Analysis of Pollution Characteristics in the Mainstream and Its Tributaries of Gongneung Stream Using Water Quality Index and Pollution Load Data (통합수질지수 및 오염부하자료를 이용한 공릉천 유입지천과 본류의 오염특성 분석)

  • In this study, we identified the major pollution-zones of the mainstream and its tributaries of Gongneung stream and investigated their pollution sources based on water quality, flowrate and pollution-load data of the stream having the characteristics of the urban-rural complex to examine the effect of the tributaries on the water quality changes in the mainstream. The water quality and flowrate data were collected for 10 months (2018 ~ 2019) at 3 points of mainstream and ten tributaries. Water quality index (WQI), load duration curve (LDC), discharge load density (DLD)and delivery ratios for each tributary were obtained so as to investigate the pollution characteristics and some of the information visualized on GIS. The main pollution-zone in the Gongneung stream was in the middle and lower streams, and the tributaries that may affect the pollution of the area were JS, JY, SL and SM. JS and SL had low WQI levels (34.7/37.5) and DLD (kg/d/㎢) of BOD and T-P were relatively high in JY (99.2/6.00) and SL (60.0/2.07). BOD and T-P delivery ratios in JS were high (0.94/0.83), suggesting that JS had significant influence on the water quality of the main pollution-zone in the Gongneung stream. Meanwhile, SM having a high T-P delivery ratio (0.97) was found to be more affected by the non-point source due to the higher LDC excess rate (%) in the low flow compared to high flow. This study provides basic data on the water quality and pollution characteristics of the Gongneung stream, and the analysis results are expected to be used as examples for identifying the main pollution-zone and tributaries of stream and their pollution sources.

A Study on Mineralogical Characteristic and Accelerated Weather Test of Red and Yellow Color Natural Inorganic Pigments (적색 및 황색 계열의 천연 무기안료의 광물학적 특성 및 촉진내후성 평가 연구)

  • We evaluated mineralogical, physical property and accelerated weather resistance of cinnabar, realgar and orpiment used as cultural heritages and traditional paintings. JB and JS are sample names of cinnabar and JH and UH are sample names of orpiment and Realgar, respectively. As a result of mineralogical property evaluation, Jinsa was identified cinnabar (HgS) and there was no difference in chemical composition according to the grade. UH and JH were confirmed realgar (AsS) and orpiment (As2S3), respectively. The polarization characteristics such as shape and color indicate that pigments using our test were natural mineral pigment observed by polarization microscope. Especially, in the case of cinnabar, it is not easy to distinguish between natural mineral pigment and synthetic pigment. But the results of polarization microscope said that cinnabar is natural occurring mineral which have authentic mineral particle and unevenly fracture. As a result of thermal analysis, JH has a higher glass transition temperature and heat stability than UH. After accelerated weather test, cinnabar, realgar and orpiment were striking result in color change because of light degradation. Red color of cinnabar turn into black and the color of realgar and orpiment became brighter than before. JB (meta-cinnabar) is more photosensitivity and faster becoming dark than JB (cinnabar). Finally light transforms realgar (red, As4S4) in arsenolite (As2O3).

The Effects of Performance-based HRM on Organizational Effectiveness : In the Case of a Public Corporation (성과주의 인적자원관리가 조직유효성에 미치는 영향에 관한 실증연구 - 공기업 직원의 인식을 중심으로 -)

  • This study aims to empirically verify the positive impacts of PB(performance-based) HRM on the organizational effectiveness in a public corporation. The independent variables are 'PB staffing', 'PB appraisal', 'PB compensation'. The dependent variables are 'JS(job satisfaction)', 'OC(organizational commitment)'. The results are as follows. 'PB staffing' shows positive impact on both 'JS' and 'OC', but 'PB compensation' doesn't show positive impact on both 'JS' and 'OC'. Also, it is found that both 'PB appraisal' and 'PB compensation' do not show positive impact on 'OC'. 'PB appraisal' shows positive impact on 'JS', though. From the empirical analysis, the positive effects of the PB-HRM on the organizational effectiveness are partially verified. It is thought that these mixed results are originated from the particular situation in which the surveyed corporation is placed. After the merger of the corporation in 2009, even though it has introduced various PB-HRM systems, it has been through a harsh time such as wage freeze and return as measures of business normalization. This organizational situation may influence the verification of effectiveness of the normal PB-HRM.

