FDG PET-CT에서 악성림프종처럼 보이는 복부 기쿠치병 (Intra-abdominal Kikuchi's Disease Mimicking Malignant Lymphoma on FDG PET-CT)

  • Kikuchi's disease is a self-limiting benign disease characterized by cervical lymphadenopathy, but it can be mistaken for malignant disease, and when involved lymph nodes are unusually located, diagnosis can be more difficult. The authors report the case of a 19-year-old man with Kikuchi's disease, who had isolated intra-abdominal lymphadenopathy and increased 18-fluoro-deoxyglucose (FDG) uptake in positron emission tomography-computed tomography (PET-CT). Although its incidence is extremely rare, intra-abdominal Kikuchi's disease with increased FDG uptake in PET-CT image should be considered in the differential diagnosis when constitutional symptoms mimic those of malignant lymphoma.

  • A formulation is proposed for modelling the process of intra-granular diffusion of fission gas during irradiation of $UO_2$ under both normal operating conditions and power transients. The concept represents a simple extension of the formulation of Speight, including an estimation of the contribution of bubble motion to fission gas diffusion. The resulting equation is formally identical to the diffusion equation adopted in most models that are based on the formulation of Speight, therefore retaining the advantages in terms of simplicity of the mathematical-numerical treatment and allowing application in integral fuel performance codes. The development of the new model proposed here relies on results obtained by means of molecular dynamics simulations as well as finite element computations. The formulation is proposed for incorporation in the TRANSURANUS fuel performance code.

Widespread Occurrence of Small Inversions in the Chloroplast Genomes of Land Plants

  • Large inversions are well characterized in the chloroplast genomes of land plants. In contrast, reports of small inversions are rare and involve limited plant groups. In this study, we report the widespread occurrence of small inversions ranging from 5 to 50 bp in fully and partially sequenced chloroplast genomes of both monocots and dicots. We found that small inversions were much more common than large inversions. The small inversions were scattered over the chloroplast genome including the IR, SSC, and LSC regions. Several small inversions were uncovered in chloroplast genomes even though they shared the same overall gene order. The majority of these small inversions were located within 100 bp downstream of the 3' ends of genes. All had inverted repeat sequences, ranging from 11 to 24 bp, at their ends. Such small inversions form stem-loop hairpin structures that usually have the function of stabilizing the corresponding mRNA molecules. Intra-molecular recombination between the inverted sequences in the stem-forming regions are responsible for generating flip-flop orientations of the loops. The presence of two different orientations of the stem-loop in the trnL-F noncoding region of a single species of Jasminum elegans suggests that a short inversion can be generated within a short period of time. Small inversions of non-coding sequences may influence sequence alignment and character interpretation in phylogeny reconstructions, as shown in nine species of Jasminum. Many small inversions may have been generated by parallel or back mutation events during chloroplast genome evolution. Our data indicate that caution is needed when using chloroplast non-coding sequences for phylogenetic analysis.

Direct characterization of E2-dependent target specificity and processivity using an artificial p27-linker-E2 ubiquitination system

  • Little attention has been paid to the specificity between E2 and the target protein during ubiquitination, although RING-E3 induces a potential intra-molecular reaction by mediating the direct transfer of ubiquitin from E2 to the target protein. We have constructed artificial E2 fusion proteins in which a target protein (p27) is tethered to one of six E2s via a flexible linker. Interestingly, only three E2s (UbcH5b, hHR6b, and Cdc34) are able to ubiquitinate p27 via an intra-molecular reaction in this system. Although the first ubiquitination of p27 (p27-Ub) by Cdc34 is less efficient than that of UbcH5b and hHR6b, the additional ubiquitin attachment to p27-Ub by Cdc34 is highly efficient. The E2 core of Cdc34 provides specificity to p27, and the residues 184-196 are required for possessive ubiquitination by Cdc34. We demonstrate direct E2 specificity for p27 and also show that differential ubiquitin linkages can be dependent on E2 alone.

