Degradation Kinetics of Carbohydrate Fractions of Ruminant Feeds Using Automated Gas Production Technique

  • The current ruminant feeding models require parameterization of the digestion kinetics of carbohydrate fractions in feed ingredients to estimate the supply of nutrients from a ration. Using an automated gas production technique, statistically welldefined digestion rate of carbohydrate, including soluble carbohydrate, can be estimated in a relatively easy way. In this study, the gas production during in vitro fermentation was measured and recorded by an automated gas production system to investigate degradation kinetics of carbohydrate fractions of a wide range of ruminant feeds: corn silage, rice straw, corn, soybean hull, soybean meal, and cell mass from lysine production (CMLP). The gas production from un-fractionated, ethanol insoluble residue and neutral detergent insoluble residue of the feed samples were obtained. The gas profiles of carbohydrate fractions on the basis of the carbohydrate scheme of the Cornell Net Carbohydrate and Protein System (A, B1, B2, B3 and C) were generated using a subtraction approach. After the gas profiles were plotted with time, a curve was fitted with a single-pool exponential equation with a discrete lag to obtain kinetic parameters that can be used as inputs for modern nutritional models. The fractional degradation rate constants (Kd) of corn silage were 11.6, 25.7, 14.8 and 0.8%/h for un-fractioned, A, B1 and B2 fractions, respectively. The values were statistically well estimated, assessed by high t-value (>12.9). The Kd of carbohydrate fractions in rice straw were 4.8, 21.1, 5.7 and 0.5%/h for un-fractioned, A, B1 and B2 fractions, respectively. Although the Kd of B2 fraction was poorly defined with a t-value of 4.4, the Kd of the other fractions showed tvalues higher than 21.9. The un-fractioned corn showed the highest Kd (18.2%/h) among the feeds tested, and the Kd of A plus B1 fraction was 18.7%/h. Soybean hull had a Kd of 6.0, 29.0, 3.8 and 13.8%/h for un-fractioned, A, B1 and B2, respectively. The large Kd of fraction B2 indicated that NDF in soybean hull was easily degradable. The t-values were higher than 20 except for the B1 fraction (5.7). The estimated Kd of soybean meal was 9.6, 24.3, 5.0%/h for un-fractioned, A and B1 fractions, respectively. A small amount of gas (5.6 ml at 48 ho of incubation) was produced from fermentation of CMLP which contained little carbohydrate. In summary, the automated gas production system was satisfactory for the estimation of well defined (t-value >12) kinetic parameters and Kd of soluble carbohydrate fractions of various feedstuffs that supply mainly carbohydrate. The subtraction approach, however, should be applied with caution for some concentrates, especially those which contain a high level of crude protein since nitrogen-containing compounds can interfere with gas production.

$TiO_2$의 광촉매 효율성에 관한 연구 (Study on the Photocatalytic Efficiencies of $TiO_2$)

  • 광촉매 특성을 지닌 $TiO_2$ 피막을 인가 전압 180 V에서 양극산화법을 이용하여 제조하고, 산화조건에 따른 구조적 차이에 대하여 조사하였다. 황산 및 황산+과산화수소 용액의 경우 $TiO_2$의 구조가 rutile과 anatase형이 혼합된 형태를 지녔으나, 황산+인산 및 황산+인산+과산화수소 혼합용액의 경우에는 대부분 anatase형의 $TiO_2$가 제조되었음을 알 수 있었다. 양극산화법에 의해 제조된 $TiO_2$는 모두 광촉매 특성율을 나타냈으며, 아닐린 블루 분해 반응의 경우 모든 산화 조건에서 반응차수가 1로 나타났으며 속도상수값이 거의 유사함을 알 수 있었다.

Changes in Serum Metabolites and Growth Characteristics of Korean Native Steers Fed Alcohol-fermented Feeds

