Physiological and Ecological Characteristics of Lipid-Producing Botryococcus Isolated from the Korean Freshwaters (한국산 고지질 미세조류 Botryococcus의 분포 및 생장 특성)

  • Recently, sustainable production of biofuel using algal biomass is being pursued because of its enormous potential. First and foremost, securing superior strains to develop an efficient production system for algal biodiesel through screening or genetic improvement of microalgae is necessary. The genus of Botryococcus is regarded as one of the superior microalgae for biodiesel production due to its ability to accumulate high amounts of lipids and hydrocarbons. However, its low growth rate is a bottleneck for large-scale production and commercialization. The purpose of this study is to obtain indigenous Botryococcus strains which possess high lipid content and biomass productivity. The Botryococcus sp. was isolated from the Seobu Reservoir in Jeju Island and identified as Botryococcus sudeticus J2 by comparative analysis of 18s rRNA gene and ITS regions. The biomass productivity and lipid content of B. sudeticus J2 were 0.116 g $L^{-1}day^{-1}$ and 40.1% of dry wt., respectively. This was higher than the value of B. braunii UTEX 572, which is widely regarded as a superior strain among Botryococcus species. The relatively high growth rate of B. sudeticus J2 was achieved under a light intensity of 240 ${\mu}mol$ photons $m^{-2}s^{-1}$ with ambient air spargingwhen compared to 120 ${\mu}mol$ photons $m^{-2}s^{-1}$ with 2% $CO_2$ supply. In summary, it is likely that the isolated B. sudeticus J2 can be used for the mass cultivation and biodiesel production.

Summary on the Dinoflagellate Cyst Assemblages of Modern Sediments from Korean Coastal Waters and Adjoining Sea (한국연안해역 와편모조류 시스트 연구에 관한 고찰)

  • To investigate the research state and characteristics of dinoflagellate cyst along the Korean coastal waters, this work analyzed 45 studies conducted in the Korean coasts and basin, the south of Jeju, East China Sea including some part of the Yellow Sea. It was found that the research on dinoflagellate cyst began in the later half of the 1980s in Korea, and that the research areas focused on eutrophied regions where a red tide occurred or on the seas where coastal development occurred. In other words, no research on the East Sea was found. In terms of research contents, there were various studies on analysis of cyst assemblage, tracking of the changes in marine environment like process of eutrophication, roles as a seed population in occurrence of a red tide, creation of cyst by use of sediment trip and tracking of its changes, morphological changes by the change of pH concentration in the hypoxia zone of eutrophied region, germination of a specific species, and other international-level studies. Species composition and cell density also varied in the Korean coastal waters. However, much research on dinoflagellate cyst in Korea is not systematic, and is fragmentary and individual and hencesuffers from great limitations. The research results so far have focused only on specific regions, and sporadic research had been conducted by specific research groups. Dinoflagellate cyst of surface sediments represents the accumulation index of pelagic environment in the sea and is a useful index to understand the marine environment efficiently. In conclusion, Korea requires organized human resources and collaborative research on dinoflagellate cyst which in turn should be considered as a component of marine ecosystem and as an essential aspect of marine biology.

Effects of the decoction water on the extraction of the bioactive compounds from rhubarb (전탕용수의 종류에 따른 대황 성분의 추출효과)

  • The quality of decoction water would be important for the preparation of herbal medicines. Four types of decoction water, S1, S2, S3 and S4, three types of mineral water from Jeju island and one type of tap water from Seoul region were evaluated. The contents of minerals in decoction water were analysed by ICP-AES and ICP-MS. There was significant difference in the mineral contents between the types of decoction water. The bioactive components from rhubarb were extracted with four types of water and the relationship between mineral contents in water and the extracted amounts of components was evaluated. The total extraction was calculated by evaporating water and weighing the residue. The bioactive components in rhubarb were determined by HPLC method. Kruskall-Wallis rank sum tests were used. Multiple factor analysis was used to analyze the relation between the contents of mineral and total extraction or bioactive components in the decocted solutions. Not only the total extraction but also the amount of extracted bioactive components showed statistically significant relationship with the contents of minerals and anions in decoction water.

