Inspections on the Food Safety of Pheasant and Mallard as a Meat Resource (식육자원(食肉資源)으로서의 꿩과 청둥오리 고기의 안전성(安全性) 검사(檢査))

  • Studies on the food-safety of pheasant and mallard, which belong to wild fowl as new meat resources. were carried out. The results were summarized as follows : 1. Food poisoning bacteria including Salmonella spp, was not detected from the inspections of small intestine, cecum, and rectum. 2. Parasite inspection tests on blood, feces, digestive organ, and thoracic organs were negative. 3. Antibiotic residues from the carcass muscle by simplified disk methods were not detected. 4. Seven different pesticide residue tests, such as DDT and BHT, on the muscle and liver were negative. 5. Four different kinds of toxic heavy metals such as Cd were much lower than the permissible concentration. Studies on the food safety tests and inspections from the pheasant and mallard were revealed that from taking this new food resources, the toxicities would be very low for the human health by the direct influences.

  • Gut microbiota is a group of microorganisms that resides in the intestine and serves many important functions in human health. Using 16S ribosomal RNA sequencing analysis, a wide variety of bacteria in human gastrointestinal tract has been identified along with intriguing findings that there is a different bacterial composition among individuals. Fecal microbiota transplantation (FMT) is a procedure of stool transplantation from healthy donors to patients suffering from various diseases. Specifically, FMT is able to alter the composition of gut microbiota of recipients and therefore could be an effective treatment for the patients with gastrointestinal diseases including recurrent Clostridium difficile infection, inflammatory bowel disease, and irritable bowel syndrome. Here we review a list of human diseases related to gut microbiota disturbance and the case studies of FMT. We also summarize medicines and diagnostic tools that are under development. Therefore, gut microbiota can be a next generation's biotherapy for promotion of health and treatment of chronic diseases.

Effect of Temperature on Mass Mortality of the Larval Toad (Bufo bufo gargarizans) in Mangwel Pond (기온변화가 망월지 두꺼비유생 집단폐사에 미치는 영향)

  • We have confirmed the first case of mass mortality which occurred in the wild populations of larval toad, Bufo bufo gargarizans during 2007~2009 at Mangwel pond, which is a long time breeding site, and is located at the south eastern area in Daegu. We have investigated through microbiological tests the cause of mass decease of the larval toad, and detected numerous bacteria from the intestine by histological examines and cultures. The pathogen isolated from the infected larval toads has been identified as Aeromonas hydrophila, which is widely distributed in aquatic environment. However, this phenomenon of mass lethal pattern of the larval toad disappeared at the breeding pond after maintaining better water levels and low temperature in spring during breeding season in 2010. We propose that the high temperatures during the 2007~2009 breeding season resulted in lower water levels and drier conditions leading to explosive propagation of A. hydrophila in the pond which impacted the larval toad's immune function.

A report on 53 unrecorded bacteria species in Korea in the class Gammaproteobacteria

  • During an investigation of unrecorded prokaryotic species in Republic of Korea, a total of 53 bacterial strains belonging to the class Gammaproteobacteria were isolated from soil, seawater, tidal flats, rhizosphere, salt ponds, beach sand, urine, manure, sediment, and animal intestine (Russian grayling butterfly [Hipparchia autonoe], mouse [Mus musculus], and sea bass [Lateolabrax japonicus]). Strains were identified to species using the 16S rRNA gene sequence, showing high similarity (>98.7%) with the closest bacterial species and forming a robust clade in the neighbor-joining phylogenetic tree. The 53 strains of Gammaproteobacteria in this study have not been report previously in Korea. Therefore, we describe 27 genera of 16 families in 7 orders: 13 strains in the order Alteromonadales, 1 strain in the order Chromatiales, 11 strains in the order Enterobacterales, 7 strains in the order Oceanospirillales, 10 strains in the order Pseudomonadales, 8 strains in the order Vibrionales, and 3 strains in the order Xanthomonadales. Gram reaction, strain ID, isolation source, and morphological and basic biochemical characteristics are described for each species.

Biotransformation of Intestinal Bacterial Metabolites of Ginseng Saponin to Biologically Active Fatty-acid Conjugates

