The beef color stability during display of two muscles, m. longissimus thoracis and m. semitendinosus, of Japanese Shorthorn steers (n=14) was compared with that of Japanese Black steers (n=14). The beef color of each carcass was evaluated according to the Japanese Grading Standards at 24 h post mortem. Steak samples from muscles were over-wrapped with PVC film and displayed under fluorescent lights at $4^{\circ}C$ for 9 days. Metmyoglobin percentages of steak samples were determined at days 0, 3, 6 and 9. The overall grade of beef color of the carcasses of Japanese Shorthorn steers was significantly (p<0.05) lower than that of Japanese Black steers. The metmyoglobin percentages during the display of two muscles of Japanese Shorthorn steers were significantly (p<0.05) lower than those of Japanese Black steers. These results suggested that though beef color evaluation of the carcasses of Japanese Shorthorn steers was lower than that of Japanese Black steers, the beef color stability during the display of the muscle of Japanese Shorthorn steers was higher than that of Japanese Black steers.
The growth performance of embryo-transferred Japanese Black calves that were born from, and suckled by, Japanese Shorthorn cows in a cow-calf grazing system (BS-group, n = 5) was compared to that of Japanese Black calves from Japanese Black cows in a cowshed (BB-group, n = 5). The daily weight gain from birth to 1 month was higher in the BS-group than in the BB-group (p<0.01), and the same trend (p<0.05) was observed at 2 and 3 months of age. This resulted in body weight that was significantly higher for the BS-group between 1 and 3 months of age than what was observed for the BB-group (p<0.05). Heart girth was significantly greater in the BS-group than in the BB-group throughout the experimental period (p<0.01), and chest depth and withers height in the BS-group were significantly greater from 2 to 4 months of age (p<0.05) and at 4 months of age only (p<0.05). No difference in body length (p>0.05) was observed between the groups. These results suggest that the maternal effect of Japanese Shorthorn cows was positive for embryo-transferred Japanese Black calf growth during the early suckling stage. As Japanese Black calves are traded at a high price on the Japanese market, we conclude that this proposed production system is likely to improve the profitability of herd management in upland Japan.

The complete mtDNA D-loop regions of Japanese and Korean cattle were analyzed for their mtDNA variations and genetic relationships. Sequencing the 30 Higo substrain and 30 Tosa substrain of Japanese Brown, respectively 12 and 17 distinct Bos haplotypes were identified from 77 polymorphic nucleotide sites. In order to focus on the relationships among Japanese and Korean cattle, two types of phylogenetic tree were constructed using individual sequences; first, a neighbor-joining tree with all sequences and second, reduced median networks within each Japanese and Korean cattle group. The trees revealed that two major mtDNA haplotype groups, T3 and T4, were represented in Japanese and Korean cattle. The T4 haplogroup predominated in Japanese Black and Japanese Brown cattle (frequency of 43.3-66.7%), while the T3 haplogroup was predominant (83.3%) and T4 was represented only twice in the Korean cattle. The results suggested that the mitochondrial origins of Japanese Brown were Japanese ancient cattle as well as Japanese Black in despite of the considerable introgression of Korean and European cattle into Japanese Brown.

In the meat industry, correct breed information in food labeling is required to assure meat quality. Genetic markers provide corroborating evidence to identify breed. This paper describes the development of DNA markers to discriminate between Japanese Black and F1 (Japanese Black${\times}$Holstein) breeds. The amplified fragment length polymorphism method was employed to detect candidate markers absent in Japanese Black but present in Holstein. The 1,754 primer combinations yielded eleven markers that were converted into single nucleotide polymorphism markers for high-throughput genotyping. The allele frequencies in both breeds were investigated for discrimination ability using PCR-RFLP. The probability of identifying F1 was 0.9168 and probability of misjudgment was 0.0066 using four selected markers. The markers could be useful for discriminating between Japanese Black and F1 and would contribute to the prevention of falsified breed labeling of meat.
Objective: The objective of this study was to identify environmental factors strongly associated with and to estimate genetic parameters of reproductive traits in Japanese Black heifers. Methods: Data included reproduction records of Japanese Black heifers born between 2004 and 2014. First service non-return rate (NRR) to 56 days from first to successful insemination (FS), number of services per conception (IN), age at first calving (AFC) and gestation length were analyzed with the use of the general linear model. Genetic parameters were estimated with the use of the univariate animal model of the residual maximum likelihood. Results: Averages of reproductive traits over eleven years were assessed, and the effects of farm, year, month, artificial insemination technician and interaction of farm×year on the traits were determined. Estimated heritability of FS was very low and that of AFC was higher than that of the other traits. A close genetic relation was observed among NRR, IN, and FS; however, their heritabilities were very low. AFC shows favorable genetic correlation with IN and FS. Conclusion: Low heritabilities of most reproductive traits in Japanese Black heifers are strongly influenced by farm management practices, and that large residual variances make genetic evaluation difficult. Among the reproductive traits, AFC is potentially more useful for genetic improvement of heifer reproductive traits because it has high heritability and favorable genetic correlations with IN and FS.
Reproductive data collected from more than 20,000 Japanese Black cows of Hyogo and Shimane Prefectures were analyzed. Averages of age at first calving, gestation length, days open and calving interval were 25.1 mo, 289 d, 112 d and 401 d, respectively. Variance components were obtained by REML procedure and the heritability estimate of age at first calving was 0.22. In gestation length the heritability estimate was 0.40 and no permanent environmental effect was estimated. Estimated variance components of calving interval were similar to those of days open and the heritability and repeatability of calving interval were 0.05 and 0.09, respectively. Random farm effects accounted for approximately 10% of phenotypic variations in all traits. Genetic and farm correlations between age at first calving and calving interval were 0.27 and 0.39, respectively. It was found that temporary environment was an important source of variation for calving intervals of Japanese Black.

