from Jeot-gal

  • Strain SA72 was isolated from Jeot-gal and identified as producer of a bacteriocin, which showed some bactericidal activity against Lactobacillus delbrueckii ATCC 4797. Strain SA72 was tentatively identified as Lactococcus lactis according to the AOI test. Lactococcus lactis SA72 showed a broad spectrum of microorganisms, tested by the modified deferred method. The activity of lacticion SA72, named tentatively as a bacteriocin produced by Lactococcus lactis SA72, was detected during the mid-lon growth phase, reached a maximum during the early stationary phase, and then declined after the late stationary phase. Lacticin SA72 also showed a relatively broad spectrum of activity against non-pathogenic and pathogenic microorganisms when assessed by the spot-on-lawn method. Its anitimicrobial activity on sensitive indicator cells disappeared completely by protease XIV treatment. The inhibitory activity of lacticin SA72 remained after treatment for 15 min at $121^{\circ}C$, 문 was stable in a pH range of 2.0 to 9.0 and all organic solvents examined. It demonstrated a typical bactericidal mode of inhibition against Lactobacillus delbrueckii ATCC 4797. The apparent molecular mass of lacticin SA72 was in the region of 3-3.5 kDa, determined by SDS-PAGE.
