Exogenous proline mitigates the detrimental effects of saline and alkaline stresses in Leymus chinensis (Trin.)

  • Proline accumulates in plants under environmental stresses including saline stress and alkaline stress. Here, we investigated the responses to two different stresses, saline stress (200 mM NaCl) and alkaline stress (100 mM $Na_2CO_3$) in two Leymus chinensis (Trin.) genotypes, LcWT07 and LcJS0107, and effects of exogenous proline on the activities of antioxidant enzymes. Both saline stress and alkaline stress significantly induced the accumulation of proline in leaves of the two genotypes after 96 h, and alkaline stress caused a transient and significant increase in LcJS0107 plants at 6 h. A reduction in the activities of catalase (CAT, EC 1.11.1.6) and ascorbate peroxidase (APX, EC 1.11.1.11), but not in the activity of superoxide dismutase (SOD, EC 1.15.1.1), was detected in plants exposed to saline and alkaline stresses. Remarkable decrease in relative water contents (RWC) was found in 144 h stressed plants. However, lipid peroxidation estimated by malonyldialdehyde (MDA) content in leaves remained relatively stable. With the addition of exogenous proline, it did not cause changes of proline levels in two genotypes, but combined with saline or alkaline stress, the exogenous application of proline significantly induced proline accumulation after even short treatment periods. Combined with salt stress, the exogenous application also increased the activities of CAT and APX. These results indicated that exogenous proline not only increases proline levels in vivo as a osmotic adjustment under stress, but mitigates the detrimental effects of saline and alkaline stresses by increasing the activities of antioxidant enzymes.

  • Eikenella corrodens is Gram-negative, facultatively anaerobic, and rod-shaped bacterium. It is a part of the normal human mucosal flora that can cause several systemic diseases such as endocarditis, liver abscess, and intracranial bacterial infection. E. corrodens KCOM 3110 (= JS217) was isolated from human subgingival dental plaque of periodontitis lesion. Here, we present the complete genome sequence of E. corrodens KCOM 3110.

Gamma-Aminobutyric Acid Production from a Novel Enterococcus avium JS-N6B4 Strain Isolated from Edible Insects

  • Gamma-aminobutyric acid (GABA)-producing strains were isolated from four edible insects and subjected to 16S rRNA sequence analysis. Among the four GABA-producing bacteria, Enterococcus avium JS-N6B4 exhibited the highest GABA-production, while cultivation temperature, initial pH, aerobic condition, and mono-sodium glutamate (MSG) feeding were found to be the key factors affecting GABA production rate. The culture condition was optimized in terms of glucose, yeast extract, and MSG concentrations using response surface methodology (RSM). GABA production up to 16.64 g/l was obtained under the conditions of 7 g/l glucose, 45 g/l yeast extract, and 62 g/l MSG through the optimization of medium composition by RSM. Experimental GABA production was 13.68 g/l, which was close to the predicted value (16.64 g/l) calculated from the analysis of variance, and 2.79-fold higher than the production achieved with basic medium. Therefore, GABA-producing strains may help improve the GABA production in edible insects, and provide a new approach to the use of edible insects as effective food biomaterials.

  • Semi-supervised learning uses both labeled data and unlabeled data. Recently consistency regularization is very popular in semi-supervised learning. Unsupervised data augmentation (UDA) that uses unlabeled data augmentation is also based on the consistency regularization. The Kullback-Leibler divergence is used for the loss of unlabeled data and cross-entropy for the loss of labeled data through UDA learning. UDA uses techniques such as training signal annealing (TSA) and confidence-based masking to promote performance. In this study, we propose to use Jensen-Shannon divergence instead of Kullback-Leibler divergence, reverse-TSA and not to use confidence-based masking for performance improvement. Through experiment, we show that the proposed technique yields better performance than those of UDA.