  • This study was carried out to assess whether feeding of alcohol-fermented feeds (AFF) affects the nutritional metabolism and growth characteristics of Korean native steers. Ten steers were randomly assigned to one of two treatment groups. The dietary treatments were AFF (50% commercial beef cattle feed+30% alcohol-fermented soybean curd dregs+20% rice straw) and control (80% commercial beef cattle feed+20% rice straw). The change of serum metabolites and growth characteristics were measured every two months during the whole twelve months experimental period and the relationships between serum metabolites and growth characteristics were simultaneously analyzed. Four hours after feeding AFF, serum alcohol concentration reached its peak with a significantly higher value than that after control feeding (11.9 and 4.9 mg/dl, respectively). Serum glucose and inorganic phosphorus (IP) concentrations (63.1 and 8.4 mg/dl, respectively) of steers fed AFF were higher than those (56.6 and 7.0 mg/dl) fed the control diet. In both treatments, the serum glucose concentration rapidly increased when body weight (BW) of the steer reached about 600kg, while IP concentrations were rapidly diminished at that BW. Lower concentrations of both blood urea nitrogen (BUN) and cholesterol were observed in steers fed AFF up to 450 kg of BW. The IP concentration was correlated with concentrations of BUN, cholesterol and glucose in AFF fed cattle but not in the cattle fed control diets. Average daily gain was higher in steers fed AFF than steers fed control, particularly during the growing stage of cattle. These findings indicated a capability of AFF to improve BW gain of Korean native steers by decreased protein degradation as well as increased fat synthesis.

Band alignment and optical properties of $(ZrO_2)_{0.66}(HfO_2)_{0.34}$ gate dielectrics thin films on p-Si (100)

  • $(ZrO_2)_{0.66}(HfO_2)_{0.34}$ thin films as gate dielectrics have been proposed to overcome the problems of tunneling current and degradation mobility inachieving a thin equivalent oxide thickness. An extremely thin $SiO_2$ layer is used in order to separate the carrier in MOSFET channel from the dielectric field fluctuation caused by phonons in the dielectric which decreases the carrier mobility. The electronic and optical properties influenced the device performance to a great extent. $(ZrO_2)_{0.66}(HfO_2)_{0.34}$ dielectric films on p-Si (100) were grown by atomic layer deposition method, for which the conduction band offsets, valence band offsets and band gapswere obtained by using X-ray photoelectron spectroscopy and reflection electron energy loss spectroscopy. The band gap, valence and conduction band offset values for $(ZrO_2)_{0.66}(HfO_2)_{0.34}$ dielectric thin film, grown on Si substrate were about 5.34, 2.35 and 1.87 eV respectively. This band alignment was similar to that of $ZrO_2$. In addition, The dielectric function (k, $\omega$), index of refraction n and the extinction coefficient k for the $(ZrO_2)_{0.66}(HfO_2)_{0.34}$ thin films were obtained from a quantitative analysis of REELS data by comparison to detailed dielectric response model calculations using the QUEELS-$\varepsilon$(k, $\omega$)-REELS software package. These optical properties are similar with $ZrO_2$ dielectric thin films.

디인터레이싱을 위한 C4.5 분류화 기법의 적용 및 구현 (The Adopting C4.5 classification and it's Application for Deinterlacing)

  • 디인터레이싱이란 두 개의 필드(짝수 필드 및 홀수 필드)로 구성된 인터레이스 영상을 프로그레시브 영상으로 변환하는 기술이다. 이는 크게 공간영역에서의 디인터레이싱과 시간영역에서의 디인터레이싱 기술로 나뉠 수 있다. 공간영역에서의 기법은 하나의 독립적인 필드만을 사용하여 디인터레이싱을 수행하는 것으로 하드웨어의 구성은 용이하지만 디인터레이싱 대상 화소의 정보가 해당 필드에 존재하지 않는 경우 화질 열화가 발생할 수 있다. 반면 시간영역에서의 기법은 메모리 사용량이 높고 하드웨어의 구성이 어렵지만 보다 높은 객관적 화질을 얻을 수 있다. 하지만 움직임 추정이 잘못된 경우 현저한 화질열화가 발생한다. 제안하는 방법은 공간영역에서의 디인터레이싱 기법으로 대상화소 주변의 통계적 특성에 따라 서로 다른 기법을 사용하여 디인터레이싱을 수행한다. 이 과정에서 최적의 디인터레이싱 방법의 선택을 위해 엔트로피 기반의 대표적인 분류 알고리즘인 C4.5 알고리즘을 적용한다. 실험결과 제안하는 알고리즘은 이전의 여러 방법들과 비교하여 높은 객관적 화질을 가지는 것을 볼 수 있었으며, 주관적 화질 또한 상대적으로 유사하거나 높은 것을 볼 수 있었다.

