Purification and Sidedness of Sarcolemma from Canine Ventricle (개 심실 형질막의 분리 및 그 방향성에 관한 연구)

  • Sarcolemmal membrane fraction from canine ventricle was isolated from the discarded pellet after the first homogenization in the isolation procedure of sarcoplasmic reticulum (Method 1) and the protein yield, purity, and sidedness of this preparation were compared to those of sarcolemmal fraction prepared by method of Lee et al. (Method 2) and a slight modification of original protocol of Jones et al. (Method 3). Method 1 differed from Method 2 essentially only in that vigorous homogenization was carried out by omnimixer and homogenization medium containing 30mM Tris-maleate was used in the first step. The sarcolemmal fraction was enriched from 45 to 50 and 29-fold in [$^3H$] ouabain, [$^3H$] DHA, [$^3H$] QNB binding and $Na^+$, $K^+$-ATPase activity, respectively, compared to homogenate. Total $Na^+$, $K^+$-ATPase activity of highly sarcolemma enriched fraction was 144.6$\pm$16.4$\mu\textrm{mol}$ Pi/mg protein/hr, which was about 85%, of total ATPase activity, and the yield of the preparation was 15.7 mg protein per 100g of starting ventricular tissue. The sarcolemmal preparation supported $^{45}Ca^{2+}$-uptake in the presence of ATP but this uptake was not dependent on oxalate. Sarcolemmal $Na^+$, $K^+$-ATPase activity and detectable [$^3H$] ouabain binding were increased about 32% and 35%, respectively, by pretreatment of sarcolemmal fraction with optimal concentration of sodium dodecylsulfate (0.3-0.4mg/mg protein), suggesting that this preparation contained about 24% of sealed rightside-out vesicles, 26% of sealed inside-out vesicles, and 5001o of freely permeable (leaky) form. This procedure showed the highest protein yield and leaky population, compared to Method 2 and 3. On the other hand, sarcolemmal fraction prepared by Method 2 and 3 showed low value in protein yield but comtained high population of inside-out (46%) and rightside-out (49%) vesicles, respectively, compared to present procedure (Method 1). The results indicate that vigorous homogenization decreases the population of sealed sarcolemmal vesicles but increases the sarcolemmal protein yield per gram tissue and that this procedure is available for further purification of sarcolemmal fraction and for the receptor binding study of sarcolemma.

A Study on the Reduction of Entry Loss by Inner Structure in Square Hood in Industrial Ventilation System (산업환기시설에서 사각형 후드의 내부 설치에 의한 유입손실 감소에 관한 연구)

  • An objective of local exhaust hood design is to design the hood to operate as efficiently as possible. The greatest loss normally occurs at the entrance to the duct, due to the vena contracta in the throat of the duct. This can be accomplished by minimizing the loss that results from the vena contracta. There have been little studied to be cost-effective approach as installing simple instrument inside the throat of the hood. The aims of this paper were to minimize entry loss using inner square, and to measure the effect of inner square when installed inside hood throat. The results of this study were as follows; First, the magnitude of vena contracta could be considered as the difference between direct measured velocity and calculated velocity, which is from Bernoulli theory. In circle hood, calculated velocity and direct measured velocity were 10.7m/sec and 10.3n/sec, respectively. And the calculated velocity and direct measured velocity in square hood were 7.7m./sec and 6.5m/sec, respectively. Second, effect of inner square by width was carried out. The widths of inner square were L/1(18cm), L/2(9cm), L/3(6cm) and L/6(3cm). In case inner square was installed with 3cm width, the entry of coefficient was 0.93, comparing with 0.85 of entry of coefficient of general hood. Third, effect of inner square by distance from hood inside surface to inner square was carried out. The distances were L/3(6cm), L/6(3cm), L/9(2cm) and L/l8(1cm). In case the distance was 3cm the best efficiency was shown (Ce= 0.93). Fourth, effect of inner square by location from hood entry to duct inside was carried out. The locations of inner square were entry(0cm), L/6(3cm), L/3(6cm), L/2(9cm) and L/l(12cm). In case the location was 0cm, 3cm and 6cm the entry of coefficients were 0.93, 0.92 and 0.90, respectively.

Fluid Flow Characteristics for Minimizing the Area of Rapid Flow Inside the Water Tank to which the Multiple Hoe Screw Nozzle Incurrent Canal is Applied, by Using the Computational Fluid Dynamics (CFD) Simulation (전산유체해석(CFD) 모의를 이용한 다공형 스크류 노즐 입수관이 적용된 물탱크 내부의 사류구역 최소화에 대한 유동특성)

  • This study was carried out for the purpose of minimizing the area of rapid flow inside the water tank. And the shape of incurrent canal was improved, and then the characteristics of fluid flow occurring inside the water tank was analyzed by using the Computational Fluid Dynamics (CFD) simulation method. better multiple hoe screw nozzle incurrent canal was used instead of conventional drop current canal used for the water tank. And according to the results of analyzing the characteristics of fluid flow, in case a screw blade was installed inside the nozzle, fluid flow was sprayed wide. And wide fluid flow was shown inside the cylindrical water tank too. Besides, a tracer simulation was carried out, in case of installing 1 and 2 multiple hoe screw nozzle incurrent canals at the cubic water tank. As a result, MODAL, MODAL index value was close to 1, in case of installing 2 canals. Therefore, it was possible to obtain the results of being close to the characteristics of plug flow.

