안정 동위원소비를 이용한 농촌하천유역 질산성 질소의 오염원 추정 (Identification of the Sources of Nitrate using Stable Isotope Mass Ratio in Rural Watersheds)

  • Pollution sources and their environmental contributions were investigated to select the best management practices for the effective control of water quality. The nitrogen isotope mass ratio was determined to estimate the nitrate sources and their contribution. Sampling sites were chosen by calculating effluent loads at each watershed. Two liters each of the surface water samples were collected from four sites at Bokha river and two sites and Gwanri river. They were sequentially prepared through distillation method and analyzed using an isotope ratio mass spectrometer(IsoPrime EA). As revealed by the experimental values obtained the mass ratio values ($\delta$$^{5}$ N) of watersheds with a large amount of nonpoint sources were less than +5 an indication that the samples were influenced by chemical fertilizers. However watersheds with large amount of point sources were influenced not only by chemical fertilizers but also by animal and municipal wastes. The mass ratio values of samples generally decreased during rainy days. But during dry days the mass ratio values well-reflected the nitrate sources and the condition of watersheds. Through this study the nitrogen isotope mass ratio has been found to be useful for estimating nitrate sources and their contribution to the rural watersheds.

Stable Oxygen and Carbon Isotope Profiles of the Bivalve Shells collected from Coastal Regions of Korea: Comparison of the Coastal Water Properties

  • Two marine bivalve shells were collected from the eastern and western coastal regions of Korea, respectively. Stable oxygen and carbon isotope profiles are constructed using the incremental sampling along the axis of maximum growth to provide the continuous ${\delta}^{18}$O and ${\delta}^{13}$C records, which register the physical, biological and chemical properties of seawater where the organisms live. Cycles in the ${\delta}^{18}$O profiles are interpreted as annual along with the identification of annual growth bands; the maximum ${\delta}^{18}$O values correspond with the coldest temperature of seawater whereas the minimum ${\delta}^{18}$O values with the warmest temperature. The primary control on the amplitude of the ${\delta}^{18}$O profiles is seasonal variation of seawater temperature. The offset of the baseline between ${\delta}^{18}$O values of the two specimens is attributed to differences in both temperature and seawater ${\delta}^{18}$O values between two localities. The ${\delta}^{13}$C profiles show the similar seasonality of carbon cycling associated with phytoplankton productivity. The offset in the ${\delta}^{13}$C profiles between two specimens may be, as in the case of oxygen isotope profile, attributed to the different ${\delta}^{13}$C value of the seawater DIC (dissolved inorganic carbon) between the western coast and the eastern coast. Relationships between the shell isotopic composition and the coastal water properties of shell growth are readily interpreted from the ${\delta}^{18}$O-${\delta}^{13}$C pair diagram of the shell isotope data, similar to the use of salinity-${\delta}^{18}$O diagram for identifying water masses. The preliminary stable isotope results of this study suggest that mollusk shell isotope geochemistry may be useful to monitor the properties of water masses in the coastal and inner shelf setting around Korea and improve the interpretation of paleoceanography, provided the fossil mollusks are well preserved.

  • This study was performed to summarize application of ${\delta}^{13}C$, ${\delta}^{37}Cl$ and ${\delta}D$ of trichloroethylene (TCE) to studies on environmental forensic field regarding identification of TCE sources and evaluation of contribution of TCE to groundwater using data collected from literatures. ${\delta}^{13}C$, ${\delta}^{37}Cl$ and ${\delta}D$ of TCE give some information regarding sources of TCE because they show specific value according to manufacturing method. Also, TCE do not show a significant isotopic fractionation owing to adsorption and dilution. The isotopic fractionation mainly occurs by biodegradation. In addition, isotopic fractionation factor for TCE is different according to a kind of microorganism participated in biodegradation. However, the isotopic data of TCE have to be applied with chemical compositions of TCE and other hydrogeologic factors because isotopic fractionation of TCE is influenced by various factors.

A Review on Metabolic Pathway Analysis with Emphasis on Isotope Labeling Approach

  • The recent progress on metabolic systems engineering was reviewed based on our recent research results in terms of (1) metabolic signal flow diagram approach, (2) metabolic flux analysis (MFA) in particular with intracellular isotopomer distribution using NMR and/or GC-MS, (3) synthesis and optimization of metabolic flux distribution (MFD), (4) modification of MFD by gene manipulation and by controlling culture environment, (5) metabolic control analysis (MCA), (6) design of metabolic regulation structure, and (7) identification of unknown pathways with isotope tracing by NMR. The main characteristics of metabolic engineering is to treat metabolism as a network or entirety instead of individual reactions. The applications were made for poly-3-hydroxybutyrate (PHB) production using Ralstonia eutropha and recombinant Escherichia coli, lactate production by recombinant Saccharomyces cerevisiae, pyruvate production by vitamin auxotrophic yeast Toluropsis glabrata, lysine production using Corynebacterium glutamicum, and energetic analysis of photosynthesic microorganisms such as Cyanobateria. The characteristics of each approach were reviewed with their applications. The approach based on isotope labeling experiments gives reliable and quantitative results for metabolic flux analysis. It should be recognized that the next stage should be toward the investigation of metabolic flux analysis with gene and protein expressions to uncover the metabolic regulation in relation to genetic modification and/ or the change in the culture condition.

