마유(Horse Fat)의 산화안정성에 대한 α-, γ-, δ-토코페롤의 첨가 효과 (Effects of α-, γ-, and δ-tocopherol on the oxidative stability of horse fat)

  • 본 연구는 $70^{\circ}C$로 저온 감압 추출한 마유에 ${\alpha}$-, ${\gamma}$-, ${\delta}$-토코페롤을 0, 30, 60, 150 mg/kg 농도로 첨가한 뒤 토코페롤 함량 변화와 지방산 조성 변화, 과산화물값, TBA값을 측정하여 저장 중산화안정성을 조사하였다. 마유를 구성하고 있는 주요 지방산은 oleic acid>palmitic acid>linoleic acid>palmitoleic acid>stearicacid>linolenic acid 순이었으며 $65^{\circ}C$, 14일 저장 동안에는 대부분의 지방산 사이에는 유의적인 변화가 나타나지 않았다. 하지만 불포화지방산 함량은 토코페롤을 첨가한 마유가 토코페롤을 첨가하지 않은 마유에 비해 감량 변화가 적어 토코페롤이 마유 유지의 산화안정성에 도움을 준 것으로 나타났다. 과산화물값의 경우, 토코페롤을 첨가하지 않은 마유는 $65^{\circ}C$에서 저장 14일 후 3.45에서 43.75 meq/kg으로 증가하였고 ${\gamma}$-와 ${\delta}$-토코페롤을 첨가한 마유의 과산화물값은 ${\alpha}$-토코페롤을 첨가한 마유의 과산화물값보다 낮아 ${\gamma}$-와 ${\delta}$-토코페롤이 ${\alpha}$-토코페롤에 비해 마유의 산화안정성을 증가시킨 것으로 나타났다. 마유의 TBA값도 저장 14일 후1.03에서 7.87 mg MA/kg으로 증가하였지만 ${\alpha}$-토코페롤을 150 mg/kg 첨가한 마유와 ${\delta}$-토코페롤을 30 mg/kg을 첨가한 경우를 제외하고 과산화물값과 같은 경향의 결과를 나타내었다. ${\alpha}$-, ${\gamma}$-, ${\delta}$-토코페롤 종류에 따른 토코페롤의 분해율은 ${\alpha}$-토코페롤>${\gamma}$-토코페롤>${\delta}$-토코페롤 순으로 결과가 나타나 ${\gamma}$-와 ${\delta}$-토코페롤을 마유에 첨가하였을 경우 마유의 산화안정성에 도움을 주었다. 마유를 재료로 생산되는 화장품 제품에는 주로 ${\alpha}$-토코페롤을 첨가하여 저장성을 향상시키는데 본 연구에서 측정한 토코페롤 종류별 산화 안정성과 불포화지방산의 저장 중 변화 및 토코페롤의 분해율을 고려하였을 때 ${\alpha}$-토코페롤에 비해 ${\gamma}$-와 ${\delta}$-토코페롤이 마유의 저장성 향상에 도움을 줄 수 있을 것으로 판단되었다.

제주 사육 말에서 말전염성빈혈 조사 (Surveillance for Equine Infectious Anemia in Jeju Island)

  • 말전염성빈혈(EIA)은 전세계의 마과동물에서 발생하는 전염병이다. 2005년부터 2011년까지 제주에서 사육되는 말을 대상으로 EIA를 조사하였다. 한국마사회 제주경마공원 동물병원에서 제주마 1,329 마리 및 한라마(제주산마) 8,324 마리 그리고 제주목장의 동물병원에서 Thoroughbred 말 387 마리 등 7년간 총 10,040마리를 효소면역측정법을 이용하여 검사하였다. 조사기간 동안 혈액에서 EIA 항체는 검출되지 않았다. 1985년 이후부터 제주에서 EIA 항체가 검출된 적이 없고, 발병 예도 없었다. 그러므로 제주에는 EIA가 없는 것으로 생각된다.

