Anti-diabetic Effects of Isaria tenuipes in OLETF Rats as an Animal Model of Diabetes Mellitus Type II (제 2형 당뇨모델 OLETF 랫드에서 동충하초의 항당뇨 효과)

  • We evaluated the anti-diabetic effects of Isaria tenuipes in diabetes mellitus type 2. For the experiments, the diabetic animal model OLETF rats were divided to 4 groups: Isaria tenuipes was administered mixed with the high fat diet 45% at dose levels of 0.0%, 0.1%, 1.0%, and 5.0% for 4 weeks. All animals have free access to water and high fat diet 45%. The diabetic clinical markers, including clinical signs, body weight and food intake, organ weights, blood glucose level, insulin level and HOMA-IR index, oral glucose tolerance test, GLUT4 mRNA and protein were measured at a time. After administration for 4 weeks, the blood glucose levels, insulin levels and HOMA-IR index of test groups were decreased compared with control group in dose-dependent manner. The body weight and diet consumptions were reduced in control group at 4 weeks. The treatments of Isaria tenuipes also showed high expression of GLUT4 mRNA and protein in the muscle of OLETF rats. The results suggest that Isaria tenuipes has anti-hyperglycemic effect attenuating blood glucose in the animal model of type 2 diabetes and might be useful as a functional diet for human diabetic diseases.

Identification and Characterization of Entomopathogenic Fungi Isolated from Bemisia tabaci in Korea (담배가루이(Bemisia tabaci )에서 분리한 곤충병원성진균의 동정 및 병원성 검정)

  • Entomopathogenic fungi were isolated from Bemisia tabaci in an Oriental melon field, and their growth characteristics, factors related to a natural outbreak, and infectivity against Bemisia tabaci, Tetranychus urticae, and Frankliniella intonsa were investigated. The isolates had erect conidiophores bearing whorls of 4-6 phialides with a swollen base where cylindrical conidia of $3.0-3.4{\mu}m$ were attached. The isolates were identified as Isaria fumosorosea on the basis of morphological characteristics and an ITS sequence with 99% similarity. I. fumosorosea IFs-08 grew well on Sabouraud dextrose agar+yeast extract medium(3.2 mm/day/$24^{\circ}C$); it grew better at $35^{\circ}C$ than at $15^{\circ}C$. The isolates of I. fumosorosea-IFs were highly infective and killed 93.9-96.7% B. tabaci, 84.9-92.0% T. urticae, and 81.5-84.4% F. intonsa in bioassay, whereas three isolates (Isaria tenuipes, Isaria farinosa, and Isaria fumosorosea) from KACC showed a low infectivity of 10-20%. To the best of our knowledge, this is the first report of I. fumosorosea isolated from B. tabaci in Korea.

Excellent Strains Selection from Wild Cordyceps spp.(II) (야생동충하초로부터 우량균주선발(II))

  • Many Cordyceps spp. were collected for excellent strains selection at near Wanju-kun from May to October, 2000. They were identified. As the resulting, They were 2 genera and 4 species. 6 strains were got through 4 species culture. 6 strains are Cordyceps militaris f. albino, C, gracilaides, C. militaris and Isaria japoica. Among them, 4 species used for induction of artifitial fruiting body. 3 strains of them were selected for excellent strains. excellent strains are Cordyceps militaris and Isaria japoica.

  • This is a forecasting report for die experiment on agent of disease to be carried on next year. 1. Muscardine of the silkworm and wild insects are supposed to be caused by a common agent, Nomuraea Pracina and Isaria Spp. 2. In the view of using the method of spreading the spores of Isaria spp to control the Dendrolimus Spectabilis Butter, the other wild insects are also supposed to be contaminated by that. 3. Hyphantria Cunea Drury does not seem to be infected by fungus of Muscardine.