Fabrication of Cu2ZnSnS4 Films by Rapid Thermal Annealing of Cu/ZnSn/Cu Precursor Layer and Their Application to Solar Cells

  • $Cu_2ZnSnS_4$ thin film have been fabricated by rapid thermal annealing of dc-sputtered metal precursor with Cu/ZnSn/Cu stack in sulfur ambient. A CZTS film with a good uniformity was formed at $560^{\circ}C$ in 6 min. $Cu_2SnS_3$ and $Cu_3SnS_4$ secondary phases were present at $540^{\circ}C$ and a trace amount of $Cu_2SnS_3$ secondary phase was present at $560^{\circ}C$. Single-phase large-grained CZTS film with rough surface was formed at $560^{\circ}C$. Solar cell with best efficiency of 4.7% ($V_{oc}=632mV$, $j_{sc}=15.8mA/cm^2$, FF = 47.13%) for an area of $0.44cm^2$ was obtained for the CZTS absorber grown at $560^{\circ}C$ for 6 min. The existence of second phase at lower-temperature annealing and rough surface at higher-temperature annealing caused the degradation of cell performance. Also poor back contact by void formation deteriorated cell performance. The fill factor was below 0.5; it should be increased by minimizing voids at the CZTS/Mo interface. Our results suggest that CZTS absorbers can be grown by rapid thermal annealing of metallic precursors in sulfur ambient for short process times ranging in minutes.

Protein Evaluation of Dry Roasted Whole Faba Bean (Vicia faba) and Lupin Seeds (Lupinus albus) by the New Dutch Protein Evaluation System: the DVE/OEB System

  • The effects of dry roasting (110, 130, $150^{\circ}C$ for 15, 30, 45 min) on potential ruminant protein nutritional values in terms of: a), rumen bypass protein (BCP); b), rumen bypass starch (BST); c), fermented organic matter (FOM); d), true absorbed bypass protein (ABCP); e) microbial protein synthesized in the rumen based on available energy (E_MP); f), microbial protein synthesized in the rumen based on available nitrogen (N_MP); g), true protein supplied to the small intestine (TPSI); h), true absorbed rumen synthesized microbial protein (AMP); i), endogenous protein losses (ENDP); j), true digested protein in the small intestine (DVE); k), degraded protein balance (OEB) of whole lupin seeds (WLS) and faba beans (WFB) were evaluated by the new Dutch DV/OEB protein evaluation system. Dry roasting significantly increased BCP, BST, TPSI, ABCP, DVE (p<0.001) and decreased FOM, E_MP, AMP, N_MP and OEB (p<0.001) with increasing temperatures and times except that when temperature was at $110^{\circ}C$. The values of BCP, BST, TPSI, ABCP and DVE at $150^{\circ}C/45min$ for WLS and WFB were increased 2.2, 3.7; -, 2.0; 1.7, 1.7; 2.3, 3.7 and 1.7, 1.7 times and the values of FOM, E_MP, AMP, N_MP and OEB at $150^{\circ}C/45min$ for WLS and WFB were decreased by 15.3, 25.8; 18.1, 25.8; 18.7, 25.8; 54.6, 41.6 and 82.3% 54.7%, respectively, over the raw WLS and WFB. The results indicated that though dry roasting reduced microbial protein synthesis due to reducing FOM, TPSI didn't decrease but highly increased due to increasing BCP more than enough for compensation of the microbial protein decreasing. Therefore the net absorbable DVE in the small intestine was highly increased. The OEB values were significantly reduced for both WLS and WFB but not to the level of negative. It indicated that microbial protein synthesis might not be impaired due to the sufficient N supplied in the rumen, but the high positive OEB values in the most treatments except of $150^{\circ}C$ for 30 and 45 min of WLS (The OEB values: 54.8 and 26.0 g/kg DM) indicated that there were the large amounts of N loss in the rumen. It was concluded that dry roasting at high temperature was effective in shifting protein degradation from rumen to intestines and it increased the DVE values without reaching the negative OEB values. No optimal treatment was found in WLS due to the too high OEB values in all treatments. But dry roasting at $150^{\circ}C$ for 30 and 45 min might be optimal treatments for WLS due to the very lower OEB values.

