Effect of Wangsuk Stream on NOM and Chlorinated DBPFPs in Han River Water (왕숙천 유입에 따른 한강본류의 천연유기물질과 염소소독부산물 생성능 변화)

  • The main purposes of this study were to compare the characteristics of fractionated natural organic matters(NOM) from Han River water and Wangsuk(W) stream water, and to investigate the relationships between NOM and the formation of disinfection by products(DBPs). Three types of resin such as XAD-4, XAD-7HP and IRC-50 were used to isolate the water samples into three organic fractions. The DOC concentrations of raw waters were relatively low($1.5{\sim}3.3$ mg/L) at all seasons. The hydrophilic was the major constituent, contributing $44{\sim}63%$ of the total NOM and hydrophobic $21{\sim}33%$, transphilic $16{\sim}31%$, respectively. The formation of trihalomethans(THMs) was highly influenced by particulated NOM especially in the rainy season, whereas haloaceticacid forming potentials(HAAFPs) depended more on the hydrophilic fraction of dissolved NOM which is known to be difficult to be removed through conventional processes. The NOM of W stream was characterized as 15% hydrophobic, 9% transphilic, and 76% hydrophilic. In the fractionation of NOM using resins, $20{\sim}40%$ of the NOM in W tributary water could not be clearly isolated, whereas, 85% of the NOM in the raw water was recovered. Although the DOC concentration of tributary water was higher than the raw waters from the Han River, the DBPFPs was approximately 40% of the raw waters. In DBPFPs aspect, W stream has less effect than Han River water itself. Bromide in tributary waters discharged from waste water treatment plants has been found to shift the distribution of THMs and HANs to the more brominated DBPs.

Studies on the Life History of Acanthoparyphium tyosenense Yamaguti, 1939 (Acanthoparyphium tyosenense Yamaguti, 1939의 생활사에 관한 연구)

  • Cercaria yamagutii Ito, 1957 (C. yamagutii) was found in Lunatia fortuni (L. fortuni) and Neverita didyma (N. didyma) collected from the tideland of Sim-po located at the estuary of the Mankyong River, Chonbuk. It was finally confirmed that the parasite is Acanthoparyphium tyosenense Yamaguti, 1939 (A. tyosenense) and its life history was clarified in this study. Mactra veneriformis (M. veneriformis) was artificially infected with C. yamagutii isolated from L. fortuni and N. didyma. It began to intrude into M. veneriformis through the inhalent canal. Five hours after infection, the tails of the cercaria began to be separated from the main body and the cercaria started to form cysts. Mature cysts were formed 340 hours (14 days) after infection. The cysts were $300\sim360{\mu}m$ in diameter and the encysted metacercarias were $790\sim800\times300\sim310{\mu}m$ in size. The metacercarias were administered orally to Larus crassiostris (L. crassiostris), and adult worms of $84.5\sim112.5\times55\sim65{\mu}m$ were found full of eggs with $2.20\~3.70$ mm long and $0.40\~0.59$ mm wide after 10 days. In a field study, it was observed that the infection rate of A. tyosenense is $99.5\%$ in M. venerifomis, $76.3\%$ Solen strictus (S. strictus), and $37\%$ Ruditapes philippinarum (R. philippinarum), No difference was found among different host sizes, It was concluded that the first intermediate hosts of A. tyosenense Yamaguti were L. fortuni, N. didyma, Tympanotonus microptera, Cerithidea (Cerithidea) largillierti, Cerithidea (Cerideopsilloa) cingulata, the second intermediate hosts M. venerifomis, S. strictus and R. philippinamn, and the final hosts L. crassiostris and Melanitta fusca stejnegeri.

  • The marine bacterial flora of seawater at Masan Bay located in southern part of Korea was investigated to provide basic information about the origin of red tides in this area. The samples were collected at 5 sampling stations (see fig.1) established once a month from July 1985 to June 1986. The results are summerized as follows: 1. Two hundred and ninety one strains of marine bacteria among 299 strains isolated were identified from 156 samples(150 seawater samples and 6 mud samples) collected in the study area. 2. During the study period, the dissolved oxygen ranged from 2.1 to 6.3 ml/l; temperature of surface seawater ranged from 4.6 to $30.1^{\circ}C$; phosphate, silicate and nitrite ranged from 1.6 to 5.6 ${mu}g-at/l$, 327.27 to 3798.38 ${mu}g-at/l$, and 1.09 to 5.5 ${mu}g-at/l$, respectively. 3. Pseudomonas spp. were the most dominant bacterial species before red tides, Ancinetobacter spp. and Flavobacterium spp. during red tides, and Flavobacterium spp., Vibrio spp. and Acinetobacter spp. after red tides. 4. Viable counts marine bacterial flora ranged $10^4{\sim}10^8cell/ml$, and the most abundant viable counts of marine bacteria occured during red tides ($10^7{\sim}10^8cell/ml$).

