Effects of Saenggantanggami-bang on Nonalcoholic Fatty Liver Disease Model Induced by Fatty Acids (지방산으로 유발된 비알코올지방간질환 실험모델에 미치는 생간탕가미방(生肝湯加味方)의 효과)

  • Objectives : This study was designed to investigate the effects of Saenggantanggami-bang (SG) on nonalcoholic fatty liver disease. Methods : HepG2 cells were used in an in vitro model. HepG2 cells were divided into three groups. The Normal group was incubated with no fatty acid. The Control group was incubated with 1mM palmitic acid to introduce fat overloading. The PA-SG group was incubated with 1mM palmitic acid and various concentrations of Saenggantanggami-bang (SG). Cell viability and cytotoxicity were analyzed by MTT assay and LDH assay. Intracellular triglyceride (TG) levels, reactive oxygen species (ROS) levels, ATP amount, and GST activity were measured. Cell death pattern and protective effect of SG on cell death were studied by DNA fragmentation and caspase-3 intensity (western blot). Results : Compared with the Control group, cell viability of the PA-SG group significantly increased (P<0.01), cytotoxicity of the PA-SG group decreased (P<0.01), and intracellular TG levels and ROS levels of the PA-SG group decreased (P<0.05). In DNA fragmentation assay, necrotic pattern was observed and DNA fragment decreased in the PA-SG group. In western blot, apoptotic pattern was observed, caspase-3 intensity of the PA-SG group was reduced significantly, but there were no significant differences in intracellular ATP amount and GST activity between the control group and the PA-SG group. Conclusion : The results suggest that Saenggantanggami-bang can be a potential candidate for the clinical treatment of nonalcoholic fatty liver disease.

Anti-inflammatory Effect of Jungcheonhwadamgangki-tang in Formalin-Injection Mouse Paw Chronic Inflammation Model (정천화담강기탕(定喘化痰降氣湯)이 formalin으로 유발된 마우스의 만성 염증에 미치는 영향)

  • In the present study, the anti-inflammatory effect of Jungcheonhwadamgangki-tang(定喘化痰降氣湯) water extract was tested in formalin-injection mouse hind paw chronic inflammation model. The test articles were dosed once a day for 10 days, and changes on the body weight, paw weights were observed with histopathology of induced paw dorsum pedis. In addition, histomorphometry was also monitored at sacrifice. The increases of absolute and relative hind paw weight detected in vehicle control compared to that of sham, were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang in the present study. A classic acute inflammatory histological changes such as subcutaneous edema, hypertrophy and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang. In addition, the increases of hind paw weight detected in the vehicle control, were also dose-dependently decreased in the all Jungcheonhwadamgangki-tang-dosing groups. Base on these aforementioned results, it is concluded that Jungcheonhwadamgangki-tang have clear anti-inflammatory effect on the chronic inflammation induced by formalin-injection.

The Effects of Forsythiae Frucus on Inflammatory Genes and Cyto-pathological Alterations in Chronic Non-Bacterial Prostatitis Rat Model (연교(連翹)가 만성 비세균성 전립선영 Rat의 염증발현인자 및 세포조직 변화에 미치는 영향)