The Influence of Dielectric Constant on Ionic and Non-polar Interactions

  • This work is focused on analyzing ion-pair interactions and showing the effect of solvent induced inter-atomic attractions in various dielectric environments. To estimate the stability of ion-pairs, SCI-PCM ab initio MO calculations were carried out. We show that the solvent-induced attraction or ‘cavitation' energy of the ion-pair interactions in solution that arises mainly from the stabilization of the water molecules by the generation of an electrostatic field. In fact, even the strong electrostatic interaction characteristic of ion-pair interactions in the gas phase cannot overcome the destabilization or reorganization of the water molecules around solute cavities that arise from cancellation of the electrostatic field. The solvent environment, possibly supplemented by some specific solvent molecules, may help place the solute molecule in a cavity whose surroundings are characterized by an infinite polarizable dielectric medium. This behavior suggests that hydrophobic residues at a protein surface could easily contact the side chains of other nearby residues through the solvent environment, instead of by direct intra-molecular interactions.

양자화학적 계산에 의한 올리고펩티드 수화물의 구조분석 (Conformational Analyses for Hydrated Oligopeptides by Quantum Chemical Calculation)

  • 이성질체의 형태는 수용액 상태에서 종종 안정성과 반응성 등의 기본상태 뿐만 아니라 사슬성장 및 접힘 과정으로 인하여 형태형성에 영향을 주기 때문에 올리고펩티드의 형태를 이해하는 것이 중요하다. 본 논문에서는 L-알라닌(LA), 글리신(G) 5량체 모델의 무수 및 수화물(수화율; h/1) 상태의 구조와 에너지를 4가지 형태이성질체 (베타-확장형;= t-/t+, $PP_{II}$형; g-/t+, $PP_{II}$-유사형; g-/g+ 및 알파-나선형; g-/g-)에 대하여 B3LYP/6-31G(d,p)를 이용하여 양자화학계산(QCC) 방법으로 분석하였다. 구조최적화는 밀도함수 이론(DFT)으로써 B3LYP를 사용하였으며, 기본설정(Basic set)으로는 6-31G(d,p)를 이용하였다. 이미노 양성자(NH)를 갖는 LA와 G에서 베타-확장형, $PP_{II}$-유사형, 알파-나선형의 3가지 형태가 얻어졌으며, 대부분 물 분자가 $PP_{II}$-유사형과 알파-나선형에서는 CO-HN 분자 내 수소결합 사이에 주로 삽입되었고, 베타-확장형은 CO기에 부착되었다. 또한, LA와 G에서 $PP_{II}$-유사형 형태이성질체가 무수 및 수화물 상태에서 가장 안정적이었으며, $PP_{II}$ 형태이성질체는 얻어지지 않았다. LA에 대한 결과는 알라닌 올리고펩티드의 안정적인 형태가 주로 $PP_{II}$라고 보고한 다른 연구의 실험적 및 이론적인 결과와는 상이했다. 올리고펩티드 형태이성질체의 생성패턴과 안정성이 CO-HN의 분자 내 수소결합의 존재 여부 또는 출발 아미노산 내 $NH_2$기의 존재 여부에 강한 영향을 받는 것을 알 수 있었다.

Simultaneous Determination of Alkoxyalcohols in Wet Wipes Using Static Headspace Gas Chromatography and Mass Spectrometry

  • Alkoxyalcohols are used as solvents or preservatives in various consumer products such as wet wipes. The metabolites of alkoxyalcohols are known to be chronically toxic and carcinogenic to animals. Thus, an analytical method is needed to monitor alkoxyalcohols in wet wipes. The aim of this study was to develop a simultaneous analytical method for 14 alkoxyalcohols using headspace gas chromatography coupled with mass spectrometry to analyze the wet wipes. This method was developed by comparing with various headspace extraction parameters. The linear calibration curves were obtained for the method ($r^2$ > 0.995). The limit of detection of alkoxyalcohols ranged from 2 to $200ng\;mL^{-1}$. The precision of the determinative method was less than 18.20% coefficient of variation both intra and inter days. The accuracy of the method ranged from 82.86% to 119.83%. (2-Methoxymethylethoxy)propanol, 2-phenoxyethanol, and 1-phenoxy-2-propanol were mainly detected in wet wipes.