RAPD-SCAR Markers Linked to Medium-Leaf Zoysiagrass Ecotypes (한국잔디 중지 변이개체와 연관된 RAPD-SCAR 마커)

  • Two medium-leaf ecotypes (CY6069, CY6097) belonging to one species (Zoysia japonica) of Korean lawngrasses were selected in sod production fields in Jang Seong, Korea. They were reported to have distinct morphological and growth rate characteristics different from the preferred medium-leaf type zoysiagrass in Korea. This study was conducted to define further the genotypic difference at the molecular level and to develop DNA marker based on the specific DNA fragment. Polymorphic DNA fragments were first explored by using randomly amplified polymorphic DNA (RAPD) primers, which were then converted into PCR-based sequence characterized amplified region (SCAR) markers. The CY6069-specific primer set amplified about 550 bp successfully, while the CY6097 marker produced the expected 690 bp band, by which those markers were nominated by CY6069_550 and CY6069_690 SCARs, respectively. Together with the reported morphological and other phenotypic features, the SCAR markers confirmed in this study will be useful to identify those medium-leaf zoysiagrass genotypes when they are cultivated with other vegetatively propagated warm-season turfgrasses in sod farms.

Current status of anisakid nematode larvae infection in marine fishes caught from the coastal area of Korea between 2010 and 2012 (2010~2012년 연안에서 서식하는 해산어에서 아니사키스 유충의 감염현황)

  • A survey was conducted to investigate infection of anisakid nematode larvae in 243 wild marine fishes caught from the southern coastal area of Korea between 2010 and 2012. The samples comprised fishes from 9 orders, 30 families and 50 species. Total infection rate of anisakid nematode larvae was 10.7% (26/243 fish), which comprised from Yeosu, 7.4% (7/95) in 2010 and 22.7% (5/22) in 2011; from Jeju, 8.2% (5/61) in 2011; from Wando, 40.9% (9/22) in 2012. Anisakid nematode larvae were not detected in Tongyoung and Wando samples in 2011. Molecular identification of the 89 worms from 26 fish was conducted by PCR-RFLP and/or sequence analysis of internal transcribed spacer (ITS) region of ribosomal DNA. From the results, 6 kinds of anisakis species were identified: Anisakis pegreffii (infection rate: 53.9%, 48/89 worms), Hysterothylacium aduncum (38.2%, 34/89), H. fabri (3.4%, 3/89), hybird (A. simplex X A. pegreffii) (2.4%, 2/89), A. simplex (1.1%, 1/89) and Raphidascaris lophii (1.1%, 1/89). The rate of single infection was 80.8% (21/26 infected fish), while 19.2% (5/26) showed mixed infection with 2 to 3 different anisakis species.

Estimation of Surplus Solar Energy in Greenhouse Based on Region (지역별 온실내의 잉여 태양에너지 산정)

  • This research was conducted to provide basic data of surplus heat for designing solar heat-storage systems. The surplus heat is defined as the heat exhausted by forced ventilations from the greenhouses to control the greenhouse temperature within setting limits. Various simulations were performed to compare the differences of thermal behaviors among greenhouse types as well as among several domestic areas by using pseudo-TMY (Typical Meteorological Year) data manipulated based both on the weather data supplied from Korean Meteorological Administration and the TMY data supplied from The Korean Solar Energy Society. Additional analyses were carried out to examine the required heating energy together with some others such as the energy balances in greenhouses to be considered. The results of those researches are summarized as follows. Regional surplus solar heats for the nine regions with 4-type were analyzed. The results showed that the ratio of surplus solar energy compared to heating energy was the highest in Jeju (about 212.0~228.0%) for each greenhouse type. And followed by Busan, Kwangju, Jinju, Daegu, Daejeon, Jeonju, Suwon and Daekwanryung. And irrespective of greenhouse types, surplus solar energy alone could cover up nearly all of the required supplemental heating energy except for a few areas.

A Method and Application of Constructing an Aggregating Indicator : Regional Descent Work Index in Korea (종합지표 작성 방법 및 적용: 우리나라 지역별 좋은 일자리 지수)

  • Job creation is the most important issue in the labor market these days, and the quality of jobs is also very important in order to resolve the mismatches that are taking place in the labor market. Kim Young-min (2014) developed the "2012 Quality of Employment Index" with twenty indicators in seven categories, including employment opportunities, to objectively assess the local labor market. This method presents the concept of the aggregate indicator, 'Quality of Work Index', and has the advantage of being easy to produce. However, it is difficult to statistically verify the adequacy of the constitutive indicators and, based on this, make them a single aggregate index through statistical techniques. Therefore, we developed an alternative '2012 Descent Work Index' and a confidence interval using Principal Component Analysis(PCA) and Unobserved Component Model(UCM) presented by Gi-Choon Kang & Myung-jig Kim (2014) and also calculated an alternative '2017 Descent Work Index' using the first half of 2017 local area labour force survey and compared its changes by region. The results of the empirical analysis show that the rank correlation coefficient between two methods of aggregating indicators, simple weight used in Young-min Kim's research, PCA method and UCM used in this study, were found to be statistically significant under 5% significance level. This implies that all methods are found to be useful. However, the PCA and UCM which determine scientific and objective weights based on data are preferred to Young-min Kim's approach. Since it provides us not only the level of aggregate indicator but also its confidence intervals, it is possible to compare ranking with the consideration of statistical significance. Therefore, it is expected that the method of constructing an aggregating indicator using UCM will be widely used in many areas in the future.