Comparison for immunophysiological responses of Jeju and Thoroughbred horses after exercise

  • Objective: The study was conducted to investigate variations in the immunophysiological responses to exercise-induced stress in Jeju and Thoroughbred horses. Methods: Blood samples were collected from the jugular veins of adult Jeju (n = 5) and Thoroughbred (n = 5) horses before and after 30 min of exercise. The hematological, biochemical, and immunological profiles of the blood samples were analyzed. Blood smears were stained and observed under a microscope. The concentration of cell-free (cf) DNA in the plasma was determined using real time polymerase chain reaction (PCR). Peripheral blood mononuclear cells (PBMCs) and polymorphonuclear cells were separated using Polymorphprep, and the expression of various stress-related and chemokine receptor genes was measured using reverse transcriptase (RT) and real-time PCR. Results: After exercise, Jeju and Thoroughbred horses displayed stress responses with significantly increased rectal temperatures, cortisol levels, and muscle catabolism-associated metabolites. Red blood cell indices were significantly higher in Thoroughbred horses than in Jeju horses after exercise. In addition, exercise-induced stress triggered the formation of neutrophil extracellular traps (NETs) and reduced platelet counts in Jeju horses but not in Thoroughbred horses. Heat shock protein 72 and heat shock protein family A (Hsp70) member 6 expression is rapidly modulated in response to exercise-induced stress in the PBMCs of Jeju horses. The expression of CXC chemokine receptor 4 in PBMCs was higher in Thoroughbred horses than in Jeju horses after exercise. Conclusion: In summary, the different immunophysiological responses of Jeju and Thoroughbred horses explain the differences in the physiological and anatomical properties of the two breeds. The physiology of Thoroughbred horses makes them suitable for racing as they are less sensitive to exercise-induced stress compared to that of Jeju horses. This study provides a basis for investigating the link between exercise-induced stresses and the physiological alteration of horses. Hence, our findings show that some of assessed parameters could be used to determine the endurance performance of horses.

Molecular Sexing and Species Identification of the Processed Meat and Sausages of Horse, Cattle and Pig

  • We developed a polymerase chain reaction (PCR)-based molecular method for sexing and identification using sexual dimorphism between the Zinc Finger-X and -Y (ZFX-ZFY) gene and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) for mitochondrial DNA (mtDNA) cytochrome B (CYTB) gene in meat pieces and commercial sausages from animals of different origins. Sexual dimorphism based on the presence or absence of SINE-like sequence between ZFX and ZFY genes showed distinguishable band patterns between male and female DNA samples and were easily detected by PCR analyses. Male DNA had two PCR products appearing as distinct two bands (ZFX and ZFY), and female DNA had a single band (ZFX). Molecular identification was carried out using PCR-RFLP of CYTB gene, and showed clear species classification results. The results yielded identical information on the sexes and the species of the meat samples collected from providers without any records. The analyses for DNA isolated from commercial sausage showed that pig was the major source but several sausages originated from chicken and Atlantic cod. Applying this PCR-based molecular method was useful and yielded clear sex information and identified the species of various tissue samples originating from livestock.

Small intestinal epiploic foramen entrapment in a seven-month pregnant Thoroughbred mare: a case report

  • A seven-month pregnant four-year-old Thoroughbred mare with colic weighing 600 kg was referred to the Jeju National University Equine Hospital. A physical examination and ultrasound suggested epiploic foramen entrapment. The patient underwent ileum resection and end-to-side jejunocecostomy. After surgery, the horse showed a consistent pain when introduced to feeding. A repeat laparotomy was performed to enlarge the stoma size with side-to-side jejunocecostomy. The repeat laparotomy was successful, and the mare gave birth uneventfully. This report describes a case of small intestinal epiploic foramen entrapment and the clinical outcomes of the 2 consecutive laparotomies a Thoroughbred mare in late gestation.

부산경남경마공원 비경주마의 행동장애 유형 (The Prevalence of Behavioral Disorders in Non-racehorses at Busan Race Park)

  • This study was conducted on 85 non-racing pleasure horses raised in Busan race park of Korea Racing Authority to investigate the prevalence of stereotypic behavioral problems in horses. the observation had been continued from March to October of 2010, and the study animals included warm-bloods, Jeju ponies and donkeys. Although there was no temperament problem detected, a wide range of well-known stable vices were observed in 15 horses. The most frequently observed stereotype was cribbing (8.2%), and it was followed by weaving (1.2%), biting (1.2%), eating too fast (1.2%), pawing (1.2%) and scraping the teeth on something (1.2%) with the equal prevalence rate. These behavioral disorders were only detected in warm-bloods, while those were absent in Jeju ponies and donkeys. In addition, primary behavioral disorders were more dominant than mimicking behaviors.