AN OPTIMUM DESIGN STUDY OF INTERLACING NOZZLE BY ANALYZING FLUID FLOW INSIDE INTERLACING NOZZLES

  • Air interlacing serves to protect the yarn against damage, strengthens inter-filament compactness or cohesion, and ensures fabric consistency. The air interlacing nozzle is used to introduce intermittent nips to a filament yarn so as to improve its performance in textile processing. This study investigates the effect of interlacing nozzle geometry on the interlacing process. The geometries of interlacing nozzles with multiple air inlets located across the width of a yarn channels are investigated. The basic interlacing nozzle is the yarn channel, with a perpendicular single air inlet in the middle. The yarn channel shapes are cross sections with semicircular or rectangular shapes. This paper presents three doubled sub air inlets with main air inlet and one of them is slightly inclined doubled sub air inlets with main air inlet. The compressed air coming out from the inlet hits the opposing wall of the yarn channel, divides into two branches, flows trough the top side of yarn channel, joins with the compressed air coming out from the sub air inlet and then creates two free jets at both ends of the yarn channel. The compressed air moves in the shape of two opposing directional vortices. The CFD-FASTRAN was used to perform steady simulations of impinging jet flow inside of the interlace nozzles. The vortical structure and the flow pattern such as pressure contour, particle traces, velocity vector plots inside of interlace nozzle geometry are discussed in this paper.

Derivation and Application of Boussinesq Equations for the Wave Field in Porous Media (공극매체에서의 파동장에 대한 Boussinesq 방정식의 유도 및 적용)

  • In the present study, the Navier-Stokes (N-S) equations delineating water flows inside porous media were derived applying Reynolds transport theorem in order to provide a basis for analyzing water wave problems inside the porous media. Then, the derived N-S equations were compared with the same species of equations in existing researches. Based on the N-S equations, a set of Boussinesq equations was then derived in such a form that the dispersiveness and nonlinearity of water waves inside the porous media can be properly reproduced. Finally, numerical analyses were carried out to demonstrate the validity of the equations. The reflection and transmission coefficients of porous breakwaters were calculated and compared with the results of existing hydraulic experiments. The numerical results showed a rather sensitive dependency on the virtual mass coefficient of grains constituting the porous media. The selection of the coefficient with zero turned out to give nice agreements with numerical and experimental results.

Numerical Analysis of Added Mass Coefficient for Outer Tubes of Tube Bundle in a Circular Cylindrical Shell (원통 내부에 배열된 외곽 전열관의 유체 부가질량계수 해석)

  • According to the wear detection history for the steam generator tubes in the nuclear power plant, the outer tubes inside the steam generator have more problems on the flow-induced vibration than inner tubes. Many researchers and engineers have used a specified added mass coefficient for a given tube array during the design stage of the steam generator even though the coefficient is not constant for entire tube in cylindrical shell. The aim of this study is to find out the distribution of added mass coefficients for each tube along the radial location. When numbers of tubes inside a cylindrical shell are increased, values of added mass coefficients are also increased. Added mass coefficients at outer tubes are less than those of inner tubes and they are decreased with increasing the gap between the outermost tube and the cylindrical shell. It also turns out when the gap between the outermost tube and the cylindrical shell approaches infinite value, the added mass coefficient converges to an asymptotic value of given tube array in a free fluid field.

  • The Real-time DBMS must provide the keeping, fabrication and control function which the data where the user requsets for processing inside the hour when it is restricted, it is quick most and safely[3]. In order lide this to take chare of the real-time control and fabrication function, details recording (log) of the control processing is very important in accomplished transactions from inside the Real-time DBMS. In this proposal, it sees recording which is like this in base and it proposes the new recovery function for transactions log. This recovery technique which it sees used the concept of the LIFO (Last-In-First-Out) method with basic. The proposed technique will becomes the technique which it will be able and to record to process efficiently with a same attribute and the continuous transactions log.

A Study on Numerical Simulation for the Work Environment Improvement of Highway Tollgate Booth (고속도로 요금소 부스 근무환경 개선을 위한 기류해석 연구)

  • Simulation study has been carried out to analyze the air flow supplying from the heat pump system inside the tollgate booth by using the Phoenix computer simulation program. Through this simulation analysis we can find the problem of present tollgate booth in terms of air flow and recommend an improved model also simulate this model. Final results as follows; It was turned out that the fresh air conditioning is not provided to the worker effectively due to the improper location of inlet and outlet in the present tollgate booth in addition to that the air curtain system applied in the booth lowered air circulation from outside. The improved model was suggested first to increase the air curtain effect by downsizing the window and by installation of the air curtain suction line to reduce the induced outdoor air second to supply the fresh air to the worker directly by relocation of the inlet and outlet of supplying air. With these improved modifications better results have been reached in terms of air flow inside the booth. Next through the air flow simulation of outside booth the contaminated outdoor air has been easuily infiltrating into the booth through the window because of its rectangle shape. Stream like shape of booth has been proposed through the computer simulation as an alternative shape of tollgate booth for a new design.

Experimental research on the failure mechanism of foam concrete with C-Channel embedment

  • An experimental investigation is carried out on the failure mechanism of foam concrete with cold formed steel double C-Channels embedment. The foam concrete is made of cement and fly ash with a compressive strength between 9 and 24 MPa with different densities. Forty-eight tests have been carried out in four groups of specimens with various embedment depths of the steel in the concrete. Four modes of failure are observed, which include the independent failure of the C-Channels with and without a concrete block inside the channel as well as the combined failure of the two channels, and the failure of the extrusion block. A theoretical model has been developed to understand the failure process. The peak compressive force applied onto the C-Channels that causes failure is calculated. It is concluded that the failure involves independent slippage between two C-Channels, and the steel and the foam concrete blocks inside the C-Channels. A method to calculate the peak force is also developed based on the test results. The calculations also show that the shear strength of the foam concrete is about 8% of the compressive strength with ${\alpha}$ coefficient of 0.4 between the steel and concrete.
