Uptake of Isolated Lyophyllum ulmarium Chromosomes by Ganoderma applanatum Protoplasts (잔나비걸상버섯 원형질체(原形質體)에 의(依)한 만가닥버섯 염색체(染色體)의 섭입(攝入))

  • The uptake of isolated Lyophyllum ulmarium chromosomes by Ganoderma applanatum protoplasts was induced with polyethylen $glycol+CaCl_2+glycine$. Uptake products of chromosomes by protoplasts showed micro-and macrotransgenome type. The former was slowly growing and unstable, the latter was outgrowing and stable mycelial colonies which made thick hyphae of width and segregation of mycelial colony on GCM containing benomyl. A comparison of macrotransgenome type was using isozyme analysis of esterase. The enzyme pattern of two transformants was distinct in position and quantity compared with parents.

Effect of Plant Hormones on the Invertase Activity in the Senescing Leaves of Phaseolus radiatus

  • Effect of plant hormones on the leaf senescence of mung bean (Phaseolus radiatus) was investigated by measuring the changes of reducing sugar contents and invertase isozyme activities in detached leaves treated with NAA, $GA_3$ or BA. During dark-induced senescence, reducing sugar contents in the detached leaves increased temporarily at 4 d, thereafter decreased rapidly and reached minimum values within 7-14 d. The pattern of soluble acid invertase activity in the senescing leaves kept in the dark was similar to that of reducing sugar accumulation, whereas the activities of alkaline and extracellular invertases were not significantly changed during leaf senescence. Therefore, these results suggest that soluble acid invertase, but not alkaline and extracellular invertases, induces the accumulation of reducing sugar during leaf senescence of mung bean plants. Exogenous NAA application had little or no effect in the increase of soluble acid invertase activity during dark-induced senescence compared to the control. However, exogenous applications of $GA_3$ and BA led to the increase of soluble acid invertase activity in the senescing leaves. Particularly, BA application was very effective in enhancing the activity of soluble acid invertase as well as in delaying chlorophyll breakdown during dark-induced senescence. These results suggest, therefore, that BA regulates the activity of soluble acid invertase, which leads to the accumulation of reducing sugar, and the stability of photosynthetic apparatus to delay leaf senescence.

  • The present researches were carried out to investigate the peroxidase activity in association with the reactions of the 4 cultivars of rice plant, Nagdong, Jinheung, Nongbaek and Taebaek to Pyricularia oryzae race KJ-I0l and KJ-301. Although the peroxidase activity was increased during the growth of the rice seedlings, the significant difference in the activity was not found among 4 cultivars. After inoculation of the fungus, the peroxidase activity was enhanced in diseased leaves, being considerably higher in the compatible than in the incompatible cultivars. The isozyme bands of peroxidases observed in mycelium of rice blast fungus were not found in the diseased leaves on the gel electrophoresis. The peroxidase activity was not affected by the increased application of nitrogenous fertilizer.

  • Horizontal starch gel electrophoresis for 29 populations (n=543) of the wrinkled frog, Rana rugosa, from Korea and Japan was peformed to assess the degree of genic variation and genetic diversity, and to understand the biogeographic pattern of distribution and speciation. A sum of 22 presumptive loci was screened from 17 enzymes and general proteins. Four loci, Aco, Est-3, Me-2, and Pgm, demonstrated high levels of polymorphism. The degree of average genetic variation of R. rugosa was P=22.7% (9.1-40.9%), Ho=0.086 (0.048-0.165) and He=0.090 (0.042-0.168). In the south-eastern region of the Korean peninsula (Chongsong, Yongchon, Ulsan, Kyongju, Pohang, yongdok and Ulchin), a few unique alleles in the Mpi locus were detected and their biogeographic implications were considered. The degree of genetic differentiation among the Korean populations was moderate (S=0.900), whereas the degree of genetic diversity between Korean and Japanese populations was notably high (S=0.687, D=0.293). This result corresponds with the data obtained by the mitochondrial cytochrome b gene sequence (Lee et al., 1999) suggesting that the Korean and Japanese R. rugosa might have evolved a specific level of genetic differentiation since their geographic isolation.

Phenoloxidases and Photomorphogenesis in Coprinus congregatus (Coprinus congregatus의 분화와 Phenoloxidase와의 관계)

  • The have been many reports that phenoloxidase are correlated with development in many fungi. C. congregatus, one of nushroom-forming basidiomycetes, which requires light for its development also has phenoloxidases. In C. congragatus, there are two sets of membrane-associated phenoloxidase (PHO I and PHO II) which are differentiated by their isozyme patterns, and each enzyme set consists of two different subtrate specific enzyme protein; o-tolidine reacting enzyme, and DOPA reacting enzyme. PHO I which is localized by a protoplast-concanavalin A technique by using a new solidifying agent, Pluronic Polyol F 127, instead of agar appears in the vegetative hyphae, and PHO II appears at the early primordial stage on agar and at the sclerotial stage of liquid shake cultures. Inhibition of PHO I with the enzyme inhibitors inhibits mushroom formation as well as melanization of the vegetative hyphae at concentrations which do not inhibit the vegetative growth. PHO I deficient mutants do not form mushrooms or melanins, and the mutants show abnormal nuclear migration patterns. PHO II has roles; possibly cementing the adjacent hyphae during the actual three dimensonal structure formation, and melanizing mushrooms and sclerotia. The possible roles of PHO I in the light reception complex and in melanin formation, the function of malanin, and possible roles of postulated post translational modifying enzymes which regulate the phenoloxidases, nuclear migration pattern, and self-nonself recognition mechanism are discussed.