  • Ginsenosides are metabolized (deglycosylated) by intestinal bacteria to active forms after oral administration. 20(S)-Protopanaxadiol $20-O-{\beta}-D-glucopyranoside$ (M1) and 20(S)-protopanaxatriol (M4) are the main intestinal bacterial metabolites (IBMs) of protopanaxadiol- and protopanaxatriol-type glycosides. M1 was selectively accumulated into the liver soon after its intravenous (i.v.) administration to mice, and mostly excreted as bile; however, some M1 was transformed to fatty acid ester (EMl) in the liver. EM1 was isolated from rats in a recovery dose of approximately $24mol\%.$ Structural analysis indicated that EM1 comprised a family of fatty acid mono-esters of M1. Because EM1 was not excreted as bile as Ml was, it was accumulated in the liver longer than M1. The in vitro cytotoxicity of M1 was attenuated by fatty acid esterification, implying that esterification is a detoxification reaction. However, esterified M1 (EM1) inhibited the growth of B16 melanoma more than Ml in vivo. The in vivo antitumor activity paralleled with the pharmacokinetic behavior. In the case of M4, orally administered M4 was absorbed from the small intestine into the mesenteric lymphatics followed by the rapid esterification of M4 with fatty acids and its spreading to other organs in the body and excretion as bile. The administration of M4 prior to tumor injection abrogated the enhanced lung metastasis in the mice pretreated with 2-chloroadenosine more effectively than in those pretreated with anti-asialo GMl. Both EM1 and EM4 did not directly affect tumor growth in vitro, whereas EM1 promoted tumor cell lysis by lymphocytes, particularly non-adherent splenocytes, and EM4 stimulated splenic NK cells to become cytotoxic to tumor cells. Thus, the esterification of IBM with fatty acids potentiated the antitumor activity of parental IBM through delay of the clearance and through immunostimulation. These results suggest that the fatty acid conjugates of IBMs may be the real active principles of ginsenosides in the body.

Effects of added chlorides and their concentrations on growth and nitrate content in leaf lettuce(Lactuca sativa L.) in hydroponics.

  • Nitrate in edible vegetables is converted to nitrite by nitrate reductase(NR) and/or bacteria in intestines. Nitrite and amino, in the intestine of some animals and human, bind to form nitrosamine, which is toxic and known as carcinogen. This study was carried out to examine the effect of added chlorides and their concentrations on growth, yield and nitrate content in leaf lettuce plants in hydroponics. Seeds of lettuce cv, "Samsunjokchukmyon" were planted on April 29, and seedlings were planted on June 2, and were cultured until July 5 in 1998. KCI and CaCl₂ were used as chloride source and their concentrations were 1, 2 and 4 me/L, respectively, in the lettuce standard nutrient solution for National Horticultural Research Institute(NHRI). Completely randomized design with 3 replications was used. Nitrate content and NR activity were measured 2 and 5 weeks after planting(WAP). The obtained results were summarized as the follows : Leaf weight per plant was difference from harvest dates and treatments, but total leaf weight was not significantly different among treatments. Number of leaves was higher in KCI 2 me/L, CaCl₂1 me/L and control at 2 WAP than the others, and was higher in KCI 1 me/L, and control at 3 WAP than others, and was higher in control at 5 WAP. Total number of harvested leaves was the highest in control with 14, which followed by KCI 2 me/L and CaCl₂1 me/L. Nitrate content was decreased by addition of chloride in nutrient solution. Nitrate content in the 3rd and 9th leaves was significantly decreased. NR activity was higher in control and CaCl₂ addition treatments, while KCI addition treatments reduced NR activity. However, no direct relationship with nitrate was observed. Growth characteristics such leaf length and leaf width were not significantly influenced by chloride addition.

Effects of soft pellet creep feed on pre-weaning and post-weaning performance and intestinal development in piglets

  • Objective: This study aimed to determine the effects of soft pellet creep feed (SPCF) on growth performance and intestinal development in piglets. Methods: A total of 18 sows and their litters of crossbred piglets (14±2 days, 3.73±0.72 kg) were assigned to one of three dietary groups receiving i) powder creep feed (PCF), ii) hard pellet creep feed (HPCF) or iii) SPCF during the pre-weaning period. After weaning, piglets were selected for continuous evaluation of the three diets on growth performance and intestinal health. Results: In the pre-weaning period, the average daily feed intake and average daily dry matter intake were significantly higher in the SPCF group than the HPCF group (p<0.05). In the post-weaning and entire experimental period, the different diets had no significant effect on growth performance. At 10 d after weaning, the serum glucose concentration was lower in the SPCF group (p<0.05) than the other groups; a higher (p<0.05) villus height and lower (p<0.05) crypt depth in the jejunum were also observed in the SPCF group than the other groups; Meanwhile, in the duodenum and jejunum, the SPCF group had a higher (p<0.05) villus height to crypt depth ratio than the other groups; Furthermore, the higher (p<0.05) threshold cycle values of lactic acid bacteria and lower (p<0.05) threshold cycle values of Clostridium, Enterobacter and Escherichia coli were also observed in the SPCF group, and the sucrase and maltase activity was higher (p<0.05) in the SPCF group than the other groups in duodenum and ileum. Conclusion: The SPCF improved pre-weaning feed intake and decreased the negative effects of weaning stress in the intestine in piglets.