Purification and Characterization of an Invertase Produced with Saccharomyces cerevisiae JS59 Isolated from Home-made Wine (포도주에서 분리한 Saccharomyces cerevisiae JS59가 생성하는 Invertase의 정제 및 특성)

  • The microorganism producing an invertase (E.C. 3.2.1.26) was isolated from wine and tentatively identified as Saccharomyces cerevisiae by cellular fatty acid analysis. The invertase was purified to homogeneity by ammonium sulfate precipitant, dialysis, ion-exchange chromatography on DEAE-Sephadex A-50, and gel chromatography on Sephadex G-200 from the culture supernatant of Saccharomyces cerevisiae JS59. The specific activity and the purification fold of the purified invertase were 7620.9 unit/mg protein and 13.9, respectively. The molecular weight of the purified invertase was estimated to be 38.5 kDa by SDS-PAGE. The optimum pH and temperature for the invertase activity were pH 5 and $55^{\circ}C$, respectively. The invertase activity was relatively stable at pH 4~6 and temperature $55^{\circ}C$. The activity of invertase was inhibited by $Ag^{2+}$ and $Hg^{2+}$, but on the contrary, activated by $Co^{2+}$ and $Mn^{2+}$. Michaelis constant ($K_m$) for invertase reaction in sucrose solution was 11.5 mM. TLC analysis of the products produced in sucrose solution during invertase reaction showed the progressive presence of glucose and fructose in accordance with sucrose hydrolysis.

Radon-222 Concentrations of Metropolitan Subway Stations and Soils in the Seoul (서울 지하철역(地下鐵驛) 승강장(昇降場) 및 토양내(土壤內) 유해(有害)라돈함량(含量) 연구(硏究))

  • The radon (Rn-222) potential of metropolitan subway stations and soils in Seoul city were delineated using alpha-track filter and EDA-200 radon detectors, respectively. The uranium (U) and thorium (Th) contents were also determined using a Multi Channel Analyzer to identify the sources of radon gas. The average U concentrations in Seoul varies according to basement rock types. For example, there is $9.40{\pm}10.11ppm$ in the Precambrian metasedimentary rock (PM), $9.08{\pm}2.85ppm$ in the Jurassic Kwanaksan granite (JK) and $4.94{\pm}1.43ppm$ in the Jurassic Seoul granite (JS). Uranium contents in soil samples are $10.30{\pm}4.74ppm$ in JK, $10.10{\pm}7.43ppm$ in PM and $6.69{\pm}3.95ppm$ in JS and these closely reflect the content of uraniferous minerals. The levels of soil radon are $604{\pm}273pCi/L$ in JK, $502{\pm}275$ in JS and $262{\pm}211pCi/L$ in PM. The soil radon concentrations are shown to reflect soil permeability and porosity rather than their U contents. The mean indoor radon contents in subway stations are $1.50{\pm}0.62pCi/L$ on the 4th line, $1.41{\pm}0.95pCi/L$ on the 3rd line, $0.84{\pm}0.13pCi/L$ on the 1st line and $0.80{\pm}0.25pCi/L$ on the 2nd line. The subway stations located in the JK have the highest average radon concentration with $2.04{\pm}0.65pCi/L$, where levels of $1.57{\pm}0.81pCi/L$ occur in the JS and $0.80{\pm}0.23pCi/L$ in the PM. The highest radon levels of 4.1 pCi/L occur mainly in Keongbokkung station on the 3rd line and these exceed 4 pCi/L of the US EPA action level.