  • To develop a beneficial microbial feed for the cultivation of rotifer, Brachionus plicatilis, an aerobic photosynthetic bacterium, Erythrobacter sp. $S\;\pi-I$ was isolated from marine structure at Haeundae beach in Pusan, Korea. Feeding effects of Erythrobacter sp. $S\;\pi-I$ on the growth of rotifer were analyzed comparing to other feeds such as PSB (purple nonsulfur bacteria), Chlorella sp. and baker's yeast. Erythrobacter sp. $S\;\pi-I$ contained more linoleic acid $(C_{18:3\omega3})$ and oleic acid $(C_{18:1\omega9})$ and amino acids than PSB (purple nonsulfur bacteria), Chlorella sp. and baker's yeast. The rotifer fed on Erythrobacter sp. $S\;\pi-I$ showed better effects than those fed on other feeds in the individual growth, size and weight. Also, the rotifer especially contained more eicosapentaenoic acid $(C_{20:5\omega3})$ and docosahexaenoic acid $(C_{22:6\omega3})$ in case of Erythrobacter sp. $S\;\pi-I$ feeding than the other feeds. In case of the feed of PSB and baker's yeast docosahexaenoic acid $(C_{22:6\omega3})$ did not show. In amino acid analysis, the rotifer fed on Erthrobacter sp, $S\;\pi-I$ showed more amino acid content comparing to those fed on other diets. Especially, arginine, isoleucine, histidine, lysine, methionine, phenylalanine, threonine, which are essential amino acid for fish growth, showed high contents. These results suggested that the aerobic photosynthetic bacterium, Erythrobacter sp. $S\;\pi-I$ would be a beneficial microbial teed for the cultivation of rotifer.

THE EFFECT OF C-FACTOR AND VOLUME ON MICROLEAKAGE OF COMPOSITE RESIN RESTORATIONS WITH ENAMEL MARGINS (법랑질 변연으로 이루어진 복합레진 수복물의 체적과 C-factor가 미세누출에 미치는 영향)

  • Competition will usually develop between the opposing walls as the restorative resin shrinks during polymerization. Magnitude of this phenomenon may be depended upon cavity configuration and volume. The purpose of this sturdy was to evaluate the effect of cavity configuration and volume on microleakage of composite resin restoration that has margins on the enamel site only. The labial enamel of forty bovine teeth was ground using a model trimmer to expose a flat enamel surface. Four groups with cylindrical cavities were defined, according to volume and configuration factor(Depth x Diameter / C-factor) - Group I : 1.5 mm ${\times}$ 2.0 mm / 4.0, Group II : 1.5 mm ${\times}$ 6.0 mm / 2.0, Group III : 2.Omm ${\times}$ 1.72 mm / 5.62, Group IV : 2.0 mm ${\times}$ 5.23 mm / 2.54. After treating with fifth-generation one-bottle adhesive - BC Plus$^{TM}$ (Vericom, AnYang, Korea), cavities were bulk flted with microhybrid composite resin - Denfill$^{TM}$ (Vericom). Teeth were stored in distilled water for one day at room temperature and were finished and polished with Sof-Lex system. Specimens were thermocycled 500 times between 5$^{\circ}$C and 55$^{\circ}$C for 30 second at each temperature. Teeth were isolated with two layers of nail varnish except the restoration surface and 1 mm surrounding margins. Electrical conductivity (${\mu}$A) was recorded in distilled water by electrochemical method. Microleakage scores were compared and analyzed using two-way ANOVA at 95% level. The results were as follows: 1. Small cavity volume showed lower microleakage score than large one, however, there was no statistically significant difference. 2. There was no relationship between cavity configuration and microleakage. Factors of cavity configuration and volume did not affect on microleakage of resin restorations with enamel margins only.

