Isolation of Angiotensin Converting Enzyme Inhibitors from Compositae Plants

  • Thirty plants of the Compositae family were screened for inhibitory activity of angiotensin converting enzyme (ACE). Among them, Chrysanthemum boreale Makino and Ixeris dentate Nakai were selected for further investigation since they had the highest inhibitory activity. Crude water extracts of the flowers of Chrysanthemum boreale Makino and the roots of Ixeris dentate Nakai were prepared by heating at 95$^{\circ}C$ and 6$0^{\circ}C$ for 2 hr, respectively, followed by centrifugation at 8000$\times$ g far 30 min. Crude extracts were then filtered using YM-3 and YM-1 membranes. The ACE inhibitors were isolated using consecutive chromatographic methods including: Sephadex G-15, ion exchange, FPLC, and reverse phase HPLC. The inhibitors were identified to have molecular masses of 204 and 283 daltons, respectively, by mass spectrometry. These results demonstrate that crude extracts of Compositae plants may be useful as functional food ingredients with anti-hypertensive properties.

Phytotoxic Effect, DPPH Radical Scavenging Activity and Chlorogenic Acid Level of Methanol Extracts from Aerial Parts of Several Korean Salad Plants

  • A series of aqueous or methanol extracts from four different Korean salad plants were assayed to determine their allelopathy and antioxidant activity. The extracts applied on filter paper in a Petri-dish bioassay significantly inhibited root growth of against alfalfa (Medicago sativa) seedlings. Leaf extracts from 40 g dry tissue $L^{-1}$ of Aster yomena was most phytotoxic to alfalfa root growth, and followed by that of Cirsium japonicum, Taraxacum officinale, and Ixeris dentate. Methanol extracts of plants dose-dependently increased DPPH free radical scavenging activity in vitro. Antioxidant activity of methanol extracts from the same plant species was investigated, and the result showed high DPPH free radical scavenging activity in Cirsium japonicum, Aster yomena, and Ixeris dentate, however, in Taraxacum officinale was least activity. By means of HPLC analysis, chlorogenic acid, p-coumaric acid, and total phenolics with 7.68, 17.47 and 18.64 mg, $100g^{-1}$, respectively, showed the highest amounts in methanol extracts from Cirsium japonicum leaves. These results suggest that Compositae salad plants contain water-soluble substances with allelopathic potential as well as antioxidant activity.

  • We used the Braun-Blanquet method to study coastal dune vegetation of South Korea. Coastal vegetation was monitored in thirty sites from April 2004 to September 2005. Seventeen plant communities were classified into five groups as follows: A. Two associations of herbaceous sand dune plants in Salsoletea komarovii Ohba, Miyawaki et Tx. 1973 (Calystegia soldanella community, Lathyrus japonicus-Calystegia soldanella community), B. Twelve associations of herbaceous perennial sand dune plants in Glehnietea littoralis Ohba, Miyawaki et Tx. 1973 (Carex pumila community, Elymus mollis community, Vitex rotundifolia-Elymus mollis community, Ixeris repens community, Elymus mollis-Ixeris repens community, Lathyrus japonicus community, Messershmidia sibirica community, Glehnia littoralis community, Carex kobomugi community, Calystegia solda-nella-Carex kobomugi community, Ishaemum anthephoroides community, Zoysia macrostachya community), C. Antioxidant and anti-diabetic effects of Ixeris strigosa extract (선씀바귀 추출물의 항산화 및 항당뇨 효과)

  • Purpose: Ixeris strigosa (IS) is a perennial plant that commonly grows in meadows. The leaves and roots of IS have been used in medicine as a sedative. This study evaluated the antioxidant and carbohydrate-digestive-enzyme inhibitory effects of IS to determine its potential as an essential antioxidant and glycemic inhibitor for type 2 diabetics. Methods: The antioxidative and α-amylase and α-glucosidase inhibitory activities were examined using the water extracts (ISW), ethanol extracts (ISE), and solvent fractions from IS. The antioxidative activities were measured using in vitro methods by measuring the 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity. Results: Investigations of the total polyphenol, flavonoid content, in vitro antioxidant activity, and α-amylase and α-glucosidase inhibitory activities of the IS extract showed that the ISE had higher total phenolic and flavonoid contents than the ISW, as well as high antioxidant activity. The ethanolic extracts of IS (70%) had an α-amylase inhibitory activity of 78.55%. The ethyl acetate fraction (90.56%) showed higher α-glucosidase inhibitory activity than the positive control, acarbose (83.01%). Conclusion: Among the ISE fractions, the ethyl acetate and butanol fractions showed the best digestive enzyme inhibitory activity. Moreover, the antioxidant activity of the extract and the carbohydrate, α-amylase, and α-glucosidase inhibitory effects showed a stronger correlation with the total phenol and flavonoid contents compared to the ISW. As a result, the antioxidant and digestive enzyme inhibitory activities of high ISE are due to the phenolic compounds, particularly the flavonoid compounds. Therefore, ethyl acetate and butanol fractions of the 70% ethanol extract are excellent anti-diabetic functional materials.