  • 종다양성 규명을 위한 계통분류학적 연구는 원시형질의 상동성에 근거한 기존의 형태형질과 효율적인 새로운 분자계통분류학적 연구방법이 병행된다면 좀 더 정확한 분류 및 계통을 추적하는데 객관적인 결론을 제시 할수 있을 것이다. 현재 활발히 연구되고 있는 미토콘드리아 DNA를 이용한 분자계통분류를 재검토할 뿐아니라 계통진화학상 조상과 자손과의 관계를 유추하는데 새로운 가설을 세울 수 있는 토대를 제공하는 기초자료로서 이용될 수 있다.

Molecular gas properties under ICM pressure : A Case study of NGC4402

  • We probe 12CO J=2-1 and 13CO J=1-0 properties of a Virgo disk galaxy, NGC 4402 which is located near the cluster center. Our goal is to study the impact of intra cluster medium (ICM) on the molecular gas of a galaxy in the cluster environment. It has been believed that cluster galaxies are deficient in atomic hydrogen gas (HI gas) compared to their field counterparts and now there is much evidence that low density ISM can be easily removed by ram pressure caused by ICM wind. Meanwhile, no significant molecular gas deficiency of the cluster galaxy population has been found yet they show overall lower star formation rate than galaxies in the field, and it is still controversy whether dense ISM can be also stripped by the ICM wind or not. NGC 4402 with truncated HI disk($D_{HI}/D_{opt}$ ~ 0.75 and only 36%of HI gas compare to field galaxies of a similar size) and a disturbed gas morphology, appears to have strong ongoing ram pressure. Using high resolution 12 and 13CO data of NGC 4402 from a Sub Millimeter Array (SMA), we probe the molecular gas properties under strong ICM pressure. We discuss how its star formation activity and hence the global color of NGC4402 would be changed in the future.

피조개, Scapharca broughtonii Schrenck RFLP 마커 개발 (Development of Molecular Detection Marks Using PCR-RFLP Technique for Arkshell (Scapharca broughtonii Schrenck))

  • 한국산과 중국산 피조개의 신속 진단, 유전적 특성 및 유연관계를 분석하기 위하여 DNA 수준에서 확인 할 수 있는 PCR-aided RFLP를 사용하였다. 피조개 mtDNA 165 rDNA gene를 제한효소로 처리하여 그 band 양상을 조사하고자 하였다. 165 rDNA gene을 분리하기 위하여 ArkF-3, ArkR-3 primer를 사용한 결과 한국산과 중국산 피조개 모두 분자량이 720 bp band가 나타났다 PCR에 의하여 증폭된 16S rRNA gene을 총 8종류의 제한효소(PvuII, BamHI, HinfI, HaeIII, EcoRI, RsaI, Ksp221, BstX21)로 절단하여 RFLP 양상을 보았다. HinfI의 제한효소 처리 시 득량, 가막, 남해, 진해, 태안산 피조개 모두 275 band절편이 관찰되었으나, 중국산은 나타나지 않았다. HinfI를 제외한 나머지 제한효소는 다형형이 관찰되지 않았고 한국산과 중국산 모두 700 bp의 동일한 band를 보였다. HaeIII에서도 득량, 가막, 태안과 남해와 진해산 PCR product가 700 bp위치에서 상이하게 보였다. 또한 한국산과 중국산 절편이 다르게 나타났다. 한국산 피조개 종내 restriction 쇼pe에 의해 유연관계의 결과에 의하면 득량, 가막, 태안은 동일한 유사도를 보였고, 남해와 진해와는 거리가 7 정도로 나타났다. 특히 한국산과 중국산 거리는 25로 나타났다. 이상의 결과를 보아서,HinfI 제한효소는 한국산과 중국산을 구별하는데 매우 유용한 molecular marker가 될 수 있을 것으로 판단되며, 유전적으로 거리가 먼 것으로 보인다. 또한 HaeIII은 한국산 종내 구분 및 중국산 신속 동정에 좋은 molecular tool로 사용될 것으로 예상된다.