  • Objective : The etiology of chronic prostatitis is likely multifactorial, resulting from either a cascade of events after an initiating factor or from a variety of etiologic mechanisms. There is substantiating evidence to support the role of the inflammatory responses in its pathogenesis, and the clinical value in the evaluation of therapeutic efficacy. Forsythiae Frucus has been traditionally used in treatment of inflammatory diseases, including of prostatitis and urinary tract inflammation. In this study, we investigated the effects of Forsythiae Frucus on inflammatory cytokines and cyto-pathological alternation in the rat model of chronic non-bacterial prostatitis induced by castration and $17{\beta}$-estradiol treatment. Methods : Two-month-old rats were treated with $17{\beta}$-estradiol after castration for induction of experimental non-bacterial prostatitis. which is similar to human chronic prostatitis in histopathological profiles. Forsythiae Frucus as an experimental specimen, and testosterone as a positive control, were administered orally. The prostates were evaluated by histopathologlcal parameters including the epithelial score and epithelio-stromal ratio for glandular damage. and the expression of inflammatory cytokine genes including interleukin (IL)-$1{\beta}$, IL-5, IL-12, tumor necrosis factor (TNF)-$\alpha$. eotaxin, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2(cox-2). Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation. the rats treated with Forsythiae Frucus showed a diminished range of tissue damage. Epithelial score was improved in the Forsythiae Frucus group over that of the control (P<0.05). The epithelia-stromal ratio was lower in the Forsythiae Frucus group when compared to that of the control (P<0.05). In the reverse transcription-polymerase chain reaction (RT-PCR) of inflammatory cytosine genes. Forsythiae Frucus inhibited the expression of IL-$1{\beta}$, TNF-$\alpha$, iNOS, cox-2 genes, while it modulated the expression of IL-5, which is an anti-inflammatory cytokine. Conclusions : These findings suggest that Forsythiae Frucus may protect the glandular epithelial cells and also inhibit stromal proliferation in association with the immune modulation including the suppression of inflammatory cytokines and increase of anti-inflammatory cytokines. From theses results. we suggest that Forsythiae Frucus could be a useful remedy agents for treating chronic non-bacterial prostatitis.

The Effects of Lygodium japonicum Treatment on Hematological and Cyto-pathological Alterations in Non-Bacterial Prostatitis Rat Model (해금사가 만성 비세균성 전립선엽 모델에서 혈액학적 및 세포조직학적 변화에 미치는 영향)

  • Objective : Although chronic non-bacterial prostatitis is a common disease, it is very difficult to treat effectively. Lygodium japonicum has been traditionally used in treatment of urinary tract inflammation and voiding disturbance. In this study, we investigated the therapeutic effects and action mechanism of Lygodium japonicum in the rat model of non-bacterial prostatitis induced by castration and testosterone treatment. Methods : Five-month-old rats were treated with 17$\beta$-estradiol after castration for induction of experimental non-bacterial prostatitis, which is similar to human chronic prostatitis in histopathological profiles. Lygodium japonicum and testosterone were administered as an experimental specimen and a positive control. respectively. The prostates were evaluated by histopathological parameters including the epithelial score and epithelio-stromal ratio for glandular damage. PCNA labeling index for cyto-proliferation and a TUNEL(deoxyuridine triphosphate biotin nick end-labeling) assay for cell apoptosis. Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation, the rats treated with Lygodium japonicum showed a diminished range of tissue damage. Epithelial score was improved in the Lygodium japonicum group over that of the control (P<0.05). The epithelio-stromal ratio was lower in the Lygodiutn japonicum group when compared to that of the control (P<0.05). Although there was no difference in PCNA and TUNEL positive cells of the glandular epithelia. we found an decreased number of PCNA positive cell and concurrent increase of TUNEL positive cells in the stroma of Lygodium japonicum treated rats (P<0.01). Conclusions : These findings suggest that Lygodium japonicum may protect the glandular epithelial cells and also inhibit stromal proliferation in association with suppression of cyto-proliferation and stimulation of apoptosis. We concluded that Lygodium japonicum could be a useful remedy agents for treating chronic non-bacterial prostatitis.

  • This article reviews the art and practice of strategic management process in hospitals today, in order to help hospital administrators for managing strategic management system in their hospitals. The strategic management process model in this article is based on an integrated approach combining traditional environmental model with resource-based model of strategy. The components of the model are consisted of five steps: (1)formulating objectives, strategic assessment by external environmental analysis, internal capability analysis, TOWS analysis and marketing audit, (3)strategy choice considering context and criteria of choice, (4)program implementation through operational planning, resource allocation, and conversion, and (5)control by monitoring and evaluating hospital outputs. This article deals with many aspects of issues inherent in every step on this strategic management process.