패스틴®첨가가 단백질 분해율과 반추위 발효 및 영양소 소화율에 미치는 영향 (Effects of Passtein® Supplements on Protein Degradability, Ruminal Fermentation and Nutrient Digestibility)

  • 본 시험은 패스틴$^{(R)}$ 을 첨가하였을 때, in vitro 상에서 단백질 fraction과 분해율에 미치는 영향과, in vivo 상에서 반추위 성상, 미생물 군집, 암모니아태 질소 농도 및 영양소 소화율에 미치는 영향을 구명하고자 실시하였다. In vitro 실험에서는 1mm로 분쇄된 대두박을 기질로 하여 패스틴$^{(R)}$ ((주)은진인터내셔날)을 첨가하여 borate-phosphate buffer와 중성세제에서의 조단백질 분해율을 측정하였으며, exogenous enzyme (Streptomyces griseus 유래 protease)를 이용하여 39$^{\circ}C$에서 0, 2, 4, 8, 12, 48 시간동안 배양 후 조단백질 분해율을 측정하였다. 반추위 발효성상과 영양소 소화율은 반추위 fistula가 부착된 평균체중 300kg의 홀스타인 수소 4두를 이용하여 무첨가구, 패스틴$^{(R)}$ 첨가구의 두개 처리구에 2마리씩 4마리를 배치하여 측정하였다. Buffer Soluble Protein fraction은 패스틴$^{(R)}$ 첨가 수준별로 차이가 없었으나, 무첨가구에 비해 패스틴$^{(R)}$ 첨가구에서 감소하는 경향을 보였다. 단백질 분해율은 배양 0 시간대에서 4시간대까지는 처리구간 유의성이 없었지만, 12 h과 48 h에서는 패스틴$^{(R)}$ 첨가로 시험구에서 감소되었다. 용해 단백질 분해율 ‘a’는 패스틴$^{(R)}$ 시험구에서 경미하게 높은 수치를 나타내었지만, 소화 가능한 단백질 분해율 ‘a+b’는 패스틴$^{(R)}$ 시험구에서 낮은 경향을 보였다. 패스틴$^{(R)}$ 첨가로 pH와 $NH_3$- N 농도는 증가하는 경향이었으며 휘발성지방산, 미생물 수 및 enzyme activity는 감소하였고 영양소 소화율은 높았으나 유의적인 차이는 없었다.

  • 한국산 고려 홍삼을 산 또는 알칼리로 가수분해하여, 여러가지 사포게닌과 프로사포게닌을 제조하였으며, 분광학적 데이터와 물리 데이터 등으로부터 이들의 화학 구조를 결정하였다. 이들 중 몇종의 분해산물은 A549, SK - OV. - 3, P388, L1210, SK - Mel - 2 및 K562 등의 암세포에 대하여 세포 독성을 나타내었다. Diol계와 triol계 모두 20번 탄소의 절대구조만이 다른 입체 이성체간의 세포독성의 차이는 인정되지 않았으며, diol 계의 물질들이 triol계 물질보다는 더 높은 활성을 나타내었다. 일반적으로 결합된 탄소의 수가 적을수록 세포독성은 강하여지는 경향??? 보였다.

토양 중 살균제 Pencycuron의 잔류 특성 (Persistence of Fungicide Pencycuron in Soils)

  • 토양 중 pencycuron의 온도, 수분함량 및 토성에 따른 흡착, 잔류 특성을 구명하고자 실내 및 포장실험과 환경의 영향에 대하여 실험을 수행하였다. 2종 토양에서 진탕 시간과 약제의 흡착량사이에 높은 유의성이 있는 power function의 상관관계가 인정되었다. 흡착속도는 진탕 1시간 이내에 사질식양토에서 최대 흡착량의 60%가, 미사질식양토에서 65%가 흡착되었고, 12시간 후에는 의사평형에 도달하였다. Pencycuron의 농도별 흡착은 Freundlich 등온식에 부합되었으며, 흡착분배계수 $K_d$값은 사질식양토에서 2.31, 미사질식양토에서 2.92었다. 토양 중 유기탄소에 대한 분배계수 $K_{oc}$는 사질식양토에서 292.9이었고, 미사질식양토에서 200.5이었다. 흡착강도 및 비선형도를 성명하는 상수값은 사질식양토에서 1.45, 미사질식양토에서 1.68이었다. 실내 실험에 있어서 pencycuron의 잔류는 1차 반응식에 부합되었고, 반감기는 $12^{\circ}C{\sim}28^{\circ}C$에서 95일${\sim}$20일로, 토양 수분함량이 포장용수량의 $30{\sim}70%$인 토양에서 38일에서 21일로 짧아졌다. 토양 종류에 따른 pencycuron의 반감기는 토성이 현저히 달랐음에도 사질식양토에서 25일, 미사질식양토에서 22일로 나타났다. 포장 실험에서도 pencycuron의 반감기는 사질식양토에서 26일, 미사질식양토에서 23일이었다. 포장에서 10%까지 잔류되는 기간은 미사질식양토에서 57일, 사질식양토에서 69일로 나타났다.