  • Smart Farm, which combines information and communication technologies with agriculture is moving from simple monitoring of the growth environment toward discovering the optimal environment for crop growth and in the form of self-regulating agriculture. To this end, it is important to collect related data, but it is more important for farmers with cultivation know-how to analyze the collected data from various perspectives and derive useful information for regulating the crop growth environment. In this study, we developed a web service that allows farmers who want to obtain necessary information with data related to crop growth to easily analyze data. Web-based data analysis serivice developed uses R language for data analysis and Express web application framework for Node.js. As a result of applying the developed data analysis service together with the growth environment monitoring system in operation, we could perform data analysis what we want just by uploading a CSV file or by entering raw data directly. We confirmed that a service provider could provid various data analysis services easily and could add a new data analysis service by newly adding R script.

A Study on the Correlation between Prefer Spicy and Physical symptoms (신미(辛味) 기호에 따른 신체증상발현의 상관성 연구)

  • Objectives : These days many people tend to prefer spicy taste. The purpose of this study was to find out the relationship between prefer spicy and physical symptoms. Methods : We examined the subjective physical condition of patients who visited L/C clinic during the period between January and June 2023 by conducting a survey. The survey was completed voluntarily, and the anonymity and confidentiality of the research data were strictly protected, and it was stated that patients could withdraw at any time if they did not want to participate. Patients who refused to complete the survey and patients with limited capacity to give consent were excluded. The survey took about 10 minutes to complete. We analyzed 248 cases who answered the survey and found that 66 patients had a high spicy preference. Results : As a result, participants with high spicy taste preference tended to have general body symptoms such as fever, sweat, and thirst; digestive symptoms such as belching, constipation, bloody stools, and abdominal distension; genitourinary symptoms such as yellowish urine, urinary retention, white fluor albus, and premenstrual tension; and neuromuscular symptoms such as edema, blepharospasm, and cold hands. Conclusion : People who have general body symptoms, digestive symptoms, genitourinary symptoms and neuromuscular symptoms described above should try to eat as little spicy food as possible and make sure they get a good balance of the five flavors.

  • Under the "Act on Safety Control of Small Public Facilities (enacted in 2015)", each local government selects and conducts annual safety inspections for small public facilities. Among small public facilities, small bridges pose high risks and are heavily utilized by local residents, making them challenging to manage due to their large numbers and limited resources. Therefore, there is a need for a rational selection method that considers the management capacity of local governments, as well as the establishment of objective risk evaluation and maintenance planning for small hazardous facilities. In this study, we propose a selection method based on structural and functional classification of small bridges, considering the management capacity of local governments. Additionally, we present quantitative evaluation indicators for ten risk evaluation criteria, aiming to facilitate objective risk evaluation.

  • Due to the outbreak of COVID-19, the domestic disinfection and quarantine market has grown, and the demand for disinfection operators who want to recruit disinfectants and disinfectants who want to work has increased, but it is not easy to find jobs. In addition, there is a need for a system that can automate or efficiently manage work by hand-recording and writing a disinfection record for disinfection work. Therefore, in this paper, an O2O disinfection service system is designed using the MVC pattern and implemented through MySQL, Ejs/BootStrap view, and Node.js. Additionally, it connects with AWS cloud services so users can use the system anytime, anywhere. Through the proposed O2O disinfection service system, we hope to solve the difficulties of recruiting personnel in the domestic disinfection station market and improve the existing inefficient disinfection work process.

New Classification of Plasmodiophora brassicae Races Using Differential Genotypes of Chinese Cabbage

