  • Regulation of inulinase biosynthesis was studied in Bacillus sphaericus 188-1 Biosynthesis of inulinase was effectively induced in the presence of 0.5% inulin for 8 hrs. Fructose (0.5%) repressed the inulinase induction by inulin and as late as addition time of fructose, inulinase formation was decreased. Catabolite repression was not reduced by the addition of CAMP for 8 hrs of induction.

  • In an attempt to develop an unique enzyme (inulinase) for fructan utilization. bacterial strains were isolated [yom soil. Stram 96-11 secreting inulinase o[ high activity was tentatively identificated as Arthrobacter protophmmiae/ranwsus. The optimum culture conditions o[the slnin for the production of the inulinase were as follow: inorganic saIl basal medium contained sources fl % (w/v) inulin, 1 % (w/v) tryptone, and 1 % (w/v) $NH_4Cl$]. $35^{\circ}C$, initial pH 7.5. aeration 1 vvm and agitation 200 rpm.

  • Inulinase of Kluyveromyces marxianus origin was produced by recombinant yeast Saccharomyces cerevisiae under the control of GAL1 promoter, to examine the expression and localization of inulinase in a repressed(galactose-free) or derepressed(galactose-containinga) medium. The inulinase gene(INU1A) was constitutively expressed at 6.7 units/ml in a repressed medium. When the cell started to utilize galactose in a derepressed medium, the INU1A gene began to be expressed, and the final expression level reached about 45 units/ml. According to be the nondenaturingPAGE analysis, inulinase produced by S. cerevisiae was found to be less glycosylated than the bakers yeast invertase. In addition, its glycosylation pattern was less heterogeneous than the K. marxianus inulinase. The supplementation of inulin or raffinose into the derepressed medium increased the cell growth rate, while the expression of INU1A was repressed. Regardless of the carbon sources examined, most of inulinase activity (more than 98%) was found in the extracellular medium, indicating excellent secretion efficiency.

  • The extracellular inulinase from Bacillus spp. was purified to a single protein through a sequence of operations including ammonium sulfate fractionation, heat treatment, DEAE Sepharose C1-6B ion exchange chromatography, Sephadex 6-100 and Sephadex 6-150 gel filtration. The purified enzyme was confirmed to be a $\beta$ -D-fructofuranosidase(EC 3.2.1.26) which was much more active on sucrose than on inulin(I/S = 0.2). The maximal inulinase activity was observed at pH 6.0 and at the temperature of $50^{\circ}C$. The mo1ecular weight of the enzyme was about 56, 000. Tryptophan and histidine residues of the enzyme molecule were found to be essential for its catalytic activity.

  • A strain of Pseudomonas sp. isolated from soil was shown to produce a high level of extracellular endo-inulinase. In this work, the endo-inulinase gene (inu1) of the bacterial strain was cloned into the plasmid pBR322 by using EcoRI restriction endonuclease and E. coli HB101 as a host strain. One out of 7, 000 transformants obtained from the above cloning experiment formed a clear zone around its colony on the selective medium supplemented with 2.0% inulin after a prolonged incubation at 37$\circ$C and subsequent cold shock treatment. The functional clone was found to carry a recombinant plasmid (pKMG50) with a 3.7 kb genomic insert containing the genetic information for the inulinase activity. The inulinase from E. coli HB101/pKMG50 was proved to be an endo-acting enzyme and produced constitutively in the recombinant E. coli cells. Zymogram of the enzyme from the recombinant cells with inulin substrate indicated that the molecular mass of the active protein was 190 Kd, while that of the endo-inulinase from the Pseudomonas strain was 170 Kd. This size discrepancy suggested that the inulinase from the recombinant E. coli HB101 cells might be the initial product of translation, not the mature form produced in the strain of Pseudomonas sp..

  • Inulin degradation was examined using patially puriiied enzyme mixtures of the Exo-inulinase from a Bacillus spp. and the Endo-inulinase from a Pseudomonas spp.. The highest synergistic xtion of the two cnzymcs was observcd when the Exo- and the Endo-inulinase werc mixed at the ratio of 1 to 13, and the rate of hydrolysis of the above process was enhanced approximately 1.6 times I1ight.1- than that of the reaction catalysed with a single enzyme of the same units. The enzymc mixture showed the maximal activity at pH 6.0 and $55^{\circ}C$, and in the prescncc of 0.5 mM each of $CO^{2+}$ and $Mn^{2+}$. Under the optimal condition described above fructosu was accumulated with the overitll concentration of 84% after 36 hours of the reiiction.