Anti-Inflammatory Effect of Ixeris dentata on Ultraviolet B-Induced HaCaT Keratinocytes

  • Human skin is the first line of defense for the protection of the internal organs of the body from different stimuli. Ultraviolet B (UVB) irradiation induces skin damage and inflammation through the secretion of various cytokines, which are immune regulators produced by cells. To prevent the initiation of skin inflammation, keratinocytes that have been irreversibly damaged by radiation must be removed through the apoptotic mechanism. Ixeris dentata (family: Asteraceae) is a perennial medicinal herb indigenous to Korea. It has been used in Korea, China, and Japan to treat in digestion, pneumonia, diabetes, hepatitis, and tumors. To gain insight into the anti-inflammatory effects of I. dentata, we examined its influence on UVB-induced pro-inflammatory cytokine production in human keratinocytes (HaCaT cells), by observing cells that were stimulated with UVB in the presence or absence of I. dentata. In the present study, pro-inflammatory cytokine production was determined by performing enzyme-linked immunosorbent assay, reverse transcription polymerase chain reaction, and western blot analysis to measure the activation of mitogen-activated protein kinase (MAPKs). I. dentata inhibited UVBinduced production of the pro-inflammatory cytokine interleukin (IL)-6 in a dose-dependent manner. Further, I. dentata inhibited the UVB-induced expression of cyclooxygenase (COX)-2. Furthermore, I. dentata inhibited the phosphorylation of c-Jun NH2-terminal kinase and p38 MAPKs, suggesting that it inhibits the secretion of the pro-inflammatory cytokines IL-6 and IL-8, and COX-2 expression, by blocking MAPK phosphorylation. These results suggest that I. dentate can potentially protect against UVB-induced skin inflammation.

Studies on the Germination Characteristics in the Several Weeds of Compositae (국화과(菊花科) 잡초(雜草)의 발아특성(發芽特性)에 관(關)한 연구(硏究))

  • This experiment was carried to investigate the germination pattern in relation to temperatures and lights, and the emergence pattern in relation to seeding depths, lights and the alpha amylase activity of Youngia sonchifolia, Lactuca indica var. laciniata, Ixeris dentata var. albiflora and Ixeris polycephala. In Y. sonchifolia, the optimum germination temperature was $25^{\circ}C$, the optimum seeding depth to emerge was 0 mm and it could emerge in 0-5mm. In L. indica var. laciniata under cool storage, the optimum germination temperatures were $19^{\circ}C-28^{\circ}C$, the optimum seeding depth was 5mm and it could emerge in 0-20mm. In L. indica var. laciniata under room storage, the optimum germination temperature was $25^{\circ}C$ the optimum seeding depth was 5mm and it could emerge in 0-10mm. In I. dentata emerge was and 0mm and it could emerge in 0-5mm. In I. polycephla, the optimum temperatures were $16^{\circ}C-19^{\circ}C$, the optimum seeding depth to emerge was 0mm and it could emerge in 0-5mm. The alpha amylase activity was lower Y. sonchifolia, L. indica var. laciniata and I. dentata var. abiflora than barley cultivar Dongbor#1. And the increased pattern of alpha amylase activity was likely to it of germination rate.

Organic Constituents in Kimchis (Ixeris sonchifolia H.) -On free amino acids- (고들빼기 김치의 유기성분(有機成分)(I) - 유이(遊離)아미노산(酸)에 관(關)하여 -)

  • The change of free amino acids was identified during fermentation of kimchis (Ixeris sonchifoliaH.) added fermented anchovy. The results were summarized as follows; In fresh roots and leaves of Ixeris sonchifolia H., 8 kinds of free amino were determined respectively. Among them, argine, cysteine and glutamic acid were abundant in fresh roots, while arginine, valine, isoleucine and phenylalanine in fresh leaves, especially arginine was dominant in fresh both roots and leaves. The amount of total amino acids in fresh leaves was about 2.5 times of that of roots. After fermentation, 15 kinds of free amino acids were determined in kimchis, and the characteristic favor of in was attributed to such amino acids as threonine, glutamic acid, alanine, leucine and cysteine. The content of total free amino acids in kimchi leaves was increased to about 5 times of that in fresh (9,435,6mg % on dry base), but in kimchi roots, 11 times of that in fresh was contained (7,079,1mg % on dry base) In kimchi'es extract, 16 kinds of free amino acids were determined, and threonine, glutamic acid, alanine, cysteine and leucine were abundant.

Dietary Fiber in Godulbaegi(Korean Lettuce, Ixeris sonchifolia H.) Kimchi (고들빼기김치 식이섬유질의 식품학적인 특성)

  • The effect of processing conditions on the changes in the contents of dietary fiber fractions and its physical properteis of Godulbaegi(Korean lettuce, Ixeris sonchifolia H.) was determined during preparation and fementation for kimchi. Water holding capacity(WHC) and oil adsorption capacity(OAC) were also checked on the subject of freeze dried powder from different stages of the kimchi processing. Neutral detergent fiber(NDF) content in young samples(leaf and root) decreased with prolonged soaking and fermentation period. Every young samples had a higher level in NDF than in ripe samples. Noticeable decrease in acid detergent fiber(ADF) without a change in ripe roots was showed after fermentation($4^{\circ}C$, 60 days). The water holding capacity of freeze dried young plants ranged from 5.78ml/g for roots to 6.31ml/g for leaves. Soaking and fermentation resulted in decreasing WHC and about 50% of WHC(raw leaves) was lowered after kimchi fermentation($4^{\circ}C$, 40 days). OAC of all samples were lower than WHC in same samples significantly and those were also decreased after soaking and fermentations as WHC.