  • Clubroot disease caused by Plasmodiophora brassicae induces severe losses of cruciferous vegetables worldwide. To control clubroot of Chinese cabbage, many CR (clubroot resistance) F1 hybrid cultivars have been bred and released in Korea, China and Japan. In this study, we determined the race of P. brassicae 12 field isolates, which collected from 10 regions in Korea, using Williams' differential varieties including two cabbage ('Jersey Queen', 'Badger Shipper') and two rutabaga ('Laurentian', 'Whilhelmsburger'). By Williams' differential varieties, 12 clubroot pathogens were assigned into one (GN2), two (HS and YC), two (HN1 and HN2), three (DJ, KS and SS) and four (GS, GN1, JS and PC) isolates for races 1, 2, 4, 5 and 9, respectively. In addition, the degree of resistance of 45 CR cultivars that were from Korea, China and Japan was tested with the 12 isolates. The 45 CR cultivars of Chinese cabbage were differentiated into three genotypes according to their resistance responses. Even though the 12 P. brassicae isolates were same race by Williams' differential varieties, three CR genotypes showed different resistance response to the isolates. These results indicate that races of P. brassicae by Williams' differentials were not related with resistance of CR cultivars, and three CR genotypes represented qualitative resistance to the P. brassicae isolates. CR genotype I including 'CR-Cheongrok' showed resistance to GN1, GN2, JS, GS, HS, DJ and KS isolates and susceptibility to YC, PC, HN1, HN2 and SS isolates. And CR genotype II such as 'Hangkunjongbyungdaebaekchae' was resistant to GN1, GN2, JS, GS, HS, YC, PC and HN1 and susceptible to DJ, KS, SS and HN2. CR genotype III including 'Chunhajangkun' and 'Akimeki' represented resistance to 10 isolates except for SS and HN2 isolates. Based on these results, we selected 'CR-Cheongrok', 'Hangkunjongbyungdaebaekchae', and 'Chunhajangkun' as a representative cultivar of three CR genotypes and 'Norangkimjang' as a susceptible cultivar. Furthermore, we investigated the resistance of 15 lines of Chinese cabbage, which were provided by seed companies, to 11 isolates except for HN1 of P. brassicae. The results showed that three lines were susceptible to all the tested isolates, whereas five, four, and three lines represented the similar responses corresponding to the CR genotypes I, II, and III, respectively; there is no line of Chinese cabbage showing different resistance patterns compared to three CR genotypes. In particular, line 'SS001' showing resistance responses of CR genotype II was a parent of 'Saerona' that have been commercialized as a CR $F_1$ cultivar of Chinese cabbage. Together, we divided 12 isolates of P. brassicae into 4 races, designated by wild type, mutant type 1, mutant type 2, and mutant type 3. Wild type including GN1, GN2, JS, GS, and HS isolates of P. brassicae was not able to infect all the cultivars of three CR genotypes, whereas, mutant type 3 such as SS and HN2 isolates developed severe clubroot disease on all the CR genotype cultivars. To mutant type 1 including DJ and KS isolates, CR genotypes I, II and III were resistant, susceptible and resistant, respectively. In contrast, to mutant type 2 including YC, PS, and HN1 isolates, CR genotypes I, II and III showed susceptibility, resistance and resistance, respectively. Taken together, our results provide the extended knowledge of classification of P. brassicae races, which is useful information for the breeding of resistant crops, with a suggestion that 'Norangkimjang', 'CR-Cheongrok', 'Saerona' and 'Chunhajangkun' cultivars of Chinese cabbage could be used as new race differentials of P. brassicae for clubroot disease assay.

Fecal Microbiota Profiling of Holstein and Jersey, in South Korea : A Comparative Study (국내에서 사육되는 Holstein 젖소과 Jersey 젖소의 대변 미생물 분석 : 비교연구)

  • In light of the complex interactions between the host animal and its resident gut microbiomes, studies of these microbial communities as a means to improve cattle production are important. This study was conducted to analyze the intestinal microorganisms of Holstein (HT) and Jersey (JS), raised in Korea and to clarify the differences in microbial structures according to cattle species through next-generation sequencing. The alpha-diversity analysis revealed that most species richness and diversity indices were significantly higher in JS than in HT whereas phylogenetic diversity, which is the sum of taxonomic distances, is not significant. Microbial composition analysis showed that the intestinal microbial community structure of the two groups differed. In the both groups, a significant correlation was observed among the distribution of several microbes at the family level. In particular, a highly significant correlation (p<0.0001) among a variety of microbial distributions was found in JS. Beta-diversity analyis was to performed to statistically verify whether a difference exists in the intestinal microbial community structure of the two groups. Principal coordinate analysis and unweighted pair group method with arithmetic mean (UPGMA) clustering analysis showed separation between the HT and JS clusters. Meanwhile, permutational multivariate analysis of variance (PERMANOVA) revealed that their microbial structures are significantly different (p<0.0001). LEfSe biomarker analysis was performed to discover the differenc microbial features between the two groups. We found that several microbes, such as Firmicutes, Bacilli, Moraxellaceae and Pseudomonadales account for most of the difference in intestinal microbial community structure between the two groups.