  • 고정화 inulinase를 이용하여 inulin으로부터 fruc-tose 시럽을 연속적으로 생산하기 위하여 Bacillus sphaericus 188-1이 생성하는 inulinase를 부분 정제한후 5시간 NaIO$_4$로 산화시킨 cellulose에 고정화시킨 다음 이들의 성질을 조사하였다. 산화 cellulose에 고정화시킨 inulinase의 활성은 g당 47 Unit 이었고 고정화수율과 활성수율은 각각 41%와 39%이었다. 고정화시킨 inulinase의 작용 최적온도와 pH는 각각 5$0^{\circ}C$, pH 9.0이었고 5$0^{\circ}C$, pH 8.0~10.0에서 비교적 안정 하였다 고정화시킨 inulinas의 활성은 K+, Ca2+, Mn2+과 Hg2+에 의하여 활성화 되었으며 EDTA(10mM)에 의하여 심하게 저해되었다.

  • The effects of medium pH and culture temperature on the expression and secretion of inulinase and invertase were investigated with recombinant Saccharomyces cerevisiae cells. These cells were obtained by transformation of 2$\mu$-based plasmids pYI10 and pYS10 which contain Kluyveromyces marxianus inulinase gene (INU1A) and S. cerevisiae invertase gene (SUC2), respectively, in the downstream of GAL1 promoter. The expression level and localization of inulinase and invertase were not affected significantly by the initial medium pH: secretion efficiencies of inulinase and invertase into the medium were about 90% and 60%, respectively, in the pH ranges of 4.0 to 6.5. However, the expression and secretion of both enzymes were strongly dependent on the culture temperature. The highest expression (7.7 units/mL) and secretion (6.7 units/mL) of inulinase were observed at 28$\circ$C and 30$\circ$C. As a consequence of decreased localization of inulinase in the periplasmic space, the secretion efficiency increased from 68% at 20$\circ$C, to 95% at 35$\circ$C,. The total expression level and secretion efficiency of invertase increased from 19 units/mL and 55% at 20$\circ$C to 25 units/mL and 68% at 35$\circ$C, respectively. Irrespective of the culture temperature, the invertase activity in the cellular fraction (periplasmic space and cytoplasmic fractions) was kept constant at around 33-45%.

  • A 11.5-kb DNA fragment containing an endo-inulinase gene was cloned from Xanthomonas oryzae #5. It contained a single open reading frame of 3,999bp, encoding a polypeptide composed of signal peptide of 32 amino acids and mature protein of 1,301 amino acids. From the comparison of amino acids sequences with fructan hydrolases, inulinase, levanase and CFTase, the sequence of the endo-inulinase had highly homology of 72% with CFTase of B. circulans, and six highly conserved regions including the ${\beta}-fructosidase$ motif were found.

  • The use of Jerusalem artichoke containing $\beta$-1, 2-fructose oligomer in the production of sorbitol that is used as food additives and precursor for the L-sorbose has been studied. Coimmobilization of both inulinase and oxidoreductase was considered for the simultaneous reaction for hydrolysis of inulin and conversion of glucose and fructose liberated from inulin to sorbitol. Both inulinase and oxidoreductase were immobilized in chitin(5%, w/v) and K-carrageenan(4%, w/v), The activity of oxidoreductase was specified by permeabilization of Zymomonas mobilis cell with 0.2% CTAB(Cetyltrimethylammonlumbromide). The use of inulinase for hydrolysis of inulin resulted in 36.65g/l of glucose and 85.32g/1 of fructose respectively. These are valuable substrates for sorbitol production. Using these hydrolyzates, accumulation of 35.64g/l for sorbitol occurred at $38^{\circ}C$ and pH6.2. When permeabilized cells and inulinase were coimmobilized, sorbitol produced at 30.15g/l although it is low compared with 35.64g/l in separated reactor system.