Use of Stable Carbon Isotope Ratios (${\delta}^{13}$C) for Identification of the Origin Organic Carbon in Benthic Food Webs in Youngil Bay, Korea

  • The analysis of stable carbon isotope ratios for benthic fauna was applied to identify the source of carbon in benthic food webs in Youngil Bay, Korea. The ${\delta}^{13}$C values of 9 invertebrate species collected in this area showed a narrow range between -20.5 and -16.3%$_o$ with a mean of-18.1 (${\pm}$1.1)"%$_o$. The results suggest that the major source of organic carbon for the benthic fauna of the lower estuarine reaches and the oceanic sites is autochthonous marine particulate organic matter. The contribution of organic matter from terrestrial and riverine sources to the diet of the benthic fauna in this area appears to be minor, despite the considerable inflow of riverine waters.

Oxygen Isotope Data of Winter Water in the Western Weddell Sea: Preliminary Results

  • In the western Weddell Sea, winter mixed layer is characterized by near-freezing temperature and higher salinity due to brine injection through sea-ice formation. This layer becomes Winter Water being capped by warmer and less saline Antarctic Surface Water during the sea-ice melt-ing season. In this study, Winter Water was preliminarily identified by the oxygen isotopic com-positions. The ${\delta}^{18}$O values of Winter Water show the progressively increasing trend from south to north in the study area. It presumably reflects the enhanced mixing with Antarctic Surface Water due to the extent of influence by low S'"0 value of sea-ice/glacier meltwater. Correlations between salinity and 6'"0 values of seawater can be used to more generally characterize Winter Water with a view to identification. However, the prediction on the degree of mixing from these relationships needs more detailed isotope data, although this study allows the oxygen isotopic composition of seawater as a tracer to identify the water mass.

Quantitative Proteomics Towards Understanding Life and Environment

  • New proteomic techniques have been pioneered extensively in recent years, enabling the high-throughput and systematic analyses of cellular proteins in combination with bioinformatic tools. Furthermore, the development of such novel proteomic techniques facilitates the elucidation of the functions of proteins under stress or disease conditions, resulting in the discovery of biomarkers for responses to environmental stimuli. The ultimate objective of proteomics is targeted toward the entire proteome of life, subcellular localization biochemical activities, and the regulation thereof. Comprehensive analysis strategies of proteomics can be classified into three categories: (i) protein separation via 2-dimensional gel electrophoresis (2-DE) or liquid chromatography (LC), (ii) protein identification via either Edman sequencing or mass spectrometry (MS), and (iii) proteome quantitation. Currently, MS-based proteomics techniques have shifted from qualitative proteome analysis via 2-DE or 2D-LC coupled with off-line matrix assisted laser desorption ionization (MALDI) and on-line electrospray ionization (ESI) MS, respectively, toward quantitative proteome analysis. In vitro quantitative proteomic techniques include differential gel electrophoresis with fluorescence dyes. protein-labeling tagging with isotope-coded affinity tags, and peptide-labeling tagging with isobaric tags for relative and absolute quantitation. In addition, stable isotope-labeled amino acids can be in vivo labeled into live culture cells via metabolic incorporation. MS-based proteomics techniques extend to the detection of the phosphopeptide mapping of biologically crucial proteins, which ale associated with post-translational modification. These complementary proteomic techniques contribute to our current understanding of the manner in which life responds to differing environment.

  • 생물조절기법 (Biomanipulation)을 이용한 수질개선 방법으로서, 이매패류인 S. woodiana 종이 여과섭식을 통해 남조류(M. aeruginosa)를 효율적으로 제어할 수 있다는 것을 안정동위원소($^{13}C$, $^{15}N$) 추적자 실험을 통하여 밝혀내었다. 이매패류를 이용하여 호소수의 남조류 번성을 억제함으로서 호소 건강성을 회복시키는 것도 중요하지만, 외래종보다는 국내 자생종을 활용하는 것이 생태계 변화를 최소화 할 수 있다는 것을 고려해야 할 것이다.

Identification of pollutant sources using water quality and stable isotope ratios of inflow tributaries in the lower reaches of the Han-River

  • Despite the expansion of sewage treatment facilities to reduce pollutants in the tributaries of the Han River, water pollution accidents such as fish deaths continue to frequently occur. The purpose of this study was to identify the pollutant sources using water quality and stable isotope ratio (${\delta}^{15}N$, ${\delta}^{13}C$, ${\delta}^{15}N-NH_4$, ${\delta}^{15}N-NO_3$) analysis results in the three inflow tributaries (Gulpocheon (GP), Anyangcheon (AY) and Sincheon (SC)) of the Han River. Water quality was analyzed in June and October from 2013 to 2017, and the results showed that the concentrations of nutrients, such as T-N, $NO_3-N$, and T-P, were increased at GP4, AY3, SC3, and SC4, which lie downstream of sewage treatment facilities. The results of ${\delta}^{15}N$ for June 2017 indicated that the source of nitrogen was sewage or livestock excreta at GP4 and SC4, and organic fertilizers at AY3 and SC3. ${\delta}^{15}N-NO_3$ results suggested that the source of nitrogen was related to organic sewage, livestock or manure at GP4, AY3 and SC4. Therefore, GP4 and SC4 were more influenced by effluent from sewage treatment facilities than by their tributaries, AY3 and SC3 were considered to be influenced more by their tributary than effluent from sewage treatment facilities. With the results of this study, the source of contamination (sewage treatment facility effluent) of river inflow downstream of Han River could be confirmed using water quality and stable isotope ratio.