EXPRESSION AND FUNCTION OF OD314, APIN PROTEIN, DURING AMELOBLAST DIFFERENTIATION AND AMELOGENESIS (법랑모세포 분화와 법랑질 형성과정에서 OD314, Apin protein의 발현 및 기능)

  • This study was aimed to elucidate the biological function of OD314 (Apin protein), which is related to ameloblast differentiation and amelogenesis. Apin protein, calcifying epithelial odontogenic (pindborg) tumors (CEOTs)-associated amyloid, were isolated from CEOTs, and has similar nucleotide sequences to OD314. We examined expression of the OD314 mRNA using in-situ hybridization during tooth development in mice. Expression of OD314 and several enamel matrix proteins were examined in the cultured ameloblast cell line up to 28 days by reverse transcription-polymerase chain reaction (RT-PCR) amplification. After inactivation and over-expression of the OD314 gene in ameloblast cell lines using U6 vectordriven RNA interference and CMV-OD314 construct, RT-PCR were performed to evaluate the effect of the OD314 during amelogenesis. The results were as follows: 1. In in-situ hybridization, OD314 mRNAs were more strongly expressed in ameloblast than odontoblast. 2. When ameloblast cells were cultured in the diffcrentiation and mineralization medium for 28 days, the tuftelin mRNA expression was maintained from the beginning to day 14, and then gradually decreased to day 28. The expressions of amelogenin and enamelin were gradually decreased according to the ameloblast differentiation. 3. Inactivation of OD314 by U6-OD314 siRNA construct down-regulated the expression of OD314, MMP-20, and tuftelin, whereas over-expression of OD314 by CMV-OD314 construct up-regulated the expression of OD314 and MMP-20 without change in tuftelin. These results suggest that OD314 is considered as an ameloblast-enriched gene and may play the important roles in ameloblast differentiation and mineralization.

Influence of Ginseng Saponins on the Isolated Aortic Contractile Response of the Spontaneously Hypertensive Rat

  • The present study was attempted to investigate the effects of total ginseng saponin (G75), panaxadiol-type (PDS) and panaxatriol-type saponin (PTS) on contractile responses of vasoconstrictors in aortic smooth muscle stripes of normotensive (NR) and spontaneous hypertensive rats (SHR). Phenylephrine (an adrenergic $\alpha$$\_$1/-receptor agonist) and high potassium (a membrane depolarizing agent) caused greatly contractile responses in both NR and AHR aorta, respectively. Phenylephrine- and high potassium-induced contractile responses were greater in NA than those in SHR aortic smooth muscle stripes. In NR, the contractile responses of high potassium (5.6$\times$10$\^$-2/ M) were not affected in the presence of GTS (300 $\mu$g/ml), PDS (300 $\mu$g/ml), and PTS (300 $\mu$g/ml), respectively whereas phenylephrine (10$\^$-6/ M)-induced contractile responses were markedly inhibited. In SHR, the contractile responses of high potassium (5.6$\times$10$\^$-2/ M) were not affected in the presence of GTS (300 $\mu$g/ml), PDS (300 $\mu$g/ml), and moderate doses of PTS (150-300 $\mu$g/ml), respectively but greatly blocked by high concentration of PTS (600 $\mu$g/ml). Phenylephrine (10$\^$-6/ M)-induced contractile responses were inhibited in a dose dependent fashion (150-600 $\mu$g/ml) by the pretreatment with PTS while not altered in the presence of GTS (300 $\mu$g/ml) and PDS (300 $\mu$g/ml), respectively. Taken together, these experimental results suggest that ginseng saponins cause vascular relaxation through blockade of adrenergic $\alpha$$\_$1/-receptors and some unknown mechanisms, and that there is some difference in sensitivity of vascular smooth muscle between NR and SHR in responses to ginseng saponins. It seems that panaxatriol type of some ginseng saponins has the greatest potency in vascular relaxation.

The taxonomic consideration of leaf epidermal microstructure in Korean Thymelaeaceae Adans (한국산 팥꽃나무과 잎표피 미세구조의 분류학적 검토)

  • To examine the leaf epidermal microstructure, nine species in five genera (Daphne L. - 4 spp., Diarthron Turcz. - 1 sp., Edgewarthia Meisn. - 1 sp., Stellera L. - 1 sp., Wikstroemia Endl. - 2 spp.) of the Korean Thymelaeaceae were investigated by light microscopy (LM) and scanning electron microscopy (SEM). The stomata of stuo야ed taxa were 'hypostomatic type' and the size range of guard cell was $13.8-34.4{\times}8.7-22.9{\mu}m$: the smallest size of stomata was found in Diathran linifolium ($15.9{\pm}2.6{\times}10.0{\pm}1.3{\mu}m$), while the largest one was measured to Daphne adara ($32.8{\pm}1.6{\times}20.7{\pm}1.3{\mu}m$). The stomatal complex was anomocytic in the most studied taxa, except Daphne kiusiana by having combined with anisocytic together. The shapes of epidermal cells are undulate anticlinal wall. The size range of epidermal cell was $20.7-61.0{\mu}m$; the smallest size of epidermal cell was found in Stellera charnaejasme ($26.0{\pm}1.9{\mu}m$), on the other hand the largest one was found in Edgeworthia chrysantha ($53.6{\pm}3.1{\mu}m$). The well-developed flaky epicuticular waxes can be divided three kinds of pattern - (1) smooth in comparison, not entire platelets and scattered, (2) isolated flake-like platelets, mostly paralleled, sparsely, (3) flake-like platelets, flat, membraneous, protruding from the surfaces at varying angles and densely. Two types of trichome are recognized; (1) Type I: uniseriate trichome of striate surface (D. genkwa, Diarthron linifalium, E. chrysantha, W. ganpi and W. trichotama), (2) Type II: multicellular trichome of papillose surface, uncinated 3-4 nodes (Diathron linifolium). Finally, the systematics significance of the leaf micromorphological features in identification and elucidation of Korean Thymelaeaceae, especially between or within the genera including among the species is also briefly discussed.