Resistance Patterns of Frequently Applied Antimicrobials and Occurrence of Antibiotic Resistance Genes in Edwardsiella tarda Detected in Edwardsiellosis-Infected Tilapia Species of Fish Farms of Punjab in Pakistan

  • Edwardsiella tarda is one of the most significant fish pathogens, causes edwardsiellosis in a variety of freshwater fish species, and its antibiotic resistance against multiple drugs has made it a health risk worldwide. In this study, we aimed to investigate the antibiotic resistance (ABR) genes of E. tarda and establish its antibiotic susceptibility. Thus, 540 fish (299 Oreochromis niloticus, 138 O. mossambicus, and 103 O. aureus) were collected randomly from twelve fish farms in three districts of Punjab in Pakistan. E. tarda was recovered from 147 fish showing symptoms of exophthalmia, hemorrhages, skin depigmentation, ascites, and bacteria-filled nodules in enlarged liver and kidney. Antimicrobial susceptibility testing proved chloramphenicol, ciprofloxacin, and streptomycin effective, but amoxicillin, erythromycin, and flumequine ineffective in controlling edwardsiellosis. Maximum occurrence of qnrA, blaTEM, and sul3 genes of E. tarda was detected in 45% in the liver, 58%, and 42% respectively in the intestine; 46.5%, 67.2%, and 55.9% respectively in O. niloticus; 24%, 36%, and 23% respectively in summer with respect to fish organs, species, and season, respectively. Motility, H2S, indole, methyl red, and glucose tests gave positive results. Overall, E. tarda infected 27.2% of fish, which ultimately caused 7.69% mortality. The Chi-squared test of independence showed a significant difference in the occurrence of ABR genes of E. tarda with respect to sampling sites. In conclusion, the misuse of antibacterial agents has led to the emergence of ABR genes in E. tarda, which in association with high temperatures cause multiple abnormalities in infected fish and ultimately resulting in massive mortality.

Comparison of the Biochemical Activities of Commercial Yogurts and Lactobacillus acidophilus-containing Yogurt (시판용 요구르트와 Lactobacillus acidophilus 요구르트의 생화학적 활성의 비교)

  • Lactic acid-producing bacteria such as Lactobacillus spp. function to ferment carbohydrates and produce ATP. Such Lactobacillus spp. are used for the production of commercial yogurts. Lactobacillus spp. are beneficial to the intestinal tract, and Lactobacillus acidophilus-containing yogurts have received considerable attention because of their preventive effects against early-stage cancer of the large intestine. In this study, lactic acid-producing bacteria were cultured from three different groups: commercial solid yogurt (for eating), commercial liquid yogurt (for drinking), and Lactobacillus acidophilus-containing yogurt. We first determined the optimum culture conditions for Lactobacillus spp. and then analyzed turbidity and pH in order to compare the growth abilities and lactic acid-production capacities among the groups. Finally, high-performance liquid chromatography was used to measure the lactic acid content in the culture supernatants, and the antibacterial activities against Staphylococcus aureus and Escherichia coli were compared among the three groups. The optimum culture conditions for Lactobacillus spp. were MRS medium at $25^{\circ}C$, for 24 h. The highest turbidity was found in L. acidophilus-containing yogurt, followed by liquid yogurt and solid yogurt. Similarly, the highest lactic acid production ability was found in L. acidophilus-containing yogurt, followed by liquid yogurt and solid yogurt. Culture supernatants from the three groups did not show any antibacterial activity towards S. aureus; however, supernatants derived from L. acidophilus-containing yogurt resulted in a 1.8 mm inhibitory zone against E. coli in a paper disk diffusion test. These results revealed the high level of lactic acid-production capacity and antibacterial activity in L. acidophilus-containing yogurt.

Development of Probiotic Products and Challenges (프로바이오틱 제품 개발 동향과 과제)

  • Probiotics beneficially affect the health of the host via various mechanisms in the intestine. Recent developments in probiotic products have mainly been made to maximize probiotic effects in human. In this regard, probiotic products containing doubly coated or encapsulated cells, multi-species probiotics, or high viable cell number (1010 viable cells/gram or more) have been developed and are already available in the market. Until now, the majority of probiotics contain live cells but little attention has been paid to other alternative products such as heat-killed cell or bacteriocin-containing ones, which could have broad applications due to advantages over live cell-based probiotics, such as safety and stability. In addition, genetically engineered lactic acid bacteria could be of great importance in the field of alimentary health if they are carefully designed for biological safety. Although a number of probiotics are marketed by claiming health benefits, regulations for health claims will be more stringent. Therefore sufficient scientific and clinical evidences supporting the safety and efficacy of the potential probiotic strain will be required by the regulatory authority for a health claim, which thus may have a huge impact on the future probiotic market.