제주마 집단의 세대 경과에 따른 모색 유전자 변화 연구 (A Study on the Changes of Coat Color-Related Genes according to Generational Changes in Jeju Horses)

  • 본 연구는 제주마 집단의 모색유전자 분포 특성 및 세대별 모색 유전자 변화 양상을 분석하기 위해 제주특별자치도에서 관리하는 제주마, 1,462두에 대한 모색 관련 유전자인 MC1R, ASIP, ECA3-inversion, STX17을 분석하였다. 흑모색과 관련된 MC1R 유전자의 $E^+/E^+$$E^+/E^e$ 유전자형 빈도는 각각 0.122와 0.447로 조사되었고, 적색(chestnut) 유전자형인 $E^e/E^e$ 유전자형 빈도는 0.429로 확인되었다. ASIP 유전자의 $A^A/A^A$, $A^A/A^a$$A^a/A^a$ 유전자형 빈도는 각각 0.46, 0.448, 0.091로 확인되어 $A^a/A^a$ 유전자형 빈도가 낮았다. 토비아노(Tobiano) 형태를 발현하는 To/To와 +/To 유전자형은 각각 0.001과 0.119로 토비아노 형태의 모색은 12%를 점유하고 있었다. 회색 모색과 관련된 STX17의 G/G와 G/g 유전자형의 빈도는 각각 0.002와 0.680로 나타나 제주마 집단에 회색마는 68.2%를 점유하였다. 세대별 모색 변화는 MC1R, ASIP 유전자에서는 큰 변화를 확인할 수 없었다. ECA3-inversion에서는 토비아노를 발현하는 To allele은 세대변화에 따라 유의적으로 감소하였고(p<0.05), 회색 모색과 관련된 STX17 G allele은 세대변화에 따라 유의적으로 증가하였다(p<0.05). 본 연구결과, 제주마 집단의 모색 다양성을 유지하기 위해서는 종마 선정 단계에 모색 유전자의 검토가 필요한 것으로 사료되었다.

제주신화 자청비(청비) 콘텐츠를 응용한 텍스타일 디자인 (Textile Design using Contents of Jeju Myth, Jacheongbi)

  • The purpose of this study is, as one of Jeju culture contents projects, to suggest a new method to develop the textile design using contents of Jeju Myth, Jacheongbi, in other words, to create mythic patterns by borrowing effectively from the mythic image of Jacheongbi on contemporary fashion. To achieve this, after extracting mythic contents from real animals from the myth (chicken, horse, cow and owl) of Jacheongbi, we draw the animal motifs from a mythic image basis of archetypal symbols and empirical awareness about animals and shamanistic imagination, and create mythic patterns. In order to achieve this design pattern, Adobe Photoshop CS5 Extended was used to design the animal motifs, and then they were arranged via bidirectional layout. And to conclude, development of the creative textile design using the mythic contents of Jacheongbi contribute to invigorating the fashion industry and regional culture contents projects in Jeju, and also become the basis of creating added value to it.

Nutritional Composition in Bone Extracts from Jeju Crossbred Horses at Different Slaughter Ages

  • The effects of slaughter age (28, 32, or 38 mon) on the proximate composition, collagen content, fatty acid composition, amino acid content, and mineral contents of horse leg bone (femur and tibia) extracts (HLBE) obtained from Jeju crossbred horses (n=14) in Korea. HLBE was extracted for 24 h with boiling water. At those ages, the respective proximate compositions of the HLBE were found as follows: 5.20-6.42% crude protein; fat 0.61-1.65% crude; and 0.10-0.22% crude ash. At 32 and 38 mon, the HLBE showed higher levels of both crude protein and collagen than at 28 mon. The major fatty acids of the horse leg bone extract powder (HLBP) were palmitic acid and oleic acid. Palmitoleic acid and the essential fatty acids were higher in the HLBP at 38 mon compared to that at 28 mon. Nearly all the amino acids were found at higher levels in the HLBP at 38 mon than at 28 mon, except histidine. The P, K, Zn, Se and Fe contents in the HLBP increased significantly with age. These results suggest that some nutrients in the HLBE increase with age, and that extracts from horses older than 32 mon would be more nutritious for human consumers.
