Isoflavanones from the Stem of Cassia siamea and Their Anti-tobacco Mosaic Virus Activities

  • Two new isoflavanones, (3R) 7,2',4'-trihydroxy-3'-methoxy-5-methoxycarbonyl-isoflavanone (1) and (3R) 7,2'-dihydroxy-3',4'-dimethoxy-5-methoxycarbonyl-isoflavanone (2), together with six known isoflavanones (3-8), were isolated from the stems of Cassia siamea. The structure of 1-8 was elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1, 2, 5-8 were evaluated for their anti-tobacco mosaic virus (Anti-TMV) activity. The results showed that compounds 1 and 6 showed potential anti-TMV activity with inhibition rates of 24.6% and 26.9%, respectively. Compounds 2, 5, 7, 8 also showed anti-TMV activity with inhibition rates in the range of 11.8-18.6%.

Cytotoxic Isoflavanones from Uraria clarkei

  • Two new isoflavanones, (3R) 5,7,3',4'-tetrahydroxy-2'-methoxyisoflavanone (1) and (3R) 5',8-di-(${\gamma}$,${\gamma}$-dimethylallyl)-2',5-dihydroxyl-4',7-dimethoxyl-isoflavanone (2), were isolated from Uraria clarkei, together with two known compounds dalbergioidin (3), 5,7-dihydroxy-2',4'-dimethoxyisoflavanone (4). The structures involving the absolute configuration of the new compounds were well elucidated by MS, IR, UV, CD, 1D and 2D NMR analyses. Cytotoxicity of the four compounds were assessed, results suggested that compound 2 possessed well cytotoxic activity, against the Hela, K562, and HL60 cell lines with $IC_{50}$ values of 28.0, 40.6 and $35.1{\mu}M$, respectively.

Structural Requirement of Isoflavonones for the Inhibitory Activity of Interleukin-5

  • Interleukin (IL)-5 appears to be one of the main proinflammatory mediators among a growing number of cytokines and chemokines that induce eosinophilic inflammation. Sophoricoside and their analogs isolated from Sophora japonica show relatively potent inhibitory activity of interleukin (IL)-5 as a small molecule. To identify structural requirements of this isoflavonone for its inhibitory activity against IL -5. isoflavonones. isoflavanones, and their glycosides were prepared and tested their inhibitory activity against IL-5. Variation of the antioxidant activity of squid jeotgal by altering the level of added soy isoflavones and storage duration

  • In this study, we propose a squid jeotgal, Korean fermented seafood, supplement with different soy isoflavones supplements, followed by fermentation for different time intervals at $4^{\circ}C$ to increase the antioxidant activity and improve the food value. In the first month, fermented jeotgals with at lowconcentration ($2mg\;g^{-1}$) of added soy isoflavones showed a significant increase in the activity of up to 55%, whereas, at high concentration ($10mg\;g^{-1}$), the activity almost doubled compared to that of the sample without isoflavones. Moreover, the squid enriched with isoflavones also exhibited significantly decreased total volatile base nitrogen, thiobarbituric acid reactive substances, and biogenic amines, indicative of higher inhibition of the formation of these substances. The changes in the microbial profile were also evaluated. This use of soy isoflavanones as an additive could aid in improving the nutritional value of fermented seafood to reduce the incidence of age-related and chronic disorders.