  • In this paper, two neural networks as a material model, which are based on the state-space method, have been proposed. One outputs the rates of inelastic strain and material internal variables whereas the outputs of the other are the next state of the inelastic strain and material internal variables. Both the neural networks were trained using input-output data generated from Chaboche's model and successfully converged. The former neural network could reproduce the original stress-strain curve. The neural network also demonstrated its ability of interpolation by generating untrained curve. It was also found that the neural network can extrapolate in close proximity to the training data.

Occupational Lifting Tasks and Retinal Detachment in Non-Myopics and Myopics: Extended Analysis of a Case-Control Study

  • Objectives: Lifting heavy weights involves the Valsalva manoeuvre, which leads to intraocular pressure spikes. We used data from a case-control study to further investigate the hypothesis that occupational lifting is a risk factor for retinal detachment. Methods: The study population included 48 cases (patients operated for retinal detachment) and 84 controls (outpatients attending an eye clinic). The odds ratios (OR) of idiopathic retinal detachment were estimated with a logistic regression model (adjusted for age, sex and body mass index). Three indexes were used to examine exposure to lifting; 1) maximum load lifted, 2) average weekly lifting, 3) lifelong cumulative lifting. Results: For all indexes, the most exposed subjects showed an increased risk of retinal detachment compared with the unexposed (index 1: OR 3.57, 95% confidence interval [CI] 1.21-10.48; index 2: OR 3.24, 95% CI 1.32-7.97; index 3: OR 2.23, 95% CI 1.27-8.74) and dose-response relationships were apparent. Conclusion: These results reinforce the hypothesis that heavy occupational lifting may be a relevant risk factor for retinal detachment.

  • In this paper, we formulate the mathematical model for a single-stand rolling mill and design control systems for the thickness control at the exit of roll stand and for the tension control of the strip in the process. We propose a thickness controller based on the Internal Model Control structure which can be an effective application when the frequency components of the thickness deviation of the entry strip are known and, show how it can be appropriately combined with BISRA AGC method for a precise thickness control while maintaining the robustness against the modeling error of the mill modulus. It is illustrated by simulations that the proposed thickness control method gives better performance than existing methods and has the robustness against the modeling error of the mill modulus as well.

  • In this paper, an improved predictive functional control (PFC) scheme for permanent magnet synchronous motor (PMSM) control system is proposed, on account of the standard PFC method cannot provides a satisfying disturbance rejection performance in the case of strong disturbances. The PFC-based method is first introduced in the control design of speed loop, since the good tracking and robustness properties of the PFC heavily depend on the accuracy of the internal model of the plant. However, in orthodox design of prediction model based control method, disturbances are not considered in the prediction model as well as the control design. A minimum-order observer (MOO) is introduced to estimate the disturbances, which structure is simple and can be realized at a low computational load. This paper adopted the MOO to observe the load torque, and the observations are then fed back into PFC model to rebuild it when considering the influence of perturbation. Therefore, an improved PFC strategy with torque compensation, called the PFC+MOO method, is presented. The validity of the proposed method was tested via simulation and experiments. Excellent results were obtained with respect to the speed trajectory tracking, stability, and disturbance rejection.

Adaptive Inverse Feedback Control of Periodic Noise for Systems with Nonminimum Phase Cancellation Path (비최소위상 상쇄계를 가진 시스템을 위한 주기소음의 적응 역 궤환 제어)

  • An alternative inverse feedback structure for adaptive active control of periodic noise is introduced for systems with nonminimum phase cancellation path. To obtain the inverse model of the nonminimum phase cancellation path, the cancellation path model can be factorized into a minimum phase term and a maximum phase term. The maximum phase term containing unstable zeros makes the inverse model unstable. To avoid the instability, we alter the inverse model of the maximum phase system into an anti-causal FIR one. An LMS predictor estimates the future samples of the noise, which are necessary for causality of both anti-causal FIR approximation for the stable inverse of the maximum phase system and time-delay existing in the cancellation path. The proposed method has a faster convergence behavior and a better transient response than the conventional filtered-x LMS algorithms with the same internal model control structure since a filtered reference signal is not required. We compare the proposed methods with the conventional methods through simulation studies.