  • Many Cordyceps rpp were collected for excellent strains selection at Mts. Moak, Younsuk and Daea-ree herbarium near Wanju-kun from mid May to October,1999. They were identified as the resulting 2 genera and 5 species, 10 strains were got through 5 species culture. Among them 4 species used for induction of artifitial fruiting body and 6 strains of them were selected to excellent strains: Cordyceps militaris (99-0007), Isaria japonica (99-0001, 99-0002, 99-0003), Cordyceps soborifera (99-0008), Cordyceps kanzashiana(99-0009).

  • Many ascomycetes were collected at Mt.Unjang, Pyonsan penisula national park, Mt.Moak Chonbuk provincial park, Daea-ri hebarium and Mt.Whaam from August 1996 to October 1999. They were identified. According to the resulting, Lanzia echinophila, Discina parma, Hypomyces auranticus, Helotium versicolor, Cordyceps militaris f. albino, Isaria sinclairii are newly to Korea. They were designed Korean common names by author.

  • Isaria sinclairii (Cicada Dongchunghacho) was studied as a potential crude natural food in powdered form. The role of tissue fatty acids in relation to the anti-obesity effects of I. sinclairii (IS) was examined by feeding the powder to SD rats ad libitum at 0, 1.25, 2.5, 5 and 10% (calculated about 8 g/kg) of the feed for a period of 3 months and 6 months. The fatty acid composition profile as indicated GC-MS, showed significantly slight dose-dependent increases in the levels of unsaturated fatty acids, particularly, arachidonic acid (C20: 4n6), oleic acid, linoleic acid, eicosadienoic acid, eicosapentaenoic acid (EPA) (C20: 5) concentration in the the ad libitum IS-fed groups compared to the control group in SD abdominal fat over 6 month period. Over viewing of the SD and Ob mice treated Isaria sinclairii powder; there were increases in the single (mono) unsaturated fatty acids ratio but decreases in polyunsaturated fatty acid. In IS-fed groups in proportion to the treatment period, this Dongchunghacho also induced an increase in the level of same result of unsaturated fatty acid in C57BL/6 obese (ob/ob) mice over a 6-month period treatment compared to those given 10% dry mulberry leaf powder (ML) or silkworm powder mixed with the standard diet.

Breeding of a Silkworm Variety for Synnemata Production of Isaria tenuipes

  • This study was conducted out to select a silkworm variety suitable for synnemata production of Isaria tenuipes. Four kinds of the mulberry silkworm varieties, Bombyx mori, were hybridized using a Japanese parental line and a Chinese parental line, and used to test for synemata formation in I. tenuipes. The larval period of normal silkworms was 22 hr longer than the silkworms inoculated with this fungus. Among the silkworm varieties tested, Hachojam had the shortest larval period with 23.02 days. The non-cocooning silkworm had a shorter larval period than the cocoon producing silkworms. The pupation rate of normal silkworms was about 9% higher than that of silkworms sprayed with I. tenuipes. Hachojam had the highest infection rate at 99.8%, but no significant difference was observed for the infection rate by silkworm variety. The production of synnemata was the best in JS171 $\times$ CS188 with an incidence rate of 99.3%, followed by Hachojam, and Chugangjam. The synnemata produced from Hachojam were the heaviest and showed white or milky-white in color.

Antioxidant Activity of N-hydroxyethyl Adenosine from Isaria sinclairii

  • The antioxidant activity of Isaria (Paecilomyces) sinclairii was determined by measuring its radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The n- BuOH extract of P. sinclairii showed strong scavenging activity to DPPH. The anti-oxidant potential of the individual fraction was in the order of ethylacetate> n- BuOH>chloroform>n-hexane. The n-BuOH soluble fraction exhibiting strong anti-oxidant activity was further purified by repeated silica gel column chromatography. N-(2-Hydroxyethyl)adenosine (HEA) was isolated as one of the active principles from the n-BuOH layer. The n-BuOH layer, particularly HEA, did not increase the level of nitric oxide (NO) production in vascular endothelial cells that might be related to vasorelaxation such as the action of viagra. In addition, the vascular endothelial growth factor (VEGF) levels showed little or no increase compared with control group with the treatment of I. sinclairii.
