생쥐 난자와 착상전 초기배아에서 IGF-1과 IGF-1 수용체 유전자 발현 (Expression of IGF-1 and Its Receptor Genes in the Oocytes and Preimplantation Embryos in Mouse)

  • 인슐린 유사 성장 호르몬 1과 2 (IGF-1 & IGF-2)는 착상 전 초기배아 발생을 조절하는 중요한 요소이다. 생쥐 착상 전 초기배아에서 IGF-1의 역할에 관한 연구를 위해, IGF-1과 IGF-1 수용체의 전사물의 존재 여부를 난자와 착상 전 초기배아에서 조사하였다. 새로이 고안된 IGF-1 primer를 이용하여 난자에서 전사물을 검출하였다. 그리고, PCR 산물을 제한효소인 Msp I으로 절단하여 확인하였다. 이 실험에서 IGF-1과 IGF-1 수용체의 전사물이 난자와 착상 전 초기배아에서 모두 검출됨을 보였다. GV-난자에 다량 존재하는 mRNA는 4- 혹은 8-세포기까지 지속적으로 감소하다가 이후에 다시 증가하는 양상을 보였다. GV-난자에서 IGF-1과 IGF-lR 전사물이 존재한다는 것은 초기배아에 존재하는 전사물이 모계유래 산물임을 암시한다. 또한, 난자와 착상 전 초기배아에 IGF-1과 IGF-1 수용체 전사물이 존재하는 것으로 보아 착상 전 초기배아에서 IGF-1은 자가 분비되어 IGF-1 수용체의 신호전달 경로를 통하여 배아발생에 작용하는 것으로 사료된다.

인슐린 비의존형 당뇨병 환자의 혈청 중 Insulin-Like Growth Factor-Binding Proteins(IGFBPs)의 분포 및 IGFBP-3의 분해 (Distribution of Insulin-Like Growth Factor-Binding Proteins(IGFBPs) and IGFBP-3 Proteolysis in Noninsulin-Dependent Diabetes Mellitus Serum)

  • 이상의 결과를 요약하연, 혈당값은 정상대조군 78{\pm}6.20mg/dl, 인슐린 비의존형 당뇨병 환자 $184{\pm}18.94mg/dl$로 인슐린 비의존형 당뇨병 환자의 혈당값이 유의적으로 높았다. IGF-I의 수준은 정상대조군 $9.32{\pm}1.48{\mu}g/ml$, 인슐린 비의존형 당뇨병 환자 $4.19{\pm}0.84{\mu}g/ml$로 인슐린 비의존형 당뇨병 환자의 IGF-I 수준이 유의적으로 낮았다. 인슐린 비의존형 당뇨병 환자의 혈청 중 IGFBP-1, -2는 증가한 반면, IGFBP-3, -4, -5는 감소하였다. IGFBP-3의 분해에 관여하는 효소는 PMSF, aprotinin, EDTA에 의해 저해되어 metal-dependent serine proteases임을 확인하였고, 효소의 크기가 대략 97,66kDa 이었으며 효소활성이 인슐린 비의존형 당뇨병 환자에게서 큼을 확인하였다.

Rat 의 포유조절이 혈중 Insulin-like Growth Factor- I 수준에 미치는 영향 (Effect of Suckling on Serum Insulin-like Growth Factor- I Levels in the Primiparous Rat)

  • 본 연구는 Wistar계 rat를 이용하여 포유기간 중 제한포유시기와 이유시기를 조절하여, 혈중 IGF- I 수준변화에 대해 검토하였다. NL군 (NL)은 포유자의 수를 8 마리로 조절하였으며, RL군 (RL)과 W군 (W)은 RL0, RL0, RL10, RLl5 및 RL20, 그리고 W0, W5, WI0, W15 및 W20으로 각각 5개 군으로 구분하여, RL군은 각 개시일에 포유자의 수를 8마리에서 4마리로 조절하였으며, W군은 각 개시일에 포유자의 수를 완전히 이유시켜 다음과 같은 결과를 얻었다. 1. NL군의 혈중 IGF-I 농도는 분만직후 (Day 0) 750.59$\pm$3.52ng/$m\ell$ 을 나타내어 15일까지 일정한 수준을 유지하다가 그 후 포유 25일에 1,690.20$\pm$4.42ng/$m\ell$ 까지 증가되었다. 2. 포유초기 RL군 (RL0, RL5)의 IGF- I 농도는 포유 10일에 RL0군과 RL5군에서 각각 1,395.90$\pm$3.45 및 1,351.73$\pm$3.23ng/$m\ell$로서 NL 군의 745.96$\pm$2.24ng/$m\ell$ 보다 유의적 (P<0.05)으로 높게 나타났으나 포유 15일부터는 차이가 없었다. 3. 포유초기 W군의 IGF- I 농도는 이유 후 일주일 정도는 NL군에 비해 높은 수준 (P<0.05)을 나타내다가 10일경에 NL군과 같은 수준으로 감소하였다. 포유자극의 강약은 난소의 생리적 변화와 함께 혈중 IGF- I의 농도를 조절하는 것으로 사료된다.