Research Advances of Leptotrombidium scutellare in China

  • Leptotrombidium scutellare is one of the 6 main vectors of scrub typhus in China. It has been found in more than 15 provinces of China. Especially in Yunnan, it was found to be mainly distributed in some mountainous areas with high altitude, low temperature and low precipitation. Rodents and some other small mammals were the most common hosts of L. scutellare. To date, more than 40 host species of L. scutellare have been recorded with very low host specificity, and the main hosts varied in different geographical regions. L. scutellare had a strong resistance against the cold environment, and the temperature and humidity were 2 important factors affecting its growth and development. Among different individuals of their rodent hosts, L. scutellare mites often showed an aggregated distribution pattern, which reflected the interspecific cooperation of the mites. The chromosome karyotype of L. scutellare was 2n=16 and all the 8 pairs of chromosomes were short rod-shaped with metacentric or sub-metacentric types. The isozyme spectrum supported that L. scutellare, L. deliense and L. rubellum were in the same species group. Based on the natural infection, experimental transmission and epidemiological evidence, L. scutellare has been eventually confirmed as the second major vector of scrub typhus in China, which is second only to L. deliense.

Transfer of Isolated Nuclei from Pleurotus florida into Protoplasts of Pleurotus ostreatus (느타리버섯 원형질체내(原形質體內)에 사철느타리버섯 핵(核)의 전이(轉移))

  • The transfer of the isolated nuclei from P. florida into protoplasts of P. ostreatus was induced with polyethylene glycol and $CaCl_2$. Three types of transfer products of nuclei were obtained when transferred to MMM. Type 1 colonies were more vigrously growing mycelium and stable on MCM. One of the type 1 colonies, appeared segregation on MCM plus benomyl. The mycelium did not form clamp connection. These results suggest that type 1 colonies were nuclear hybrids or allodiploids. Type 2 was main products of nuclear transfer. The mycelium formed clamp connection and fertile on sawdust media. Type 3 was very slow growing or non-viable colonies after debris of nuclei or chromosomes transfer into recipient protoplasts. Isozyme pattern of esterase in type 1 produced a new band. Type 2 and type 3 could be characterized by parental bands.

Comparison of lsozymes and Growth Characteristics of Cultivars in Dioscorea batatas DECNE (재배마의 동위효소와 생육특성 비교)

  • This study was carried out to compare isozyme band pattern, growth characteristics and tuber yield of Dioscorea batatas to get basic information for varietal classification. It could be identify clearly by protein and esterase band pattern of leaf, petiole and stem tissue in three cultivar but with peroxidase band it was difficult to identify because of similar pattern in leaf and petiole tissue. Three cultivars has different leaf shape as Dan-ma and Jang-ma were lanceolate, Sukunea was long-heart shape in upper part of plants. In phyllotaxis, Dan-ma and Jang-ma shows alternate and opposite but Suwon 2 shows opposite in upper parts of stem. Root length shows significant difference from 9.7cm to 51cm respectively as 30cm, 51cm and 9.7cm in Dan-ma, Jang-ma and Sukunea. Tuber yield of three cultivars were 27.5M/T in Dan-ma, 22.8M/T in Jang-ma and 22.8M/T/ha in Sukunea.

Characteristics of Alkaline and Acid Phosphatase in Echinostoma hortense (호르텐스극구흡충에서 Alkaline Phosphatase 및 Acid Phosphatase의 특성)

  • This study was aimed to investigate the enzyme-histochemical localization and characteristics of alkaline and acid phosphatase extracted from adult of Echinostoma hortense. Using the Gomori calcium stain and the Gomori lead nitrate satin method, we found that the alkaline and acid phosphatases were localized mostly in the intestine, vitellaria and pharynx of Echinostoma hortense. The three isozymes of alkaline phosphatase and two isozymes of acid phosphatase were separated from Echinostoma hortense by electrophoresis. The isozymes of alkaline phosphatase were 145.9, 207.5, 220.8 kDa and the isozymes of acid phosphatase were 179.5 and 209.4 kDa. The activity of alkaline phosphatase was denatured completely after heating at 9$0^{\circ}C$ for 12 seconds. The optimum pH and temperature for activity of alkaline phosphatase were about pH 9 and 4$0^{\circ}C$, while the optimum pH for activity of acid phosphatase was about pH 5. The maximum activity of alkaline phosphatase was at 189 unit, but maximum activity of acid phosphatase was at 71 unit As the result from above, we observed that alkaline and acid phosphatases funtion mainly in the alimentary tract and vitellaria. Echinostoma hortense performs the parasitism in the intestine of host by using proper isozyme of phosphatase.