Differences in isolates of Tomato yellow leaf curl virus in tomato fields located in Daejeon and Chungcheongnam-do between 2017 and 2018

  • To follow up on a 2017 survey of tomato virus diseases, samples with virus-like symptoms were collected from the same areas (Buyeo-gun, Chungchungnam-Do and Daejeon, Korea) in 2018. While in 2017 mixed infections of Tomato mosaic virus with either Tomato yellow leaf curl virus (TYLCV) or Tomato chlorosis virus were detected, only TYLCV was detected in symptomatic samples in 2018. TYLCV amplicons of c.777 bp representing the coat protein (CP) coding region were cloned from the TYLCV positive samples, and the sequence data showed a 97.17% to 98.84% nucleotide and 98.45% to 99.22% amino acid identity with the 2017 Buyeo-gun isolate (MG787542), which had the highest amino acid (aa) sequence identity of up to 99.2% with four 2018 Buyeo-gun sequences (MK521830, MK521833, MK521834, and MK521835). The lowest aa sequence identity of 98.45% was found in a 2018 Daejeon isolate (MK521836); the distance between Buyeo-gun and Daejeon is about 45 km. Phylogenetic analysis indicated that the currently reported CP sequences are most closely related to Korean sequences from Masan (HM130912), Goseong (JN680149), Busan (GQ141873), Boseong (GU325634), and the 2017 isolate TYLCV-N (MG787543) in the 'Japan' cluster of TYLCV isolates and distinct from the 'China' cluster isolates from Nonsan (GU325632), Jeonju (HM130913) and Jeju (GU325633, HM130914). Our survey data from 2017 and 2018 suggest that TYLCV has become established in Korea and may be spread by whitefly vectors from weed reservoirs within the farm environment.

A Study on Human Thermal Comfort of Residential Development Districts in Summer Season (여름철 택지개발지역의 열쾌적성에 관한 연구)

  • This study measured the Physiological Equivalent Temperature (PET) of the hottest day time in a day, in order to verify the characteristics of human thermal comfort in case of heat wave during summer time in each region, by subdividing the urban areas in accordance with the climatic characteristics with the use of Local Climate Zone (LCZ) as a method of classifying the type of urban climate and the land cover map, targeting the Homaesil residential development district in Suwon. In the results of measurement, the forest and paddy field showed the moderate heat stress while the urban park showed the strong heat stress. Other developed areas showed the extreme heat stress. Such results show the possibility of institutional utilization for the improvement of human thermal comfort through the verification of climatic characteristics and differences in each type of urban areas, and the efficient placement of green infrastructure and the planning of land use to cope with the heat wave even in the planning stage for the establishment of urban planning.

Study on Chinese poems written by Gusadang Kim, Nak-Haeng (구사당(九思堂) 김낙행(金樂行)의 한시(漢詩) 연구(硏究))

  • Gusadang Kim, Nak-Haeng is a scholar of 18th century in Yeongnam region who wrote about 130 Chinese poems. In this study, I searched Gusadang's inner world by interpreting his Chinese poems. His life is closely related to his father Jesan Kim, Seong-Tak who was exiled. The fact that he devoted himself to his father for 10 years shows he had strong standards in making decisions in life. In short, Gusadang was a person who looked gentle but was tough inside and he remained firm in his faith even with outside pressure. He could not achieve glory because he spent time serving his father in his thirties. Although he heard compliments from others that he was talented enough to succeed as a scholar, he lacked time and mental energy to study. Also, he was a moralist and wrote some poems about impressive events in his life even though he did not fully devote himself to writing poems. In this study, I searched his inner world focusing on how he felt and thought about outside world by analyzing his poems. In conclusion, I found three characteristics from his poems. Firstly, depressed feelings are shown based on excessive self-consciousness in the poems related to his father. Secondly, his will to keep balance in life is shown because he wanted a harmonious life as a seeker after truth. Thirdly, a sense of isolation is shown because he had to keep a distance from outside world.