Sleep Quality and Attention May Correlate With Hand Grip Strength: FARM Study

  • Objective To determine the socio-demographic, psychologic, hematologic, or other relevant factors associated with hand grip strength in Korean farmers. Methods A total of 528 healthy Korean farmers were enrolled. Hand grip strength was measured in both hands using a hydraulic dynamometer. Socio-demographic characteristics were assessed and anthropometric measurements were obtained. Psycho-cognitive measurements such as sleep quality (Pittsburgh Sleep Quality Index) and Go/No-Go test response time were conducted. In addition to physical measurements, serologic parameters including insulin-like growth factor 1 were measured. The factors associated with hand grip strength were analyzed using multiple linear regression analysis after adjusting for age, height, and weight. Results The mean hand grip strength was associated with the Pittsburgh Sleep Quality Index total score (${\beta}=-0.12$, p=0.01), the Go/No-Go test response time (${\beta}=-0.18$, p=0.001), vitamin D (${\beta}=0.12$, p=0.02), and insulin-like growth factor 1 levels (${\beta}=0.1$, p=0.045). In female farmers, hand grip strength was only associated with the Pittsburgh Sleep Quality Index total score (${\beta}=-0.32$, p<0.001). Conclusion The results of this study demonstrate that hand grip strength was associated with sleep quality and attention in Korean farmers.

Ethanol extract of Sinsun-yukza-hwan, a Korean medicinal prescription, promotes hair growth in C57BL/6 mice, an alopecia animal model

  • Objectives : In Korean medicine, a prescription of Sinsun-yukza-hwan (Shenxian-liuzi-wan, SSY) has been used in clinic for treatment of alopecia via oral. This study was performed to determine transdermal effects of the ethanol extract from SSY on hair growth and -related gene expressions in mice. Methods : We analyzed index compound, 5-hydroxy-methyl-2-furaldehyde (HMF), in SSY extract by ultra performance liquid chromatography (UPLC). 6 weeks old C57BL/6 mice with removed hair were used as an alopecia animal model. Mice were divided into 3 experimental groups including normal (3 water: 1 ethanol: 2 polyethylene glycol mixture as a vehicle), SSY extract and 5% minoxidil (as a positive control), treated groups. SSY was applied topically on the hair-shaved skin of C57BL/6 mice every day for 15 days. The color, thickness and density of hair were monitored every $5^{th}$ day by naked eye, photograph and phototrichogram using folliscope. Also hair growth-associated gene expressions were measured by immunoblotting assay. Results : Hair density of minoxidil or SSY-treated group was significantly increased compared to that of vehicle application on the $15^{th}$ day, respectively. And hair thickness of minoxidil and SSY groups was increased compared to that of vehicle treated group on the $15^{th}$ day, respectively. Induction of insulin-like-growth factor 1(IGF-1) and vascular endothelial growth factor (VEGF) were also significantly accelerated by SSY extract compared to those of vehicle-applied group. Conclusions : These results provide scientific evidence to support the potent multi-application of SSY as a cosmeceutical material for promoting hair growth.

Effects of Epidermal Growth Factor and Insulin-like Growth Factor-I on Placental Amino Acids Transport Activities in Rats

  • Epidermal growth factor (EGF) and insulin-like growth factor-I (IGF-I) have been shown to stimulate proliferation and differentiation of various somatic cells, including placental trophoblasts and also to enhance fetal growth and development when maternally administered. Since an increase of the expression of placental EGF and IGF-I receptors in rat, mouse, and human with the gestation advanced, both EGF and IGF-I were considered to play pivotal roles on fetal growth by regulating some function of placental cells. Amino acids are crucial importance for both maternal and fetal requirements of energy source and essential constituent of fetal mass during pregnancy. Impaired fetal and placental uptake of amino acids has been observed in several models of growth retardation in the rat. Amino acid is concentrated in the fetal side through active transport by amino acid transporters and is one of the important metabolic fuels for the fatal growth. Therefore, at first plasma amino acid concentrations in mothers and fetuses were measured as an index of uphill transport across the placenta associated with EGF and IGF-1. The EGF administration at the concentration of 0, 0.1, or 0.2 $\mu\textrm{g}$/g to pregnant rats from day 18 to 21 of gestation apparently increased fetal/maternal ratio of serum proline concentration and also fatal growth in EGF dose-dependent manner. When IGF-I in doses of 0, 1, 2, and 4 $\mu\textrm{g}$/g were administrated, the ratio of leucine, isoleucine, tryptophan, phenylalanine, tyrosine and also fetal growth significantly increased with a dose-dependent manner. These results suggested that EGF and IGF-I enhanced fatal growth by, as one of its possible mechanisms, promoting placental activity to transfer some amino acid supplies from the mother to the fetus in late pregnancy.