Pathogenicity of Five Strains of Toxoplasma gondii from Different Animals to Chickens

  • Toxoplasma gondii is a protozoan parasite with a broad range of intermediate hosts. Chickens as important food-producing animals can also serve as intermediate hosts. To date, experimental studies on the pathogenicity of T. gondii in broiler chickens were rarely reported. The objective of the present study was to compare the pathogenicity of 5 different T. gondii strains (RH, CN, JS, CAT2, and CAT3) from various host species origin in 10-day-old chickens. Each group of chickens was infected intraperitoneally with $5{\times}10^8$, $1{\times}10^8$, $1{\times}10^7$, and $1{\times}10^6$ tachyzoites of the 5 strains, respectively. The negative control group was mockly inoculated with PBS alone. After infection, clinical symptoms and rectal temperatures of all the chickens were checked daily. Dead chickens during acute phage of the infection were checked for T. gondii tachyzoites by microscope, while living cases were checked for T. gondii infection at day 53 post-inoculation (PI) by PCR method. Histopathological sections were used to observe the pathological changes in the dead chickens and the living animals at day 53 PI. No significant differences were found in survival periods, histopathological findings, and clinical symptoms among the chickens infected with the RH, CN, CAT2, and CAT3 strains. Histopathological findings and clinical symptoms of the JS (chicken origin) group were similar to the others. However, average survival times of infected chickens of the JS group inoculated with $5{\times}10^8$ and $1{\times}10^8$ tachyzoites were 30.0 and 188.4 hr, respectively, significantly shorter than those of the other 4 mammalian isolates. Chickens exposed to $10^8$ of T. gondii tachyzoites and higher showed acute signs of toxoplasmosis, and the lesions were relatively more severe than those exposed to lower doses. The results indicated that the pathogenicity of JS strain was comparatively stronger to the chicken, and the pathogenicity was dose-dependent.

Growth Inhibition of Toxic Cyanobacterium Microcystis aeruginosa by Various SNPs (Silver Nanoparticles) (여러 가지 은나노 물질의 유해 남조 Microcystis aeruginosa 생장억제)

  • The effect of various SNPs (silver nanoparticles) on the growth of Microcystis aeruginosa was investigated in laboratory and field experiment. Four SNPs, namely JS47N, JS47N-K2, JS47N/3-1 and JS47N/3-2 were used to this study. The Ag size, concentration and color of these solutions were about $20{\sim}40nm$, $200mg\;L^-1$ and brown, respectively. At 0.01 and $0.1mg\;L^-1$, SNPs inhibited the growth of unicellular M. aeruginosa by 99.4% and 99.9%, respectively. However, SNPs of $1mg\;L^-1$ inhibited the growth of colonial M. aeruginosa by 98.5%, whereas the other three concentrations (0.001, 0.01 and $0.1mg\;L^-1$) had little inhibitory effect. In experimental enclosures from eutrophic lake, cyanobacteria including M. aeruginosa were found to be more sensitive to the SNPs than green algae and diatoms. In conclusion, our study indicates that SNPs has a selective cyanocidal potential when used to M. aeruginosa. We believe that future studies need to test on various other organisms, and determine minimum concentration for field application.

A study on the selection of the optimal smoke control mode in train platform through quantitative risk assessment (정량적 위험도 평가를 통한 열차 승강장 화재시 최적 제연모드 선정에 관한 연구)

  • In the case of train stations, due to the specificity of underground spaces with limited smoke emissions, if appropriate removal equipment is not equipped, the damage caused by fire smoke may increase in the event of a fire. As a result, the need for measures to ensure the safety of evacuation of underground stations has been highlighted, and research for safe evacuation of platform users in case of fire is continuously being conducted at home and abroad. However, although the smoke removal area is currently divided by smoke boundary walls and platform screen doors (PSD) and installed in the train platform, standards for smoke removal methods (air supply or exhaust) for each fire removal area, that is, smoke removal mode, are not presented. In this study, fire analysis and evacuation analysis were performed to estimate the number of deaths and to derive F/N guidance in order to quantitatively evaluate the fire risk according to the fire station fire, and the total risk was the lowest in the case of fire area exhaust and supply to adjacent areas.