To evaluate direct and maternal effects on calf market weight (CMW) and carcass weight (CW) in Japanese Black cattle under an animal model, genetic parameters were estimated using 51,320 records of CMW and 11,944 records of CW, respectively. Direct and maternal heritabilities, and direct-maternal genetic correlation were estimated to be 0.22, 0.06 and 0.27 for CMW, and 0.23, 0.12 and -0.40 for CW, respectively. Correlation coefficient between maternal breeding values for CMW and CW was 0.521 for 157 sires appeared in both CMW and CW data sets. These results suggest that the maternal genetic effect on pre-weaning growth carries over to carcass weight. Maternal breeding values for both calf market weight and carcass weight could be used as the indicator traits of maternal ability in Japanese Black cattle.
A tetraparental chimeric bull was successfully produced by aggregating bovine IVF embryos of F1 (female Holstein${\times}$male Japanese Black) and F1(female Japanese Brown${\times}$male Limousin) and culturing in vitro without the zona pellucida at Yamaguchi Research Station in Japan. In the microsatellite genotyping, 12% (28/228) microsatellite primer sets ware potentially useful for this parentage analysis in the chimeric bull, 78.6% (22/28) of microsatellite present in the chimeric bull were uniquely contributed from the Japanese Black and 21.4% (6/28) from Limousin. This chimeric bull semen was used in producing IVF embryos. The chromosome preparations were made from peripheral lymphocytes. Based on chromosome analysis the Chimera had apparently normal chromosomes (29 acrocentric pairs, one large sub metacentric X chromosome and one small sub metacentric Y chromosome). The proportion of acrosome reacted spermatozoa after 1 h of incubation was higher (p<0.01) with the Chimera than with the Holstein and in Japanese Brown bulls. But did not differ from Japanese Black and Limousin bull sperm. Fertilization rates observed after 5 h of sperm-oocyte incubation with Chimera sperm were higher (p<0.05) than with Japanese Brown and (p<0.01) than with Holstein sperm, but did not differ from Japanese Black and Limousin sperm. The cleavage rates of IVF oocytes inseminated with Chimera sperm were also higher (p<0.001) compared with Holstein, (p<0.01) Japanese Brown and (p<0.05) Limousin, but did not differ from Japanese Black sperm. The blastocyst rates of IVM oocytes inseminated with sperm were higher (p<0.05) than in Limousin, Japanese Brown and Holstein, but did not differ from Japanese Black. Chimeric cattles were produced by aggregation of parthenogenetic (Japanese Brown) and in vitro fertilized (Holstein) bovine embryos at the Yamaguchi Research Station in Japan and by aggregation of parthenogenetic (Red Angus) and in vitro fertilized (Holstein) embryos at the St. Gabriel Research Station in Louisiana. The aggregation rate of the reconstructed demi-embryos cultured in vitro without agar embedding was significantly lower than with agar embedding. The aggregation was also lower when the aggregation resulted from a whole parthenogenetic and IVF-derieved embryos cultured without agar than when cultured with agar. The development rate to blastocysts, however, was not different among the treatment. To verify parthenogenetic and the cells derieved from the male IVF embryos in blastocyst formation, 51 embryos were karyotyped, resulting in 27 embryos having both XX and XY chromosome plates in the same sample, 14 embryos with XY and 10 embryos with XX. The viability and the percentage of zonafree chimeric embryos at 24 h following cryopreservation in EG plus T with 10% PVP were significantly greater than those cryopreserved without PVP. Pregnancies were diagnosed in both stations after the transfer of chimeric blastocysts. Twin male and single chimeric calves were delivered at the Yamaguchi station, with each having both XX and XY chromosomes detected. Three pregnancies resulted from the transfer of 40 chimeric embryos at the Louisiana station. Two pregnancies were Jost prior to 4 months and one phenotypically chimeric viable male born.
Individual management of the animal is the first step towards reaching the goal of precision livestock farming that aids animal welfare. Accurate recognition of each individual animal is important for precise management. Electronic identification of cattle, usually referred to as RFID (Radio Frequency Identification), has many advantages for farm management. In practice, however, RFID implementations can cause several problems. Reading speed and distance must be optimized for specific applications. Image processing is more effective than RFID for the development of precision farming system in livestock. Therefore, the aim of this paper is to attempt the identification of cattle by using image processing. The majority of the research on the identification of cattle by using image processing has been for the black-and-white patterns of the Holstein. But, native Japanese and Korean cattle do not have a consistent pattern on the body, so that identification by pattern is impossible. This research aims to identify to Japanese black cattle, which does not have a black-white pattern on the body, by using image processing and a neural network algorithm. 12 Japanese black cattle were tested. Values of input parameter were calculated by using the face image values of 12 cows. The face was identified by the associate neural memory algorithm, and the algorithm was verified by the transformed face image, for example, of brightness, distortion, noise and angle. As a result, there was difference due to a transformation ratio of the brightness, distortion, noise, and angle. The algorithm could identify 100% in the range from -30 to +30 degrees of brightness, -20 to +40 degrees of distortion, 0 to 60% of noise and -20 to +30 degree of angle transformed images.