정상 어린이에서 혈청 인슐린양 성장인자-I과 인슐린양 성장인자 결합단백-2 및 -3의 농도 분석 (Evaluation of Serum Insulin-Like Growth Factor(IGF)-I, Insulin-Like Growth Factor Binding Protein(IGFBP)-2 and IGFBP-3 Levels in Healthy Korean Children)

  • 목 적 : 혈청 IGF-I, IGFBP-2, IGFBP-3 농도 및 IGF-I/IGFBP-3 molar ratio는 소아의 성장 발달, 당뇨병, 비만 및 악성종양의 진단, 예후 예측과 치료후 추적에 유용한 지표이지만 성별, 연령별, 인종별 변이가 있다. 저자들은 우리나라 어린이의 성별 및 연령군별 혈청 IGF-I, IGFBP-2, IGFBP-3 농도 및 IGF-I/IGFBP-3 molar ratio의 정상 표준치 확립을 위하여 본 연구를 시행하였다. 방 법 : 총 92명(남자 42명, 여자 50명)의 건강한 어린이를 대상으로 하였고, 신생아군 9명(남자 3명, 여자 6명), 영아군 19명(남자 9명, 여자 10명), 유아군 24명(남자 13명, 여자 11명), 학령군 30명(남자 12명, 여자 18명), 청소년군 10명(남자 5명, 여자 5명)으로 분류 후, ELISA 방법을 이용하여 혈청 IGF-I, IGFBP-2 및 IGFBP-3 농도를 측정하였고, 각 군간의 IGF-I, IGFBP-2, IGFBP-3 및 IGF-I/IGFBP-3 molar ratio를 비교분석하였다. 결 과 : 1) 남자에서는 IGF-I과 IGFBP-3는 출생과 더불어 증가하여 각각 유아군(P<0.05), 학령군(P<0.05)에 최고값에 도달한 후 청소년군으로 갈수록 감소하였다. IGFBP-2와 IGF-I/IGFBP-3 molar ratio는 각각 학령군과 영아군에 최고값을 보이고 감소하였으나 출생시에 비해 통계적으로 유의한 증가를 보이지 않았다. 각 군간의 비교에서 IGFBP-2만이 유의한 차이를 보였다(P<0.05). 2) 여자에서는 IGF-I과 IGFBP-2, IGFBP-3는 출생과 더불어 증가하여 각각 청소년군, 학령군(P<0.05), 학령군(P<0.05)에 최고값에 도달하였으나 IGF-I은 출생시에 비해 통계적으로 유의한 증가를 보이지 않았다. IGF-I/IGFBP-3 molar ratio는 출생시에 최고값을 보였으나 통계적으로 유의한 차이를 보이지 않았다. 각 군간의 비교에서 IGFBP-2 및 IGFBP-3가 유의한 차이를 보였다(P<0.05). 3) 동일연령군별 남녀 비교에서 IGF-I과 IGFBP-2, IGFBP-3 및 IGF-I/IGFBP-3 molar ratio 어느 것도 남녀 간에 통계적으로 유의한 차이를 보이지 않았다. 4) 상관관계 조사시 IGF-I의 농도가 높을수록 IGFBP-3의 농도 역시 의미있게 증가하였고(r=0.504, P=0.000), IGF-I/IGFBP-3 molar ratio와 비만도와의 상관 관계를 조사해 본 결과 통계적으로 의미있는 음의 상관 관계를 보이지 않았다(r=0.051, P=0.629). 또한 비만도와 IGF-I, IGFBP-2 및 IGFBP-3 사이에 통계적으로 유의한 상관 관계는 없었다. 결 론 : IGF-I과 IGFBP-3는 사춘기 진행에 따라 증가하며, 본 연구에서 IGFBP-3가 학령군에서 최고 수준을 보이는 것은 사춘기가 일찍 오는 요즘 어린이들의 성장 추세를 반영하는 것으로 추정되어진다. IGFBP-2는 신생아군에서 가장 높고 영아군에서 감소하는 데 이것은 IGFBP-2가 출생 전 발달 과정에 중요한 물질임을 뒷받침해주는 소견이다. 향후 더 많은 정상아를 대상으로한 연구를 통하여 보다 정확한 우리나라 어린이의 성별 연령군별 정상 표준치 확립이 필요하겠다.