Extraction of Acetylcholinestrase from the Housefly and Three Other Insect Species for In Vitro Anticholinesterase Screening (In Vitro Anticholinesterase 스크리닝을 위한 집파리 및 3종 곤충으로부터의 Acetylcholinesterase의 추출)

  • The optimal pH of the extraction buffer was 7.5 considiering AChE stability and its buffer capacity when AChE was isolated and extracted from the housefly(Musca domesitca L.)and three other insect species with 0.01 M sodium phosphate buffer. Also, the optimal pH of the reaction buffer was 7.5 considering enzyme activity and its buffer capacity when AChE activity was measured with the substrate in 0.1 M sodium phosphate buffer. The Potter Elvehjem type homogenizer with Teflon pestle was used to homgenize the tissues. When preparing a AChE suspension by centrifuging the homogenate, 700 g supernatant of adult head for the housefly, 700 g supernatnat of 5th instar nymphal whole body for the brown planthopper, lipid-eliminated 10,000 g supernatant of 5th instar larval whole body for the diamondback moth, and 700 g supernatant of 4th instar larval head for the tobacco cutworm were considered satisfactory as enzyme sources in view of mass preparation, extraction efficiency and stability of enzyme activity during evaluation. When AChE suspensions of 4 insect species were stored at $-18^{\circ}C$, more than 90% of activity was maintained up to 3 weeks. Km values of AChEs of the housefly, the brown planthopper, and the diamondback moth were 0.042, 0.037 and 0.043 mM, respectively and AChE-specific substrate inhibition was observed at high concentration. Km value of the tobacco cutworm ChE was 1.15 mM and BuChE characteristics was observed, though further study is needed. The optimal substrate concentration for the AChE inhibition tests was 0.5 mM for the housfly, the brown planthopper, and the diamondback moth and 12 mM for the tobacco cutworm.

Geographic Variation in Pond Smelt (Hypomesus nipponensis) by RAPD Analysis (RAPD 분석에 의한 빙어 (Hypomesus nipponensis)의 지리적 변이)

  • Genomic DNA isolated from two geographical populations of pond-smelt (Hypomesus nipponensis) was amplified for RAPD (randomly amplified polymorphic DNA) analysis. The populations were obtained from Chungju (CJ), in the inland area, and Dangjin (DJ), in the vicinity of the West Sea in Korea. Seven arbitrarily selected primers, OPB-06, OPB-10, OPB-13, OPB-17, OPC-09, OPC-17 and OPC-20, were used to generate the shared loci, polymorphic, and specific loci. Three hundred and eighty-three loci observed per primer were identified in the CJ population, and 287 were identified in the DJ population. Among them, 91 polymorphic loci or 23.8% were polymorphic in the CJ population, and 47 (16.4%) in the DJ population. The number of shared loci observed was 198 in the CJ population and 176 in the DJ population. Forty-four and 75 specific loci were detected in the CJ and DJ populations, respectively. Especially, 99 numbers of shared loci by the two populations, with an average of 14.1 per primer, were observed in the two pond-smelt populations. The average bandsharing value between the two geographical pond-smelt populations was $0.700{\pm}0.008$, ranging from 0.600 to 0.846. Compared separately, the bandsharing value of individuals within the CJ population was higher than that of the DJ population. The dendrogram obtained using the data from the seven primers indicated three genetic clusters: cluster 1, CJ 01, 02, 03, 04, 05, 06, 07, 08, 09, 10, and 11; cluster 2, DJ 01, 02, 03, 04, 05, 06, 07, 08, and 09; and cluster 3, DJ 10 and 11. The genetic distance between the two geographical populations ranged from 0.040 to 0.545. Thus, RAPD-PCR analysis revealed a significant genetic distance between the two pond-smelt populations.