Role of the insulin-like growth factor system in gonad sexual maturation in Pacific oyster Crassostrea gigas

  • Background: The IGF system plays important roles in controlling growth, development, reproduction, and aging of organisms. Methods: To estimate maturation of the Pacific oyster Crassostrea gigas, we investigated the expression of insulin-like growth factor (IGF) system components and sex-specific genes. To determine the role of the IGF system in the growth and spawning period of female and male oysters, we examined mRNA expression levels of the C. gigas insulin receptor-related receptor (CIR), IGF binding protein complex acid labile subunit (IGFBP_ALS), and molluscan insulin-related peptide (MIP), as well as those of vitellogenin (Vg) and receptor-type guanylate cyclase (Gyc76C) in gonads of C. gigas collected between April and October, when sex can be determined visually in this species. Results: We found that MIP, IGFBP_ALS, and CIR mRNA expression levels were dependent on sex and month and were greater in males than in females. CIR and Vg mRNA expression levels were very similar among females, whereas IGF system components and Gyc76C were very similarly expressed among males. The highest expression values were observed in May, when oysters are mature; CIR and Vg mRNA expression levels were highest in females, and those of MIP, IGFBP_ALS, CIR, and Gyc76C were highest in males. Interestingly, we observed a 1:1 proportion of females to males during this period. Conclusion: Our results suggest that IGF system components, as well as Vg and Gyc76C, are associated with sexual maturation in C. gigas.

Molecular Cloning of Insulin-like Growth Factor-I (IGF-I) and IGF-II Genes of Marine Medaka (Oryzias dancena) and Their Expression in Response to Abrupt Transfer from Freshwater to Seawater

  • Growth hormone (GH) is known as one of the main osmoregulators in euryhaline teleosts during seawater (SW) adaptation. Many of the physiological actions of GH are mediated through insulin-like growth factor-I (IGF-I), and the GH/IGF-I axis is associated with osmoregulation of fish during SW acclimation. However, little information is available on the response of fish IGF-II to hyperosmotic stress. Here we present the first cloned IGF-I and IGF-II cDNAs of marine medaka, Oryzias dancena, and an analysis of the molecular characteristics of the genes. The marine medaka IGF-I cDNA is 1,340 bp long with a 257-bp 5' untranslated region (UTR), a 528 bp 3' UTR, and a 555-bp open reading frame (ORF) encoding a propeptide of 184 amino acid (aa) residues. The full-length marine medaka IGF-II cDNA consists of a 639 bp ORF encoding 212 aa, a 109 bp 5' UTR, and a 416 bp 3' UTR. Homology comparison of the deduced aa sequences with other IGF-Is and IGF-IIs showed that these genes in marine medaka shared high structural homology with orthologs from other teleost as well as mammalian species, suggesting high conservation of IGFs throughout vertebrates. The IGF-I mRNA level increased following transfer of marine medaka from freshwater (FW) to SW, and the expression level was higher than that of the control group, which was maintained in FW. This significantly elevated IGF-I level was maintained throughout the experiment (14 days), suggesting that in marine medaka, IGF-I is deeply involved in the adaptation to abrupt salinity change. In contrast to IGF-I, the increased level of marine medaka IGF-II mRNA was only maintained for a short period, and quickly returned a level similar to that of the control group, suggesting that marine medaka IGF-II might be a gene that responds to acute stress or one that produces a supplemental protein to assist with the osmoregulatory function of IGF-I during an early phase of salinity change.

Expression Characteristics of Proteins of the Insulin-like Growth Factor Axis in Non-small Cell Lung Cancer Patients with Preexisting Type 2 Diabetes Mellitus

  • Background: Preexisting type 2 diabetes mellitus (T2DM) affects the prognosis and mortality of patients with some cancers. Insulin like growth factor (IGF) and insulin receptor (IR) signaling axes play important roles in both cancer and diabetes development. We aimed to explore the expression characteristics of proteins in IGF/IR axis in non-small cell lung cancer (NSCLC) cases with preexisting T2DM. Methods: Fifty-five NSCLC patients with preexisting T2DM were retrospectively included and matched by 55 NSCLC without diabetes at a 1:1 ratio. The expression of proteins in IGF/IR axis was detected by immunohistochemical staining. Clinicopathological data were collected to analyze their relationship with the protein expression. Results: Both IGF 1 receptor (IGF-1R) and insulin receptor substrate 2 (IRS-2) showed higher expression in the NSCLC with T2DM group, compared with those without T2DM. The high expression of IGF-1R and IRS-2 were found to be negatively associated with lymph node metastases and T staging in the T2DM group, respectively, and IRS-2 expression was also found more in the subgroup whose T2DM duration was more than 4 years. No difference was detected in the expression of IRS-1, IGF-1, IGF-2, IGFBP3, IR and mTOR between groups with or without T2DM. Conclusion: Our study found higher expression of IGF-1R and IRS-2 proteins in NSCLC patients with preexisting T2DM, and that there was an association with early stage NSCLC, which suggested that IGF signaling may play an important early event in development of NSCLC associated with diabetes.
